Metagonimus yokogawai 세르카리아의 감각유두에 관한 연구 (Study on Sensory Papillae of Metagonimus yokogawai Cercaria)

  • A number of studies on the papillae of cercariae of trematodes reported that the papillar patterns (or chaetotaxy) of cercariae might be an excellent method to attain better understanding of the digenetic trematodes (Richard, 1971 ; Short and Cartrett, 1973; Bayssade-Dufour, 1979) . The present study was aimed to determine the number, distribution pattern and structure of the sensory papillae of Metagonimus yokogawai cercariae, and to elucidate the chaetotaxy of this digenetic trematode. M. yokogawai cercariae were pipetted from a vial in which infected snails (Semisulcospira libertina) had been kept for 3 hours. The snails were collected from an endemic area of M. yokogawai, Boseong river in west-southern part of Korea. Observations of papillae were based on light microscopy of those stained with silver nitrate, and on scanning electron microscopy The results are summarized as follows: 1, All papillae observed were uniciliated. 2. Cilia in anterior tip were shorter than the others in other portions. 3. The body papillae were arranged in essentially symmetrical patterns, Total number of the papillae was 126(63 pairs) in average; anterior tip 40(20 pairs), ventral 20(10 pairs), lateral 42(21 pairs), and caudal 8(4 pairs). 4. 충주호 및 그 상류 지역의 Echinostoma spp.에 대한 연구 (Studios on Echinostomu spp. in the Chungju Reservoir and upper streams of the Namhan River)

  • The present study was performed to know the epidemiological statug of echinostomiasis in the Chungju Reservoir and upper streams of Namhan River, together with an experimental study on the life history o( Echinostoma hortense. The stool specimens of 169 inhabitants and 473 junior high school students from 5 different villages revealed 3(0.5%) echinostomatid egg Positive cases. E. hortense adult worms were recovered from one patient after a treatment and purgation. For the other two patients, it was presumed that one had 2. hortense and another E. cinetorchis infection, based on the morphology of eggs. Five kinds of freshwater snails(168 Radix auricularia coreane, 534 Physa arutp, 144 Hippeutis cnntori, 56 Cipcngopaludina chinensis malleata and 125 Semiiulcospira nediila glebus) examined for the cercariae of echinostomes showed negative results. Ten kinds of freshwater fishes examined for E. herten-'emetacercariae revealed positive rates as Misgurnus anguillicaudatus 40.5%, Odentobutis obscura interrupta 20.3%, Moroco onycephalus 3.9%, and Coreoperca hawamebari 2.0%. In the experimental study, the metacercariae of E. horlense were infected to rats, eggs were collected from adult worms and cultivated, and miracidia were obtained. The miracidia were artificially infected to freshwater snails (R. auricularia), and cercarial shedding was studied. It was revealed that, when the snails were kept at a low temperature ($24^{\circ}C$), only 523 cercariae (on average) were produced during 24 hrs, while they were at a high temperature ($30^{\circ}C$), as many as 9, 990 cercariae (on average) were shed during the same time. The experimental infection of E. hortense cercariae to freshwater fishes was successful in 0. obscura interrupta 52.0%, M. anguillicaudatus 30.3%, C. kawamebari 27.0%, Cobitis lutheri 15.0%, M. cxycephazus 7.3%, Pseudogobio esocinus 4.3%, Squalidus cereanus 2.0%, Zccco platypus 1.3%, and Pungtungia herzi 1.3%. However, infection was not successful to snails, C. chinensis. It has been proved that the Chungju Reservoir and upper streams of Namhan riyrr are endemic areas of echinostomiasis, especially of E. horfense, and snails such as R. auritulerie coreana and fishes such as O. obscura interrupts, M. gnguillicgudgtus, and others are taking the role of 6rst and second intermediate hosts, respectively.

  • The present studies were undertaken to determine accurately whether Korean native fresh-water shrimps can serve as the second intermediate host of Clonorchis sinensis. The experimental results obtained were as follows; 1. Metacercaria of Clonorchis sinensis was not discovered in 36 Macrobranchium nipponensis (De Hann), 1992 Palaemon paucidens (De Hann) and Palaemon madestus Heller, and 860 Caridina leucastica Stimpson, collected in Kimhae district, which were digested in artificial gastric juice, using binocular dissecting microscope. 2. Cercariae of the Chinese liver fluke could not approach the body wall of young shrimps due to streaming caused by their motion. 3. Young Palaemon spp. of 247 were divided into two groups and placed in two different room aquariums, one is at high temperature, the other is normal. Then they were contacted with the cercariae. Only 11 degenerated metacercariae were found in 9 Palaemon spp. It was suggested that they had died immediately or within a few days after penetration. Furthermore the temperature did not affect the infective ratio. 4. At the field experimental aquarium, no metacercaria of Clonorchis sinensis was discovered tn 100 Palaemon spp. which had been brought into contact with 5000 Parafossarulus manchouricus two trials. 5. Specific changes in the cercariae mixed with the extract of shrimps meat were not observed through the dissecting microscope. 6. These results suggest that the Korean native shrimps cannot serve as the intermediate host of Clonorchis sinensis.

Freshwater Snail Diversity in Mae Lao Agricultural Basin (Chiang Rai, Thailand) with a Focus on Larval Trematode Infections

  • The aim of this study was to conduct a freshwater snail survey in Mae Lao agricultural basin to assess the diversity with a focus on habitat types and their larval trematode infections. Snails were collected and examined in 14 sites of Mae Lao agricultural basin from August 2016 to October 2017. A total of 1,688 snail individuals were collected and classified into 7 families, 8 genera, and 12 species. Snail diversity and habitat types were higher in rice paddies than irrigation canals and streams. The most abundant species was Bithynia siamensis siamensis, representing 54.6% of the sample. Three species of snails act as first intermediate host were found with cercarial infections. They were Filopaludina sumatrensis polygramma, B. s. siamensis, and Melanoides tuberculata. The cercariae were categorized into 7 types; echinostome, monostome, gymnocephalous, virgulate, parapleurolophocercous, pleurolophocercous and megalurous cercariae. Parapleurolophocercous cercariae constituted the most common type of cercariae recovered, contributing 41.2% of all infections in snails. Echinostome metacercariae infections were found in 6 snail species with 7.6% prevalence. In addition, the metacercaria of avian trematode, Thapariella sp. were found in Filopaludina spp. snails and B. funiculata with a prevalence of 0.5%. This is the first report for Thapariella metacercariae in the snail host, B. funiculata, and also confirmed that viviparid and bithyniid snails act as the second intermediate hosts of this trematode. This work will provide new information on the distribution and intermediate host of trematode in this area.

Development of the head collar and collar spines during the larval stages of Isthmiophora hortensis (Digenea: Echinostomatidae)

  • It is uncertain when the head collar and collar spines of Isthmiophora hortensis (Digenea: Echinostomatidae), a zoonotic echinostome species in Far Eastern Asia, develop during its larval stages. In this study, the appearance of the head collar and collar spines was studied using light and scanning electron microscopy in cercariae and metacercariae experimentally obtained from freshwater snails (Lymnaea pervia) and tadpoles (Rana nigromaculata), respectively. The cercariae were shed from the snail on day 30 after exposure to laboratory-hatched miracidia. Metacercariae were obtained from the experimental tadpoles at 3, 6, 12, 15, 20, 24, 26, and 30 h after exposure to the cercariae. The head collar was already visible in the cercarial stage, although its degree of development was weak. However, collar spines did not appear in the cercarial stage and even in the early metacercarial stage less than 24 h postinfection in tadpoles. Collar spines became visible in the metacercariae when they grew older than 24 h. It was concluded that the head collar of I. hortensis developed early in the cercarial stage, but the development of collar spines did not occur until the worms became 24-h-old metacercariae in our experimental setting. Counting the number of collar spines was concluded as an unfeasible diagnostic method for I. hortensis cercariae when they are shed from the snail host.

남강유역 간흡충감염의 역학적 조사연구 (Epidemiological Studios on Clonorchis sinensis Infection along the Nam-river in Gyeongnam Province, Korea)

  • An epidemiological study on Clonorchis sinensis infection along the Nam-river (total length ; 186km) flowing in Gyeongsang-nam-do, southern part of Korea, was carried out. Formalin-ether concentration technique and Stoll's egg counting method were employed to figure out the prevalence of C. sinensis infection. For the detection of cercariae from Parafossarulus sp., the snail host of C. sinensis, each snail was placed in aerated tap water, and examined for expelled cercariae. For observing the metacercariae the fresh water fish favorably eaten in raw conditions wore prepared by means of pressing the muscles between two slide glasses and/or by digesting them with artificial gastric juice. The fresh water fish were fed to the rabbits to get the worms and to identify the morphology of adult C. sinensis. The results are summarized as follows: 1. Overall Prevalence of C. sinensis infection was 38.75 from 5,291 examinees; 44.15 (1,408 out of 3,196) in male and 30.4% (637 out of 2,095) in female. 2. The prevalence rates were 42.0% at the upper stream, 41.2% around the vicinity of Jinyanglake, 34.2% at Jinju city, 34.2% at middle stream and 40.3% at down stream regions, respectively. 3. By age, the highest positive rate (53.4∼54.3%) was observed in 30 to 59 years of age. In this age group, the rate in males was 59.7∼62.2%, and in females 42.2∼44.4%, In the age group of less than 19 years it was 7.5∼20.9%. 4. By social strata, the positive rate was 16.5% in the primary school children, 22.6% in school students, 46.2% in teachers and local officers and 49.6% in the general inhabitants. 5. The quantitative examinations with the stool collected from clonorchiasis cases revealed that the light infection (less than 4,000/EPG) was 53.6%, moderate infection (4,001∼10,000 /EPG) 30.3% and heavy infection (more than 10,001/EPG) 16.1%, respectively. More than half of total cases examined were light infection, and 73.2% of female examinees were lightly infected with this cuke. 6. The average value of EPG was 4,963 (male, 6,057; female, 2,557 and the highest value was obtained from the age group of 30 to 59 years (5,240∼6,454). 7. The prevalence of Metagonimus yokogawai infection in humans was 5.5%, and 89.8% of metagonimiasis cases wore double-infected arith C. sinensis. The highest prevalence rate was observed in Jinju city (11.2%). 8. Total of 5,005 Parafossarulus sp., the snail intermediate host of C. sinensis, were examined for the detection of cercariae. The cercarial expulsion rate was 0.34%, and the snails collected in Jin-yang-lake side and in the down stream expelled mainly the cercariae of C, sinensis. 9. About 788 cercariaejday (range: 127∼1,503) were expelled daily from a snail naturally infected with C. sinensis. The snails which released more than 1,000 cercariae/day were 30.8% out of total collected. A snail uniquely released 5,840 cercariae/day in this study. 10. The other trematode cercariae besides C. sinensis were also detected, and the rates out of total snails were the cercariae of Losogenes liberum 6.71%, Cyathocotyle orientalis 0.26%, Notocotylus attenuatus 2.52% and Mucobucaris 0.54%, 11. Ten out of 18 species of fresh water fish caught along the river harbored the metacercariae of C, sinensis. The highest rate of metacercarial infection in fish was detected in Pseudorasbora larva (85.9%). The fish mainly eaten by the inhabitants along the Nam-river, and the metacercarial infection rates were: Zacco platypus 8.0%, Hemibarbus sp. 18.2∼26.7%, Gnathepcgcn sp. 37.5%, Ischikauia steenackeri 42.9% and Pseudogobio esocinus 16.7%. 12. Out of 36 P. larva, the number of metacercariae were about 109 (ranEe; 18∼446) per fish and 27 per gram of flesh. The fish caught in spring harbored the highest number of metacercariae. As indicated above, the prevalence of C. sinensis infection in the inhabitants residing around the Nam-river was relatively high. The farther toward the upper stream areas, the higher was the positive rate of C. sinensis infection, and most of clonorchiasis cases were lightly infected. The snail hosts of C. sinensis distributed all around the Nam-river. Several species of freshwater fish were infected with the metacercariae of C. sinensis and the infection rates were relatively high. To prevent C. sinensis infection in the endemic areas, the effective health education system is suggested as a control measure, although mass treatment is also expected to be useful, using chemotherapeutic agents such as "praziquantel", a recently developed anthelminthic for C. sinensis inftraction.

Tilapia mossambica의 간흡충에 대한 방어기전 (Defence Mechanism of Java Tilapia, Tilapia mossambica, to Clonorchis sinensis)

  • 틸라피아(Tilapia mossambica)에 대한 간흡충의 제2중간정도로서의 역할 여부를 구명하기 위한 일환으로 우선 간흡충 유미유충의 틸라피아 치어에 대한 인공감염실험을 수행하여 그 역할을 할 수 없다는 사실을 천명한 다음 그 방어기전을 규명하기 위하여 그 체표면 점액으로부터 살충성 물질을 순수분리하여 기기분석을 통하여 그 화학적 구조를 확인한 결과를 요약하면 다음과 같다. 1. 입체현미경하에서 틸라피아 치어를 유미유충에 조우를 시도한 바 일반적으로 유미유충은 수분 후에 미부가 체부로부터 분리하기 시작하여 시간이 경과함에 따라 그 수가 증가하여 10여분 후에는 $80\%$에 이르렀으나 극히 소수의 유미유충은 어체표면에 부착한 채 체부만이 어체내로 침입하고 미부는 분리되었다. 2. 틸라피아 치어를 실내수조에서 유미유충에 24시간 노출시킨 바 일부 유미유충은 어체내로 침입하였으나 그 대부분이 피낭한 극소수도 4.2 시간 이내에 거의 모두 사감하였다. 3. 틸라피아의 체표면 점액의 에테르 추출물을 여러 단계의 화학적 처리를 거쳐 제3단계에서 분획한 유백색 상청액을 박막크로마토그라휘하여 얻은 2개의 반점물질을 간흡충의 탈낭유충에 in vitro에서 직접 접촉시켜 살충시험을 수행한 바 그 중에서 제1반점인 Rf. 0.2966 값의 물질이 다른 것에 비교하여 살충성의 월등하게 강력하였다. 4. 틸라피아의 체표면 점액의 에테르 유출물 66.50g으로부터 138mg의 살충성 순수물질이 회수되었으며, 그 수율은 $0.2075\%$, 그 정제도는 71배이었다. 5. 순수하게 분리된 살충성물질에 대하여 적외선, 자외선 및 핵자기공명 분광분석을 이용하여 구조확인을 하였던 바 이 물질은 linoleic acid로 판명되였다. 이상의 실험결과로 미루어 보아 틸라피아는 자연계에 있어서 간흡충의 제2중간숙주 역할을 할 수 없으며, 그 체표면 정액내의 살충성물질인 linoleic acid가 그 방제기전에 관여하고 있다고 생각한다.

Corbicula fluminea (Bivalvia: Corbiculidae): a possible second molluscan intermediate host of Echinostoma cinetorchis (Trematoda: Echinostomatidae) in Korea

  • More than 1,500 clams of Corbicula fluminea, the most favorable food source of freshwater bivalves in Korea, were collected from 5 localities to examine cercarial and metacercarial infection with Echinostoma cinetorchis. Although 3 clams infected with suspicious E. cinetorchis metacercariae out of 200 specimens collected at Kangjin, Chollanam-do were detected, no cercarial and metacercarial infections with E. cinetorchis were observed in field-collected Corbicula specimens. In the susceptibility experiments with laboratory-reared clams, those infected with miracidia of E. cinetorchis did not release their cercariae up to 60 days after infection. To confirm the identity of second intermediate host of E. cinetorchis experimentally, a total of 30 clams were exposed to the cercariae from Segmentina hemisphaerula that had been infected with miracidia of E. cinetorchis. The clams were susceptible to cercariae of E. cinetorchis with an infection rate of 93.3%. Metacercariae from clams taken more than 7 days after cercarial exposure were fed to rats (S/D strain), and adult worms of E. cinetorchis, characterized by 37-38 collar spines on the head crown, were recovered from the ileocecal regions. This is the first report of C. fluminea as a possible second intermediate host of E. cinetorchis.

"이전고환극구흡충" 패류중간숙주로서의 "애기물달팽이" (Austropeplea ollular (Pulmonata: Lymnaeidae): a new first intermediate host of Neodiplostomum seoulense ( Trematoda : Diplostomatidae ) in Korea)

  • Some planorbid snails such as Hippeutis cantori and Segmentina hemisphaerula have reported as the molluscan intermediate hosts of Neodiplostomum seoulense, one of important snail-borne human intestinal trematodes in Korea. However, one of the Korean lymnaeid snail species, Austropeplea ollular was also found to be the first intermediate ho of N. seoulense. In field-collected Austropeplea snails from Sorae and Kimpo out of se collected localities, the bifurcated cercariae of N. seoulense were shed (infection rat 0.3%), whereas no Radix auricularia and Fossaria truncatula were found shedding cercariae. Each of 12 tadpoles of Rana nigromaculata, known as the second intermedia host of N. seoulense, were exposed to 200 cercariae shed from field-collected A ollula. F tadpoles of R. nigromaculata were found to be massively infected with metacercariae o N. seoulense (recovery rate: 62.1%). Each of five rats (Sprague-Dowley strain) was or fed with 200 metacercariae, and eggs of N. seoulense were detected in the rat feces on week later. These rats were killed 4 weeks after postinfection and adult worms of N seoulense were recovered from the small intestines (recovery rate: 9%). This is the f report of A. ollula as the first molluscan intermediate host for N. seoulense in Korea.

경상남도 남강유역의 의용패류 분포 및 흡충류 유충 감염실태 조사 (Distribution of medically important freshwater snails and larval trematodes from Parafossarulus manchouricus and Semisulcospira libertina around the Jinyang Lake in Kyongsang-Nam-Do, Korea)

  • The Jinyang Lake is dammed up Nam River, and surrounded by Jinju city and four counties; Jinyang, Sanchong, Hading, and Sachon in kyongsang-Nam-Bo, Korea. The areas around this man-made lake have been known as an endemic focus of clonorchiasis in Korea. The present study was first aimed to know the distribution of freshwater mollusks including medically important snails, and larval trematodes shed from Parafossarulus manchouricus and Semisulcospira libertina. In addition to above studies, water analyses in each snail habitat were carried out in order to figure out a part of their environmental factors. This malaco-ecological survey was done at the sirs areas around upper, middle and lower parts of the lake for 4 months, August-November, 1983. Total nine species of freshwater mollusks were collected throughout the study: 4 species of gastropods: Semisulcospira libertina, Cipangopaludina chinensis, Parafossarulus manchouricus and Radin auricularia, and 5 species of bit.alves; Unio dcuglasiae, Ancdonta woodiana, Lamprotula gottschei, Corbicula yuminea and Limncperma lucustris. Out of nine species of freshwater mollusks, three species of gastropods; S. Zibertina, p. manchouricus and R. auricularia were medically important in terms of the transmission of digenetic trematodes to humans. P. manchouricus and R. auricularia were mainly collected from the shallow ponds and the irrigation channels with the muddy basin, but S. libertine and the bivalves were only collected from the stream of Nam River where the gravels and rocks were dominant. The levels of dissolved oxygen (D.O.) and biochemical oxygen demand \(B.O.D._5)\ of the water specimens sampled from the study areas ranged from 6.0 to 9.6 ppm and from 0.4 to 1.6 ppm respectively. As a result, it is considered that water system around the Jinyang Lake might be relatively clean without any heavy pollution of aquatic microorganisms and organic materials during the period of this study. On the other hand, eight metalic constituents from the water samples were also assayed, and all metalic ions detected were remarkably low below the legal criteria. However, calcium ion in the water samples from the habitats of P. manchouricus was considerably higher than others. Infection rates of digenetic trematodes in the snails were 6.9% in P. manchouricus and 4.8% in S. libertine, respectively. P. manchouricus snails harboring with thc cercariae of Clonorchis sinensis were only 0.14% among the snails examined, and other trematode cercariae except cercaria of C. sinensis were: furcocercus cercariae, cercaria of Loxogenes liberum type I and II. S. libertine snails parasitized with the cercariae of Metagonimus yokogawai were 1, 5% out of the snails examined, and no cercaria of Paragonimus westermani was found in S. libertine snails in the present study. Digenetic trematode cercariae other than M. yokogawai observed in S. libertina snails were: Cercaria yoshidae (B type) , Cercaria cristata, Cercaria innominatum, Cercaria of Centrocestus formosanus and Cercaria nipponensis.
