Manufacturing Conditions for Rice Porridge with Optimum Properties after Microwave Range Reheating (마이크로웨이브 레인지 재가열 후 최적 특성을 갖는 쌀죽 제조조건)

  • The purpose of this study was to derive the conditions for manufacturing rice porridge with optimum properties after reheating. The characteristics of rice porridge according to the soaking time, water addition rate, heating temperature, heating time, and cooling conditions were compared using the 'Samkwang' cultivar. In Step I, as the heating temperature increased, the weight change decreased and the viscosity increased, and the temperature known as the main factor of the gelatinization also appeared to affect the viscosity increase. In Step II, the viscosity and the texture properties was not significantly different as the soaking time was reduced, and 10 minutes was suitable because of due to the shortening effect of the total process time. In Step III, the residual heat was lowered by cooling after the rice porridge production, so the viscosity could be greatly reduced. Also, it was confirmed that the water addition rate of 900% and the heating temperature of 15 minutes were optimal manufacturing conditions. The next study will investigate the porridge processability of rice cultivars using these results.

INTRACEREBROVENTRICULARLY ADMINISTERED PHENYLALANINE AND TYROSINE: EFFECTS ON FEEDING BEHAVIOUR AND NOREPINEPHRINE CONCENTRATIONS OF SPECIFIC BRAIN SITES IN THE CHICKEN

  • A study was carried out to investigate the action of central L-pheylalanine (Phe) and L-tyrosine (Tyr) on food intake of the chicken. In the first trial, Phe ($200{\mu}g/10{\mu}l$) or saline was acutely administered into the right lateral ventricle (i.c.v.) of chickens (5 birds per each group). Birds (4 birds per each group) were administered with the i.c.v. Tyr ($200{\mu}g/10{\mu}l$) or saline in the second trial. The brains of the birds were removed for catecholamine assy 30 min postadministration. Catecholamine concentrations were measured at specific sites of the brain (LH: lateral hypothalamus, PVN: paraventricular nucleus, and VMH: ventromedial hypothalamus). No significant effect of amino acids on the concentration of norepinephrine of brain sites investigated was detected. Food intake and rectal body temperature were also monitored for 6 h after central administrations of Phe, Tyr or saline (5 birds per each group). Both Phe and Tyr, up to $1mg/10{\mu}l$, failed to modulate food intake or rectal body temperature.

Optimization of Curd Yogurt Production Using Saccharified Rice Solution by Response Surface Methodology (쌀당화액을 활용한 호상요구르트 제조의 최적화)

  • This study was carried out to establish the optimal conditions for curd yogurt using saccharified rice solution. A central composite design was applied to investigate the effects of pectin (2.8~8.4 g), skim milk (14~70 g) and oligosaccharide (28~84 g) ratio on the physicochemical and sensory characteristics of curd yogurt using saccharified rice solution. pH, total acidity, and color value, were analyzed and sensory evaluation was performed. The pH of curd yogurt decreased with decreasing pectin and oligosaccharide ratio. The addition of pectin had a significant effect on the viscosity while skim milk had a significant effect on the color value (a and b value). The results of sensory evaluation showed that, oligosaccharide and skim milk had significant effects on sweetness and sour taste. Oligosaccharide and skim milk masked the sour taste of curd yogurt. The optimum range of ingredients for curd yogurt using saccharified rice solution was predicted to be 4.27~4.90 g of pectin, 30.80~41.30 g of skim milk, and 28.00~36.10 g of oligosaccharide. Based on the overlapped results of physicochemical and sensory evaluation, the optimal amounts of pectin, skim milk and oligosaccharide were 4.59 g, 36.50 g and 32.05 g, respectively.

Production of Glutaminase (E.C. 3.2.1.5) from Zygosaccharomyces rouxii in Solid-State Fermentation and Modeling the Growth of Z. rouxii Therein

  • Glutaminase production in Zygosaccharomyces rouxii by solid-state fermentation (SSF) is detailed. Substrates screening showed best results with oatmeal (OM) and wheatbran (WB). Furthermore, a 1:1 combination of OM:WB gave 0.614 units/gds with artificial sea water as a moistening agent. Evaluation of additional carbon, nitrogen, amino acids, and minerals supplementation was done. A central composite design was employed to investigate the effects of four variables (viz., moisture content, glucose, corn steep liquor, and glutamine) on production. A 4-fold increase in enzyme production was obtained. Studies were undertaken to analyze the time-course model, the microbial growth, and nutrient utilization during SSF. A logistic equation ($R^2$=0.8973), describing the growth model of Z. rouxii, was obtained with maximum values of ${\mu}_m$ and $X_m$ at $0.326h^{-1}$ and 7.35% of dry matter weight loss, respectively. A goodfit model to describe utilization of total carbohydrate ($R^2$=0.9906) and nitrogen concentration ($R^2$=0.9869) with time was obtained. The model was used successfully to predict enzyme production ($R^2$=0.7950).

  • Under the circumstances of China's slower economic growth, the first document ofthe central committee of the CPC continue to focus on "three agricultures" problems, agriculture play a basic role on China's economic. Since 2007, the first document directly stresses the important role of agricultural and fisheries every year. Central Government Working Report of 2015 also stresses that under the new normal of economy, it is important to improve quality and efficiency of agriculture. Agricultural focus going forward will be on improving capacity of competitiveness, innovation and sustainable development.The fishery as an important part of agriculture plays a vital role in the protection of national food security, the prosperity of the rural economy and the optimization of national food structure. However, the situation faced on accelerating the speed of Chinese fisheries is still grim. As an important fishery breeding city in my country, Weihai has achieved remarkable results in the development of fisheries. Based on the input-output indicators of Weihai City from 2010 to 2020, this article uses the DEA model method to conduct a comprehensive analysis of the factors affecting the fishery production efficiency in Weihai City. This paper calculates the two stages of comprehensive efficiency, pure technical efficiency and scale efficiency, and comprehensive compares the two stages. The research results show that: From 2010 to 2020, the average comprehensive technical efficiency of Weihai fishery was 0.928, the average scale efficiency was 0.963, and the average pure technical efficiency was 0.963. The comprehensive technical efficiency of Weihai fishery production showed an upward and downward trend, the pure technical efficiency showed a downward and then upward trend, and the pure technical efficiency showed a fluctuating trend.

Identification and Characteristics of Lactic Acid Bacteria Isolated from Shellfishes (패류로부터 젖산 세균의 분리 및 특성)

  • Lactic acid is an important product arising from the anaerobic fermentation by lactic acid bacteria (LAB). It is used in the pharmaceutical, cosmetic, chemical, and food industries as well as for biodegradable polymer and green solvent production. The poly lactic acid (PLA) is an important material for bio-plastic manufacturing process. For PLA production by new LAB, we screened LAB isolates from shellfish. A total of 28 LAB were isolated from various shellfishes. They were all Gram positive, oxidase and catalase negative. Based on API 50CHL kit, 7 strains among the 28 isolates were identified as Lactobacillus plantarum, 6 strains as Lactobacillus delbrueckii, 5 strains as Leuconostoc mesenteroides, 3 strains as Lactobacillus brevis, 2 strains as Lactococcus lactis, 1 strain as Lactobacillus salivarius, 1 strain as Lactobacillus paracasei, 1 strain as Lactobacillus pentosus, 1 strain as Lactobacillus fermentum and 1 strain as Pediococcus pentosaceu. Also, we examined the amount of total lactic acid produced by these new strains by HPLC analysis with Chiralpak MA column. One strain E-3 from Mytilus edulis was indentified as Lactobacillus plantarum and found to produce 20.0 g/L of D-form lactic acid from 20 g/L of dextrose. Further studies are underway to increase the D-lactic acid production by E-3.

Nitrogen Harvest Index in Some Varieties of Mulberry, Morus spp.

  • Mulberry being the only food of silkworm, Bombyx mori L., is of great economic importance to the silk industry, The success in cocoon production mainly depends on the supply of quality leaves in sufficient quantity. In mulberry, where the economic product is leaf, the uptake of nitrogen from soil is very heavy and high responses to application of nutrients have been reported. Nitrogen supports vegetative growth particularly the leaf biomass. Variation in nitrogen harvest index and other physiological and yield contributing traits were estimated in five mulberry genotypes. Considerable variation was observed for nitrogen harvest index, protein yield per plant and harvest index. The correlation studies indicated the protein yield per plant was significantly correlated with leaf yield, nitrogen content in leaf, nitrogen harvest index and harvest index. The broad sense heritability estimates revealed that harvest index showed highest heritability (88.07%) followed by nitrogen content (82.52%), protein yield (70.28%) and nitrogen harvest index (66.52%).

  • Various antimicrobial peptides (AMPs) from microalgae have shown antibacterial, antiviral, antifungal, anticancer, and antioxidant effects, and play crucial roles in medical applications, aquaculture-related disease management, and the food industry. Magainin 2 (MAG2), an AMP, exhibits high antibacterial and antitumor activity, necessitating an efficient recombinant expression system for low-cost, large-scale production. To enhance MAG2 secretion efficiency in Chlorella, we constructed the SS:MAG2:His vector using the known Chlamydomonas reinhardtii CA1 signal sequence (SS) and obtained a stable transformant via an Agrobacterium-mediated transformation method and RT-qPCR. ELISA results revealed that the MAG2 content secreted into the medium by the SS:MAG2:His transformants increased proportionally with mRNA expression. These findings offer a strategy for high MAG2 secretion in the Chlorella vulgaris platform, potentially minimizing downstream processing costs.

Japanese Apricot in Taiwan (대만의 매실산업)

  • Japanese apricot (Prunus mume Sieb. et Zucc.) is the national symbol and was once the most important temperate fruit crop in Taiwan. Fruiting cultivars were originally introduced from southern China but commercial production was not significant until the 1970s. Currently 6,400 ha of Japanese apricot orchards distribute on shallow mountain hills in the central and the southern part of the island. Taiwanese commercial fruiting cultivars are plausibly chance seedlings or sports from the early introduction and are very low chilling required for budbreak. Ornamental cultivars have been mainly introduced from Japan but cultivations have been limited in high altitude area due to their high chilling requirement. In 2009, Taiwan Agricultural Research Institute's breeding program released the first low chill ornamental cultivar 'Tainung No.2' with a great ornamental potential in subtropical regions. Cultivation and production of Japanese apricot fruit in Taiwan continue to dwindle due to the declining Japanese market share. Ongoing industry transformation to increase domestic consumption and consumer's interest will sustain the future of Japanese apricot in Taiwan.

Improvement of Lutein and Zeaxanthin Production in Mychonastes sp. 247 by Optimizing Light Intensity and Culture Salinity Conditions

  • In this study, we sought to improve lutein and zeaxanthin production in Mychonastes sp. 247 and investigated the effect of environmental factors on lutein and zeaxanthin productivity in Mychonastes sp. The basic medium selection and N:P ratio were adjusted to maximize cell growth in one-stage culture, and lutein and zeaxanthin production conditions were optimized using a central composite design for two-stage culture. The maximum lutein production was observed at a light intensity of 60 μE/m2/s and salinity of 0.49%, and the maximum zeaxanthin production was observed at a light intensity of 532 μE/m2/s and salinity of 0.78%. Lutein and zeaxanthin production in the optimized medium increased by up to 2 and 2.6 folds, respectively, compared to that in the basic medium. Based on these results, we concluded that the optimal conditions for lutein and zeaxanthin production are different and that optimization of light intensity and culture salinity conditions may help increase carotenoid production. This study presents a useful and potential strategy for optimizing microalgal culture conditions to improve the productivity of lutein and zeaxanthin, which has applications in the functional food field.
