Development of an Onion Vinegar Beverage Containing Yuza (Citrus junos Sieb ex Tanaka) and Its Biological Activity (유자 첨가 양파발효음료의 제조 및 생리활성)

  • Onion vinegar has an undesirable flavor and taste that results from alcohol and acetic acid production from fermentation. In this study, we have used onion vinegar to develop an onion vinegar beverage with better sensory quality. The objective of this study was to determine the optimum blending ratio by using response surface methods to produce an onion vinegar beverage containing Yuza (Citrus junos Sieb ex Tanaka). The optimal formula for a fermented onion beverage was determined using a central composite design by the response surface methodology. The independent variables were obtained by regression analysis of the reaction surface of brown sugar, apple extracts and Yuza extracts. The optimum mixing ratio for onion vinegar:water:brown sugar:apple extracts:Yuza extracts was 6.0:77.6:4.9:9.2:2.3 (w/w). The actual overall acceptance was 7.08 under optimum conditions, which was close to the maximum predicted value of 6.96. The concentration of phenolic compounds, total flavonoids, and quercetin present in the onion vinegar beverage was 14.8 mg/100 g, 2.6 mg/100 g and 1.4 mg/100 g, respectively. The onion vinegar beverage showed antimicrobial activity against Staphylococcus aureus, Escherichia coli, Salmonella typhimurium and Enterobacter aerogenes. It also showed antioxidant effects, with a DPPH radical inhibition rate of 18.2% and superoxide dismutase (SOD)-like activity of 11.5%. In conclusion, the onion vinegar beverage described here seems to have nutritional value and potential biological activity.

  • The ways of preservation of chotkal which are classified by the principles of fermentation, are analyzed and interpreted. Chotkal is preserved mainly as two forms; one is chot and the other is shikhae. The fact that shikhae is preserved only in the eastern area can be attributed to two main reasons; (1) raw materials are available throughout four seasons and (2) relatively less production of salt. Chotkal is further classified into nine different ways of preservation and shikhae into fourteen. Regional characteristics for chot include that (1) in western part, Kechot is preserved in salty water (2) in central part Origuljot is fermented either with salt and powdered red pepper, or with salt, powdered red pepper and cooked cereal, (3) in southern part, freshwater shrimp with (1) salt only, (2) salt and powdered red pepper, (3) salt and cooked cereal, (4) salt, powdered red pepper and cooked cereal, (4) in northern part, fish with salt and powdered red pepper. Those for shikhae include that malt is used in southern part, shredded radish in northern part and cooked cereal more in southern part than in northern part.

Diffusion of New Technology on Rice Farming and Linkages between Agricultural Education and Extension (벼농사 신기술 전파와 농업교육 및 농촌지도의 연계)

  • The purposes of the study were to; 1) examine the diffusion and adoption of new agricultural technology in rice farming, 2) survey the status of agricultural education institutions on technology transfer, and 3) explore the linkages between agricultural education and extension for more effective reinforcement strategies to further the diffusion and adoption of new agricultural technology in Korea. Literature review, field survey and panel discussions were employed to strengthen the linkages among agricultural education, research and extension in Korea. As the results of analysis of the diffusion and adoption of new agricultural technology in rice farming following recommendations were offered; 1. Extension services have helped Korean farmers in achieving self-sufficiency in rice and year-round supply of green vegetables as well as upbringing rural youth, however, government changed the status of extension educators from central government staff to county/city government staff since 1997. This brought about various problems such as: decreased morale and number of extension educators, thus weakening the extension education function, weakened linkages between national and local extension offices, less opportunity for in-service education of extension educators, and weakened linkages between agricultural research and extension, etc. 2. In order to insure that extension educators are high caliber professional individuals, it is crucial to establish a system that nationally recognizes these individuals as such, and that provides a professional development path. It is important to restore the morale of extension educators to continually achieve the national goals of food production, balanced national development and preservation of environment, at the same time to meet increasing needs of farmers. 3. Because of the critical importance of diffusing innovations to agricultural producers in order to ensure quality and steady food supply, it is crucial that these issues be addressed before the extension service further deteriorates. Further research activities on agricultural extension should be conducted for more effective agricultural extension system in Korea to achieve a greater rural, national and regional development. 4. Research and implementation activities should be emphasized and encouraged to improve the cooperation and collaboration among agricultural education research and extension to better serve farmers' educational and information needs. National level research activities should be encouraged to improve the effectiveness of agricultural technology transfer including internet education programs for farmers in Korea.

  • The Mongolian milk processing technology has a specific characteristic as a result of the living habits of Central Asian nomads. There are many kinds of milk products in Mongolia due to their processing activity used milk from various dairy animal species. Mongolia has over 30 kinds of dairy products and fermented milk products which are occupied more than 36% of the Mongolian dairy products. Herdsmen who engaged extensive livestock production have specific methods to conserve and prepare the starter culture of fermented milk products in nomadic condition. Specifically, Mongolian lactic starter cultures were prepared from keeping milk products and specific wild plants. Nowadays, over 5 kinds of wild plants such as Rheum undulatum L. Rheum ribes L, Rumex acetosa L. Artemisa sibersena L, Artemisa vulgare are used for lactic starter preparing. Traditional processing methods of the Mongolian fermented milk products are based on the scientific basis and there are easy to learn and operate.

Social Perception on Biotechnology in Korea (생명공학에 대한 사회적 인식)

  • Understanding of the social perception of biotechnology would facilitate the public awareness and debate over the social implications of biotechnology, leading to strengthened basis for social consensus. As a part of the ELSI (Ehical, Legal and Social Implications) project in Korea, the authors have launched a series of social surveys on the social perception of biotechnology. This article is based on the analysis of the first survey, conducted in October 2001. The data were collected through telephone survey on 500 adult respondents nationwide selected by a stratified sampling method. The survey addresses the following questions: What is the present state of public awareness and attitude toward a variety of medical and social applications of biotechnology, such as genetic testing, prenatal genetic screening and testing, xenotransplant, genetic screening for employment, central collection and management of genetic information, and GM food? What factors are related with this perception? The analysis shows that a majority of respondents are in favor of the medical applications. Concerning the social applications and GM food, however, the respondents express a high level of negative attitude a with significant portion of 'do not know' responses. The public perception of the biotechnology is not crystallized in coherent manner yet. The public perception is strongly influenced by mass media, which tend to deliver rather positive information on biotechnology. The analysis suggests that the production and dissemination of diverse information should be activated to reach a sound decision on controversial issues surrounding the development of biotechnology both at individual and societal level as well.

Optimization for the Phytosterol Extraction and Production of Structured Lipids from Safflower seed (홍화씨로부터 Phytosterol 추출의 최적화 및 재구성지질의 합성)

  • Response surface methodology was used for monitoring and optimizing the extraction conditions of campesterol, stigmasterol, ${\beta}$ -sitosterol, and total sterols from the safflower seed. The conditions of phytosterol extraction were optimized by using central composite design with the temperature(35∼75$^{\circ}C$, X$_1$), the time (1∼11hr, X$_2$), and the preheating temperature(60∼100$^{\circ}C$, X$_3$) as three variables. The extraction conditions for maximum campesterol content were 59.01$^{\circ}C$(X$_1$), 2.88hr(X$_2$), and 75.04$^{\circ}C$(X$_3$). But stigmasterol, ${\beta}$ -sitosterol and total sterols were not significantly different under designed extraction condition in this study. Besides, oil was extracted from safflower seed at various conditions and yields were 23.44% at 35$^{\circ}C$ and 20.05% at 80$^{\circ}C$, respectively. Total tocopherol content increased from 0.172% to 0.207% as the extraction temperature increased from 35$^{\circ}C$ to 80$^{\circ}C$. A structured lipids(SL) was synthesized enzymatically by extracted safflower oil and conjugated linoleic acid(CLA). After 24hr reaction, 31.79 mol% CLA was incorporated into the extracted safflower oil.

Effects of Ulva lactuca Extracts on Cytotoxicity of Cancer Cell Lines and Immune Stimulation (갈파래(Ulva lactuca) 추출분획의 암 세포주에 대한 세포독성 및 면역활성 효과)

  • Extracted fractions of the green seaweed Ulva lactuca were studied to verify the cytotoxicity and immunostimulating activity. The fractions from the ethanol extract of U. lactuca were prepared by the systematic extraction procedure with solvents such as hexane, ethyl ether, methanol, butanol and $H_2O$. The cytotoxic effects of U. lactuca fractions against human leukemia cell line U937, mouse neuroblastoma cell line (NB41A3), human hepatoma cell line (HepG2) and rat glioma cell line (C6) were investigated. Ethyl ether fraction showed the highest cytotoxicity against all four cell lines tested. In addition, $H_2O$ fraction also showed relatively high cytotoxicity. Dose dependent patterns were observed on all four cell lines. The immune-stimulating effects of U. lactuca fractions on rat macrophage cell line (RAW 264.7) were also investigated. All five fractions of U. lactuca extract stimulated NO production with concentration dependant manner. These results suggest that U. lactuca may be a useful candidate for a natural cancer preventing and immune-stimulating agents.

Sorghum Extract Enhances Caspase-dependent Apoptosis in Primary Prostate Cancer Cells and Immune Activity in Macrophages (수수 추출물에 의한 primary 전립선 암세포의 caspase 의존성 apoptosis 유도 및 대식세포 면역활성 증가)

  • Sorghum bicolor L. is one of the important minor cereals in Asia, Africa, and the central United States, and it is considered a rich source of polyphenols, flavonoids, and dietary fiber. However, there is a lack of data on the anti-cancer activity of Sorghum in prostate cancer cells and immune activity in macrophages. This study aims to investigate the potential effects of an ethanol extract of S. bicolor L. (SE) on inducing apoptosis in RC-58T/h/SA#4 cells and immunomodulatory activity in RAW 264.7 cells. SE significantly inhibited the viability of RC-58T/h/SA#4 primary prostate cancer cells in a dose-dependent manner. The morphology of RC-58T/h/SA#4 cells treated with SE was shrunken and involved the formation of an apoptotic body and nuclear condensation. In addition, SE markedly activated caspase-8, -9, and -3; increased the protein levels of Bax, p53, cleaved PARP, and cytosolic cytochrome c; and decreased Bcl-2 protein expression. Furthermore, the inhibition of caspases in RC-58T/h/SA#4 cells with z-VAD-fmk attenuated SE-induced cell growth inhibition. The production of nitric oxide (NO) was also elevated by SE treatment, as revealed by immune response parameters. These results suggest that SE inhibits growth and induces apoptosis in primary human prostate cancer cells in a caspase-dependent manner, and it modulates the immune functions in macrophages. Therefore, Sorghum bicolor L. may be used as a functional food to prevent prostate cancer and enhance immune activity.

The Influence of Environmental Characteristics on the Fatness of Pacific Oyster, Crassostrea gigas, in Hansan-Koje Bay (한산${\cdot}$거제만의 환경특성이 양식 굴의 비만에 미치는 영향)

  • Long line suspended culture of oysters has been started commercially in Hansan-Koje Bay since 1969. However, its Annual production has been decreased and culturing periods extended in recent years. So, we investigated environmental parameters and food organisms to identity the causes of poor fatness of oysters in Hansan-Koje Bay from February to November, 1994. As the result, the Water quality of Hansan-Koje Bay was found to be good for culture. For example, the mean concentration of COD was $1.35mg/\ell$, phosphate phosphorus was $0.30{\mu}g-at/\ell$ and dissolved inorganic nitrogen was $4.68{\mu}g-at/\ell$. However, the Hwado island and the inner part of the Hansan-Koje Bay were found to be eutrophicated due to various contaminants transported by land-based activities. But in the central pan of the Hansan-Koje Bay where the oyster farms Have been developed densely, the level of nutrient concentration was very low. During the study period, the dominant species of phytoplankton was Chaetoceros spp. with the percentage of $72.6\%\~87.8\%$ and the mean values of Chlorophyll-a concentration and phytoplankton standing crops were $2.05mg/m^3\;and\;188ind./m\ell$, respectively. The distribution of these parameters also showed similar trends those of nutrients. Especially, chlorophyll-a contents was very low with the concentration of below $0.5mg/m^3$ at central part of the Bay, Juklimpo. The fatness of oysters and the eutrophic index in this area were $18.1\%$ and 0.54, respectively. These values were lower than those of other culturing farms in the southern coastal areas in Korea. Therefore, we estimated that the insufficient food supply due to the low level of nutritional status was the major factors affecting the poor fatness of the Pacific oysters in Hansan-Koje Bay.

Dietary Tea Catechin Inclusion Changes Plasma Biochemical Parameters, Hormone Concentrations and Glutathione Redox Status in Goats

  • The beneficial effects of tea catechins (TCs) are related not only to their antioxidant potential but also to the improvement of animal meat quality. In this study, we assessed the effects of dietary TC supplementation on plasma biochemical parameters, hormone responses, and glutathione redox status in goats. Forty Liuyang goats were randomly divided into four equal groups (10 animals/group) that were assigned to four experimental diets with TC supplementation at 4 levels (0, 2,000, 3,000 or 4,000 mg TC/kg DM feed). After a 60-day feeding trial, all goats were slaughtered and sampled. Dietary TC treatment had no significant effect on blood biochemical parameters, however, low-density lipoprotein cholesterol (p<0.001), triglyceride (p<0.01), plasma urea nitrogen (p<0.01), and glucose (p<0.001) decreased and total protein (p<0.01) and albumin (p<0.05) increased with the feeding time extension, and day 20 was the turning point for most of changes. Interactions were found in glutathione (p<0.001) and the ratio of reduced and oxidized glutathione (p<0.05) in whole blood between treatment and feeding time. Oxidized glutathione in blood was reduced (p<0.05) by 2,000 mg TC/kg feed supplementation, and a similar result was observed in longissimus dorsi muscle. Though plasma glutathione peroxidase (p<0.01) and glutathione reductase (p<0.05) activities were affected by treatment and feeding time interactions, and glutathione S-transferases activity increased with feeding day extension, no changed values appeared in longissimus dorsi muscle. In conclusion, dietary TC supplementation affected the concentrations of some blood metabolites and accelerated GSH depletion in the blood of goats. In terms of less high-density lipoprotein cholesterol, the highest insulin and IGF-I concentrations, the highest ratio of reduced and oxidized glutathione in plasma, the dosage of 2,000 mg TC/kg feed might be desirable for growing goats to prevent glutathione depletion and keep normal physiological metabolism.
