Investigation of the Molding Conditions to Minimize Residual Stress and Shrinkage in Injection Molded Preform of PET Bottle (PET 병용 프리폼 사출성형에서 잔류응력과 수축 최소화를 위한 성형조건의 연구)

  • PET bottle is manufactured by blow molding the preform, which is molded by injection molding. The neck part of the preform of PET bottle for juice or grain-based beverage is crystallized before blowing to improve heat resistance at the entrance of the bottle. However, residual stress, developed during injection molding of preform, prevents the crystallization. In order to release the residual stress in the preform, the preform is annealed after the injection molding. If the residual stress is reduced by optimizing the injection molding conditions of preform the annealing time would be shortened. In this study, the optimum conditions for minimizing the residual stress and increasing dimensional accuracy of the injection molded preform are suggested through CAE analysis. In order to optimize the molding conditions, minimizing residual stress and shrinkage, computer simulations have been carried out with help of design of experiment scheduling. Injection temperature, initial packing pressure and filling time were selected for control parameters. Residual stress was affected by injection temperature and filling time. Shrinkage was affected by injection temperature. It was found that maximum residual stress, distribution of residual stress and shrinkage were decreased by 22%, 40% and 25%, respectively at an optimum molding condition compared with the results of previous molding condition.

Identification of the Protein Function and Comparison of the Protein Expression Patterns of Wheat Addition Lines with Wild Rye Chromosomes (야생 호밀 염색체 첨가 밀 계통의 단백질 발현 양상 비교 분석)

  • The objectives of this study were to compare the protein expression patterns and degrees and identify the protein function of disomic addition lines (DAs) in Leymus racemosus, in order to improve the quality of wheat. Upon SDS-PAGE, L. racemosus showed two major protein bands whereas Chinese Spring (CS) had four major protein bands of high molecular weight. The DA(s) generally showed a similar protein expression pattern to that of CS, because 42 chromosomes were from CS and two chromosomes were from L. racemosus. However, only the L.r[J] line showed two protein bands of between 15 and 20 kDa, like L. racemosus. Image analysis based on 2-DE revealed that L.r[F] had the most upregulated protein spots, whereas L.r[N] had the least upregulated protein spots. For L.r[I], the frequency of the downregulated protein spots was higher than that of the upregulated ones. Using MALDI-TOF MS, the protein function was identified for each protein spot on the 2-DE polyacrylamide gel. The protein spots were classified into 11 groups according to protein function. Among the 11 groups, most protein spots of the DA(s) were identified as proteins related to metabolism. Additionally, unique protein spots of the DA(s) were related to abiotic stressors such as cold and heat. Those proteins are useful for improving wheat quality with resistance against abiotic stressors.

  • The round bale wrappers are generally used for rice straw after the harvesting of low land rice by combine harvester. In this situation, the bale wrappers should be well adapted under the travelling over raised borders and temporary ditches in soft soil of narrow rice fields. The study was conducted to improve the performance of bale wrapper through the new design for compact size, lowered gravity center and lowered power consumption. The prototype of round bale wrapper had been designed and assembled to tractor with three point hitch mounted. The machine type is one roll driving system with one roll for rotating and one roll for wrapping. The driving torque and work performance of the machines were measured and analysed. The torque requirement of the prototype and conventional type was 6kgf-m and 12kgf-m, respectively. The prototype shaved less friction resistance between bale driving roll and round bale. and the power requirement can also be reduced from 12kgf-m in the conventional to 6kgf-m in the prototype. The work efficiency of the new bale wrapper was $45\%$ higher than the conventional wrapper, and the working cost of the prototype can be reduced $17\%$ than that of the conventional.

Factors Affecting Basilar Artery Pulsatility Index on Transcranial Doppler (뇌혈류 초음파 검사에서 기저동맥 박동지수에 영향을 미치는 인자)

  • Transcranial doppler is a non-invasive method that measures the blood flow velocity and the direction of cerebral blood vessels through the doppler principle. The pulsatility index is an index for measuring the transcranial doppler that reflects the distal vascular resistance and is used as an index for the presence and diffusion of cerebral small vessel diseases. The purpose of this study was to evaluate the risk factors affecting the basilar artery pulsatility index in ischemic stroke patients. From January 2014 to May 2015, 422 patients were selected by measuring the transcranial doppler pulsatility index, considering their basilar artery pulsatility index. Univariate analysis was performed using the basilar artery pulsatility index as a dependent variable. Multiple regression analysis was performed considering the factors affecting the pulsatility index as variables. Univariate analysis revealed age, presence of hypertension, presence of diabetes mellitus, presence of hyperlipidemia, and hematocrit (P<0.1) as factors. Multiple regression analysis showed statistically significant results with age (P<0.001), presence of diabetes (P=0.004), and presence of hyperlipidemia (P=0.041). The risk factors affecting the basilar artery pulsatility index of transcranial doppler were age, diabetes, and hyperlipidemia. Further research will be needed to increase the cerebral pulsatility index as a surrogate marker of the elderly, diabetes, and hyperlipidemia.

A Comparative Study on Strength Development, Chloride Diffusivity and Adiabatic Temperature Rise of Marine Concrete Depending on Binder Type (결합재 종류에 따른 해양 콘크리트의 강도 발현, 염화물 확산 및 단열온도 상승 특성에 대한 비교 연구)

  • Recently, in order to reduce a damage of chloride attack and hydration heat in marine concrete structures, blended cement in mixing the marine concrete is widely used. Long term strength development is distinct in concrete with blended cement and it also has excellent resistance to chloride attack and reduction of hydration heat. However, blended cement has a characteristic of relatively low compressive strength in early age of 28 days. On the other hand, a high level of compressive strength is required in the Standard Specification for marine concrete mix design. Such concrete mix design satisfying Standard Specification is effective to chloride attack but disadvantageous for hydration heat reduction due to large quantity of binder. In this study, the material properties of marine concrete considering water-binder ratio and binder type are experimentally investigated. Through the research results, compressive strength in blended cement at the age of 56 days is similar although it has smaller compressive strength at the age of 28 days compared with result of OPC (ordinary portland cement). Even though blended cement has a large water-binder ratio and small unit of binder content, chloride ion diffusion coefficient is still small and hydration heat is also found to be reduced. For meeting the required compressive strength in Standard Specification for marine concrete at 28 days, the increased unit content of binder is needed but the increased hydration heat is also expected.

The Influence of Specimen Volume on the Adiabatic Temperature Rise of Concrete (콘크리트 단열온도 상승량에 미치는 시험체 용적의 영향)

  • To secure the thermal crack resistance of mass concrete, researches and the field applications of low heat portland cement (LPC), ternary blended cement (TBC) which is produced by blending ordinary portland cement with blast furnace slag and fly ash, and early strength low heat blended cement (EBC) increased in recent years. Although the model for adiabatic temperature rise is necessary for estimating the risk of thermal cracking of concrete structures, sufficient data have not been accumulated for these mixtures. In addition, the differences in adiabatic test results have been reported for the volume of test specimens. Therefore, the present study evaluated the characteristics of adiabatic temperature rise based on the type of binder and the volume of the adiabatic test specimen. Test results indicated that the maximum temperature rise ($Q_{\infty}$) and the reaction factor (r) of TBC were the lowest. Test results also showed that $Q_{\infty}$ and r changed with respect to the volume of test specimen. $Q_{\infty}$ and r obtained from 6l equipment were lower than those of 50l equipment. Therefore, corrections with respect to this phenomenon was confirmed and the corrections factors are presented.

Potential of fascaplysin and palauolide from Fascaplysinopsis cf reticulata to reduce the risk of bacterial infection in fish farming

  • Marine natural products isolated from the sponge Fascaplysinopsis cf reticulata, in French Polynesia, were investigated as an alternative to antibiotics to control pathogens in aquaculture. The overuse of antibiotics in aquaculture is largely considered to be an environmental pollution, because it supports the transfer of antibiotic resistance genes within the aquatic environment. One environmentally friendly alternative to antibiotics is the use of quorum sensing inhibitors (QSIs). Quorum sensing (QS) is a regulatory mechanism in bacteria which control virulence factors through the secretion of autoinducers (AIs), such as acyl-homoserine lactone (AHL) in gram-negative bacteria. Vibrio harveyi QS is controlled through three parallel pathways: HAI-1, AI-2, and CAI-1. Bioassay-guided purification of F. cf reticulata extract was conducted on two bacterial species, i.e., Tenacibaculum maritimum and V. harveyi for antibiotic and QS inhibition bioactivities. Toxicity bioassay of fractions was also evaluated on the freshwater fish Poecilia reticulata and the marine fish Acanthurus triostegus. Cyclohexanic and dichloromethane fractions of F. cf reticulata exhibited QS inhibition on V. harveyi and antibiotic bioactivities on V. harveyi and T. maritimum, respectively. Palauolide (1) and fascaplysin (2) were purified as major molecules from the cyclohexanic and dichloromethane fractions, respectively. Palauolide inhibited QS of V. harveyi through HAI-1 QS pathway at 50 ㎍ ml-1 (26 μM), while fascaplysin affected the bacterial growth of V. harveyi (50 ㎍ ml-1) and T. maritimum (0.25 ㎍). The toxicity of fascaplysin-enriched fraction (FEF) was evaluated and exhibited a toxic effect against fish at 50 ㎍ ml-1. This study demonstrated for the first time the QSI potential of palauolide (1). Future research may assess the toxicity of both the cyclohexanic fraction of the sponge and palauolide (1) on fish, to confirm their potential as alternative to antibiotics in fish farming.

  • Presently, for the cement industry, studies that seek to reduce $CO_2$, because of the development of the plastic industry and demand for reduction of energy use, have been actively conducted among them, studies attempting to use Gamma-$C_2S({\gamma}-C_2S)$ to fix $CO_2$ have been actively conducted. The ${\gamma}-C_2S$ compound has an important function in reacting to $CO_2$ and stiffening through carbonatization in the air. The ${\gamma}-C_2S$ compound, reacting to $CO_2$ in the air, generates $CaCO_2$ within the pore structure of cement materials and densifies the pore structure this leads to an improvement of the durability and to the characteristic of resistance against neutralization. Therefore, in this experiment, in order to synthesize ${\gamma}-C_2S$, limestone sludge and waste foundry sands are used these materials are plasticized for 30 or 60 minutes at $1450^{\circ}C$, and are prevented from being cooled in the temperature range of $30{\sim}1000^{\circ}C$ when they are about to be cooled. XRD analysis and XRF analysis are used to determine the effects of this process on ${\gamma}-C_2S$ synthesization, the temperature at which a thing is plasticized, and the conditions for cooling that obtain in the plasticized clinker also, in order to confirm the $CO_2$ capture function, analysis of the major hydration products is conducted through an analysis of carbonatization depth and compressive strength, and through MIP analysis and XRD Rietveld analysis. As a result of these analyses, it is found that when ${\gamma}-C_2S$ was synthesized, the clinker that was plasticized at $1450^{\circ}C$ for one hour demonstrated the highest yield rate the sample with which the ${\gamma}-C_2S$ was mixed generated $CaCO_3$ when it reacted with $CO_2$ therefore, carbonatization depth and porosity were reduced, and the compressive strength was increased.

  • ${\beta}-Lactamase$, secreted from Bacillus sp. J105 strain was purified to a single band on SDS-PAGE by ammonium sulfate precipitation, ion exchange column chromatography and gel-filtration. The molecular weight of the purified enzyme was 31 kDa on SDS-PAGE and its isoelectric point was 7.35. Optimal pH and temperature for enzymatic reaction were 5 and $40^{\circ}C$, respectively. As a result of total amino acid composition analysis of the purified enzyme, Gly and Ala were occupied 14.1 and 13.3 mole %, respectively. Km and Vmax value of purified enzyme were 1.33 mM and 0.36 mM/ml using ampicillin as a substrate, respectively.

Gene Promoter Variation of Phosphoglycerate Kinase, a Glucose Metabolism Enzyme, is a Biomarker for Selection of Disease-resistant Sea Squirt, Halocynthia Roretzi (당 생합성 효소 PGK 유전자 프로모터 변이와 물렁증 저항성 멍게의 선별)

  • The sea squirt, Halocynthia roretzi, has experienced mass mortality due to softness syndrome. The identification of disease-induced genes can provide insights into the development of this syndrome. To identify the genes, we performed differentially expressed gene (DEG) analysis. The expression of the phosphoglycerate kinase (HrPGK) gene was significantly decreased in diseased sea squirts compared to normal ones. We confirmed the result of the DEG analysis through RT-PCR and real-time PCR. In addition, we detected single nucleotide polymorphisms at position -106 (A/T) and -254 (G/T) in the HrPGK gene promoter by genotyping analysis. At the -106 site of the HrPGK gene, the frequency of the AA allele in disease-resistant sea squirts was about two-fold higher than that of sensitive ones, and the frequency of the TT allele in the disease-resistant sea squirts was about six-fold lower. At the -254 site of the HrPGK gene, the frequency of the GT and the GG allele was approximately two-fold higher and two-fold lower, respectively, in the disease-resistant sea squirts compared to the disease-sensitive ones. Analysis of the relationship between the genotypic variation at the -106/-254 promoter and the expression of HrPGK mRNA showed that HrPGK mRNA expression was higher in the -106/-254 AA/GT genotype samples than in the -106/254 TT/GG genotype ones. These results show that sea squirts harboring the AA/GT genotype may have more resistance to mortality than the sea squirts with other genotypes.
