세포막 정보전달 과정에 참여하는 효소로 inositol triphosphate($InsP_3$)를 분해하는 $InsP_3$ kinase를 소의 뇌 조직으로 부터 새로운 정제방법을 개발하고 isozyme들의 특성을 관찰하여 다음과 같은 결과를 얻었다. 분쇄한 소의 뇌조직을 PEG로 단백침전하고 DEAE cellulose chromatography 하여 $InsP_3$ kinase I, II의 두 isozyme을 얻었으며, 각각의 isozyme 분획을 Matrix green gel 및 calmodulin-Affigel 15 column으로 chromatography하였다. Phenyl-TSK HPLC하여 정제하였으며 I은 3,103배, II는 2,310 배의 정제를 나타내었다. 정제 단계에서 specific activity를 비교해 볼 때 Matrix green gel chromatography가 DEAE cellulose에서 보다 I이 30배, II가 약 2.3배로 나타났고 정제배수도 I이 17.2%에서 62.1%로 II가 16.6%에서 38%로 나왔으나 calmodulin-Affigel 15과 비교시는 큰 차이가 없었다. 그러므로 Matrix green gel이 정제에서 매우 중요한 단계라 할 수 있다. 효소의 분자량을 알기 위하여 DEAE HPLC로 두 개의 isozyme을 분리하여 전기영동한 결과 I은 환자량 145,000, 85,500 및 69,500이 3개의 단백질을 얻었고 II는 분자량 79,000 및 57,000의 2개의 단백질을 얻었다.
The polymer electrolyte membranes, CL-SPEEK/Cellulose composite membrane I, II, III with the improved electrochemical and mechanical properties were prepared and characterized. The engineering plastic polyether ether ketone (PEEK) and cellulose were sulfonated and cross-linked. The membranes were prepared by sol-gel casting method with different amount of cross-linking reagent. In conclusion, the composite membranes I, II, III showed improved thermostability, tensile strength and oxidative durability. Proton conductivity of the membranes was also improved and the composite membrane I showed 0.1312 S/cm at $80^{\circ}C$ which was the best of those composite membranes.
Ternary solutions that cellulose triacetate (CTA) and polyethylene terephthate (PET) were mixed in a solvent trifiuoroacetic acid (TFA)/methylene chloride (MC) (6/4 : v/v) showed phase separation and mesophase formation. The ternary systems which were mesomorphic were spun into a methanol both and relatively strong cellulosic fillaments were successfully produced. Analysis showed that CTA/PET fibers have fibrillar structure and high orientation parallel to the fiber axis. These fibers proved to be molecular composite and have relatively high strength and modulus as spun.
The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. Major cellulase components, such as three endoglucanases (endoglucanases I, II, and III) and one exoglucanase (exoglucanase II), were isolated from a commercial cellulase (Meicelase TP 60) derived from the fungus Trichoderma viride by a series of chromatography procedures. These procedures were the gel filtration on Bio-Gel, the anion exchange on DEAE-Bio-Gel A, the cation exchange on SP-Sephadex C50, and the affinity chromatography on Avicel cellulose. The average molecular weights determined by SDS-polyacrylamide gel electrophoretic analysis were 51,000, 59,000, 41,000 and 62,000 Da for endoglucanases I, II and III and exoglucanase II, respectively. The extinction coefficients, ${\varepsilon}^{1%}$ 280 nm, of these enzymes were 11.7, 3.3, 7.2 and 11.3, respectively. Among them, the endoglucanase II showed the very low value of the coefficient compared with the others. On the other hand, it was found that endoglucanase II and III were of more random hydrolytic mode on carboxymethylcellulose as compared with those of endoglucanase I and exoglucanase II. Especially, endoglucanase I showed less random action than that of exoglucanase II. In the hydrolysis of insoluble cellulose by the enzyme components, cellobiose was the major product, but glucose was the major product by endoglucanase III.
This study was undertaken to dtermine whether dietary fibers had an effect on postprandial plasma lipoprotein and liver lipid composition in rats. Each experimental diet ocntained 10 % dietary fiber by weight. All animals were sacrificed in postprandial state. Sodium alginate-fed animals has significantly lower plasma cholesterol, VLDL-, LDL-cholesterol, TG, and protein levels than did the fiber-free control group. In addition, liver cholesterol and TG concentrations were the lowest in sodium alginate -fed animals. There was no significant change in HDL-cholesterol levels among experimental groups. Cellulose-fed animals also have lower plasma cholesterol and TG levels than fiber-free controls, but liver TG concentrations were not different from those in fiber-free controls. This study demonstrates that dietary fiber included in the diet of rats is able to alter postprandial lipoportein cholesterol and TG, and that sodium alginate, a soluble fiber, was the most effective in lowering plasma and liver cholesterol and TG.
Regenerated cellulose fibers were prepared from three pulps containing different degree of polymerization(DP) and $\alpha$-cellulose contents by dry-jet wet spinning technique with cellulose dope in N-methylmorpholin N-oxide (NMMO). The effect of antioxidant, n-propyl gallate (PG) on the properties of different regenerated celluloses was studied using X-ray diffraction, copper number calculation, and viscometry. The degradaqtion of regenerated cellulose from pulp containing higher DP and lower $\alpha$-cellulose content was occurred more seriously. The tensile strength and initial modulus of regenerated cellulose fiber obtained from NMMO dope with PG were higher than those of fiber obtained from NMMO dope without PG. All fibers showed the round shape cross section and typical cellulose II crystalline structure.
Effect of a nonionic surfactant, Tween 20 on the adsorption and kinetic mechanism for the hydrolysis of a microcrystalline cellulose, Avicel PH 101, by endoglucanase Ⅰ (Endo Ⅰ) and exoglucanase Ⅱ (Exo Ⅱ) isolated from Trichoderma viride were studied. The Langmuir isotherm parameters, amount of maximum adsorption (Amax) and adsorption equilibrium constant (Kad) for the adsorption, were obtained in the presence and the absence of nonionic surfactant. On the addition of Tween 20, the Kad and Amax values of Exo Ⅱ were decreased, while those of Endo Ⅰ were not affected. These indicate that the adsorption affinity of Exo Ⅱ on the cellulose is weakened by nonionic surfactant, and the surfactant enhanced desorption of Exo Ⅱ from insoluble substrate. The enzymatic hydrolysis of the cellulose can be described by two parallel pseudo-first order reactions using the percentages of easily (Ca) and hardly (Cb) hydrolyzable cellulose in Avicel PH 101 and associated rate constants (ka and kb). The Ca value was increased by adding Tween 20 for all enzyme samples (Exo Ⅱ, Endo Ⅰ and their 1:1 mixture) implying that the low-ordered crystalline fraction in the cellulose may be partly dispersed by surfactant. The ka value was not affect by adding Tween 20 for all enzyme samples (Exo Ⅱ, Endo Ⅰ and their 1:1 mixture). The kb value of Exo Ⅱ was increased by adding Tween 20, while that of Endo Ⅰ was not affected. This suggests that the surfactant helps the Exo Ⅱ desorb from microcrystalline cellulose, and increase the hydrolysis rate. These results were show that the increase of hydrolysis of cellulose by the nonionic surfactant is due to both the activation of Exo Ⅱ and partial defibrillation of the cellulose.
The effects of dietary cellulose to utilize the intake nutrients in the albino rats were investigated by the two consecutive experiments in this study. The author wanted to observe the effects of the age of animals and different level of dietary cellulose, high and low in the first experiment and the effects of dietary cellulose from three different sources, rice bran, vegetable and pure-cellulose at high level in the second experiment. The results were summarized as follows. (1) The obvious effect of dietary cellulose caused to increase feces weight. Consequently apparent digestibilities of intake nutrients in the diet decreased. It was noteworthy that fecal glucose excretion was significantly affected by cellulose intake. Since three nutrients, protein, fat and carbohydrate serve as energy sources in the body, this result can be interpreted to be lowered in the efficiency of energy utilization out of intake from the diet. (2) The nutrients loss due to dietary cellulose resulted in reduction of skeletal muscle and bone weights which lead eventually to affect in reduction of body weight. (3) The effects of dietary cellulose showed differently according to the amount of cellulose ingested, dietary nutrients composition, the kinds of dietary cellulose supplemented and the age of experimental animals. When the intake diet was proportionately imbalanced in three different energy sources, carbohydrate, fat and protein, the cellulose intake was adversely affected in the utilization of the nutrients in the body. Cellulose from rice bran decreased body weight gain, cellulose from vegetable maintained medium level in body weight and pure-cellulose increased body weight. Growing rats showed more sensitive effects of cellulose tolerance in the body than grown rats in every concerns of this experiment.
Chitosan(CS) and cellulose acetate(CA) composite films were prepared using formic acid as a cosolvent by casting, solvent evaporating and neutralization method. This study examines if the blending method, which uses formic acid as a cosolvent is efficient in improving the mechanical properties of CS film, especially wet strength and elongation. Formic acid is an effective cosolvent for the blend of CS and CA. Under wet condition, tensile strength and elongation of the composite films were obviously higher than those of the films made from pure CS. FTIR, DSC, and X-ray diffraction showed that the composite films exhibit a high level of compatibility and that strong interaction between the CS and CA was caused by intermolecular hydrogen bonding. The affinity series of composite film to transition metal ions are Cu(II) > Cd(II) > Cr(III). The adsorption of Cu((II) ion was shown to be highly pH sensitive.
