  • 本實驗은 Vernal알팔파의 callus 誘導에 미치는 몇가지 要因들에 對하여 糾明하고자 逐行되었으며 얻어진 結果는 다음과 같다, Callus 誘導에서 auxin 源으로서 2,4-D를 2-5mg/1 單用處理한 것이 가장 좋았으며, 여러가지 基本 培地 中에서는 B5와 SH가 callus 誘導에 가장 效果的이었는데, PC와 MS 培地에서 生成된 callus는 friable 하였다. 培地#의 pH는 5.8이 가장 좋았으며 7.0 以上에서는 callus는 거의 생장하지 않았다. 組織片은 9日 齡된 植物體의 것이 가장 좋았으며, 이를 前後하여 callus의 生成量은 減少하였다. Callus 誘導時, 光條件의 有無는 큰 影響을 나타내지 않았으나, 빛 條件에서는 callus 組織은 푸르게 되었다.

Mass Production of Yeast Spores from Compressed Yeast

  • Saccharomyces yeast spores are more resistant to drying and storage than vegetative cells. For the mass production of yeast spores, compressed yeast was directly inoculated into a sporulation medium (SM). The effects of inoculum size and the addition of rice wine cake (RWC) into SM on the sporulation were examined using flasks. With $1\%$ inoculum of compressed yeast, $1.45{\times}10^8/ml$ of asci was obtained. The addition of $0.5\%$ RWC into SM improved the cell growth and spore yield, and the number of asci formed was $2.31{\times}10^8/ml$. The effects of culture temperature, temperature-shift, and concentrations of inoculum, potassium acetate, and RWC on the sporulation were also evaluated using a jar fermentor. The optimum temperature for spore formation was $22^{\circ}C$ where the number of asci formed was $2.46{\times}10^8/ml$. The shift of culture temperature from initial $30^{\circ}C$ for 1 day to $22^{\circ}C$ for 3 days increased the number of asci formed to $2.96{\times}10^8/ml$. The use of $2\%$ (w/v) inoculum of compressed yeast, $2\%$ potassium acetate, and $1\%$ (w/v) RWC in SM with the shift of culture temperature of initial $30^{\circ}C\;to\;22^{\circ}C$ resulted in $90\%$ sporulation ratio and formation of $6.18{\times}10^8\;asci/ml$.

Effect of Broussonetia papyrifera L. (paper mulberry) silage on dry matter intake, milk composition, antioxidant capacity and milk fatty acid profile in dairy cows

  • Objective: This study was carried out to investigate the possible application of Broussonetia papyrifera (B. papyrifera) silage as a functional feeding stuff in dairy cattle. Methods: Seventy-two Holstein cows were divided into four groups randomly and allocated to 6 pens with 3 individuals in each group and fed the original total mixed ratio (TMR) in the dairy farm or the new TMR with 5%, 10%, and 15% B. papyrifera silage, separately. Feed intake were recorded, milk and blood samples were collected, and milk composition, blood metabolites and milk fatty acids composition were measure at the end of the experiment. Results: Dry matter intake of cows decreased when they fed on diet with B. papyrifera, but no differences were observed in body condition score, milk yield, milk protein and lactose, feed efficiency and serum metabolites between groups. Both 10% or 15% of B. papyrifera silage in the diet significantly increased the immunoglobulin A (IgA) and IgG in serum, 15% of B. papyrifera silage increased the content of serum catalase, superoxide dismutase, total antioxidant capacity, and decreased the content of 8-hydroxy-2'-deoxyguanosine. Furthermore, 10% or 15% of B. papyrifera silage resulted in a significant decrease in the milk somatic cell count, and increased the polyunsaturated fatty acids content in the milk. Conclusion: The diets with 10% to 15% of B. papyrifera silage might enhance the immune and antioxidant function of dairy cows and increase the polyunstaturated fatty acid concentration in the milk.

  • The tissue inhibitor of metalloproteinase-2 (TIMP-2) inhibits matrix metalloproteinases activity and modulates cellular proliferation and apoptosis. The human serum albumin-TIMP-2 with folic acid conjugate (termed HT2-folate) was synthesized to promote uptake through folate receptors (FRs), and a corresponding radio-labeled compound was prepared for tumor diagnosis by positron emission tomography (PET). $^{68}Ga$-NOTA-HT2-folate was synthesized from $^{68}Ga$ and the NOTA chelator with HT2-folate. The fusion protein was identified using MALDI-TOF mass spectrometry. The radioligand was prepared with a high radiochemical yield. Cell-surface association of $^{68}Ga$-NOTA-HT2-folate significantly increased over time in FR-positive tumor cells. In animal PET and biodistribution studies, tumor uptake was very high as early as 1 h after radioligand injection. Folate conjugation enhanced the selective receptor-targeting efficacy of HT2 in FRexpressing tumors, and its radioligand will be useful as an in vitro tool and for in vivo tumor diagnosis by PET imaging.

비타민 B1 유도체(Thiamine Di-lauryl Sulfate:TDS)의 나노입자화를 통한 고추탄저병균의 항진균 활성 증진 (Enhancement of Antifungal Activity of Anthracnose in Pepper by Nanopaticles of Thiamine Di-lauryl Sulfate)

  • This study was performed to enhance antifungal activity of anthracnose in chili pepper by nanopaticles of thiamine di-lauryl sulfate (TDS) through high pressure homogenization process. Yield of TDS was 79.14% by reaction of thiamine hydrochloride and sodium lauryl sulfate. TDS nanopaticle solution was manufactured through high pressure homogenization process. The turbidity of nanoparticles solution was increased with increasing the concentration of TDS, and nanoparticles solution of 100 ppm was showed the highest turbidity with absorbance of 3.212. The size of nanoparticles solution was measured as average 258.6 nm by DLS. Nanoparticles solution of 100 ppm showed growth inhibition activity with higher than about 80% compared to the control group against Colletotrichum gloeosporioides. Finally, nanoparticles solution was increased effectively the penetration of the TDS nanopaticles on attached cell membrane of hyphae and started to destruct the cells under microscope observation. Consequently, we suggested that the TDS nanoparticle solution by high pressure homogenization process might be suitable biochemical pesticides for improving the antifungal activities against anthracnose in pepper.

  • 본 논문은 구조물의 미시적 측면에서 유효평균탄성계수를 결정하기 위한 균질화기법인 점근적 방법을 적용하였고, 탄성값을 조사하기 위하여 유한요소법으로 정식화하였다. 수치 예로서 물성치가 각기 다른 등방성 재료를 적층한 부재의 임의 단면에서 단위요소를 해석영역으로 설정하고 산출된 탄성계수를 기존의 해석방법으로부터 산출된 값과 비교하였다. 균질화기법으로 산출된 탄성계수는 과소평가되어 나타나며, 이는 해석영역을 유한요소 정식화하는 과정에서 수정항만큼 차이가 난다는 것을 증명하였다. 기존 해석방법으로는 복합재료의 탄성계수가 단순히 재료의 산술적 평균값으로 계산되는 것과는 달리, 미시적으로 복합재 단위요소의 반복성을 고려함으로써 제안된 해석방법이 보다 유용하다는 것을 보여 주었다.

질소 화합물이 담배연기성분 및 안전성에 미치는 영향 (Effect of nitrogen compounds on the chemical composition and biological activity of mainstream smoke)

  • The objective of this study was to investigate the effect of nitrogen compounds such as protein on the chemical composition and toxicity of cigarette mainstream smoke. BSA protein was treated into the tobacco leaf of original 2R4F cigarette at 1~4 % level. The studies were performed which included a bacterial mutagenicity assay and a mammalian cell cytotoxicity assay for total particulate matter(TPM), and glutathione(GSH) consumption assay for gas/vapor phase(GVP) and determination of smoke chemical constitute. Cigarettes treated with protein were observed dose-dependent increase in yield of volatiles, semi-volatiles and aromatic amines compared with control cigarette. However, carbonyl compounds such as acrolein was lower than that of control cigarette when calculated on an equal TPM basis. The cytotoxicity of TPM obtained from the protein-added cigarettes was not different from that of control cigarette. However, the mutagenicity of the TPM from protein-treated cigarettes(1~4 %) was up to 10-27 % higher than that of control. On the other hand, toxicity of GVP from protein-treated cigarette(4 %) was significantly decreased compared with control cigarette. An overall assessment of our data suggests that nitrogen compounds such as protein should be important for the chemical composition and biological activity of cigarette mainstream smoke.

난각막 분해물의 식품 소재로서 기능적 특성 (Functional Properties of Egg Shell Membrane Hydrolysate as a Food Material)

  • 본 연구는 난각막(egg shell membrane)에 keratinase를 생성하는 Bacillus licheniformis를 배양하여 제조한 효소분해 난각막분해물(enzyme-degradation egg shell membrane hy-drolysate, EESMH)과 IN NaOH-Ethanol로 분해한 알칼리분해 난각막분해물(alkalic-degradation egg shell membrane hy-drolysate, AESMH)을 제조하여 식품소재로 활용하고자 성분분석 및 분해물의 유화성, 거품형성력, 항산화력 등의 식품학적 기능을 확인하였다. 즉, EESMH의 생산수율은 약 15% 였고 AESMH의 경우 약 70%의 수율로 가수분해물이 생성됐으며 아미노산 분석 결과 EESMH와 AESMH의 아미노산 구성은 주성분이 aspartic acid, glutamic acid, cystine으로 큰 차이가 없었으나 EESMH의 경우 histidine이 18.69%로 AESMH의 2.56%에 비해 6배 정도 함량이 높게 나타났다. 단백질의 기능적 특성 측면에서 용해성은 EESMH와 AESMH 모두95%이상의 높은 용해성을 나타내었다. 유화 활성의 경우 EDESDBB가 더 효과가 있었으며, 유화 안정성도 EESMH가 AESMH보다 약 8% 안정하였다. 거품 형성력은 AESMH가 EESMH보다 모든 pH범위에서 약 2배 정도 높았으며 안정성도 우수하였다. 항산화력의 경우AESMH가 항산화력이 높은 것으로 확인하였고 EESMH의 경우 전체적으로 매우 약하게 나타났다.

  • 소와 양의 Rumen에서 Hungate's roll tube 방법에 의해 통성 혐기성 섬유소 분해세균을 분리하였다. 가장 강한 섬유소 분해력을 갖는 균주를 선발하여 형태학적 배양학적 생리학적 특성 및 전자현미경사진을 검토하여 Cellulomonas fimi C-14으로 동정(同定)하였다. 분리된 C. fimi C-14의 균생육 및 효소생산은 $30^{\circ}C$, pH6.5에서 최대이었으며, 그의 조효소(組酵素)는 pH6.0, $40^{\circ}C$에서 최대 역가를 보였으며 그 $C_1,\;C_x,\;{\beta}-glucosidase$의 Activity 는 20.6, 227.6, 0.56 $(unit{\times}10^3/ml)$ 이었다. 곰팡이 Cellulase의 첨가로 Enzyme Activity가 증가하였으며, 현사시나무의 톱밥을 기질로 하여, 분리균과 S. cerevisiae DY2를 접종하여 Ethanol 발효생산시 ${\beta}-glucosidase$의 첨가로 상승효과를 보였으며, 이단계 발효법보다 동시 당화 발효법에 의한 Ethanol 생산이 더 좋았다.

Penicillium verruculosum의 원형질체 형성과 재생 (Formation and Regeneration of Penicillium verruculosum Protoplasts)

  • 섬유소 분해효소를 생산하는 P. verruculosum F-3 의 세포융합에 관한 연구의 일환으로 균사체로부터의 원형질체 형성과 재생 최적조건을 검토하였다. 세포벽 분해효소로 유효한 각종시판효소제제중 0.5%(w/v) Novozym 234가 가장 효과적이었고 PDY 배지로 3$0^{\circ}C$ 20시간 배양한 균사체 400mg을 3$0^{\circ}C$ 1시간 반응시 가장 많은 원형질체가 형성되었다. 원형질체 형성을 위한 최적 삼투압 조절제는 0.7M sorbitol(pH5.6)이었고 원형질체 재생을 위한 최적 삼투압 조절제는 0.6M MgSO$_4$(pH 5.6)이었으며 RCM에서의 재생율은 4.6~27.8%였다. 원형질체를 RLM에 배양시 균사체로 환원되는 형태적 변화가 관찰되었다.
