Effects of Pluronic F-68 on Cell Growth of Digitalis lanata in Aqueous Two-Phase Systems

  • The effects of Pluronic F-68, a non-ionic surfactant, on the growth and physical characteristics of Digitalis lanata suspension cultures were investigated in aqueous two-phase systems (ATPSs) composed of $4.5\%$ polyethylene glycol (PEG) 20,000 and $2.8\%$ crude dextran. In the range of 0.1-10.0 g $1^{-1}$, Pluronic F-68 enhanced the maximum cell density in a medium with ATPSs, even though Pluronic F-68 did not affect cell growth in a normal growth medium. In terms of physical properties of ATPSs with cell suspension cultures, 0.2 g $1^{-1}$ of Pluronic F-68 reduced viscosity by up to $40\%$, while 0.1 g $1^{-1}$ of Pluronic F-68 significantly enhanced the oxygen transfer rate. In addition, we successfully performed aqueous two-phase cultivation in a 5-1 stirred tank bioreactor with 0.5 g $1^{-1}$ of Pluronic F-68, and discovered that cell growth in ATPSs was similar to that in normal growth medium.

An Experimental Study on Growth Pattern and Ultrastructure of Human Gingival Fibroblasts Treated by Low Level Laser (저출력레이저가 성인의 치은섬유아세포의 성장양상과 미세구조에 미치는 영향에 관한 실험적 연구)

  • In order to verify the acceleration effect of low level laser (LLL) on oral mucosal wound healing process at cell biological level, the author studied growth pattern and ultrastructure in human gingival fibroblasts flashed by GaAlAs LLL-830 nm, 15mW for 10 minutes/flash one to three times at interval of 3-4 days through the evaluation of cell growth rate, protein conent/cell, DNA content/cell and ultrastructural changes for 14 days. The results were as follows : 1. The growth rate in gingival fibloblasts treated by LLL showed 4 orderly stages-decreasing stage after LLL treatment, acute increasing stage 3 days after LLL treatment, restring stage and recovering stage. 2. The effect of multiple flashes on LLL at interval of 3 days more or less was not proportional to times of flash on acceleration of growth in gingival fibroblasts. 3. The total protein content per gingival fibroblast was not significantly changed by LLL treatment in comparison with control group. But some kinds of protein which might be cell growth promoting factors were decreased immediately after LLL treatment, thereafter were acutely increased in cellular protein profile. 4. In ultrastructural changes of gingival fibroblasts treated by LLL, more prominent rough endoplasmic reticulum, mitochondrial hyperplasia/hypertrophy and increased extracellular fibrillar matrix were observed in comparison with control group under same experimental period.

On the mutual action of boron, calcium and sucrose in pollen cell growth and pollen bursting (화분세포생장과 파열에 미치는 붕소, 석탄 그리고 자당의 상호작용에 관하여)

  • The mutual relationships of boron, Ca and sucrose were studied in relation to in vitro pollen growth and pollen bursting, by using conventient pollen from Crinum asiaticum for experiment. Crinum pollen are paticularly sensitive to Ca. Addition of very small amount of boron to cultural media was apparently synergistic to the action of sucrose and Ca in pollen germination and tube elongation. This action was extended to a higher level of boron concentraton. Combined application of boron, Ca and sucrose always gave the better results in pollen growth and protection against pollen bursting much more than when used singly. This indicated that there is a direct relationship between better growth of pollen and increased rigidity of pollen cell wall. A higher level of Ca concentration tended to increase bursting rate of pollen grains and decrease that of pollen tubes, while boron always depressed the rate of bursting. This was considered due to increased failure in pollen germination at high level of Ca that favors pollen tube elongation. The fact that Ca show an antagonistic effect on the suppressive action of high level of boron in pollen growth and shows different effect in response to pollen bursting from boron, suggested mode of Ca and boron action in the presence of sucrose is quiate different, although to increase in rigidity of pollen cell wall by them is in common nature. It was postulated therefore that Ca acts on pectins of pollen cell wall largely as "non-metabolic" and boron as "metabolic" promoter is pollen growth and protecting pollen bursting, since boron and Ca have common nature in strengthening the pollen cell wall but act differently.but act differently.

Effect of Agitation and Aeration Rate on Nicotiana tabacum Suspension Cell Culture in Bioreactors (Bioreactor를 이용한 담배세포 현탁배양에서 교반형태와 통기량이 미치는 영향)

  • For the optimization of operating conditions for plant cell suspension culture in bioreactors, effects of bioreactor types, various kinds of impellers, and aeration rates were examined using Nicotiana tabacum cells as a model system. Stirred tank bioreactor and airlift bioreactor were used for the comparison of bioreactor type. Growth rates in both bioreactors were lower than in shake flasks. In terms of final cell concentration, stirred tank bioreactor supported a little bit better growth compared to airlift bioreactor. Impeller type did not affect cell growth significantly, but it was apparent that cell size index decreased in the case of using hollowed paddle impeller. When the aeration rate was maintained at 0.3 vvm, cell growth was the best. At above 1.0 vvm, growth inhibition as well a browning was noticed. In addition, it was found that cell size index reduced proportionally to the increased of aeration rate.

Comparative Characterization of Growth and Recombinant Protein Production among Three Insect Cell Lines with Four Kinds of Serum Free media

  • Three insect cell lines, Sf9, Sf21 and Tn5Bl-4, and four different kinds of serum free media (SFM), Sf 900 II, EX-CELL 420, EX-CELL 405 and Express Five, were used to compare the nutrient consumption, byproduct formation, production of recombinant protein and protease activity in suspension cultures. The Sf 900 II SFM was a ppropriate for the cell growth and protein production of the Sf9 and Sf21 cell lines. When the Tn5Bl-4 cell line was grown in the Express Five SFM, the specific growth rate was 1.6 fold higher than those of either the Sf9 or Sf21 cell lines. The glucose and glutamine consumption rates per cells, were 4 and 2.3 times higher than those of the Sf9 cell line, respectively. The overall yield coefficients of the lactate and ammoniumion were 2.8 and 1.5 times higher compared to those of the Sf9 cell line. respectively. The maximum specific ${\beta}$-galactosidase production rate was 4.5 fold that of the Sf9 cell line, a 3 times higher protease activity per cell.

COX-2 INHIBITOR INDUCED APOPTOSIS IN ORAL SQUAMOUS CELL CARCINOMA CELL LINE THROUGH AKT PATHWAY (COX-2 억제제에 의한 AKT 경로를 통한 구강편평세포암종 세포주의 세포사멸 유도)

  • The objectives of this study was to check up the effect of celecoxib, COX-2 inhibitor, on the pathogenesis of oral squamous cell carcinoma. After mefenamic acid, aspirin and celecoxib, COX-2 inhibitor, were inoculated to HN 22 cell line, the following results were obtained through tumor cell viability by wortmannin, growth curve of tumor cell line, apoptotic index, PGE2 synthesis, total RNA extraction, RT-PCR analysis and TEM features. 1. When wortmannin and celecoxib were given together, the survival rate of tumor cells was lowest about 47 %. So wortmannin had an effect on the decrease of survival rate of tumor cells. 2. In growth curve, the slowest growth was observed in celecoxib inoculated group. 3. The synthesis of PGE2 was decreased in all group and the obvious suppression and highest apoptotic index was observed in celecoxib inoculated group. 4. Suppression of expression of COX-2 mRNA was evident in celecoxib inoculated group. But that of COX-1,2 mRNA was observed in mefenamic acid inoculated group and aspirin inoculated group. 5. In celecoxib inoculated group, mRNA expression of AKT1 was decreased and that of PTEN & expression of caspase 3 and 9 was evidently increased. Depending on above results, when celecoxib was inoculated to oral squamous cell carcinoma cell line, an increase of mRNA expression of caspase 3,9 and PTEN is related to a decrease of mRNA expression of AKT1. Wortmannin had an effect on the decrease of survival rate of tumor cells. Celecoxib might induce apoptosis of tumor cell by suppression of AKT1 pathway and COX-2 inhibition. This results suggested that COX-2 inhibitor might be significantly effective in chemoprevention of oral squamous cell carcinoma.

Effect of Initial condition on the Characteristics of Ehtanol Fermentation (발효 초기조건이 에탄올 발효 특성치에 미치는 영향)

  • The effect of initial cell concentration on the characteristics of ethanol fermentation was investigated in the batch fermentation of Saccharomyces cerevisiae ATCC 24858. The characteristics were investigated in the range of 60 to 230 g/l of the initial sugar concentrations and 0.5 to 85 g/l of the initial cell concentrations. When the initial cell concentrations were 27 g/l for 60 g/l of the initial sugar and 85 g/l for 230 g/l, the fermentation time required for the complete consumption of the initial sugar was one hour, respectively. The ethanol productivity increased with the initial cell concentration so that, in the case of 100 g/l of initial sugar, the productivity rose up to 55 g/l/hr at 55 g/l of the initial cell concentration. The specific growth rate decreased according to the increase in the initial biomass concentration and finally became zero at around 25 g/l of the cell concentration regardless of the initial sugar concentration. The specific ethanol production rate was constant as 1.02 g/l/hr up to 150 g/l of the initial sugar. However, the rates decreased sharply with the augmentation of concentration of the initial sugar above 160 g/l. The overall ethanol yield represented a constant value, 0.475 g/g irrespective of the initial cell and sugar concentrations. The overall biomass yietd showed a trend to diminish in values with the biomass and ultimately to reach zero more than 25 g/l of the initial cell concentration.

Effect of Water Temperature, Rearing Density, Salinity, and Food Organisms on The Growth and Survival Rate in Early Juvenile Hard Shelled Mussel, Mytilus coruscus (수온, 사육밀도, 염분 및 먹이생물에 따른 참담치(Mytilus coruscus) 초기 부착치패의 성장과 생존)

  • For the effective seedling production of the hard shelled mussel, Mytilus coruscus, this study assessed the effects of the dietary value of live food, density, water temperature and salinity on growth and survival rate of the larvae. The optimal survival rate and growth rate were examined under differing conditions of water temperature, salinity, and rearing density for 30 days. The three groups were provided different feeding organisms, such as Isochrysis galbana and Teleaulax suecica. The mixtures were provided at a rate of $5{\times}10^4cell\;mL^{-1}$. The best growth was observed in the group with conditions $21^{\circ}C$ water temperature ($16.2{\pm}9.1{\mu}m$), 33 psu of salinity ($16.82{\pm}3.9{\mu}m$), $2500individual\;m^{-2}$ ($17.2{\pm}5.9{\mu}m$), and fed with $5{\times}10^4cell\;mL^{-1}$ of I. galbana and T. suecica mixture ($16.0{\pm}7.3{\mu}m$). The highest survival rate was found in the group at conditions $18^{\circ}C$ water temperature (66.4%), 33 psu of salinity (24.4%), $2500individual\;m^{-2}$ (65.8%), and fed with $5{\times}10^4cell\;mL^{-1}$ of I. galbana and T. suecica mixture (58.8%). We therefore conclude that the suitable culture conditions for the stable production of hard shelled mussel artificial seedlings was at 18 to $21^{\circ}C$ of temperature, 30 to 33 psu of salinity, 2500 to $5000individual\;m^{-2}$ of rearing density, and feeding supplement of $5{\times}10^4cell\;mL^{-1}$ of I. galbana and T. suecica mixture under semi running water system.

Low Serum Medium for CH07E02 Hybridoma (하이브리도마 배양을 위한 저혈청배지의 조성 결정)

  • Up to now, 10% Fetal Bovine Serum(FBS(V/V)) was added to basal medium for the cultivation of hybridoma. For the cultivation of hybridoma cell line, CH07E02, against colon cancer, serum concentration was reduced to 3% FBS without influence on cell growth and maximum cell concentration. By the addition of cell growth promoting substances-insulin (I), pyruvate (P), oxaloacetate(O), Pluronic F-68(P) and 2-mercaptoethanol(2-ME)-to 1% FBS medium, a cell density higher than that with 1% FBS medium alone was achieved. FBS 3% medium was replaced by very cheap 2% Calf Serum (CS) medium without influence on cell growth rate and concentration. Cells grew vigorously in 0.5% CS+IPOP medium. This composition was used during suspension culture and exhibited good viability and high specific growth rate.

  • The pullulan production and morphological change of Aureobasidium pullulans ATCC 9348 were investigated both in batch fermentation and in continuous fermentation. The best carbon source for pullulan production was sucrose among seven different carbon sources. The pullulan production of A. pullulans was increased with increasing the carbon to nitrogen ratio of the medium using sucrose as a carbon source. In batch fermentation, production of pullullan occurred following exhaustion of the nitrogen source from the medium. The continuous fermentation showed that the pullulan production was closely parallelled with cell growth and was most effective at a dilution rate of 0.06~0.07 hr$^{-1}$-. The ratio of yeast-like cells(blastospores) of A. pullulans increased with the increase of growth rate, and reached 100% over the growth rate of 0.07 hr$^{-1}$. The growth rate, within a certain range, affected not only on the cell morphology, but on the specific pullulan productivity of A. pullulans.
