Electrostatic Immobilization of D-Xylose Isomerase to a Cation Exchanger for the Conversion of D-Xylose to D-Xylulose (D-xylose에서 D-xylulose로의 전환을 위한 D-xylose Isomerase의 정전기적 고정화)

  • Since D-xylose is not fermentable in Saccharomyces cerevisiae, its conversion to D-xylulose is required for its application in biotechnological industries using S. cerevisiae. In order to convert D-xylose to D-xylulose by way of an enzyme immobilized system, D-xylose isomerase (XI) of Escherichia coli was fused with 10-arginine tag (R10) at its C-terminus for the simple purification and immobilization process using a cation exchanger. The fusion protein XIR10 was overexpressed in recombinant E. coli and purified to a high purity by a single step of cation exchange chromatography. The purified XIR10 was immobilized to a cation exchanger via the electrostatic interaction with the C-terminal 10-arginine tag. Both the free and immobilized XIR10 exhibited similar XI activities at various pH values and temperatures, indicating that the immobilization to the cation exchanger has a small effect on the enzymatic function of XIR10. Under optimized conditions for the immobilized XIR10, D-xylose was isomerized to D-xylulose with a conversion yield of 25%. Therefore, the results of this study clearly demonstrate that the electrostatic immobilization of XIR10 via the interaction between the 10-arginine tag and a cation exchanger is an applicable form of the conversion of D-xylose to D-xylulose.

  • Retention behavior of lanthanides was studied on the dynamic system using dinonylnaphthalene sulfonic acid cation exchanger and tartaric acid as a complexing agent. The relations between logk' and log$[NH^{4+}]$/log[tartarate], and between logk' and logR showed good linearity in isocratic and gradient elution, respectively. In gradient elution of tartaric acid the slopes were increased as the initial concentrations were decreased. Column efficiencies and resolution of lanthanides on dinonylnaphthalene sulfonic acid equilibrated cation exchanger were found to be better in ${\alpha}$-hydroxyisobutyric acid eluent than in tartaric acid.

  • Prediction of retention times in transition metal-mandelate and transition metal-tartrate complex systems were studied on the cation exchanger. Plots of k' vs [mandelate] and k' vs [tartrate] were obtained under the condition of a constant competing cation concentration. The equation to predict the retention time of transition metal ion was derived from the ion exchange equilibria. Individual capacity factors (${k_1}',\;{k_2}'$) and stability constants ($K_1,\;K_2$) of the complexes were calculated from the non-linear least square method. Good resolution of the transition metals was predicted by the stepwise equation in the gradient method. The values of retention times from the calculation and the experiment agreed well each other.

  • A method for calculating the main characteristics of a potentiometric titration curve in a carboxylic ion exchanger has been investigated. The potentiometric titration curves of simple electrolyte and ion exchangers (polyelectrolytes) showed a great difference between them. The acidity parameters of the ion exchangers, the thermodynamic constant (pK0), apparent equilibrium constant (K), and correction for the apparent equilibrium constant (b), were introduced and used to express the characteristics of the carboxylic ion exchanger. A characteristic equation related to the acidity parameters of the ion exchangers systems was derived. A fibrous carboxylic cation exchanger was used and potentiometric titration curves at different concentrations of the supporting electrolyte were obtained . To prove the validity of the characteristic equation, the concentration of the supporting electroyte was varied. In the present study, good agreement between the data points and the fitted curves was found in all the cases. The g (number of moles of alkali to 1 g of ion exchanger) of carboxylic ion exchanger was calculated from the concentration of supporting electrolyte (C), pH of the solution, and degree of neutralization of ion exchanger (x).

  • Extended use of porcine blood in food ingredients depends on the decolorization of red blood cell concentrates and the modification of its functional properties. The purpose of this study is to compare the relative effect of cation ion exchanger for decolorization of porcine red blood globin. The globin extract is freeze-dried for determination of various functional properties, such as solubility, emulsion capability and foaming ability. Since the isoelectric point of blood globin is located at pH 6.8, which is the neutral pH ranges (6-8), so its functionalities are inferior around these pHs. This weakness has been the main reason, which limit the extended use of blood globin in food industry. Acetylation and succinylation of blood globin can be an alternative way to improve its functionalities. These results may provide new information to understand the decolorization mode by cation ion exchanger for the blood globin. With chemical, the functionalities of blood globin could be obviously improved. The above findings could enable food industry to extend the use of blood globin as a food ingredient.

  • The amount of nitrate in the tobacco leaf has been shown to be correlated with the levels of alkaloids and nitrosamines. Also the nitrate content of the tobacco correlated closely with the smoke delivery of nitric oxide and tobacco-specific nitrosamines (TSNAs). These are related with the effect of the reconstituted tobacco leaf(Recon) using the tobacco stems. Adsorption process is gaining interest as one of the effective processes of advanced liquid treatment for liquid containing unnecessary materials. This study is focused on the evaluation of four anion exchangers, a cation exchanger and an activated carbon, as adsorbents for reduction of nitrate anion from Recon extract. In order to analyze the nitrate anion, the IC method used in this work was carried out with a Dionex ICS-2000 system. The effects of dosages of adsorbents and concentration of extract on the removal of nitrate anion were examined. Experimental results showed that for nitrate-anion exchanger, nitrate-cation exchanger and nitrate-activated carbon adsorption system, approximately 70 %, 10 %, and 4 % removal efficiencies were achieved at the Brix 10 and the 20 % addition. Although the activated carbon was little efficient for removal of nitrate ion, the removal of nicotine was very efficient at given conditions.

Investigation of Adsorption Mechanism and Selective Adsorption of Carbonyl Compounds of Mainstream Tobacco Smoke by Ion Exchangers (이온교환체에 의한 담배 주류연 중 카보닐 화합물의 선택적 흡착 특성 및 흡착 메카니즘 구명)

  • This work has been conducted to select appropriate filter materials for removing carbonyl compounds in mainstream tobacco smoke. To investigate of the usability of this filter materials, two types of bead ion exchangers were synthesized and their adsorption characteristics for carbonyl compounds were investigated. Sulfonic acid group-containing cation exchanger and ammonium group-containing anion exchanger were synthesized by the suspension polymerization of glycidylmethacrylate(GMA) and divinylbenzene(DVB) followed by the subsequent functionalization, respectively. The removal efficiency of carbonyl compounds by these two ion exchangers increased in the presence of moisture. However, the amount of carbonyl compounds adsorbed on the anion exchanger was larger than that on the cation exchanger under two levels of water contents tested. This phenomenon seems to arise from the electron delocalization in carbonyl group of the anion exchangers. There was not any significant relationship between the amount of carbonyl compounds adsorbed on ion exchangers and the length of adsorption column. From the large ion exchange capacity and rapid ion exchange reaction rate of the anion exchanger, it is suggested that the anion exchanger may be a good filter material for removing carbonyl compounds in the mainstream tobacco smoke.

  • Copper is a well known alloying element that is used to improve the resistance to general corrosion of stainless steel And also Cu cation have the anti-fouling effect to inhibit adhesion of the marine algae and shellfish to the surface of heat exchanger cooling pipe or outside wall of the ship, Therefore there are some anti-fouling methods such as anti-fouling Paint mixed with copper oxide or MGPS(Marine Growth Preventing System) by using Cu cation dissolved to the sea wather solution. Cu cation can be dissolved spontaneously by galvanic current due to Potential difference between Cu and cooling pipe of heat exchanger with Ti material, which may be one of the anti-fouling designs. In this study the effect of annealing heat treatment to galvanic current and Polarization behavior was investigated with a electrochemical points of view such as measurement of corrosion Potential, anodic polarization curve. cyclic voltammetric curve, galvanic current etc The grain size of the surface in annealed at $700^{\circ}C$ was the smallest than that of other annealing temperatures. and also the corrosion Potential showed more positive potential than other annealing temperatures. The galvanic current between Ti and Cu with annealed at $700^{\circ}C$ was the largest value in the case of static condition. However its value in the case of flow condition was the smallest than the other temperatures. Therefore in order to increase anti-fouling effect by Cu cation, the optimum annealing temperature in static condition of sea water is $700^{\circ}C$, however non- heat treated specimen in the case of flow condition may be desirable.

The Characteristic Calculation of a Phosphoric Acid Ion Exchanger using the Potentiometric Titration (전위차 적정법을 이용한 인산형 양이온교환수지의 특성 계산)

  • We calculated the characteristics of a phosphoric cation exchanger and studied on an accurately computable method to determine the ion exchange capacity for type of potentiometric titration curve. The ion exchanger was prepared by phosphorylation of a styrene-divinylbenzene copolymer with 4% crosslinking. The ion exchange capacity is 5.7 meq/g. The experimental pK values versus ${\mathit{x}}$ in phosphoric cation exchanger can be expressed as a linear equation. The ${\Delta}pK$ values were obtained from the slope of linear equation. The ${\Delta}pK$ values are the differences of antilogarithms(pK) values of the apparent equilibrium constant at complete and zeroth neutralization of the ion exchanger. Also the experimental pK values at ${\mathit{x}}=0.5$ were accorded well with theoretical data. And when it is titrated with NaOH and $Ba(OH)_2$ solutions, a good agreement between experimental and theoretical pK values for various ${\mathit{x}}$ was seen in all the potentiometric titration curves. We knew that the inflection point of potentiometric titration curve in the case of divalent ions are changed much large than that for monovalent ions. If the relation between g values and ${\partial}pH/{\partial}g$ was plotted to the Lorentz distribution curve, ion exchange capacity can be accurately evaluated.
