Activities of scavenging enzymes of oxygen radicals in early maturation stages of Paragonimus westermani (산소 라디칼 관련 효소의 폐흡충 발육 단계별 활성도 변화)

  • In early maturation stages of Paragonimus westermani (metacercariae, 4-, 8-, 12-week old worms), activities of antioxidant enzymes, such as superoxide dismutase, catalase, peroxidase and glutathione peroxidase, were examined. Specific activity of catalase was the highest in metacercariae and decreasing with age. That of superoxide dismutase was higher in metacercariae and 4-week worms. Specific activity of peroxidase was at its peak in 4-week worms while that of glutathione peroxidase was in 8-week worms. Specific activities of all these antioxidant enzymes were decreased to their lowest in 12-week old adults.

Effects of Biphenyldimethyl Dicarboxylate(DDB) on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in Mercuric Chloride-induced Hepntotoxic Rats (Biphenyldimethyl dicarboxylate(DDB)가 염화 제2수은-유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거효소 활성도에 미치는 영향)

  • In an attempt to define the effects of biphenyldimethyl dicarboxylate (DDB) on the lipid peroxidation and oxygen free radical scavenging enzymes activities in mercuric chloride-induced hepatotoxic rats, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase (SOD) in liver of the rats at 24 hr after the injection of mercuric chloride. Sprague-Dalwey albino rats were injected subcutaneously with mercuric chloride (5 mg/kg) only and mercuric chloride (5 mg/kg) plus. DDB (200 mg/kg/day, p.o) is administered for 4 days prior to 3 days from the injection of mercuric chloride. The group treated with mercuric chloride showed significantly higher MDA level and lower catalase and SOD activities as compared with that of control group. The group treated with mercuric chloride plus DDB showed significantly lower MDA level and catalase activity and higher SOD activity as compared with that of mercuric chloride-treated group. These results suggest that the excessive oxygen free radicals resulting from the depression of superoxide dismutase activity is an important determinant in the pathogenesis of mercuric chloride-induced hepatotoxicity and DDB has antioxidant effects.

  • Srrepromycec. corlirolar produces at least 4 catalase activity bands with different electrophoretic mobilities on polyacrylamide gel which vary during development. Spores and mycelia at stationary phase produced all the activity bands(Cat1. 760 kr); Cat3-I, 170 kD: Cat3-2, 140 kD: Cat3-3. 130 kD; Cat4, 70 kD) except for Cat2 (300 kD). Mycelia at mid-logarithmic phase produced only Cat2 and Cat3-2 bands, and mycelia at late-logarithmic phase produced bands except Catl and Cat\ulcorner. Catalase-deficient mutants were screened in S. coelicalur by H201 bubbling test following NTG mutagenesis. Wc tested sevcral non-bubbling or slow-bubbling mutants for their catalase activities. The overall activities in cell extracts decreased more than 5 fold. Activity bands in native gel selectively decreased in intensity or disappeared. In all the non-bubbling mutants testcd, Cat3-2 band decreased significantly or disappeared. suggesting that Cat3-2 is the major catalase. The selective disappearance of bands in mutants suggest that each band is governed by different genes. We purified catalase activity from -:ell extracts obtained at late-logarithmic phase. Following chromatographies on Sepharose CL-4B. DEAE Sepharose CL-6B. Phcnyl Sepharose CL-4B. and hydroxylapatite columns. only the Cat3-2 activity was obtained. The native form of Cat3-2 has molecular weight of approximately 140 kD, judged by gel electrophoresis. Thc electrophoretic mobility on SDS-polyactylamide gel suggests that this enzyme contains 2 identical subunits of 67 kD.

  • Plants are exposed to wide range of different stresses. As plants have only limited mechanism for stress avoidance, they require flexible means for adaption to changing environmental conditions. This study was carried out to reasearch the changes of antioxidant enzymes activities as caused by water stress in four lettuceUactuca sativa) lines. Four lettuce lines exposed to water stress showed premature senescence as evidenced by the consistenent reduction in the content of total soluble protein and total lipid. Water stress also caused decreased activities of superoxide dismutase, catalase, ascorbate peroxidase, but decrease rates were different. Catalase activity was decreased much more than that of ascorbate peroxidase that suggest catalase reacted with hydrogenyperoxide directly not with ascorbate peroxidase.

Dose-Response Relationship between Catalase and Superoxide Dismutase Activity in Testes of Acutely Intoxicated Rats by Cadmium (급성 카드뮴중독시 랏트의 고환조직내 Catalase 및 Superoxide Dismutase 활성도와의 양-반응관계)

  • Dose-response relationship among blood cadmium concentrations, catalase and superoxide dismutase activities were studied with acutely intoxicated rats by cadmium. The Sprague-Dawley male rats to which single dose of $1{\sim}32mg/kg\;CdCl_2$ were administered into peritoneal cavity were sacrificed by decapitation at $3{\sim}36$ hours after the administration. Cadmium concentrations in blood increased significantly with dose of $CdCl_2$ administered and reached peak level at 3 hours later. Catalase activities in rats' testes were not correlated with esposure time elapsed after the administration in rats to which $1{\sim}2mg/kg\;of\;CdCl_2$ were administered, but they showed linear relationship with time in groups to which $4{\sim}32mg/kg\;of\;CdCl_2$ were administered. Cu, Zn-SOD activities in testes of acutely intoxicated rats by cadmium were not altered either by dosage or by time elapsed after the administration. Mn-SOD activities in the testes were also not influenced by dosage of $1{\sim}2mg/kg\;CdCl_2$, but remarkably inactivated by dosage of $4{\sim}32mg/kg\;CdCl_2$ with time elapsed after the administration. Neither catalase, Cu, Zn-SOD nor Mn-SOD activities of testes were correlated with blood cadmium concentrations in acutely intoxicated rats by cadmium.

  • In an attempt to define the effects of Biphenyldimethyl dicarboxylate(DDB) on the lipid peroxidation, oxygen free radical scavenging enzymes activities and hepatic functions in ethanol-induced hepatotoxic rats, we studies malondialdehyde(MDA) level and the activities of catalse, superoxide dismutase(SOD), glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) in liver of the rats at 24, 48 and 72 hr after the injection of ethanol and DDB. Sprague-Dalwey albino rats weighing 250 to 280gm were injected intraperitoneally with ethanol(2.5 gm/kg ) only and ethanol plus DDB(300mg/kg ). The result obtained can be summarized as follows : 1) The group treated with ethanol showed significantly higher MDA level and lower catalase and SOD activities at 24, 48 and 72hr after the injection as compared with that of control group. 2) The group treated with ethanol showed significantly higher GOT and GPT activities at 24, 48 and 72hr after the injection as compared with that of control group. 3) The group treated with ethanol plus DDB showed significantly lower MDA level and higher catalase and SOD activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. 4) The group treated with ethanol plus DDB showed significantly lower GOT and GPT activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. These results suggest that the excessive oxygen free radicals resulting from the depression of the activities of catalase and superoxide dismutase is an important determinant in pathogenesis of ethanol-induced hepatotoxicity and DDB has antioxidant effects.

Effects of Hyperbaric Oxygen Treatment on the Malondialdehyde Level and Activities of Catalase and Superoxide Dismutase in the Kidney of the Rats Exposed to Carbon Monoxide (일산화탄소 폭로후 고압산소 투여가 흰쥐 신장에서의 malondialdehyde 함량과 catalase 및 superoxide dismutase 활성에 미치는 영향)

  • In an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen free radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde (MDA) level and activities of catalase and superoxide dismutase in the kidney of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260 gm were used. Experimental groups consist of Control group (=breathing with air), HBO group (=exposed to hyperbaric oxygen 〔HBO, 3ATA, 100%〕 after air breath), CO group (=exposed to CO〔3,970 ppm〕after air breath), CO-Air group (=exposed to CO after air breath followed by air breath) and CO-HBO group (=exposed to CO after air breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of CO-induced nephrotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the kidney of the rats exposed to carbon monoxide.

  • Mycobacterium sp. strain JCl DSM 3803 grown in methanol showed no methanol dehydrogenase or oxidase activities found in mast methylotrophic bacteria and yeasts, respectively. Even though the methanol-grown cells exhibited a little methanol-dependent oxidation by cytochrome c-dependent methanol dehydrogenase and alcohol dehydrogenase, they were not the key enzymes responsible for the methanol oxidation of the cells, in that the cells contained no c-type cytochrome and the methanol oxidizing activity from the partially purified alcohol dehydrogenase was too low, respectively. In substrate switching experiments, we found that only a catalase-peroxidase among the three types of catalase found in glucose-grown cells was highly expressed, in the methanol-grown cells and that its activity was relatively high during the exponential growth phase in Mycobacterium sp. JCl. Therefore, we propose that catalase-peroxidase is an essential enzyme responsible for the methanol metabolism directly Of indirectly in Mycobacterium sp. JCl.

Conjugated Linoleic Acid Supplemented to Dietary Fat Has an Antioxidant Activities, but It Depends on the Type of Fat in Diet (쥐에서 식이에 보충한 Conjugated Linoleic Acid가 식이지방 종류에 따라 항산화작용에 미치는 영향)

  • The study was designed to observe an antioxidant activities of conjugated linoleic acid(CLA) in rat liver by determining the activities of antioxidative enzymes(superoxide dismutase, glutathione peroxidase, catalase) and the levels of tocopherol and thiobarbituric acid reactive substance(TBARS). Male Sprague Dawley rats at weeks-old were divided into groups according to the types of dietary fat(beef tallow and fish oil) and then each group was subdivided into groups depending on CLA supplement. All rats were fed experimental diet containing 12% total fat including 1% CLA by weight for 30 weeks. CLA supplemented to beef tallow diet did not have significant effect on the level of TBARS and tocopherol. The level of TBARS was significantly increased in fish oil diet(highly unsaturated fat diet), but its level was significantly reduced by increasing SOD and GSH-Px activities when CLA was supplemented to fish oil diet so that CLA showed a sparing action of tocopherol in tissue. CLA did not have significant effect on peroxisomal catalase activities, but its activity was significantly increased when TBARS production was high in the fish oil diet. CLA could be incorporated into phospholipid of microsomal membrane, and interfered the conversions of C18 : 0 into C18 : 1 and C18 : 2 into C20 : 4 in liver. In conclusion, CLA had an antioxidant activities depending on the type of fat in diet. Therefore, it could be recommended to use CLA when highly unsaturated fat was used in meal preparation.

In vivo Antioxidant Effects of Aralia elata Seemann Ethanol Extract Administered with Benzo($\alpha$) pyrene

  • This study has examined the effects of Aralia elata Seemann ethanol extract on antioxidant enzyme systems inrats along with benzo($\alpha$) pyrene(B(a)P) administration . The ethanol extract of Aralia elata Seemann (50mg/kg body wt.) was fed to rats for 4 weeks by stomach tubing. The extract administration increased antioxidant activities of glutathione sulfur transferase(GST) comparing to the control. also total superoxide dismutase(SOD) and Cu, Zn-SOD activities were stimulated. Catalase activities were increased by 50% with the extract feeding compared to the control . Combined administration of B($\alpha$)P and the extract increased GST activity in B($\alpha$) P group. Although total SOD acitivity was decreased , Cu, Zn-SOD was greately increased from 0.10unit to 0.18 unit and catalase activity also was increased compared to the group of B($\alpha$) P. GST activity in CLE group was 1.32 unit, increased by 33% comparing to the group CL of 0.99unit. Cu, Zn-SOD and catalase activities in thegroup fed high fat and ethanol extracts were increased by 25% and 39%, respectivley comparing to the group of high fat. In addition , total SOD was decreased but, Cu, Zn-SOD acitivity was increased from 0.09 unit to 0.18unit. Catalase activity was 76.05 unit in the group of B($\alpha$) P and extract comparing to 65.26 units in B($\alpha$)P group. Serum $\alpha$-tocopherol of rat was markedly increased by theextract. Administration of B9$\alpha$)P reduced $\alpha$-tocopherol levels in the serum, on the other hand, lard in the diet increased $\alpha$-tocopherol levels in the serum. The above results indicate that Aralia bud exerts antioxidant functions in vivo against B($\alpha$)P. Further research may be necessary for the identification fo the biologically active material.
