Studies on the Anti-inflammatory Agents. Synthesis of Carboxamides and Their Anti-inflammatory Activity

  • Four compounds, benzamide, N-(3-trifluoromethylphenyl) benzamide, N-(2,3-dimethylphenyl) cinnamamide and N-(3-trifluoromethylphenyl) cinnamamide, were synthesized and evaluated their activity by the method of the inhibitory effect on the carrageenin-induced rat paw edema, compared with phenylbutazone. All of these exhibited anti-inflammatory activity, and N-(2,3-dimethylphenyl) benzamide exhibited 45.7% edgema inhibition vs. control when 100mg/kg of dosage was administered, whereas phenylbutazone, 51.4% for 50mg/kg.

  • From the leaves of Lindera sericea Bl., kaempferol-3,7-0-di-a-L-rhamnopyranoside was isolated and characterized by spectral data and acid hydrolysis. This compound is reported for the first time from this plant. The content of kaempferol glycoside was high in May and June, and low in the fall. And the methoanolic extract of title plant showed the inhibitory effect on carrageenin-induced edema.

  • The anti-inflammatory effect of the methanolic extracts of some medicinal plants was investigated on the hydrolysis of N-benzoylarginine-p-nitroanilide by trypsin in vitro, the writhing syndrome induced by acetic acid in mice, and the carrageenin-induced paw edema in rats. Among tested medicinal plants, the methanoilc extract of Angelica keiskei, Rosa rugosa and Cudrania tricuspidata showed significant inhibitory effect. And we also found that adenosine, isolated from A. keiskei was identified as one active components.

  • A rare flavonol glycoside identified as $eupalitin-3-O-{\beta}-D-glucoside$ (I) was isolated from Tephrosia spinosa (Leguminosae) and its anti-inflammatory activity was evaluated against carrageenin induced paw edema. The compound exhibited significant activity when compared to the standard drug indomethacin.

Pharmacological Studies on Butanol Fraction of Puerariae Radix (갈근 Butanol Fraction의 약리 작용에 관한 연구)

  • Effective BuOH fraction was extracted from Puerariae radix (Pueraria thunbergiana), and several pharmacological activities were examined. From several pharmacological examinations, it was found that BuOH fraction has an antipyretic activity against typhoid vaccine pyrexia in rabbit, a papaverine-like antispasmodic activity on isolated ileum and jejunum of rat, an analgesic effects on mice treated by 0.7% HAc, and an anti-inflammatory activity of carrageenin edema in rat. It was found that BuOH fraction has detoxication effects on acute and subacute toxic rat induced by alcohol in consequence of biochemical tests of serum. Therefore it was considered that BuOH fraction has preventive and therapeutic effects in alcohol intoxication.

  • Anti-infammatory, analgesic and ulcerogenic activities of fentiazac were investigated in comparison with those of acetylsalicylic acid, fenbufen, naproxen and phenylbutazone. On the anti-inflammatory activity in carrageenin-induced rat paw edema and the analgesic activity on writhing syndrome induced with acetic acid in mice, fentiazac displayed more potent effect than acetylsalicylic acid, fenbufen and pbenylbutazone. But the ulcerogenic action of fentiazac on gastrointestinal tract in fasting rats was less than that of reference drugs. From these investigation, fentiazac seemed to indicate a poor correlation between the extent of anti-inflammatory activity and ulcerogenic action.

  • In the previous report, it was described that the butanol fraction of the root bark extract of Aralia elata had potent antigastritic and antiulcerative effects in rats, In this pharmacological profile, the fraction revealed low acute toxicities in mice and rats, no central nervous system activities, moderate analgesic action, weak inhibitory action in carrageenin edema of rats and no direct effects on isolated smooth muscles of rats and guinea-pigs. Slight lowering of blood pressure as given i.v. in rabbit was observed.

  • To observe the diuretic action in mice, the anti-inflammatory action by carrageenin method and the anti-pyretic action by yeast method in rats, the Bittern was administered. Diuretic effort of Bittern was studied by measuring the urine flow, sodium, potassium in urinary excretion. The results in this work were summarized as follows: 1. The Bittern showed significant diuretic action. 2. The Bittern showed significant anti-inflammatory effect. 3. The Bittern showed significant anti-pyretic effect. According to the above results, the Bittern seems to be applicable to the treatment of edema.

  • Alpha-viniferin was isolated from Carex humilis (Cyperaceae), and showed anti-inflammatory effects on carrageenin or histamine-induced paw edema in mice. To understand mode of the anti-inflammatory action. effects of alpha-viniferin on cyclooxygenase (COX)-2, iNOS, oxygen radicals and proinflammatory cytokines have been analyzed. Alpha-viniferin showed selective inhibitory effect with an IC50 value of 5 $\mu\textrm{m}$ on COX-2 activity but showed weak inhibitory effect on the synthesis of COX-2 transcript which was identified by RT-PCR.   • 'Sochungryoung-Tang' has been known to be used in acute edema, bronchitis, pneumonia, nephrosis, and arthritis etc. in traditional chinese medicine. In this study, it was examined on the antimicrobial and anti-inflammatory activities. The following results were obtained; 1) In the antibacterial activities, the ethanol extract of 'Sochungryoung-Tang' showed to be more active than water extract aginst ten bacterials (Table I). Especially, antibacterial activities against Staphylococcus sp. showed the highest among the strains tested. But in the fungi, ethanol extract and water extract showed slightly significant effects (Table II). 2) The water extract showed significant anti-inflammatory effects against the rat hind paw edema induced by 1% carrageenin in the scab and carbuncle in the mice in fected by Staphylococcus aureus CUCC 74371.
