Anti-inflammatory and Analgesic Activities of the Root of Caragana koreana ("참골담초"근(根)의 소염(消炎) 및 진통작용(鎭痛作用)에 관(關)한 연구(硏究))

  • The studies on the anti-inflammatory and analgesic activities were carried out to develop the domestic natural sources of the underground parts of Caragana koreana Nakai (Leguminosae), which have been used for the treatment of neuralgia, arthritis, and gout etc. in folk remedies. The methanol extract of the root was shown significant antiinflammatory effect in the paw edema induced by carrageenin, and also indicated remarkable preventive and therapeutic effects to chronic arthritis model induced by Freund's complete adjuvant in rats. It also has inhibited significantly the increase in vascular permeability and the number of writhings induced by acetic acid in mice.

  • Antiinflammatory action of phytolaccosides B and E isolated from the roots of Phytolacca americana was investigated and compared with that of aescin. Phytolaccoside B, when it was administered intravenously, exerted a potent inhibitory effect on the transudation caused by absorptive sponge pellet implanted s.c. in rats and on carrageenin induced edema in rats. Phytolaccoside E showed a weaker action than Phytolaccoside B did. Hemolytic potency of the saponins was decreased in the order of aescin, phytolaccosides B and E showing a same tendency as in antiinflammatory action. The experimental result suggests that the mode of action of phytolaccosides is the same as that of aescin.

  • The recombinant gamma-interferon (LBD-001) which was produced by yeast as host system was investigated on the pharmacological activities. This gamma-interferon exhibited potent inhibitory effect on adjuvant induced arthritis, but no effect on carrageenin induced paw edema in rats. It did not show any sedative, anticonvulsive, analgesic and hypothermic activities in animals. It also had no influences on isolated tracheal muscle and ileum of guinea pig, isolated uterus and fundus strip of rats, and on blood pressure and respiration in situ experiments of rabbits.

  • The anti-inflammatory and analgesic actions of Cedrela sinensis, Oenanthe javanica and ARtemisia princeps var. orientalis were carried out. The methanolic extract of Artemisia princeps var. orientalis showed the inhibitory effect on carrageenin-induced edema as well as vascular permeability in mice. And we also found the alagesic activity in these plants.

  • Protocollagen proline hydroxylase (PPH) activity in the course of rebound of rat carrageenin granuloma was assyed to investigate its relationship with collagen blosynthesis. The specific activity of PPH which was inhibited significantly by treatments with hydrocortisone acetate 93mg/kg/day, 3days) was recovered near to the normal level by 48hr after the cessation of the corticoid treatments. The total enzyme activity of the granuloma of treated group was not yet recovered even on the 3rd day, still showing a significant difference from control. PPH activity expressed in terms of granuloma cells between control and the drug treated group, however, was changed showing no apparent difference throughout the experimental period. These results suggested that the synthesis of protocollagen(collagen precursor) rather than PPH activity was directly affected by administration and withdrawal of corticoid.

Anti-inflammatory Activity of Propolis

  • Propolid (bee-glue), known as a folk medicine, is a lipo;hilic material found in honeybee hives. In the present study on the anti-inflammatory effect of Korean propolis, it was extracted with ethanol, and used as a test material. The $LD_{50}$ value with the oral administration of ethanolic extract of Korean propolis (EEKP) was higher than 2g/kg in mice. The oral administration of the propolis extract (100mg/kg) significantly inhibited the development of hind paw edema induced by carrageenin in rats. the oral pretreatment of the propolis extract markedly inhibited the increase in vascular permeability and the number of writhing induced by acetic acetic acid in mice. Propolis extract, 50 and 100 mg/kg p.o. per day for 7 days, produced a significant inhibitory effect on granuloma and exudate formation in rats. This inhibitory effect was enhanced with the concomitant use of prednisolone (2.5 mg/kg). These results suggest that Korean propolis apparently has a strong anti-inflammatory activity.

  • Besides (-) pimara-8(14), 15-dien-19-oic acid [I] which had already been isolated as an active anti-inflammtory principle of Aralia continentalis, (-) kaur-16-en-19-oic acid [II] was separated as another active component of the plant, by tracing albumin stabilizing activity. $IC_{50}$ of [II] for the protein stabilizing activity was 0.026mg/3ml, when those of [I] and phenylbutazone were 0.032 and 0.32 mg/3ml, respectively. Being investigated employing carrageenin-induced edema test in rat hind paw, the anti-inflammatory activity of [II] administered s. c. was slightly lower than that of phenylbutazone, whereas the activity of [II] administered p. p. was three times greater than that of phenylbutazone. These results of [II] were contrary to those of [I] in the aspect of administration routes.

  • Stephania hernandifolia belonging to the family Menispermaceae is the biggest storehouse of diphenylbisbenzylisoquinoline (DBBI) alkaloids. Exhaustive chemical processing of the bulb of S. hernandifolia by the application of modern separation techniques yielded a DBBI alkaloid which was identified as cycleanine using spectroscopic methods (UV, IR, $^1HNMR$. $^{13}CNMR$, Mass). Cycleanine showed significant anti-inflammatory activity against carrageenin induced paw oedema, comparable to that produced by diclofenac sodium, used as standard drug. It exhibited potent analgesic effects against chemical and thermal noxious stimuli. It was also found to possess anticonvulsive activity in the strychnine induced convulsion model.

Biological Activities of Pedicularis resupinata var. oppositifolia (마주송이풀의 생리활성(生理活性))

  • The plant of Pedicularis resupinata var. oppositifolia(Scrophulariaceae) is described to be used for treatment of rheumatoid arthritis, malignant abscess (tumor), urolith and diuretics in oriental medicinal books. This study is concerned with some general pharmacological activities. It was observed that the methanol extract of the aerial part of this plant showed some effects on acute toxicity, carrageenin induced edema in rat paw, bile juice secretion and antibacterial effect. From this result it was obtained that the methanol extract revealed the antiinflammatory activity and bile juice secretion increasing effect. Thus, it was fractionated and buthanol fraction has obvious inhibition effect on edema and bile juice secretion increasing effect. The total extract showed low acute toxicity i.e., the minimum lethal dose was more than 8000mg/kg in oral administration in mice.

  • Three components, fraxetin-8-glucoside, esculetin-6-glucoside and mannitol, were isolated from the stem bark of Fraxinus chiisanensis var. stenophylla, F. japonica var. intermedia and F. densata. The MeOH extract of the cortex of F. densata had the antiinflammatory activity on the carrageenin-induced paw edema in rat. The MeOH extracts of the cortex of all three Fraxinus spp. have the potent analgesic activity on the HOAc-induced writhing syndrome in mouse and the hepatoprotective activity on the $CCl_4-induced$ fatty liver in rat: protection of ballooning formation and inhibition of sGPT and sGOT increased by $CCl_4$.
