Development of Fermented Isotonic Beverage with Anticariogenic Activity using Bacteriocin-Producing Lactic Acid Bacteria (박테리오신 생성 젖산균을 이용한 항충치 활성을 지닌 발효이온음료 개발)

  • The fermented fruit and vegetable mixed broth was prepared by using bacteriocin-producing lactic acid bacteria to evaluate the possibility of developing isotonic beverage with anticariogenic activity. Optimum conditions were also established to produce bacteriocin by a mixed culture system consisting of Lc. lactis and Leu. mesenteroides in a fruit and vegetable mixture. Production of bacteriocin was not observed when both strains were simultaneously innoculated, but pH adjustment of the broth fermented by Leu. mesenteroides to nearneutral pH stimulated the production of bacteriocin by Lc. lactis. The concentration of sodium of the fermented broth was higher than those of commercial products. Color change of the fermented broth was not observed during storage. The fermented broth showed strong inhibitory effect against Streptococcus mutans which is an oral inhabitant with a cariogenic activity. Bacteriocin activity in the fermented broth was retained very stable for 4 weeks at 4$\^{C}$. The results indicated that bacteriocin-producing lactic acid bacteria can be used for the preparation of a Korean style thirst-quenching beverage containing bacteriocin.

  • The purpose of this study was to compare the growth-inhibiting effect of sodium fluoride and stannous fluoride on a cariogenic strain of Streptococcus mutans exposed to the different concentration of $SnF_2$ and NaF The result were as follows: 1. The growth rate of the Streptococcus mutans was unaffected by 75 ppmF, slowed by 150 ppmF, and immediately arrested by 300 or 600 ppmF where increments of NaF were added to actively growing 12-hour broth culture. 2. $SnF_2$ slowed the growth rate at 75 ppmF, was to bactericidal initially at 150 and 300 ppmF, and was to totally bactercidal at 600 ppmF. 3. $SnF_2$ has been shown to be more effective than NaF in inhibiting the growth activity of Streptococcus mutans. 4. The inhibitory effect of fluoride compounds on the growth of Streptococcus mutans was increased in the decreased pH at concentration of 600 ppmF.

Antimicrobial Effect of Polyphenon 60 against Streptococcus mutans and Streptococcus sobrinus

  • Polyphenon 60 refers to the mixture of catechins present in green tea. The aim of this study was to investigate the antimicrobial activities of polyphenon 60 against 4 strains of Streptococcus mutans and 2 strains of Streptococcus sorbrinus, which are the major causative bacteria of dental caries. The minimum bactericidal concentration (MBC) values of polyphenon 60 for S. mutans and S. sobrinus were determined and the effect of biofilm formation inhibition of that was evaluated. The MBC value of polyphenon 60 against the bacterial strains was 2.5 mg/ml except for one particular strain, S. mutans KCOM 1128 for which the value was 1.25 mg/ml. The results of biofilm formation inhibition assay revealed that polyphenon 60 inhibited biofilm formation more than 90% at a concentration of 2.5 mg/ml. It was apparent that polyphenon exhibited biofilm formation inhibition activity along with bactericidal effect against S. mutans and S. sobrinus. Therefore, it is proposed that polyphenon 60 as one of the components of bactericidal agents could be useful in developing oral hygiene products, toothpaste or gargling solution.

EFFECT OF RESIN MONOMERS ON THE ACTIVITY OF CARIOGENIC BACTERIA (수종의 레진 모노머가 우식 유발성 세균의 활성에 미치는 영향)

  • The purpose of this study was to examine the effects of resin composite monomers (Bis-GMA, TEGDMA, EGDMA, UDMA, HEMA, Camphorquinone) on the growth of the two cariogenic bacteria, Streptococcus mutans and Streptococcus sobrinus. We obtained the following results : 1. The growth rate of S. mutans and S. sobrinus was decreased significantly in the group of all composite resin monomer at a concentration of 0.03mmo1/L(P<0.01). 2. The growth rate of S. mutars in the group of UDMA at a concentration of 0.01 mmol/L and the group of CQ at a concentration of 0.005 mmol/L, 0.01 mmol/L was decreased significantly compared to the control group(P<0.01). 3. The growth rate of S. sobrinus in the group of HEMA, UDMA at a concentration of 0.01 mmol/L and the group of CQ at a concentration of 0.005mmol/L. 0.01mmo1/L was decreased significantly compared to the control group (P<0.01). 4. The growth rate of S. sobrinus in the group of EGDMA at a concentration of 0.001, 0.01, 0.03mmo1/L was decreased significantly compared to the control group(P<0.01) and were showed to be statistically significant difference between experimental groups(P<0.01).

A Study on Artificiality Salivary pH and Sugar Fermentation Test of Caries Potentiality Foods (우식유발식품에 의한 인공타액내 pH 변화와 당 분해 효소 활성에 관한 연구)

  • This study was designed to provide basic data for developing a standardized caries potentiality index to help to choose snack foods with a low score on the index by investigating adolescents' snack intakes and measuring them in total saccharinity, pH, and acidogenic potential and glucosidase activity of Streptococcus mutans (S. mutans). Total 28 snack foods were selected and measured for total saccharinity (Pocket refractometer PAL-1, ATAGO) and pH with a pH meter (Mentor, Seoul, Korea). Artificial saliva was added to each sample. The experiment group was administered with S. mutans (ATCC 3692), cultured in a constant temperature incubator at $37^{\circ}C$, and measured for pH changes over five times including after 10 minutes, 30 minutes, 1 hour, and 24 hours. Each sample of 0.3 ml was added to tubes containing sucrose, galactose, or glucose to measure the glucosidase activity of S. mutans. They were then observed for glucosidase activity with colorimetry after 24 hours of culture in a constant temperature incubator at $37^{\circ}C$. The mean pH by the acidogenic potential of S. mutans was pH 5.33. The experiment group dropped in pH more than the control group due to the increasing acidogenic potential of S. mutans by glucosidase activity, recording pH 5.27 after 10 minutes, pH 5.21 after 30 minutes, pH 5.15 after 1 hour, and pH 4.80 after 24 hours. The observation results of glucosidase activity of S. mutans with colorimetry show that most of the samples were positive in orange and yellow with glucose, sucrose, and glactose recording activity of 78.58%, 75%, and 71.42%, respectively.

Effect of the Antimicrobial Peptide $\small{D}$-Nal-Pac-525 on the Growth of Streptococcus mutans and Its Biofilm Formation

  • Streptococcus mutans is the primary etiological agent of dental caries. The antimicrobial peptide $\small{D}$-Nal-Pac-525 was designed by replacing the tryptophans of the Trp-rich peptide Pac-525 with $\small{D}$-${\beta}$-naphthyalanines. To assess the effect of $\small{D}$-Nal-Pac-525 on cariogenic bacteria, the activity of $\small{D}$-Nal-Pac-525 on the growth of S. mutans and its biofilm formation were examined. $\small{D}$-Nal-Pac-525 showed robust antimicrobial activity against S. mutans (minimum inhibitory concentration of 4 ${\mu}g/ml$). Using scanning electron microscopy and transmission electron microscopy, it was shown that $\small{D}$-Nal-Pac-525 caused morphological changes and damaged the cell membrane of S. mutans. $\small{D}$-Nal-Pac-525 inhibited biofilm formation of S. mutans at 2 ${\mu}g/ml$. The results of this study suggest that $\small{D}$-Nal-Pac-525 has great potential for clinical application as a dental caries-preventing agent.

THE INHIBITORY EFFECT OF CHLORHEXIDINE VARNISH AND POLYURETHANE SEALANT ON STREPTOCOCCUS MUTANS IN THE PRIMARY DENTITION (유치열에서 Chlorhexidine Varnish와 Polyurethane Sealant의 Streptococcus mutans억제 효과에 대한 연구)

  • Numerous chemical agents have been developed to reduce the activity of cariogenic bacteria. Of these, chlorhexidine is acknowledged as the most effective. Gel and mouthrinse have been the traditional method of its application in the mouth. It has been reported that chlorhexidine varnish has prolonged inhibitory effect on the number of streptococcus mutans in saliva and plaque. Recently, chlorhexidine varnish and polyurethane sealant have been developed to promote prolonged anticariogenic effect of chlorhexidine. Products containing 10% chlorhexidine varnish and polyurethane sealant have been developed to prevent caries by reducing the number of streptococcus mutans in the oral cavity. The purpose of this study is to investigate the inhibitory effect of chlorhexidine varnish and polyurethane sealant on streptococcus mutans in the primary dentition. Children with primary dentition containing no active carious lesion were divided into two groups. To the experimental group (n=11), chlorhexidine varnish and polyurethane sealant ($Chlorzoin^{(R)}$, Knowell Therapeutic Technologies, Inc. Canada.) was applied once a week for four weeks according to the manufacturer's instruction. Only oral prophylaxis was performed on the control group(n=7). Caries activity was measured after using $Cariescreen^{(R)}$SM (Knowell Therapeutic Technologies, Inc. Canada,) to incubate streptococcus mutans before and 5, 12, 24 weeks after initial varnish application. The following results were observed.; 1. There was statistically significant decrease in the number of streptococcus mutans in the experimental group for 5 weeks(P<0.01), 12 weeks(P<0.05) after the initial application. but, by 24 weeks significant difference had disappeared. 2. As the inhibitory effect of chlorhexidine varnish and polyurethane sealant application is not everlasting, reapplication at 12-24 weeks should be needed.

Effects of the Medicinal Plants Against Streptococcus Sobrinus B13 in Hexane Fraction (수종의 생약 헥산분획 추출물에 의한 Streptococcus sobrinus 억제 효과)

  • This study was carried out to assess anticariogenic activity from natural products. The hexane extracts of 11 medicinal plants were tested for the antimicrobial activities against a cariogenic bacterium, S. sobrinus B13. The hexane extracts of 11 medicinal plants such as Alisma orientale, Cyperus rotundus, Saussurea lappa, Atractylodes japonica, Kalopanax pictus, Angelica gigas, Salvia miltorrhiza, Lonicera japonica, Gardenia jasmoides, Aurantii nobilis percarpium and Bupleurum lorgiradiatum were used for this study, and S. sobrinus B13 was cultured with 0.5% extracts of the medicinal plants, in trypticase soy broth medium under 5% $CO_2$ for 0, 6, 12, 24 hours at $37^{\circ}C$. The obtained results were as follows. Immediately after culturing for inoculation, Kalopanax pictus and Cyperus rotundus showed better antimicrobial activities than those of others. After culturing for 6 hours, the order of effective antimicrobial activity was Salvia miltorrhiza, Angelica gigas. After incubation for 12 hours, Salvia miltorrhiza showed the best antimicrobial activity. After incubation for 24 hours Salvia miltorrhiza, Cyperus rotundus, Saussurea lappa, Atractylodes japonica had better antimicrobial activity than others. In conclusion, among Hexane extracts of 11 medicinal plants, Cyperus rotundus, Saussurea lappa, Atractylodes japonica showed effective antimicrobial activity.

Antibacterial Activity of Sea-mustard, Laminaria japonica Extracts on the Cariogenic Bacteria, Streptococcus mutans (충치균 (Streptococcus mutans)에 대한 다시마 추출물의 항균활성)

  • In previous reports the authors have screened the inhibition effects of marine algae extracts on halitosis, and demonstrated that a brown algae, Eisenia bicyclis (' Daehwang') possess not only strong deodorant effect bug also considerable anticariogenic activities. In this study, we screened antibacterial effects of various marine algae, and measured minimum inhibitory concentration (MIC) value of them against mutans streptococci in vitro. Among the 27 species of marine algae, $80\%$ ethanol extract of dried sea-mustard, Laminaria japonica ('Dasima') showed the strongest inhibition activity against Streptococcus mutans KCTC 3300. The extracts of Ulva lactuca ('Galparae'), Codium fragile ('Cheonggak'), Ecklonia cava ('Gamtae'), E. stolonifera ('Gompi') and Undalia Pinnatifida ('Miyeok') showed slightly weaker inhibitory potency than L. japonica. Differences of MIC values in $80\%$ ethanol extract of some species of marine algae were observed depending on test bacterial species, i.e., S. mutans KCTC 3300 or S. sobrinus KCTC 3307. Eighty percent ethanol extract of dried L japonica was fractionated with diethyl ether, chloroform, ethyl acetate, n-buthanol and water successively, The ether-soluble fraction had inhibitory effect on S. mutans KCTC 3300, however the inhibitory effects were not found in the other fractions. The MIC values of $80\%$ ethanol extract and ether fraction were 180 and 105 $\mu$g/mL respectively, while no significant inhibition activity of water-soluble fraction was found even when the fraction was added up to 5,500 $\mu$g/mL.

Effect of Gargle Containing Citrex and Cetylpyridinium Chloride on the Oral Pathogenic Bacteria (수종약물성분이 함유된 양치액의 구강질환 유발균 및 구강환경에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate that the effect of Citrex and CPC on the growth of oral microorganism such as Streptococcus mutans KCTC 3065, Candida albicans KCTC 7122, and Staphylococcus aureus KCTC 1916 which were cariogenic, candidiasis(bleeding gum, dry mouth and tongue, thrush) and angular cheilitis inducing bacteria, respectively. The efficacy of complex of Citrex and CPC was determined in assays measuring Halo Test. The gargle containing Citrex(0.02%) and CPC(0.02%) demonstrated broad-spectrum anti-microbial properties, with activity against both Gram-positive and Gram-negative and a yeast(Candida albicans).
