Purification of Capsular Polysaccharide Produced by Streptococcus pneumoniae Serotype 19A

  • Streptococcus pneumoniae is a major cause of invasive infection in young infants and older adults. There are currently 90 capsular serotypes identified and 23 serotypes (1, 2, 3, 4, 5, 6B, 7F, 8, 9N, 9V, 10A, 11A, 12F, 14, 15B, 17F, 18C, 19F, 19A, 20, 22F, 23F, and 33F) are responsible for about 90% of invasive disease. Among the more than 90 different S. pneumoniae serotypes, serotype 19A is globally very prevalent. A simplified purification procedure including adjustment of cell lysate pH to 4.5, fractionation with 50. 80% ethanol, and dialysis rendered capsular polysaccharide (CPS) in a yield of $31.32{\pm}3.11$ mg from 1 l culture (75% recovery after lyses). The product contained only 69.6 ${\mu}g$ of protein (99.78% purity) and 0.8mg (sum of the precipitants from 50~60%, 60~70%, and 70~80%) of nucleic acid (97.45% purity). The purified CPS was conjugated with bovine serum albumin; the product size ranged from 100 to 180 kDa.

Prevalence and serotypes of Streptococcus suis from pigs in Korea (국내 돼지의 Streptococcus suis 감염율 조사 및 혈청형 동정)

  • To investigate the prevalence of Streptococcus suis in pig industry in Korea, we isolated S suis from pigs during two years. The isolates were identified by biochemical and coagglutination test. Serotypes and antimicrobial susceptibility of the isolates were determined. Fourteen strains(12.2%), 11 strains(27.5%), 58 strains(7.8%) and 4 strains(11.1%) of S suis were isolated from 115 nasal swabs, 40 tonsils of healthy pigs, 745 pneumonic lungs and 36 meningitis of diseased pigs, respectively. The isolates were highly susceptible to ampicillin, cephlothin and ofloxacin while they were resistant to oxytetracycline. Among the isolates, 63(75.9%) strains were to the S suis capsular type 1 to 10 and 11(13.3%) strains were untypable. Capsular type 2 was the most prevalent with 32(38.6%) strains of all isolates.

Capsular Polysaccharide Serotypes Among Staphylococcus aureus Isolates from Cases of Bovine Mastitis and Dogs (젖소 유방염과 개에서 분리된 황색포도상구균에 대한 Capsular Polysaccharide형의 동정)

  • 본 연구는 임상형 및 준임상형 유방염에 이환된 젖소의 유즙에서 분리된 24주의 황색포도상구균에 대한 capsular polysaccharide(CP)형을 확인하고, 1% 토끼 혈청이 함유된 serum-soft agar 배지에서 집락의 모양을 관찰하였다. 또한 동물에 따라 우세한 CP형의 차이가 있는지를 비교하고자 개에서 분리된 13주에 대하여 동일한 실험을 수행하여 다음과 같은 결과를 얻었다. 집락모양을 관찰한 결과 젖소 유래 24주 중 16주(66.7%)는 diffuse, 5주(20.8%)는 compact, 나머지 3주(12.5%)는 분류가 불가능(indeterminate)하였다. CP형 확인결과 9주(37.5%)는 type 5, 2주(8.3%)는 type 8, 13주(54.2%)는 분류가 불가능(non-typeable)하였다. 한편, 개에서 분리된 균주 중 1주(type 8)를 제외한 12주(92.3%)는 type 5로 분류되었으며, 13주 중 8주(61.5%)가 diffuse형의 집락을 보였다. 본 실험에 사용한 균주의 수가 충분하지 못한 문제가 있지만 동물에 따라 우세한 CP형이 다를 수 있으며 분류 불가능한 균주가 상대적으로 높은 비율을 차지하였는데 이는 새로운 CP형의 분류형이 필요함을 시사하는 것으로 판단된다. 또한 type 5와 type 8만을 포함한 유방염 백신은 한계가 있을 것으로 사료된다.

Studies on Pasteurella multocida isolated from pneumonic lungs of slaughter pigs (도축돈의 폐렴병소에서 분리한 Pasteurella multocida 에 대한 연구)

  • P multocida was isolated from 80(17.7%) of 450 pneumonic lungs of slaughter pigs. The majority of the biochemical and cultural characteristics of P multocida isolates were identical to those of the reference strains employed. Seventy seven strains(96.3%) among 80 isolates were capsular serotype A while the remaining 3(3.8%) were serotype D. All isolates were very susceptible to ampicillin, ceftiofur, cephalothin, ciprofloxacin and penicillin-G although some of them were resistant to sulfamethoxin and/or streptomycin. Sixty one(76.3%) of all 80 P multocida isolates were dermonecrotic toxin producers. Out of 77 isolates of serotype A and 3 isolates of serotype D, 59(76.6%) and 2(66.7%) were toxigenic, respectively. No difference was noted in dermonecrotic toxigenicity of the isolates in relation to capsular serotypes.

  • Streptococcus pneumoniae (pneumococcus) is the most frequent causative agent of acute bacterial pneumonia. Outstanding characteristic of pneumococcus is an ample polysaccharide capsule that is highly anti-genic agent and is the major factor for classification of pneumococcus into more than 94 serotypes. In this study, production of capsular polysaccharide from Streptococcus pneumoniae type 23 was optimized by supplementation of metal ions or by modulation of culture conditions. When brain heart infusion (BHI) broth was supplemented with 1 mM concentration of $CaCl_2$, $CuSO_4$ and $MgSO_4$, growth of pneumococcus as well as polysaccharide yield were stimulated. Also slight aeration gave rise to better polysaccharide yield.

  • The mating types and serotypes of 10 clinical isolates of Cryptococcus neoformans have been investigated. Seven isolates were serotype A and three were serotype D and thus they fell in C. neoformans var. neoformans. Mating types of six isolates were found $\alpha$ and two were $\alpha$ but another two isolates were untypable. Enzyme linked immuno-sorbent assay(ELISA) using rabbit hyper-immune serum to cryptococcal polysaccharides was well adapted to the analysis of capsular polysaccharides in sera of the patients with cryptococcal meningitis.

  • Haemophilus somnus, Mycoplasma bovis and Pasteurella multocida were responsible for respiratory diseases in bovine. Methods for identifying these bacteria had poor sensitivity and specificity. In this paper, PCR assays were applied for rapid identification of H. somnus, M. bovis, P. multocida B:2 and P. multocida capsular types. The specific PCR products were amplified from H. somnus, but not from other bacteria. Ten-fold diluted H. somnus were mixed with P. multocida and then the mixed cultures were inoculated on agar plates. After incubation, PCR was performed with harvest from agar plates and could detect as few as 3.4 CFU/ml of H. somnus. The primers MboF and MboR produced an amplification product unique to M. bovis and sensitivity of PCR was as low as 100 pg of DNA. Only serotype B:2 of P. multocida, the causal agent of haemorrhagic septicemia in bovine, was specifically amplified in PCR among the 16 reference serotypes. The multiplex capsular PCR typing for P. multocida was produced the P. multocida specific product as well as the capsular serogroup-specific product. The present PCR assays should be useful for the rapid identification of bacterial pathogens from bovine respiratory diseases.

  • Streptococcus(S.) suis is a pathogen, causing meningitis, septicemia and sudden death in weaning piglets as well as fattening pigs. Using multiplex PCR method based S. suis capsular genes, 61 S. suis isolates was classified as serotypes 2, 7, 9 and untypable. Genotyping of S. suis isolates was analysed by PFGE pattern with treated Sma I restricted enzyme. Of the 61 S. suis, 25 (40.9%) were serotype 2, 6 (9.8%) were serotype 7, 5 (8.2%) were serotype 9, and 25 (40.9%) were untypable, respectively. Twenty four PFGE patterns were detected in this study and also PFGE patterns were classified according to serotype; serotype 2 was classified as 6 genotypes, serotype 7 was 5 genotypes, serotype 9 was 3 genotypes, and untypable was 11 genotypes, respectively.

  • The present study was conducted to investigate the incidence of Streptococcus suis infection in the pneumonic lungs of slaughter pigs from December 1992 to April 1994 and to examine the biochemical and cultural properties, the antimicrobial susceptibility and serotype of the isolates. Fifty-six(7.95%) strains of S suis was isolated from 704 pneumonic lungs of slaughter pigs, of which 33 cultures were isolated alone, while remaining 18 were complicated wide Pasteurella multocida. Of the 56 S suis isolates, 18(32.1%) were serotype 2, 5(8.9%) were serotype 1/2, 2 were serotype 1, 1 each for serotype 3 & 5, and the remaining 29 were untypable. The twenty five strains(44.6%) of all isolates produced hyaluronidase while 15 strains(88.3%) of 18 serotype 2 isolates were hyaluronidase producers. All of isolates were highly susceptible to ampicillin, amoxicillin, ceftiofur, cephalothin, ciprofloxacin and penicillin G, although the majority of them were resistant to kanamycin, colistin, erythromycin, oxytetracycline.

Pneumococcal Surface Protein A of Streptococcus pneumoniae Isolates from Koreans (한국인에서 Streptococcus pneumoniae 분리주의 폐구균 표면 단백 A)

  • Purpose : Pneumococcal protein vaccine based on pneumococcal surface protein A (PspA) is in development with the potential to offer a broad range of protection against different strains. PspA elicits protection in mice against fatal sepsis as well as carriage and lung infection. This study was performed to investigate the frequency of PspA families among Streptococcus pneumoniae recovered from Korean children and adults. Methods : A total of 89 pneumococcal isolates was included in the study. They were capsule serotyped by the slide agglutination assay with commercial antisera. PspA families were determined with polymerase chain reaction using the pair of primers for family 1 and family 2. Results : Seventeen pneumococcal serotypes were found in a total of 89 isolates. PspA typing was able to ascertain 79 of the 89 isolates (88.8 percent). Among these, 20 (22.5 percent) isolates were family 1 PspA, 59 (66.3 percent) were family 2. Moreover, because 9 (10.1 percent) isolates were of positive reactions for both, families 1 and 2 primers, the potential coverage of PspA vaccine was 98.9 percent. PspA families were not associated with age group, source of isolates, or penicillin susceptibility. However, the relative distribution of family 1 isolates to family 2 isolates was significantly different over capsular serotypes. Conclusion : The finding that 98.9 percent of Korean isolates belonging to PspA families 1 and 2 support the hypothesis that a human PspA vaccine covering a few PspA families could be broadly effective. The monitoring of the PspA families derived from large population-based isolates will be necessary in the context of vaccine development.
