개에서 피부세정제 적용빈도가 정상피부에 미치는 영향 (Functional Assessment of Frequency of a Commercial Shampoo for Normal Canine Skin)

  • Canine skin is a highly dynamic organ that is constantly adapted to changes in its environment. It provides structural, sensory, immunologic, and physiologic functions and contributes an essential barrier function against potential environmental insults. We assessed the changes of canine skin hydration, pH, and protein contents by noninvasive method according to the frequency of application with canine commercial shampoo. Canine commercial shampoo was applied topically every other day, 5 days, and 8 days on different sites, respectively in 8 dogs. Saline-applied site was as a control. Skin hydration was increased and skin surface pH was decreased significantly in canine commercial shampoo-applied site every other day (p < 0.05). Also, skin protein concentration measured by D-squame$^{(R)}$ tape stripping method was increased significantly in canine commercial shampoo-applied site every other day (p < 0.05). There were alterations on every 5 days and 8 days, but not significantly. These results may be recognized that the frequent use of canine commercial shampoo cause abnormalities of the skin barrier function and alteration of stratum corneum integrity/cohesion. Consequently it was suggested that bathing the dog with canine commercial shampoo was appropriate every 5 or 8 days.

아세톤에 의해 손상된 개 피부장벽 모델에서의 경표피수분소실도 및 각질층보습도 평가와 전자현미경적 관찰 (Transepidermal Water Loss, Stratum Corneum Hydration and Transmission Electron Microscope Observation on Acetone Damaged Canine Skin Barrier Model)

  • 본 연구는 실험적으로 개의 피부장벽 손상 모델을 평가하기 위해 아세톤을 이용하여 개 피부장벽의 손상을 유도하고 손상의 정도를 경표피수분소실과 각질층보습도 수치를 측정하였다. 손상 후의 각질층세포간 지질구조는 전자현미경을 이용하여 평가하였다. 피부 및 임상병리학적 검사에서 건강한 2~4세령의 수컷 비글 개 6마리를 사용하였으며 아세톤에 의해 피부는 삭모 후 48시간후 에 손상되었다. 경표피수분소실도와 각질층보습도는 아세톤 손상전보다 손상후에 유의성 있게 저하되었다. 아세톤 손상표피의 전자현미경학적 평가에서는 손상된 각질세포간 비정상적이고 불완전한 지질층판이 관찰되었다. 본 연구를 통하여 아세톤을 이용한 개의 피부장벽손상은 개 아토피나 건성피부 연구를 위한 손상모델 가능성이 있는 것으로 판단된다.

Effect of Methyl Gallate on 1-Nitropyrene-Induced Keratinocyte Toxicity in a Human and Canine Skin Model

  • The skin, which is the largest organ of the human body, is in direct contact with pollutants in the surrounding atmosphere. Meanwhile, 1-nitropyrene (1-NP), the most abundant nitro-polycyclic aromatic hydrocarbon found in particulate matter, is known to have carcinogenic effects; however, studies on its toxicity in human and canine skin are still needed. In this study, we investigated 1-NP-induced apoptosis and inflammatory pathways in HaCaT cells. In addition, we also measured the cytoprotective effect of methyl gallate (MG), which is widely distributed in medicinal and edible plants and is well known for its anti-inflammatory and antioxidant properties. MG inhibited 1-NP-induced cell death and apoptosis pathways, including the cleavage of PARP and activation of caspase-3, -7, and -9. MG also suppressed 1-NP-induced COX-2 expression and phosphorylation of mitogen-activated protein kinases (MAPKs) and MAPK kinases (MAPKKs). Our findings suggest that 1-NP induces skin toxicity in human and canine through apoptosis and inflammatory responses, and moreover, that this can be prevented by treatment with MG.

국내 유기견의 개 옴 감염 상황 (Prevalence of canine scabies in the Korean stray dogs)

  • Recently, the number of stray dogs is proportionate to the increase of the number of the companion dogs. Sarcoptic scabiei var canis, causing scabies, is one of the most important canine zoonotic arthropods in Korea and around the world. Thus, we have tried to know the prevalence of canine scabies in the stray dogs in Korea. A total of 565 stray dogs were collected from the rescue centers all over the country from Jan. 2006 to Dec. 2006. They were grouped with euthanasia or natural death and examined for the gender and age estimated by dental formula. To identify the lesions, the whole body was grossly examined and tested pathologically. Thirty two (5.66%) of 565 dogs were diagnosed as canine scabies. Dogs from urban areas had fewer scabies (0.62%) than those of rural areas (12.5%). Prevalence of scabies in male and female dogs was no difference as 5.96% and 5.25%. Euthanasia group showed higher prevalence (6.48%) than natural death group (2.44%) in scabies. Old dogs over five years showed lower infestration (1.82%) in scabies. In histopathological findings, there were mites in the burrows formed in the subcorneal space. Acanthosis, hyperkeratosis with crust, and vascular dilatation were main findings. One hundred thirteen (20%) of 565 stray dogs were diagnosed to have skin disease. Among them, canine scabies is the most prominent ectoparasite as 5.66 %. With previous reports on human infection in Korea, canine scabies must be regarded as the important zoonotic canine skin disease. Accordingly, for the human and canine hygiene it is imperative that stray dogs with skin problems are segregated and tested for the parasites to treat properly as soon as arriving at rescue shelter.

개에서 피부손상에 의한 표피내 칼슘이온 분포상 (Visual Imaging of Calcium Ion Distribution in Acetone and Tape Stripping Damaged Canine Epidermis)

  • 본 연구의 목적은 개 피부장벽 손상모델을 실험적으로 구현하기 위해 개의 정상피부, 제모 된 피부 그리고 아세톤 및 테잎 스트리립핑에 의한 손상된 피부에서의 표피내 칼슘이온의 분포를 관찰하였다. 피부손상에 따른 표피의 칼슘이온 변화도는 화학적 표시자(Calcium Green-1)가 포함된 gel blotting을 적용하여 피부장벽손상 효과를 시간에 따라 형광현미경으로 관찰하였다. 아세톤 손상 후 3분 및 1시간에 표피와 모낭의 칼슘이온의 차이를 나타내는 형광도차는 보다 밝게 관찰된 후 48시간 후에 소실되었다. 이와는 대조적으로, 테잎스트립핑손상 후 3분 및 1시간의 표피 칼슘이온의 형광도차는 아세톤 손상에서보다 더 밝게 보였다가 48시간 후에 소실되었다. 본 실험상의 방법을 통해 피부 손상 방법에 따른 표피 칼슘이온의 가시적 이미지를 관찰할 수 있었고, 표피내 농도차이를 확인할 수 있었다. 따라서 본 화학표시자를 이용한 염색법은 개 피부장벽 복구기전에 대한 칼슘이온의 역할을 규명하는데 유용할 것으로 판단되며, 추후 표피칼슘이온 농도의 정량분석법에 관한 연구가 필요할 것으로 사료된다.

In Vitro: Antimicrobial Effect of Lactobacillus salivarius on Staphylococcus pseudintermedius

  • Lactobacillus spp. are the bacteria most commonly used as probiotics and it has been proven that they inhibit pathogenic bacterial growth and improve skin repair in humans. This study was conducted to investigate the growth inhibitory effect of Lactobacillus on Staphylococcus pseudintermedius, the most commonly isolated pathogen in canine pyoderma, and whether Lactobacillus could inhibit the adhesion capability of S. pseudintermedius to canine corneocytes. For this study, L. salivarius and S. pseudintermedius were isolated from healthy beagle fecal samples and the skin surface of dogs with skin infection, respectively. S. pseudintermedius was co-cultured with L. salivarius to assess the inhibitory effect. For the adhesion assay, corneocytes were collected from healthy beagle ventral abdominal skin. Both bacterial species attached to corneocytes and were assessed in number. As a result, L. salivarius significantly inhibited the growth of S. pseudintermedius in the culture medium. Moreover, L. salivarius reduced attachment of S. pseudintermedius in the adhesion assay. These results suggest that L. salivarius has an inhibitory effect on S. pseudintermedius and may be effectively used in the topical therapy of canine skin infections.

  • 표피유분측정기를 이용하여 정상적인 개 피부에 상용샴푸를 적용한 후 유분을 측정하였다. Sebocalm이 표피유분에 가장 많은 영향을 미쳤으며 Sebolytics와 Benzoyl peroxide는 중등도의 영향을 주었다. 물비누의 영향이 가장 낮았다. 따라서 Sebumeter는 표비유분측정에 간단하고 편리한 측정기로 샴푸이외의 피부적용 약물의 표피유분에 미치는 영향을 조사하는데 유용할 것으로 판단된다.

공포영화에 나타난 공포 캐릭터의 분장표현 연구 (A Study on Make-up Expression of Horror Character in Horror Movie)

  • The purpose of the study was to examine make-up expression elements of horror characters as an evil spirit, a vampire, a zombie, a monster and a psycho murderer through make-up, focused on horror movies of the United States after1960s. First, an evil spirit was expressed that the pupil grew dim without a focus and the skin was torn by going bad and rot completely. Also, it was expressed that its teeth was changed to pointed canine teeth as a wild beast and its pale and bloodless face showed blood vessels clearly. To express its bloodless and pale skin, air brush was swept into the skin over several times. Its canine tooth was made acryl powder into its shape and was harden to make its shape smooth. Also, it was colored to show people like the real thing. Second, a vampire was expressed by make-up elements as long nails, a sharp canine tooth, a pale skin with a blood vessel and dead eyes. Its rough skin and long nails were manufactured with latex foam, which was colored like the skin. Third, a monster image as a werewolf was expressed by make-up elements as fur, sharp teeth, changed nails and toenails, a rough skin and a face changed to a wolf. Rubber mask and rubber body suit were manufactured as a make-up of special character and so all its body was changed. Fourth, a psycho murderer and body-deformed man were expressed by make-up elements as a distorted mask, swelled-up and deformed skull and face form, terribly deformed body, discolored and hang down skin and teeth with an indeterminate form. Its body and face were manufactured by the foam latex technology according to make-up design as a make-up of special character.

개에서 개선충(Canine Scabies) 감염에 대한 월 2회 Selamectin 적용 일례 (Efficacy of Application of Selamectin for Canine Scabies Infestation in a Dog)

  • 3연령 잡종편이 소양감, 탈모의 증상으로 내원하였다. 신체검사에서 전신적으로 발적, 구진, 탈모, 비듬 그리고 가피의 병변이 관찰되었다. 전반적인 피부검사결과 개선충, Staphylococcus spp과 Malassezia spp의 혼합 감염으로 진단되었다. Selamectin 6 mg/kg을 2주간격으로 2회 경부피부에 적용하였다. 그리고 한 달간 구균과 Malassezia 치료를 위해 amoxicillin trihydrate/clavulanate pattassium과 ketoconazole 10 mg/kg을 경구투여하고 5% chlorhexidine 샴푸를 주2회 실시하였다. 첫 치료 후 7,14,21 그리고 28일에 피부를 재검사 하였을 때 개선충은 관찰되지 않았으며 피부 증상은 현저하게 개선되었다. 피모는 거의 정상으로 회복되었으며 Staphylococcus와 Malassezia spp는 거의 관찰되지 않았다. 그러므로 이 치료절차에 따른 selamectin 적용은 개선충이 감염된 개에서 매우 효과적이라고 사료된다.

Evaluation of the Agreement between Immunodot Assays and Intradermal Skin Testing or Favrot Diagnostic Criteria in Canine Atopic Dermatitis

  • This study was undertaken to identify differences between atopic and non-atopic dogs in three rapid screening immunodot assays as well as the ability of the assays to predict the results of intradermal skin testing (IDST) or Favrot diagnostic criteria (FDC). Twenty-nine dogs diagnosed with canine atopic dermatitis (CAD) were selected as the atopic group. Twenty-five dogs without CAD were included as the non-atopic group. Three types of immunodot assays were conducted on all serum samples from both groups: Allercept E-screen 2nd generation (ES2G), Canine Allergic Tendency Reference Test (ALERT), and Asan Easy Test Canine IgE (AETC). IDST, which included 39 allergens, and immunodot assays were performed concurrently in 13 dogs from the atopic group and compared. While there were no significant differences in positivity between the two groups in the evaluation of ALERT (P = 0.435) and AETC (P = 0.313), positivity in ES2G testing was significantly higher in the non-atopic group than the atopic group (P = 0.038). The ES2G, ALERT, and AETC results showed fair (${\kappa}=0.235$), slight (${\kappa}=0.133$), and slight (${\kappa}=0.014$) accordance with IDST, respectively. The outcomes of ES2G, ALERT, and AETC indicated poor (${\kappa}=-0.211$), slight (${\kappa}=0.106$), and slight (${\kappa}=0.087$) agreement with FDC. In conclusion, rapid screening immunodot assays were not useful for the diagnosis of CAD. These assays may provide a supplementary method for predicting the results of IDST in atopic dogs.
