The effect of phytohormones, light and phosphate on in vitro production of ubiquinone 10 from the suspension cultures of Nicotiana tabacum cv. Xanthi callus was investigated. The inoculum size and cultured time in the suspension culture had to be at least over 2 % of medium volume at 15 days for the excellent growth of Xanthi callus. The growth of Xanthi callus in the suspension culture was improved by addition of NAA and 2,4-D, especially NAA 1.0mg/1 alone, at the light condition. The optimal concentration of phytohormone was 0.1 mg/l 2.4-D and 1.0 mg/l NAA for productivity of ubiquinone 10 in the suspenseion of Xanthi callus. Addition of 3mM KH$_2$PO$_4$ to the medium was more effective in promoting ubiquinone-l0 formation than other concentration in the light condition. Content and production of ubiquinone-l0 in the suspension cultures of Xanthi callus were the highest at the MS media containing 0.5mg/L kinetin, 0.5mg/L 2,4-D, 1.0mg/1 NAA, 3mM phosphate and 2 % inoculum in the light.

We confirmed conditions for callus formation and plant regeneration of five varieties of orchardgrass (Dactylis glomerma L.). Among five varieties of orchardgrass, "Hapsung 19" expressed the highest rate for both of callus formation and plant regeneration. Otherwise, among SH (Schenk and Hildebrandt), MS (Murashige and Skoog) and N6 medium (Chu), MS and N6 medium were highest degree of efficiencies in callus formation and plant regeneration, respectively. In this study, we determined volume of hormones appended in media; $3\;mg/\;{\ell}$ ofdicamba for callus formation and $1\;mg/\;{\ell}$ of NAA (I-naphtalene acetic acid) and $5\;mg/\;{\ell}$ kinetin (6-furfurylaminopurine) for plant regeneration were appended in their media. We obtained orchardgrass plants from callus about 50~80 days after transferred to regeneration media.ation media.

For callus formation, only $5\;mg/\;{\ell}$ of 2,4-D (2,4-dichlorophenoxy acetic acid) was appended in their media. For plant regeneration, we used MS medium containing $1.0\;mg/\;{\ell}$ of BA and $0.1\;mg/\;{\ell}$ of NAA.
We investigated the effect of safflower-seed on fracture healing of fracture model in rat. Fracture healing was evaluated by examining the degree of wound healing macroscopically, radiography, bone histomorphometry and biochemical examination. After 1, 3, 5, 7 days, the would healing was accelerate in safflower-seed diet group. Radiography does not reveal the difference in fracture healing between two group. After 2 weeks, safflower-seed had a significant, stimulatory effect on external callus formation (p<0.05). But after 4, 6, 8 weeks, no difference was observed between normal and safflower-seed dietgroup in callus size. Urinary hydroxyproline, osteocalcin and total alkaline phosphatase decreased significantly (p<0.05) in safflower-seed treated group at 2 week after tracture.

Among SH (Schenk and Hildebrandt), MS (Murashige and Skoog) and N6 medium (Chu), SH medium was highest degree of efficiencies respectively in callus formation and plant regeneration. In this study, we determined volume of hormones and other compounds appended in media. For callus formation, only $3mg/{\ell}$ of 2,4-D (2,4-dichlorophenoxy acetic acid) was appended in their media. For plant regeneration, we used BOi2Y medium (Bingham et al.). We obtained birdsfoot trefoil plants from callus by regeneration, about sixty days later transfer calli to regeneration media.
In order to know the growth of suspended cells by explant sources, the change of nitrogen contents of cultured cells following the growth periods, capability of micro-callus formation according to cell plating methods, growth of suspended cells on various media, and efficiency of micro-callus formation by using growth regulators and different N strengths were investigated. 1. When suspension culture was tried by using the callus induced from internode and petiole, cell fresh weight and packed cell volume increased with similar way and the growth reached at stationary phase after 12 culture days. 2. N-contents of cultured cells increased upto 3 days and decreased around 6days. But the values increased again upto 9 days, after that they showed gradual decreases. 3. Of cell plating methods, embedding method was the best for micro-callus formation. 4. Growth of suspened cells showed the rest performanoes, when they were cultured on LM medium with 1/2N strengths and BAP 0.01.2.4-D 0.1, and NAA $1.0mg/{\ell}$, after 15 cultured days(upto 76.9 folds). LM medium was better than MS or GD. The combination of auxin and cytokinin was better for cell growing than auxin-treatment only. 5. Micro-callus from single cell and small cell aggregates was formed only on MS and LM media with 2,4-D $1.0mg/{\ell}$.
In maize, immature embryos (IEs) are highly regenerative explants most suitable for producing high frequencies of plantlet regeneration in vitro. Apart from media, explants, and hormones, genotypic variation also influences in vitro characters to a great extent. In the present study, IEs were used to study the distinctive effect of variation of size/stage and hormones in different genotypes on five in vitro characters viz., frequency of callus induction, growth rate of total callus, frequency of E. callus induction, and volume and number of regenerated plantlets. LS medium with different concentrations of 2,4-D (0.5, 1.5, 2.5, 4.0 and 5.0 mg/L) were used to study the former four in vitro characters, and medium with 6-benzylaminopurine and kinetin (0.5 mg/L, each) was used for plantlet regeneration. IEs of 1.0, 1.5, 2.0, 2.5 and 3.0 mm in size were isolated from four inbred lines viz., NM74C, NM81A, NM5883 and NM5884. Two-way ANOVA revealed that explant size and genotypes, as well as hormonal concentrations showed significant effects on in vitro characters. Two millimeter IEs were found to be suitable for in vitro cultures. LS medium with 1.5 mg/L 2,4-D and LS with BAP and Kn (0.5 mg/L, each) were found to be the best hormonal concentrations for callus induction, maintenance, and regeneration, respectively. Among the four genotypes, NM81A and NM5883 yielded more non-embryogenic and Type I E. calli. In contrast, NM74C and NM5884 yielded more highly regenerative Type II calli. Inbred line NM5884 was found to be the best among these four genotypes.

The regenerated-bulblets placed in liquid free media resulted in good formation of roots and bulblets. On 1/4 MS free medium, roots and bulblets were predominantly induced. The 1/4 MS liquid medium supplemented with plant growth regulators was the best suitable condition for elongation of leaves and roots. Somatic embryos were frequently developed from embryogenic callus in liquid media with 2,4-D 1mg/ l . On free liquid media, the viability of callus reduced. As the salt strength of MS media reduces, the viability of callus reduced significantly. However, Leaves were induced from several callus clumps. When leaves, roots and bulb-scale segments were placed on MS media containing NAA 1mg/ l or 2,4-D 1mg/ l and various sucrose concentration, the best result about the differentiation, growth of leaf and the differentiation of leaf was obtained on MS media added 1.5% sucrose and 2,4-D 1mg/ l, 3% sucrose and NAA 1mg/ l, and 1.5% sucrose and NAA 1mg/ l, respectively. Also the better result differentiation, growth of root and differentiation of bulb was obtained on MS media with 6% sucrose and NAA 1mg/ l. Spermidine promoted the growth of leaf and the differentiation of bulb. However, spermine promoted the differentiation of leaf, the differentiation and the growth of root in MS solid media. On the MS liquid media, both spermine and spermidine stimulated organogenesis from bulb-scale segments. Regenerated plantlets were acclimatizated and grown in greenhouse in vermiculite + perlite (1 : 1 by volume) well. The optimal soil condition of rooting for plantlets regenerated was in peat moss.

Introduction : In recent years, vascularized, i.e., living bone grafts, have been widely applied in the field of oral and maxillofacial surgery, as a method of treatment of congenital or acquired non-unions, and a large defects in mandible. The vascularized fibular graft has been especially used for this purpose because of its shape and mechanical strength. The postoperative hypertrophy of grafted fibula is of particular interest to us. Material and methods : This study was undertaken to determine the volume change(indirect methods) and radiographic appearance of a free vascularized fibular graft as it responds to the mechanical and physiologic features of its new environment. In order to elucidate the long term effect on fibular mass after mandibular reconstruction, change in various method of volume change was utilized as indirect measure of change in long-term. Results : The younger the patient, the more prominent and rapid the hypertrophy of the graft. the hypertrophy of the graft never exceeded the diameter of the recipient bone, except for callus enlargement after stress fracture of the grafted bone. Conclusion : Etiologic explanations for this phenomenon have not been clarified in the previeous literature. some of the factors implicated include a periosteal reaction or new bone formation, as seen at the onset of bone union after a fracture in a child, a reaction to the mechanical loading on the graft and a reaction to the circulatory changes resulting from the grafting procedure.
