Histopathologic Lesion of Mannheimia haemolytica pneumonia in calves (송아지에서 발생한 Mannheimia haemolytica 폐렴의 병리조직학적 소견)

  • Four 5 month old calves were died after showing respiratory distress after long-distance transportation at winter season. They were diagnosed as fibrinous lobar pneumonia caused by Mannheimia (M.) haemolytica. Grossly, lungs were attached onto the pleura by fibrin, with a rich yellowish fluid in thorax. The cut surface of the lung was showed marbled pattern of the reddish or greyish consolidation and widened interlobular septa by fibrin deposition. Histopathologically, parenchymal necrosis was delineated by a band of the degenerated inflammatory cells, and distended interlobular septa with serofibrinous exudates and vascular thrombosis with alveolar capillaries degeneration and abundant serofibrinous exudates in alveoli. M. hemolytica were isolated from all calves, and bovine viral diarrhea virus and parainfluenza type 3 virus in one calf were detected by RT-PCR. Thus, it was concluded that this case was diagnosed as pneumonic mannheimiosis suggested by complex infection with viruses after long-distance transportation and coldness.

Hemorrhagic Enteritis in Two One-month-old Dairy Calves Infected with Beta2-toxigenic Clostridium perfringens and Coccidium

  • Two one-month-old dairy calves which have Eimeria oocysts in their bloody diarrhea died acutely a few days after showing the first clinical signs. At necropsy, hemorrhagic and congestive gastrointestinal organs were observed in both calves, and abomasal ulcerations existed. As a prevalent agent in all of the collected intra-intestinal specimens, Clostridium perfringens was isolated and the strain was identified as type A possessing alpha and beta2-toxins. In these clinical cases, intercurrent infection by C. perfringens type A and Eimeria through contaminated environment may be responsible for acute hemorrhagic enteritis.

Substituting oat hay or maize silage for portion of alfalfa hay affects growth performance, ruminal fermentation, and nutrient digestibility of weaned calves

  • Objective: The impact of forage feeding strategy on growth performance, ruminal fermentation and nutrient digestibility in post-weaning calves was investigated. Methods: Forty-five female Holstein calves (body weight [BW] = $79.79{\pm}0.38kg$) were enrolled in the 35-d study at one week after weaning and randomly assigned to one of three dietary treatments. All diets were fed as total mixed ration containing 60% (dry matter [DM] basis) of basal starter feed and 40% (DM basis) of forage, but varied in composition of forage source including i) alfalfa (40% DM, AH); ii) alfalfa hay (26.7% DM)+oat hay (13.3% DM; OH); iii) alfalfa hay (26.7% DM)+corn silage (13.3% DM; WS). Results: Dry matter intake was not different among treatment groups (p>0.05). However, BW (p<0.05) and average daily gain (p<0.05) of calves fed AH and OH were greater than WS-fed calves, whereas heart girth was greater in OH-fed calves than those fed AH and WS (p<0.05). Ruminal fermentation parameters including proportion of butyric acid, acetated-to-propionate ratio, concentration of total volatile fatty acid, protozoal protein, bacterial protein, and microbial protein in rumen were the highest in OH (p<0.05) and the lowest in WS. Compared with the AH and WS, feeding oat hay to postweaning calves increased crude protein digestibility (p<0.05), and decreased duration of diarrhea (p<0.05) and fecal index (p<0.05). Conclusion: Our results suggested that partially replacing alfalfa hay with oat hay improved ruminal fermentation, nitrogen utilization, and reduced incidence of diarrhea in post-weaning dairy calves.

Evaluation for detection of Cryptosporidium oocysts in diarrheal feces of calves (야외 송아지 설사변에서 작은와포자충 검출에 대한 평가)

  • For the detection of Cwptospori,mum oocysts, fecal samples were collected from 201 calves which showed diarrhea. Among the 201 samples, 29 samples (14.4%) were positive for Cwptosporinium spry. by the DMSO-modified acid-fast stain (MAFS) , 23 samples (11.4%) were positive by commercial kit (Meridian Diagnostics, Cincinnati, Ohiol and 23 by the indirect immunofluorescence antibody (IFA )assay employing the monoclonal antibody (mAb C6). When tested by both IFA and MAFS, 20 fecal samples were positive for Cwptosporinium oocysts whereas 169 fecal samples were negative. If the MAFS is considered a standard method for oocyst detection, the IFA showed 69% of sensitivity and 98% of specificity. When tested by both IFA and commercial kit, 22 fecal samples were positive for Cwptospori,mum oocysts while 177 samples were negative. One sample tested by IFA was found to be false negative, when compared with the results by commercial kit. The sensitivity of IFA was calculated as high as 96%; the specificity as 99% and the predictive value was also 99%. In the present study, IFA employing the nAb C6 revealed that 23 samples (11.4%) were positive among the 201 calves showing diarrhea. Of 23 IFA positive samples, 4 samples (5%) showed cryptosporidial oocysts more than 105 OPG Therefore. it is concluded that the calves showing cryptosporidial oocysts more than 105 OPG in the feces were highly associated with clinical cryptosporidiosis.

Simultaneous Detection of Major Pathogens Causing Bovine Diarrhea by Multiplex Real-time PCR Panel (Multiplex real-time PCR을 이용한 송아지 설사병 원인 주요 병원체의 동시검출)

  • Bovine diarrhea is a major economical burden to the bovine industry in Korea. Since multiple infectious agents can be involved in bovine diarrhea, differential diagnosis is essential for effective treatment. Therefore, a panel of two multiplex real-time PCR assays which can simultaneously detect six major bovine enteric pathogens [i.e., bovine viral diarrhea virus (BVDV), bovine coronavirus (BCoV), group A bovine rotavirus (BRV), Salmonella spp., Escherichia coli (E. coli) $K99^+$, and Cryptosporidium parvum] was developed and applied to test 97 fecal samples collected from cattle farms in Korea. In addition, microscopic examination was also preformed on the samples to detect Coccidium oocyst. The estimated sensitivity of the multiplex PCR was 0.1 $TCID_{50}$ for BVDV, BCoV and group A BRV, 5 and 0.5 CFU for E. coli $K99^+$ and Salmonella, respectively, and 50 oocysts for Cryptosporidium. The amplification efficiency of the multiplex PCR ranged between 0.97 and 0.99 for each pathogen. Among 97 samples, 36 samples were positive for at least one of the 6 major pathogens and 6 samples were simultaneously positive for 2 pathogens by the multiplex PCR assay. Coccidium oocysts were also detected in 48 samples, which were all collected from over 1 month old calves. In conclusion, the multiplex real-time PCR panel can be a useful tool for fast and accurate diagnosis of calf diarrhea associated with BVDV, BCoV, group A BRV, E. coli $K99^+$, Salmonella, and/or Cryptosporidium and Coccidium may be an important target which needs to be included in the multiplex PCR panel in the future.

Effects of Supplementing Herbs on Growth Performances, Blood Composition and Diarrhea in Hanwoo Calves (한방제재 첨가급여가 한우송아지의 성장, 혈액성상 및 설사에 미치는 영향)

  • The current study was conducted to determine the effects of herbal supplement on growth performances and diarrhea in Hanwoo calves. Total 24 Hanwoo calves (3 treatments: 8 calves per treatment) were randomly assigned to either Control (no treatment), Treatment 1 (0.3% herb supplement), or Treatment 2 (0.5% herb supplement) diets for 150 days. Total body weight gain and total feed intake per head in Treatment 1 was 142.8 kg and 545.9 kg, respectively, and it was higher (p<0.05) compared to other groups. The blood total cholesterol range was 86.43~97.00 mg/dl, triglycerides 13.26~13.86 mg/dl, GOT 76.97~79.60 mg/dl, GPT 19.54~20.97 mg/dl, WBC 8.75~9.95 k/${\mu}{\ell}$, RBC 10.14~11.91 M/${\mu}{\ell}$, and hemoglobin 10.74~11.20 g/$d{\ell}$, respectively, with no significant (p>0.05) differences among treatments. The blood immunoglobulin G levels were 5.74~6.05 mg/ml which tended to decrease as experimental period extended. Total number of pathogens in feces showed peaks at 1~2 months after the initiation of experiment, and tended to decrease thereafter. Total number of Eimeria spp., E. coli and BVDV in feces showed no significant differences but control group showed higher counts than both treatment groups. During overall period, the incidence of pathogenic diarrhea in calves of 2 treatment groups was much lower than control group calves (C: 24, vs T1: 9, T2: 13 heads), however, it was not significant (p>0.05). In conclusion, supplementation of herbs in Hanwoo calf diets might be beneficial to improve growth performances and prevent diarrhea.

Effect of acidified milk feeding on the intake, average daily gain and fecal microbiological diversity of Holstein dairy calves

  • Objective: To evaluate the effect of feeding acidified milk on the growth and fecal microbial diversity of dairy calves. Methods: Twenty healthy 3-day-old female Holstein calves with similar body weights were selected and randomly divided into two groups. One group was fed pasteurized milk (PM, Control), while the other was fed acidified milk (AM) ad libitum until weaned (day 60). The experiment lasted until day 180. Results: There was no difference in the nutritional components between PM and AM. The numbers of Escherichia coli and total bacteria in AM were lower than in PM. At 31 to 40 and 41 to 50 days of age, the milk intake of calves fed AM was higher than that of calves fed PM (p<0.05), and the solid feed intake of calves fed AM was higher than that of calves fed PM at 61 to 90 days (p<0.05). The average daily gain of calves fed AM was also higher than that of calves fed PM at 31 to 60, 61 to 180, and 7 to 180 days (p<0.05). The calves fed AM tended to have a lower diarrhea rate than those fed PM (p = 0.059). Bacteroides had the highest abundance in the feces of calves fed AM on day 50, while Ruminococcaceae_UCG_005 had the highest abundance in the feces of calves fed AM on day 90 and calves fed PM on days 50 and 90. At the taxonomic level, the linear discriminant analysis scores of 27 microorganisms in the feces of calves fed AM and PM on days 50 and 90 were higher than 4.0. Conclusion: Feeding AM increased calf average daily gain and affected fecal bacterial diversity.

Studies on immunotherapy to calves with failure of colostrum passive transfer using bovine immune sera (소 면역혈청을 이용한 초유 이행 부전 송아지의 치료 효과)

  • The efficacy of bovine immune sera to correct the calves with failure of passive transfer(FPT) was evaluated. Immune sera were produced from 14 one-year-old Holstein cattle which were inoculated commercial combined viral vaccine, administered by the challenge of some main enteric or respiratory viruses, aseptically filtered and stored at $4^{\circ}C$ before used. After the treatment of bovine immune sera, Mean transfer factor($mg/d{\ell}$, of IgG administered/kg of body weight) was $5.46{\pm}2.74,\;11.17{\pm}1.27,\;1.40{\pm}0.21$ in K-IP, H-IP and K-IV group, respectively. The corrective effect of bovine immune sera to FPT calf without any clinical signs showed that intravenous route was more effective than intraperitoneal administration(P<0.01). FPT calves with severe signs were not effective response to the immunotherapy used and consequently died within 10 days after the treatment. Ten percentage of controls appeared the clinical signs including diarrhea. On the contrary, there were not any clinical signs in K-IP and H-IV group. There was significant increase of the neutralizing titer against bovine viral diarrhea virus and bovine coronavirus as well as of cell population including CD2, CD4, and monocyte in K-IP and H-IV group after the immunotherapy(P<0.05). Also, K-IP and H-IV group showed the successful correction to FPT within one week after the immunotherapy, but controls had kept the FPT two-four weeks even after the same treatment. Consequently, the results were suggesting that the bovine immune sera could be used the corrective tool to young calves with FPT.

Effects of Agricultural By-Product Feeds on Growth and Carcass Characteristics of Korean Native Steer (농산 혼합 부산물 첨가급여가 송아지 질병 및 거세 한우 성장 및 도체특성에 미치는 영향)

  • This study was carried out investigate the effects of dietary supplementation of agricultural by-product feed(persimmon-pell+barley bran+fermented rice bran+activated carbon) on calves disease, growth and carcass characteristics of Korean native steer. A total of twenty Korean native steer were allocated into four feeding group and assigned to four dietary treatments: Control(normal concentrate as a basal diet), T1(1% addition of by-product feeds), T2(3% addition of by-product feeds) and T3(5% addition of by-product feeds). The calf diarrhea was the highest at Control, while T3 was the lowest. The breath disease was the lowest at T2, occurrence with diarrhea and breath disease was the highest at Control. The daily gain was orderly ranked as T2(0.76kg)>T1(0.7kg), T3(0.70kg)>Control(0.67kg). The back fat thickness was orderly ranked as T2>T3>Control>T1, eye muscle area was orderly ranked as T3>T1>Control>T2. Grade of meat quantity was the highest at T1(2.3), but fat color, firmness and maturity tends to have a similar result. The marbling score and quality grade of T2 were the highest at 5.0 and 3.5, respectively. Based on the study, agricultural by-product feeds are able to many used for various purposes at disease reduce, increment of meat quantity and improvement of quality grade.

Immuno-specificity of Egg Yolk Antibodies against Bovine Rotavirus and Bovine Coronavirus causing Calf Diarrhea (송아지 설사병 주요원인체인 소로타바이러스와 소코로나바이러스에 대한 난황항체 생산 및 면역특이성 분석)

  • This study was performed to produce specific egg yolk antibodies against bovine rotavirus (BRV) and bovine coronavirus (BCV) that are major pathogens causing diarrhea in calves. Chickens were immunized with BRV and BCV intramuscularly in the breast muscle by injection 5 times at two weeks interval. At 6 weeks after the first immunization of BRV or BCV, cross reactivity of each serum derived from BRV- or BCV-immunized hens was tested. Each serum antibody against BRV or BCV was reacted with only specific BRV or BCV antigens. Serum and egg yolk-antibody titers of hens against BRV or BCVwere highest at 8~12 weeks after first immunization. Specific serum and egg yolk-antibody titers against BRV were about 104,000 and 107,000, respectively, and those against BCV were about 145,000 and 155,000, respectively. Hemagglutination inhibition titers in the immunized egg yolk antibodies against BRV and BCV were 5,120 and 1,280, respectively, and were ${\geq}8$ times higher than that in non-immunized control. These results suggested that the immunized egg yolk antibodies could effectively neutralize BRV and BCV.
