On the Accumulation of Minerals with the Plant Species in a Reclaimed Land (干拓地 植物의 無機營養素 蓄積에 對하여)

  • For reaserching the factors of plants micro-distribution, accumulation of 5 minerals-total-nitrogen, phosphorus, potassium, sodium and calcium-for 19 plant species was investigated in reclaimed land, in western coast of Korea, In the five minerals, sodium contents were quite different among the species. Plant species were divided into 4 groups based on the sodium accumulation and accumulation site in plant tissue: Na accumultion type above-ground part>below-ground part : Triglochin maritimum, Chenopodium virgatum, Atriplex subcordata, Salicornia herbacea, Suaeda japonica, suaeda asparagoides, Limonium tetragonum, Aster tripolium, Artemisia scoparia, Sonchus brachyotus above-ground partbelow-ground part : Zoysia sinica, amagrostis epigeiosa bove-ground part

A rare case report of neonatal calcinosis cutis induced by distant and delayed extravasation of intravenous calcium gluconate

  • A 3,480 g male neonate showed tachypnea symptom with a serum ionized calcium level of 0.66 mmol/L by routine clinical analysis. He was injected calcium gluconate intravenously through femoral vein catheter to treat the hypocalcemia. On second day after the injection, he started to show erythema in the flank area. The lesion became firm and changed into whitish crust consist of small crystals. Abdominal X-ray and ultrasonography showed the accumulation of calcium deposit in the subcutaneous layer of the lesion. Surgical debridement was performed to remove the crust with calcium deposit and acellular fish skin graft rich in omega-3 (Kerecis) was applied to the defect site for secondary intention of the defect wound. After 2 months, the skin and soft tissue defect were fully covered with healthy normal skin without depression or contracture. This report is a first case of iatrogenic calcinosis cutis without extravasation symptom.

An Electron Microscopic Study on the Effect of Calcium-free Reperfusion in Isolated Perfused Guinea Pig Heart after Global Ischemia (허혈후 칼슘 결핍 용액의 재관류가 적출 관류 기니픽 심근 세포에 미치는 영향에 관한 전자현미경적 관찰)

  • The effect of calcium-free reperfusion for 5, 10, and 15 minutes, respectively, followed by continuous reperfusion with normal Tyrode solution containing 1.0mM calcium chloride, after global ischemia in the isolated perfused guinea pig heart by Langendorff techniques was examined with transmission electron microscope. Compared to the nomal Tyrode solution-perfused control hearts, the 5 minute calcium-free-reperfused hearts showed loss or thickening of Z lines, focal sarcolemmal disruption, mitochondrial swelling, clumping of chroma-tin, intracellular fluid accumulation, and some separation of cell junctions, especially the fasciae adherentes. These changes became more severe in the hearts of 10 minute calcium-free reperfusion. Subsarcolemmal larger bleb and near complete separation of cell junctions were noticed. In the 15 minute calcium-free-reperfused hearts, irreversible ultrastructural changes including contraction bands, biazrre mitochondria, and sarcolemmal destruction were widely distributed. The severity of myocardial changes were in accordance with the duration of calcium-free reperfusion. These changes indicate that calcium-free reperfusion regardless of its duration could not salvage the post-ischemic myocardium probably due to development of calcium paradox.

Experimental Intervention to Reverse Inhibition of Nitric Oxide Production by Cyclosporin A in Rat Aortic Smooth Muscle Cells (혈관평활근세포에서 Cyclosporin A에 의한 Nitric Oxide 생성억제를 길항하는 실험적 중재법)

  • The inhibitory effect of cyclosporin A (CsA) on nitric oxide production is not related to the immunosuppressive action of the drug, but to the renal toxicity and arterial hyper-tension. In this study the experimental interventions to reverse the inhibition of nitric oxide production by cyclosporin A in rat aortic smooth muscle cells were examined. CsA inhibited the accumulation of nitrite, the stable end product of nitric oxide, in culture media in a concentration $(0.1{\sim}100{\mu}g/ml)-dependent$ manner. The inhibitory effect of CsA on nitrite accumulation were not antagonized by arginine (10 mM), a substrate of nitric oxide synthase, nor by calcium ionophore A23187 $(7{\mu}M)$. Forskolin, an activator of adenylate cyclase, which enhanced iNOS induction at transcriptional level, completely reversed the inhibitory action of CsA on nitrite accumulation. However, PMA (2 nM) and PDB (50 nM), PKC activators, increased the inhibitory action of CsA on nitrite accumulalion. From these results, it is suggested that cyclic AMP-elevating agents may be candidates of therapeutic agents in prevention and treatment of renal toxicity and arterial hypertension induced by CsA. Among conventional antihypertensive drugs, calcium channel blockers and ${\alpha}-blockers$ are preferred to ${\beta}-blockers$.

Inhibitory Effect of Fermented Spanish Extract on Inorganic phosphate-induced Vascular Calcification in ex vivo Aortic Rings (발효 시금치 추출물의 무기인산염에 의해 유도된 혈관 석회화 저해 효과)

  • Spinach (Spinacia oleracea L.), a green leafy vegetable, is well known as a functional food due to its biological activities. Vascular calcification is associated with several disease conditions including atherosclerosis, diabetes, and chronic kidney disease (CKD), and is known to raise the risk of cardiovascular diseases related morbidity and mortality. However, there are no previous studies that have investigated the effects of fermented spinach exract (FSE) against aortic and its underlying mechanisms. Therefore, this study investigated the effects and action of possible mechanisms of FSE on inorganic phosphate (PI)-induced vascular calcification in ex vivo mouse aortic rings. PI increased vascular calcification through calcium deposition in ex vivo aortic rings. FSE inhibited calcium accumulation and osteogenic key marker, runt-related transcription factor 2 (Runx2), and bone Morphogenetic Protein 2 (BMP-2) protein expression in ex vivo aortic rings. And, FSE inhibited PI-induced extracellular signal-regulated kinase (ERK) and p38 phosphorylation in ex vivo aortic rings. These results show that FSE can prevent vascular calcification which may be a crucial way for the prevention and treatment of vascular disease association with vascular calcification.

Changes in Cell Ca2+ Distribution in Loquat Leaves and Its Effects on Cold Tolerance

  • Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.

Strontium Metabolism in Higher Plants: Effect of Strontium of the Polyamine Biosynthesis during Germination of Wheat(Triticum aestivum L.) (식물체 내에서 Strontium의 대사 : 밀(Triticum aestivum L.)의 발아과정중 Polyamine 생합성에 미치는 Strontium의 영향)

  • Wheat (Triticum aestivum L.) seeds were used to study a possible relationship between strontium and polyamines (PAs) in the coleoptile, root, and endosperm during germination. When $Sr^{2+}$ (0.001 mM + 10 mM) was applied to the incubation medium with $10\;{\mu}M$ $GA_3$, great increases in putrescine (Put) were observed in root and spermidine (Spd) in the coleoptile, depending on the concentration. In germinating seeds, putrescine accumulation was induced even at a low concentration (0.01 mM Sr), whereas spermidine accumulation was stimulated considerably at a high concentration (10 mM Sr). The putrescine levels, on a gram fresh weight (g-fr-wt) basis, in the roots which were growth-inhibited by 1 and 10 mM $Sr^{2+}$ were 22.4 and 15.3 fold higher respectively than at the same concentrations of $Ca^{2+}$. The accumulation of total polyamine (TA), in particular Put and Spd, induced by Sr seemed to be an important physiological response not only on a g fr wt basis but also on an RNA basis. In contrast, the levels of agmatine (Agm) and cadaverine (Cad) were notably enhanced by 10 mM $Ca^{2+}$ in the coleoptile and root. Cadaverine was detected only in $Ca^{2+}-treated$ seedlings. However, $Ca^{2+}-treatment$ in the range of 0.001 mM to 1.0 mM resulted in reduction of TA content. The distinction of accumulated polyamines and the change in diamine (DA) / TA and tri- and tertiary (tPA) / TA ratios were likely to be a physiological difference between $Sr^{2+}$ and $Ca^{2+}$ during germination in wheat.

The effect of dehydroepiandrosterone administration on intestinal calcium absorption in ovariectomized female rats

  • [Purpose] Dehydroepiandrosterone (DHEA) administration reportedly recovers osteoporosis, a bone disorder associated with bone deficiency in postmenopausal women. However, the physiological mechanism of DHEA in osteoporosis remains elusive, especially in terms of intestinal calcium absorption. Therefore, we investigated the effect of DHEA administration on calcium absorption in ovariectomized (OVX) female rats using an estrogen receptor antagonist. [Methods] Female Sprague-Dawley rats (n=23, 6 weeks old) were randomized into three groups: OVX control group (OC, n=7), OVX with DHEA treatment group (OD, n=8), and OVX with DHEA inhibitor group (ODI, n=8) for 8 weeks. [Results] Intestinal calcium accumulation, as well as the rate of absorption, demonstrated no significant differences during the experimental period among investigated groups. The bone mineral density (BMD) of the tibia at the proximal metaphysis was higher in the OD group than that in the OC group (p<0.05); however, BMD of the ODI group showed no significant difference from investigated groups. Furthermore, the BMD of the tibia at the diaphysis did not significantly differ among these groups. [Conclusion] We revealed that DHEA administration does not involve intestinal Ca absorption, although this treatment improves BMD levels in OVX rats. These observations indicate that the effect of DHEA on the bone in postmenopausal women is solely due to its influence on bone metabolism and not intestinal calcium absorption.

Effects of Nicotine on mineralization in human fetal osteoblasts (니코틴이 사람태아골모세포의 광물화 과정에 미치는 영향)

  • Nicotine is one of the major components of cigarette smoking which causes various systemic and local diseases to human body. The purpose of the present study was to investigate the effects of nicotine on bone mineralization in human fetal osteoblasts cell line(hFOB1). To compare the alkaline ph-osphatase(ALP) synthesis, hFOBl were cultured with DMEM/F-12 1:1 Mixture and 100 pg/ml, 1 ng/ml, 10 ng/ml, 100 ng/ml, 1 ${\mu}g$/ml, 10 ${\mu}g$/ml, 100 ${\mu}g$/ml of nicotine. And to compare the calcium accumulation, hFOB1 cultured for 23 days were quantified and photographed. ALP activity of hFOB1 exposed to nicotine was not significantly changed at a lower concentrations of nicotine, but was significantly decreased at a higher concentrations (10 ${\mu}g$/ml, 100 ${\mu}g$/ml) of nicotine (p<0.05). A quantified calcium acculation in hFOB1 was significantly decreased at 1,10, and 100${\mu}g$/ml of nicotine (p<0.05). Significantly decreased calcium deposition was observed at 1, 10, and 100${\mu}$/ml of nicotine. These results indicate that a higher concentration of nicotine show a negative effects on mineralization of hFOB1.

Role of Intracellular Taurine in Monensin-induced $Na^+,\;Ca^{++}$ Accumulation and Mechanical Dysfunction in Isolated Rat Hearts

  • It has been postulated that the intracellular taurine is co-transported with $Na^+$down a concentration gradient and prevents the intracellular accumulation of sodium. It is therefore, expected that an elevated level of intracellular taurine prevents the sodium-promoted calcium influx to protect the cellular damages associated with sodium and calcium overload. In the present study, we evaluated the effects of intra- and extracellular taurine on the myocardial $Na^+$and$Ca^{++}$ contents and the cardiac functions in isolated rat hearts which were loaded with sodium by monensin, a $Na^+-ionophore$. Monensin caused a dose-dependent increase in intracellular $Na^+$ accompanied with a subsequent increase in intracellular $Ca^{++}$ and a mechanical dysfunction. In this monensin-treated heart, myocardial taurine content was decreased with a concomittent increase in the release of taurine. The monensin-induced increases in intracellular $Na^+$, $Ca^{++}$ and depression of cardiac function were prevented in the hearts of which taurine content had been increased by high-taurine diet. Conversely, in the hearts of which taurine concentration gradient had been decreased by addition of taurine in the perfusate, the monensin-induced increases in $Na^+$, $Ca^{++}$ and functional depression were accelerated. These results suggest that taurine, depending on the intra-extracellular concentration gradient, can affect intracellular sodium and calcium concentrations, and that an increased intracellular taurine may play a role in protection of myocardial dysfunction associated with the sodium and calcium overload.
