Microbiological Assessment of Home-Delivered Meals for Children from Low-income Families during Production and Delivery (결식아동을 위한 가정배달 도시락의 생산과 배달과정 중 미생물적 평가)

  • The purpose of this study was to assess the microbiological quality of home-delivered meals during production and delivery for children from low-income families. Production flows from a facility in Seoul that provides home-delivered meals were analyzed and the time-temperature of the food was measured. Microbiological assessment was performed for the production environment, personal hygiene, and food samples at each production and delivery step based on the process approach. It took 2 hours or longer from completion of production to meal delivery. An aerobic colony count (ACC) and coliform were not detected at knives, cutting boards, and dish towels. However, ACC (at pre-preparation, preparation, and packing areas) and coliform (at the preparation area) were detected on the hands and gloves of employees. Air-borne bacterial counts varied according to day and preparation area (ND~6 CFU/plate/15 min). Food temperatures, on the completion of production and meal delivery, fell into temperature danger zones. ACC and coliform counts of raw ingredients did not decrease after pre-preparation (washing and sanitizing) for menus involving food preparation with no cook step. ACC decreased after cooking step for menus of food preparation with cook step, but the ACC of the stir-fried and seasoned dried filefish fillet on the completion of cooking was too numerous to count due to improper heating. The ACC of seasoned young Chinese cabbages (a menu with complex food preparation) increased during delivery (from 2.5 log CFU/ml to 5.0 log CFU/ml). This qualitative assessment of foodborne pathogens revealed that B. cereus was detected in vegetable and meat product menus. These results suggest time-temperature control is necessary during production and delivery and management guidelines during production of home-delivered meals are provided for safe production.

Effects of Mashed Maesil and Maesil Extract on the Fermentation Characteristics of Kimchi (매실 첨가가 김치의 발효 특성에 미치는 영향)

  • This study investigated the effects of mashed maesil and maesil extract on Kimchi during fermentation. The mashed maesil and maesil extract were added at 5% of the weight of salted cabbages to the Kimchi sauce, and then physiochemical and sensory characteristics were examined over 28 days of fermentation at 4$^{\circ}C$. During the entire fermentation process, the mashed maesil Kimchi had a consistent pH of 4.4 to 4.2, and the mashed maesil controlled fermentation better than the maesil extract. The mashed maesil Kimchi had the lowest acidity ($0.43{\pm}0.01{\sim}0.42{\pm}0.01$), despite showing a similar pH level to the maesil extract Kimchi. The mashed maesil Kimchi presented the highest hardness level. And in the early stage of fermentation the control Kimchi had the lowest hardness; however, at the end of storage, hardness increased in every Kimchi and no differences were shown. In terms of color value changes, the L-values of every Kimchi increased as the fermentation period increased; but on the $28^{th}$ and final day of fermentation L-values had decreased in all groups. The group with mashed maesil had the highest a-value, but there were no significant differences in b-values among the groups. In evaluating sensory characteristics and acceptability of flavor, the maesil extract Kimchi was higher in intensity with regard to sourness, umami taste, sweetuess, maesil taste, carbonated taste, and fresh taste; it also had higher overall acceptability. Therefore, a fixed amount of maesil extract(5%) can be added to Kimchi with good affects on quality, by controlling fermentation and increasing flavor.

Development of a Cabbage Loader

  • Cabbage is one of the most important vegetable in Korea. The cabbage production was mainly based on human labor A comprehensive research fur substituting the human labor by machines has been performed until now. In general, cabbage is cultivated on hillside in Korea. Picking up the harvested cabbages in field and carrying to a vehicle fur transportation are very laborious work. Manual transportation of cabbage is likely to damage the quality and is also a cause to increase the cost of cabbage production. This study was to develop and evaluate a prototype cabbage loader fur efficient and safe transportation of cabbage. The developed loader was a semi-tracked vehicle operated by a hydraulic system, allowing the safe transportation and loading of cabbage in a steep field. The maximum loading weight of the loader was 1.0 ton. By using two sets of safety devices attached to the loader to avoid the roll-over in a steep field, the static rollover slopes were increased up to 34.0% and 37.4% fur the left and the rear direction, respectively The maximum field speed was about 6km/hr with two cabbage pallets of 750kg at a 25% inclined field. The loading capacity of the loader was about 35 pallets/hr when picking up, carrying, and unloading two cabbage pallets for one loading operation. The loading capacity was more than 8 times in comparison with the conventional human labor. The developed loader could be used fur loading and carrying the other vegetables. The study suggested a possible approach for designing the field machines operated on hillside.

Development of a Rapid Detection Method for Pectobacterium carotovorum subsp. carotovorum Using the Loop-Mediated Isothermal Amplification (LAMP) (Loop-Mediated Isothermal Amplification (LAMP)법을 이용한 Pectobacterium carotovorum subsp. carotovorum의 신속 진단법 개발)

  • Pectobacterium carotovorum subsp. carotovorum is the causative agent of soft rot in crops such as potato and cabbages. Loop-mediated isothermal amplification (LAMP) is a simple DNA amplification method, as well as isothermal PCR technique. In this study, a new method for the rapid detection of Pectobacterium carotovorum subsp. carotovorum was developed using LAMP that named PCC-LAMP. Based on lytic murein transglycolase gene of Pectobacterium carotovorum subsp. carotovorum, a set of four primers for LAMP was designed. The optimal PCC-LAMP reaction temperature was established at $61^{\circ}C$. Under standard conditions, PCC-LAMP amplified $1{\times}10^3$ copies of clone PCC-pBX437 per reaction. Further, this method can also assay directly by SYBR Green I without electrophoresis. Amplification was not detected for five other bacterial species. In conclusion, PCC-LAMP may be a useful method for the detection Pectobacterium carotovorum subsp. carotovorum in the field.

Growth Response and Total Coliform Distribution of Spinach and Chinese Cabbage and Soil Quality by Irrigation of Domestic Wastewater (하수종말처리장 방류수를 밭작물 관개용수로 처리시 시금치와 배추의 생육, 대장균 분포 및 토양의 질 평가)

  • The experiment was carried out to determine the distribution of total coliform the translocation of heavy metals and the salt accumulation in soils following the supply of ground water, the domestic wastewater and the ultraviolet treatment of domestic wastewater during the spinach and the chinese cabbage cultivation. There were not much changes in the Total-N, Total-P and cations in soils following the ultraviolet treatment of the domestic wastewater. However, the density of total coliform was dramatically reduced from 894MPN/100mL to 5MPN/100mL. The diagnosis of composition of soil after the harvest of chinese cabbages and spinach has shown that the concentration of Na$^+$ was 3-4 times higher in plot using domestic wastewater than in plot using ground water. When domestic wastewater and ground water were used in growing spinach md chinese cabbage at 50% each, the application of chemical fertilizers reduced about 25% to 50% compared to the criteria set down by the Rural Development Administration and there was not significant difference in terms of harvest. Using non-ultraviolet treatment of domestic wastewater directly on spinach and chinese cabbage has resulted in excessive density of total coliform at 25,000MPN/100mL. Even when ultraviolet treatment, the density was still high at 2,000MPN/100mL. The high density of total coliform even following ultraviolet treatment of domestic wastewater is considered to be caused by photo-reactivation of micro-organism. When reusing domestic wastewater, the application of sterilization such as ultraviolet, ultrasonic and electron-beam for public health and hygiene reasons may provide safe supply of agricultural water.

  • Croping pattern, fertilizer application, Soil chemical characteristics, plant nutritional condition and growth status were investigated in three major vinyl house farms near Yesan. Croping pattern changed from tomato to pumpkin due to tomato diseases such as wilting and blossom-end rot. Wilting seemed to be closely related with high EC, nitrogen and root cyst nematodes. Calcium deficiency seemed to be due to high potassium, EC in soil and high uptake of iron. Chinese cabbages in summer showed poor growth (80% inhibition) due to high EC(1.8mmho/cm) and easily got wenny root disease that might due to high phosphorus (1055ppm) in soil. Summer radish showed poor growth (50% inhibition) due to high EC(1.6mmho/cm), K and Mg resulting in base imbalance. Farmers used 5 kinds of compound fertilizer as basal application and one without P for top dressing. Urea and KCL were used for top dressing. Heavy application of livestock manure and chemical fertilizer for every crop made eutrophic even in subsoil.

Influence of Commercial Antibiotics on Biocontrol of Soft Rot and Plant Growth Promotion in Chinese Cabbages by Bacillus vallismortis EXTN-1 and BS07M

  • We investigated influence of three commercial antibiotics viz., oxolinic acid, streptomycin, and validamycin A, on biocontrol and plant growth promoting activities of Bacillus vallismortis EXTN-1 and BS07M in Chinese cabbage. Plants were pre-drenched with these strains followed by antibiotics application at recommended and ten-fold diluted concentration to test the effect on biocontrol ability against soft rot caused by Pectobacterium carotovorum SCC1. The viability of the two biocontrol strains and bacterial pathogen SCC1 was significantly reduced by oxolinic acid and streptomycin in vitro assay, but not by validamycin A. In plant trials, strains EXTN-1 and BS07M controlled soft rot in Chinese cabbage, and there was a significant difference in disease severity when the antibiotics were applied to the plants drenched with the two biocontrol agents. Additional foliar applications of oxolinic acid and streptomycin reduced the disease irrespective of pre-drench treatment of the PGPRs. However, when the plants were pre-drenched with EXTN-1 followed by spray of validamycin A at recommended concentration, soft rot significantly reduced compared to untreated control. Similarly, strains EXTN-1 and BS07M significantly enhanced plant growth, but it did not show synergistic effect with additional spray of antibiotics. Populations of the EXTN-1 or BS07M in the rhizosphere of plants sprayed with antibiotics were significantly affected as compared to control. Taken together, our results suggest that the three antibiotics used for soft rot control in Chinese cabbage could affect bacterial mediated biocontrol and plant growth promoting activities. Therefore, combined treatment of the PGPRs and the commercial antibiotics should be carefully applied to sustain environmental friendly disease management.

  • The cDNA clone of Brassica rapa WRKY7 (BrWRKY7) was obtained from EST collection in Brassica genomics team and its DNA sequence was determined. The cDNA clone is 1,037 bp long in nucleotides and encodes an open reading frame of 307 amino acids. Based on a phylogenetic tree, BrWRKY7 belongs to group IId. BrWRKY7 was induced by wound and SA. It was also induced by pathogen attack such as Xanthomonas campestris pv. campestris (Xcc), suggesting that this BrWRKY may play an essential role in defense response of chinese cabbages.

Consumed-Power and Load Characteristics of a Tillage Operation in an Upland Field in Republic of Korea

  • Purpose: This study derived the consumed power and load characteristics of a tillage operation performed in an upland field located in Seomyeon, Chuncheon, Rep. Korea, where potatoes and cabbages were cultivated in two crops. Methods: A plow and rotavator were mounted on a tractor with 23.7 kW of rated power to perform the tillage operation. The work conditions were determined, considering the actual working speed of the tillage operation performed by the local farmers. The power consumption of the rear axle, engine, and power take-off (PTO), PTO torque, and tractive force were measured under each work condition. The consumed power and load characteristics were analyzed using their average values. Results: The rotary-tillage operation consumed more engine power than the plow operation for the same tractor-transmission gear condition. The PTO in the rotary-tillage operation and the rear axle in the plow operation consumed the most power. The power consumption of the engine and the PTO for the rotary-tillage operation tended to increase as the transmission gears of the tractor and the PTO became higher. In contrast, the rear-axle power consumption was insignificant. In addition, the PTO torque tended to rise as the tilling pitch increased. For the plow operation, the drawbar power and the rear axle power accounted for 68-90% of the engine power. The engine and rear axle power, drawbar power, and tractive force tended to rise as the working speed increased. Conclusions: The power consumption and load characteristics differed for the plow and rotary-tillage operations. They may also differ depending on the soil conditions. Therefore, the power consumption and load characteristics in various work environments and regions should be analyzed, and reflected in the design of tractors and working implements. The results derived from this study can be used as a reference for such designs.

Metagenomic Approach to Identifying Foodborne Pathogens on Chinese Cabbage

  • Foodborne illness represents a major threat to public health and is frequently attributed to pathogenic microorganisms on fresh produce. Recurrent outbreaks often come from vegetables that are grown close to or within the ground. Therefore, the first step to understanding the public health risk of microorganisms on fresh vegetables is to identify and describe microbial communities. We investigated the phyllospheres on Chinese cabbage (Brassica rapa subsp. pekinensis, N = 54). 16S rRNA gene amplicon sequencing targeting the V5-V6 region of 16S rRNA genes was conducted by employing the Illumina MiSeq system. Sequence quality was assessed, and phylogenetic assessments were performed using the RDP classifier implemented in QIIME with a bootstrap cutoff of 80%. Principal coordinate analysis was performed using a weighted Fast UniFrac matrix. The average number of sequence reads generated per sample was 34,584. At the phylum level, bacterial communities were composed primarily of Proteobacteria and Bacteroidetes. The most abundant genera on Chinese cabbages were Chryseobacterium, Aurantimonadaceae_g, Sphingomonas, and Pseudomonas. Diverse potential pathogens, such as Pantoea, Erwinia, Klebsiella, Yersinia, Bacillus, Staphylococcus, Salmonella, and Clostridium were also detected from the samples. Although further epidemiological studies will be required to determine whether the detected potential pathogens are associated with foodborne illness, our results imply that a metagenomic approach can be used to detect pathogenic bacteria on fresh vegetables.
