Nosema sp. isolated from Cabbage White Butterfly(Pieris rapae) Collected in Korea

  • A microsporidium, from cabbage white bntteflies, Pieris rapae, collected in Korea, was purified and characterized according to its gene structure, spore morphology and pathogenicity. From the observation of the isolate by SEM and TEM, the endospores, exospores and nuclei, about 12 polar filament coils of the polar tube and posterior vacuoles were all identified. The nucleotide sequence was determined for a portion of genomic DNA which spans the V4 variable region of the small subunit rRNA gene. Comparison with the GenBank database for 15 other microsporidia species suggests that this isolate is most closely related to Nosema species. The pathogenicity against cabbage white butterflies was quantified by inoculating variable doses of spores to the second instar larvae. Peroral inoculation at a dosage of 10$\^$8/ spores/ml resulted in the death of all larvae prior to adult eclosion, but at lower spore dosages of 10$\^$4/-10$\^$5/ spores/ml, many adults successfully emerged. The median lethal dose (LD$\_$50/) was deter-mined to be 4.6$\times$10$\^$6/ spores/ml and the isolate also transmitted transovarially to the progeny eggs at a frequency of 92%.

  • This study has been performed to investigate normal synaptic organizations in the subesophageal ganglion and terminaiion of antennal receptor cells in the ipsilateral subesophageal gan91ion of Pieris rapae. The various normal synaptic organizations in subesophageal ganglion could be differentiated into the five types. The proximal removal of a left antenna resulted in the weakly-dark, semidark and dark degenerations in the type I bostons of the ipsilateral subesophageal ganglion. Therefore, it was concluded that the axon terminals of the receptor cells projecting from the antenna into the brain form the type 1 synapses together with the dendrites in the ipsilateral subesophageal ganglion.

  • 배추흰나비의 전용기 혈림프에서 $\beta$-N-Acetylglucosaminidase를 DEAE-cellulose ion exchange 및 Sephadex G-200 column chromatography 법을 이용하여 정제하였다. 정제된 효소는 7% acrylamide gel 전기영동에서 단일 band를 나타내었으며, 당을 함유한 복합단백질이었다. 최적 pH는 5.0, 최적반응온도는 6$0^{\circ}C$이었으며, 각 온도에서 10분간 incubation한 열안정성에 있어서는 6$0^{\circ}C$까지 비교적 높은 활성을 나타내었지만 7$0^{\circ}C$ 이후에는 거의 활성을 나타내지 않았다. 또한 Hg$^2$+ 처리는 심한 효소활성의 저해 현상을 보였지만, Mn$^2$+, $Mg^2$+ 등은 활성의 증가를 나타내었으며, Km 값은 6.67$\times$10-$^3$M, pl 값은 5.8, 분자량은 4.1$\times$105 daltons 이었다.

  • The study on the nerve cells in the pars intercerebralis(IP) of 5-day-old cabbage butterfly Pieris rapae L. was performed to observe their ultrastructures and classify them on the basis. of the differences in size, shape and relative distribution cf cell organelles. The brain-subesophageal ganglion complex was fixed in 1% paraformaldehyde-1% gluaraldehyde mixture and embedded in araldite mixture. The transverse thin sections of IP were stained with uranyl acetate and lead citrate and examined by Hitachi 500 and ]EM 100B electron microscope. Five distinct types. of nerve cells are recognized and are arbitrarily designated as Type I, Type II Type III, Type IV and Type V. Type I neurone: These neurones are neurosecretory cells. Several neurosecretory cells are. recognized in the pars intercerebralis. They are roughly round or peach-shaped cells measuring $13{\sim}25{\mu}m$ in diameter. The rounded nucleus shows about $5{\sim}10{\mu}m$ in diameter. The chromatin is predominantly diffused with only occasional dense patches. The perikaryon contains numerous. mitochondria, free polyribosomes and neurosecretory granules. The neurosecretory granules are relatively uniform in electron density, and each one is about $100{\sim}400{\mu}m$ in diameter and surrounded by a single membrane. The granules are also observed mostly as in groups. In one group of neurones the cisternae of endoplasmic reticulum are distended or in other group of neurones are not distended. Golgi saccules are slightly dilated at their lateral extremities and contains. frequenty dense rounded materials. Type II neurone: Thes have the largest soma in the pars intercerebralis about $30{\sim}35{\mu}m$ in diameter. They also show roughly polygonal in shape. The nucleus is elongated or sickle-shaped. The chromatin is mainly in the euchromatin form. The perikarya in these cells are well populated with populated with free ribosomes and contain numerous mitochondria and Golgi bodies. The cisternae of granular endoplasmic reticulum are also well distributed. Type III neurone: They are oval or spindle-shaped and also medium-sized. neurones approximately $15{\sim}17{\mu}m$ in length. The nucleus is oval or slightly elongated in shape and $8{\sim}9{\mu}m$ in length. The chromatin occurs in diffused form. The cytoplasm contains many filamentous or oval mitochondria. The perikaryon has also numerous free polyribosomes and cisternae of granular endoplasmic reticulum. Type VI neurone: They are roughly polygonal in shape probably due to the close approximation of the adjacent cells. The soma is about $7{\sim}8{\mu}m$ in diameter. The nucleus is round or oval in shape and $5.0{\sim}5.8{\mu}m$ in diameter. The necleus also occupies a large proprion of the cell body. The perikaryon is well populated with free ribosomes and contains several mitochondria and cistenae of granular endoplasmic reticulum. Type V neurone: These neurones are similar to Type VI neurones in various respects such as cell size and cell inclusion, but they differ from Type IV neurones in shape. The soma is oval or slightly elongated. The cell body contains several filamentous and oval mitochondria.

한국의 초등 과학 교과서 생명영역에 나오는 탐구 주제 및 생물 재료에 대한 초등 예비교사들의 인식 분석 (The Analysis of Pre-service Elementary Teachers' Perception on Biology Inquiry Subjects, and biology Materials in the Korea Elementary School Science Textbook)

  • The aim of this study was to analyze a group of pre-service elementary teachers experience on the cognitive terms of biology inquiry subjects, and experience on living materials related with the domain of life in the Korea elementary school science textbook. Firstly, elementary school pre-service teachers had the confidence of inquiry subjects related with the domain of life such as 'when we take breath what will happen in human body?' and 'Can we make the model of human body?' But they did have the most diffidence of 'The search for pieris rapae, a cabbage butterfly's life', 'Can we grow the little creature?' The reason why pre-service elementary teachers had diffidence of comprehending experiment procedure and planning ability was that 'they focused on understanding the scientific concepts instead of planning experiments by themselves', and 'they carried out the least amount of experiments' Secondly, elementary school pre-service teachers had never experienced biology materials related with the domain of life in the Korea elementary school biology science textbook such as 'tradescantia reflexa', 'Caddis larva', 'hydrilla verticillata', and 'Plantain lily.' According to the findings obtained from the study, the reason why they had never seen the biology materials was that they had the least opportunity of immediately observing life creatures due to memorization-intensive classes.

  • The oval shaped pericardial cells are clustered along the lateral sides of the heart and irregularly connected with the heart. The cells are bounded by a basement membrane. The basement membranes of the connected two peicardial cells are irregularly linked each other there-fore funnels are formed. The multiple invaginations of the cell membrane are observed and septate junctions develope at the part of enterance of the cell membrane. The coated pits are appeared in the inner side of the invaginated cell membrane. The coated vesicles, tubular and spherical shaped vesicle, Golgi complex containing high electron densed material in the cisternae and mitochondria are observed in the cytoplasm and lysosomes are remarkably well developed. The whirled membrane structures in the multiformed complex bounded by single membrane are linked with low electron densed granules and spherical shaped small granules having high electron density with $0.03{\mu}m$ in diameter are located between the whirled membrane in a row and gradually secretes the granules and then they produced the multilamellar body. The lysosomal regions of cytoplasm of pericardial cell are appeared negative reaction to the acid phosphatase and according to the results of the electrophoresis, lipoproteins having acid phosphatase activity are contained. The axon is contacted with the pericardial cells.

  • A observation of the ovarian development and oogenesis of Pieris rapae Linne has been carried out during metamorphosis using stereo-microscope, light microscope and electron microscope. The results obtained through this experiment are as follows: 1. The ovarian development and vitellogenesis begin at the 3-day old pupa and the 6-day old pupa respectively, and the adult ovary right after their emergence contains a few mature eggs. 2. The species described above are further observed at six different stages in oogenesis, and the results are summarized as follows. 1) Pieris rapae has polytrophic ovarioles. The cell organelles of the nurse cells are transfered to the oocyte through the ring canal at the early oogenesis. 2) At stage 2, the nuclear envelope of oocyte nucleus is less infolding than that of nurse cell nucleus. In the oocyte cytoplasm a large number of ribosomes are observed. 3) At stage 3 and 4, many micropinocytotic vesicles are observed in the oocyte cytoplasm. These vesicles are fused together to form large proteid yolks. 4) At stage 5, the vitelline membrane is laid down in the intercellular space between the follicle cells and oocyte. 5) At stage 6, the chorion is formed by the follicle cells. 6) A micropyle and a number of aeropyle are observed on the surface of a mature egg.

배추흰나비 (Pieris rapae L.) 성충 복안에서의 기관지 미세구조 (The Ultrastructure of Trachea of the Compound Eye in Adult Pieris rapae L.)

  • The ultrastructure of the trachea of the compound eye in adult cabbage butterfly, Pieris rapae L., was studied. Trachea under the basement membrane is divided into tracheoles of the ommatidia which blanched 4 tracheoles during 2 divisions. Tracheoles of the ommatidia originated from one tracheolar cell and its nuclear is located under the basement membrane and its cytoplasm enveloped the retinular cell and the retinular pigment cell. The modified structure of tracheoles is located at the first division area of the ommatidia and its size is $8{\mu}mX4.5{\mu}m$. And the length of its taenidium is $0.1{\sim}0.6{\mu}m$, diameter $0.15{\mu}m$, and interval $0.3{\mu}m$.
