쪽(Polygonum tinctortium) 세포의 형질전환 및 쪽 세포에서 Sodium Butyrate가 Green Fluorescent Protein 발현에 미치는 영향 (Effect of Sodium Butyrate on GFP Expression Level in Transgenic PoIygonum tinctorium Cells)

  • 쪽(Polygonum tinctortium) 세포에서 외래 단백질 발현을 검토하기 위하여 green fluorescent protein(GFP)가 내재하는 pCAMBIA1302를 형질 전환시켰으며 Western blot 분석에 의해 GFP의 발현을 확인하였다. Sodium butyrate가 GFP생성에 미치는 효과를 검토한 결과, 10 mM에서 세포성장이 지연되었으며, 15 mM 이상에서는 정지되었다. 세포 내 GFP 생성량은 세포 접종 후 3일째 5 mM sodium butyrate를 첨가하였을 때가 0일째 처리에 비해 120% 증가하였다. 또한 접종후 3일 후 5 mM의 sodium butyrate를 처리한 경우가 10 mM의 경우보다 GFP의 수율이 50% 증가하였다. 본 실험을 통하여 세포 접종 후 3일째, 5 mM의 농도로 처리한 sodium butyrate가 외래 단백질의 발현을 효과적으로 증가시키는 결과를 확인하였다.

Growth of Clostridium thermobutyricum: a Cellulolytic Thermophile

  • Increased concentrations of yeast extract led to increased growth yields and faster growth rates of the newly isolated Clostridium thermobutyricum. This species produced butyrate as its main fermentation product from glucose as well as from yeast extract. In the presence of peptone or tyrptone and during growth on agar, up to 70% of the cells sporulated. Growth yields were 30 and 55 g per mole glucose in the presence of 0.05 and 2.0% yeast extract, respectively. The Arrhenius graph was biphasic, exhibiting an intermediary plateau around $38^{\circ}C$ with a concomitant change in the Arrhenius energy. The optimum temperature was $55^{\circ}C$. An unusually sharp decline in the growth rate occurred above $59^{\circ}C$ .

  • Fibers of microbial polyesters, poly(3-hydroxy butyrate) (PHB) and poly(3-hydroxy butyrate-co-3-hydroxy valerate) (HB-co-HV) were prepared by electrospinning method. The obtained fibers were evaluated by differential scanning calorimetry, scanning electron microscopy, and oil absorption. The formation of fibers was strongly dependent on a concentration of solution. At a low concentration, the fibers contained beads which is from aggregation of polymer due to short evaporation time. The fine fibers with $2-5{\mu}m$ diameter were obtained at 20 wt% concentration. The contact angle measurement showed that the fiber had higher water contact angle than the film due to the lotus-like effect. Oil absorbency showed that the fiber had higher than the film. Specially, the HB-co-HV fiber which was spinned from 20 wt% absorbed 65% oil which is much higher than that of a normal polypropylene-based oil paper.

식물성유 첨가가 면양의 반추의 발효특성과 소화율 및 지방산염 형성에 미치는 영향 (Effects of Supplemental Vegetable Oils on Ruminal Characteristics, Digestibility and Formation of Fatty Acid Soaps in Sheep)

  • 본 시험은 oil 첨가시 알팔파건초를 기질로 하여 반추위 내에서 유리된 지방산과 양이온이 결합하여 형성되는 지방산염, 반추위 발효성상 및 소화율에 미치는 영향에 대하여 살펴보고자 시험을 실시하였다. 면양 3두를 이용하여 $\circled1$ oil 무첨기구, $\circled2$ 대두유 7% 첨기구 및 $\circled3$ 옥수수유 7% 첨가구로 하여 3${\times}$3 라틴방각법으로 수행하였다. 면양에게는 알팔파건초와 농후사료를 70 : 30의 비율로 급여하였다. 위액은 오전사료 급여 전 1시간, 사료 급여 후 1, 3, 6 및 9시간째 채취하였다. pH는 사료급여 후 3시간까지 감소하다가 그 이후에는 증가하였다{P<0.0001). $NH_3$-N는 pH 와는 반대의 경향을 나타내었다(P<0.0001). 총 휘발성 지방산, acetate, propionate 및 butyrate의 농도에는 변화가 없었다. A/P 비율은 배양시간이 증가함에 따라 감소하다가 9시간에서는 사료급여 1시간 수준이었다{P<0.0001). 건물, total-N, 조섬유, 회분, 기용무질소물, NDF 및 ADF의 소화율은 차이가 없었다. FAS의 형성은 사료급여 1시간에 비하여 사료급여 후에 증가하였으며(P<0.05), 대조구에 비해 oil 첨가구가 높았다(P<0.05). 장쇄지방산이 주로 지방산염을 형성하였다. 그래서 oil 첨가가 반추위 발효특생과 소화율에 미치는 유해한 영향을 방지할 수 있었던 것으로 사료 된다.

투과증발을 이용한 부탄올 추출발효 시스템의 모델링과 분석 (Modeling and Analysis of Extractive Butanol Fermentation with Pervaporation)

  • Results from experiments and mathematical modeling were compared for pervaporative butanol fermentation. The developed model includes expressions to predict characteristics of butanol fermentation, such as, microbial growth, solvent (butanol, acetone, and ethanol) formation and organic acid (acetate and butyrate) production. Butanol diffusivity was 1.15${\times}$10(sup)-7 ㎡/hr at 1.5 L/min-tubing of air flow rate using a pervaporative module. The model correlated well with experimental results (cell growth, glucose consumption and concentrations of solvents and organic acids) for batch fermentation with and without pervaporation. Larger surface area and thinner module tubing resulted in an increased glucose consumption and a decreased residual butanol concentration. Optimum membrane area and thickness were 0.34 ㎡ and 120 $\mu\textrm{m}$, respectively.

  • Cobalt(Ⅲ) complexes of trifunctional amino acid and N,N'-diethylethylenediamine-N,N'-di-α-butyrate(deedba), s-cis-[Co(deedba)(L-aa)] (L-aa=S-methyl-L-cysteine, L-aspartic acid, L-glutamic acid) have been prepared from the reaction between the s-cis-[Co(deedba)(Cl2)]- complex and the corresponding amino acid. The amino acids have been found to coordinate through the amine and carboxylate groups. The S-methyl-L-cystene is coordinated not by the sulfur donor atom, but by the nitrogen and oxgen donor atoms, and the L-aspartic and L-glutamic acids are coordinated to the cobalt(Ⅲ) ion via formation of the five-membered glycinate chelate ring. Relatively small optical activity shown by the complexes is due to the chiral center present in the amino acids.

음식물쓰레기의 수소발효 타당성 평가 (Feasibility Test of Biohydrogen Production from Food Waste)

  • 그동안 유기성폐수에 대해서는 다양한 연구가 시도되었지만, 유기성고형폐기물에 대해서는 보도된 바가 없다. 그러므로 본 연구는 열처리된(heat-shocked) 혐기성슬러지를 식종한 침출상(leaching-bed) 반응조에서 다양한 희석율(dilution rate; D)에 대하여 음식물쓰레기로부터의 수소생산 효율을 살펴보았다. 반응조의 성공적인 운전은 적절한 희석율의 선택에 달려있는데, 높은 희석율에서는 미생물의 유실이 발생하는 반면 낮은 희석율에서는 유기산의 축적으로 product inhibition이 발생한다. 따라서 이러한 요인들이 높은 효율의 수소생산을 제한하게 된다. 모든 반응조들은 희석율에 관계없이 다음과 같은 경향을 보여주었다. 운전 초기 3일 동안, 수소생산이 활발하였으며 유기산의 농도가 에탄올보다 높게 유지되었다. 유기산 중에는 락트산이 초기 잠깐동안 높은 값을 유지했을지라도, 전체 운전기간 내내 뷰틸산과 초산이 유기산의 주요성분이었다. 다른 희석율에 비해 희석율 $4.5d^{-1}$는 발효가 진행되는 동안 가장 높은 뷰틸산/초산 (B/A) 비율을 보여주었는데, B/A 값의 경향은 수소생산의 경향과 매우 유사하여 뷰틸산의 형성이 수소생산에 매우 호의적임을 나타내준다. 에탄올은 수소생산이 멈춘 4일째부터 크게 증가하였는데, 이것은 열처리된 슬러지가 대사흐름을 수소 및 유기산생성경로(hydrogen-and acid-producing pathway)에서 알콜 생성경로(alcohol-producing pathway)로 전환시킬 수 있음을 보여준다. 희석율 $4.5d^{-1}$의 COD 제거효율은 58%로 다른 희석율(2.1, 3.6 및 $5.5d^{-1}$)의 제거율(51.5, 55.3 및 53.7%)보다 높았으며, 이때 제거된 COD는 수소(10.1%), 유기산(30.9%), 에탄올(17.0%)로 전환이 되었다.

  • The carbon metabolism of newly isolated Clostridium tyrobutyricum JM1 was investigated at varying initial glucose concentrations (27.8-333.6mM). Because an understanding of metabolic regulations was required to provide guidance for further effective metabolic design or optimization, in this case, maximizing hydrogen production, carbon and energy balances by C. tyrobutyricum JM1 were determined and applied in anaerobic glucose metabolism. The overall carbon distribution suggested that initial glucose concentrations had strong influence on the stoichiometric coefficients of products and the molar production of ATP on the formation of biomass. C. tyrobutyricum JM1 had a high capacity for hydrogen production at the initial glucose concentration of 222.4 mM with high concentrations of acetate and butyrate.

Effects of the Redox Potential of the Acidogenic Reactor on the Performance of a Two-Stage Methanogenic Reactor

  • Distillery wastewater was used in a thermophilic laboratory-scale two stage anaerobic digester to test the effects of the redox potential of the first acidogenic reactor on the performance of the system. The digester consisted of first a acidogenic reactor and the an upflow anaerobic sludge blanket (UASB) reactor. The digestor was operated at a hydraulic retention time (HRT) of 48 h. Under these conditions, about 90% of the chemical oxygen demand as measured by the chromate method ($COD_{cr}$) was removed with a gas production yield of 0.4 l/g-COD removed. The redox potential of the acidogenic reactor was increased when the reactor was purged with nitrogen gas or agitation speed was increased. The increase in reduction potential was accompanied by an increase in acetate production and a decrease in butyrate formation. A similar trend was observed when a small amount of air was introduced into the acidogenic reactor. It is believed that the hydrogen partial pressure in the acidogenic reactor was decreased by the above mentioned treatments. The possible failure of anaerobic digestion processes due to over-loading could be avoided by the above mentioned treatments.
