Application of a Gas Chromatography/Mass Spectrometric Method for the Determination of Butyltin Compounds in Sediment

  • A gas chromatography/mass spectrometric (GC/MS) method has been developed for the determination of trace mono-n-butyltin (MBT), di-n-butyltin (DBT), and tri-n-butyltin (TBT) compounds in sediments. Samples were extracted by 10% acetic acid in methanol containing 0.03% tropolone and were then derivatized for GC/MS analysis. Ethylation by sodium tetraethylborate and phenylation by sodium tetraphenylborate were evaluated as a derivatization reaction of the organotins in sample extract. n-Hexane was added into reaction media in the beginning of the reaction for the continuous extraction of derivatized organotins. Ethylation requires less than 2 hours to get proper derivatization yields for MBT, DBT, and TBT altogether and produces relatively low amounts of side reaction products. Compared to ethylation, phenylation requires much longer time but provides relatively lower yield and produces considerable amounts of side reaction products. Therefore, the ethylation reaction was applied for the analysis of organotin compounds in sediment. An isotope dilution mass spectrometric (IDMS) method based on GC/MS has been applied to the accurate determination of DBT compounds in the sediments. The IDMS results from the analyses of sediment samples showed a reasonable repeatability and a good agreement with the values obtained by IDMS based on liquid chromatography/induced coupled plasma/mass spectrometry.

한국연안의 참굴 및 바지락 속에 존재하는 부틸주석화합물의 분석 (Analysis of butyltin compounds in Crassostrea gigas and Tapes philippinarum on the korean coast)

  • IMO (International Maritime Organization)와 한국은 2003년을 기점으로 하여 TBT의 사용을 전면 금지 하였다. 이 금지를 통하여 몇몇 나라에서는 해양환경(바닷물, 퇴적물, 굴)에서 TBT의 오염도가 차츰 감소되고 있는 것을 확인 하였다. 본 연구에서는 2003년 규제 이후 TBT의 오염도를 확인 하기 위하여 Crassostrea gigas(굴) 및 Tapes philippinarum(바지락)을 선택 하여 부틸주석화합물(butyltin compounds)의 농도를 측정하였다. Crassostrea gigas 및 Tapes philippinarum 가 생산되는 농장 및 주요 TBT 오염이 예상되는 지점에서 시료를 직접 채취하여 농도를 측정하였다. 각각 측정된 농도는 Crassostrea gigas의 경우 MBT, DBT와 TBT가 각각 N.D. -N.D., N.D. -N.D., N.D. -60.21 ng Sn $g^{-1}$ (wet wt)으로 측정 되었다. Tapes philippinarum의 경우 MBT, DBT와 TBT가 N.D. -12.27, N.D. -45.95, N.D.-7.30 ng Sn $g^{-1}$ (wet wt)로 각각 측정이 되었다. 본 연구에서는 TBT 오염도 감소를 확인 하기 위하여 1994년도에 측정된 동일 지역의 Crassostrea gigas의 부틸주석 화합물의 농도 대하여 비교조사를 해보았다. 조사결과 비교대상 7개 지점 샘플 중에 모든 샘플에서 TBT가 감소 추세를 보이는 것으로 나타났다. 이 중 4개 지점 샘플은 butyltin compounds가 검출한계 이하로 측정이 되었다. Tapes philippinarum의 경우도 외국 연구자료와 비교하여 볼 때 높지 않은 것으로 나타났다.

TBTCl (tri-n-butyltin chloride)과 bisphenol A에 의한 단기노출이 등줄쥐의 번식에 미치는 영향 (Effects of Short-Term Exposure with Tri-n-Butyltin Chloride (TBTCl) and Bisphenol A on the Reproduction of the Striped Field Mouse)

  • TBTCl과 bisphenol A (BPA)가 등줄쥐의 번식에 어떠한 영향을 미치는지 알아보기 위하여, 등줄쥐를 비번식기에 포획하여 번식기 직전에TBTCl과 BPA를 근육 주사한 후, 4개월 간 사육하였다. 그 결과, 암 수 모두 체중 증가량에 차이가 없었고, 수컷 체내의 각 화합물의 체내 잔류량, 성체와 새끼(F1) 수컷에 대한 생식소중량지수(GSI) 및 정소와 부정소의 광학 및 전자현미경 조직상이 대조군과 별다른 차이점을 보이지 않았다. 그러나 TBTCl 투여군에서는 출생한 태아의 사망 빈도가 높고, BPA 투여군에서는 유산율이 대조군에 비해서 높았다. 이로부터, 비번 식기의 성체 등줄쥐에게 TBTCl과 BPA를 단기간 투여했을 때, 이들 물질은 체내에 장기간 잔류하지 않으며, 성체 및 다음 세대 수컷의 생식기관에 영향을 미치지 않지만, 수태된 태아의 발생을 저해하는 물질임이 밝혀졌다.

  • This study find out the effect of red Ginseng extract(1.0g/kg) against TBTO(10, 20 and 40mg/kg) poisoning on each organs, hematological, serum biochemistry in rat for 3 weeks. 1. Each organ weight per body weight ration in treated group, all rats liver were significantly increased. (P<0.05,P<0.01)2. Iin TBTO treated group, the hematological parameters such as WBC, RBC, Hgb, Hct, PLt, MCV, MCH and MCHC in serum were remarkably decreased in comparison to that of control group. Only added red Ginseng extract group were slightly decreased but not significanted. 3. In TBTO treated group, the biochemical parameters such as AST, ALT in serum were remarkable elevated in comparison to that control group (P>0.05).4. In TBTO treated group, the activity of Triglyceride in serum of male rat were significantly increased in comparison to that of control group (P<0.05, P<0.01).5. In case of B.U.N activity in treated group(10, 20 and 40 mg/kg) were almost increased in comparison to that of control group, but not added red Ginseng extract.

넙치의 혈청, 간 및 근육 중 Tributyltin 축적 경향 (Accumulation of Tributyltin (TBT) in Blood, Liver and Muscle of Olive Flounder)

  • Accumulation of tributyltin (TBT) in serum, liver and muscle of olive flounder (Paralichtys olivaceus) was investigated in a 60-d static renewal exposure at $0.1{\mu}g$ TBT/L. Tributyltin accumulated rapidly from 83 ng Sn/g to 2,227 ng Sn/g on a wet weight basis in the serum of the olive flounder and to a greater extent than in the other tissues. The accumulated TBT concentrations in tissues were in the order of serum>liver>muscle on wet or dry-weight basis. High concentrations of dibutyltin (DBT: 990 ng Sn/g dry wt) and monobutyltin (MBT: 141 ng Sn/g dry wt), degradation products of TBT were found in liver of olive flounder at the end of exposure. On the other hand, DBT and MBT was below the detection limits in muscle during the exposure, and only low concentration of DBT (56 ng Sn/g) were detectefd in serum. Butyltin compounds were also quantitatively determined in feral fine-spotted flounder (Pleuronichthys cornutus) collected from Gwangyang Bay as one of polluted area and Sori Island as a reference site. All three butyltin compounds were detected from the fine-spotted flounder from Gwangyang Bay up to 3,107 ng Sn/g of total butyltins in liver, while 120 ng Sn/g of total butyltin concentration was found in the liver of fish from Sori Island.
