Analysis of Gene-specific Molecular Markers for Biotic and Abiotic Stress Resistance in Tropically adapted Japonica Rice Varieties

  • Since 1992, the Rural Development Administration (RDA), Republic of Korea in collaboration with International Rice Research Institute (IRRI) has developed 6 japonica rice varieties(MS11, Japonica 1, 2, 6, 7 and Cordillera 4) that are adaptable to tropical regions. However, these varieties show moderate resistance or susceptibility to certain biotic and abiotic stress. The development of varieties with more stable forms of resistance is highly desirable, and this could be possibly achieved through rapid introgression of known biotic and abiotic resistant genes. In this study, we analyzed the allele types of major biotic stress resistant genes including Xa5, Xa13, Xa21 and Xa25 for bacterial leaf blight, Pi5, Pi40, Pish and Pita2 for blast, tsv1 for rice tungro spherical virus, and Bph6, Bph9, Bph17, Bph18 and Bph32 for brown planthopper by using gene-specific molecular markers. In addition, seed quality related genes Sdr4 for preharvest sprouting and qLG-9 for seed longevity were also analyzed. The results revealed that2h5 and Xa25 resistance alleles showed in all varieties while Pi5 resistance allele showed only in MS11. The Pish resistance allele were present in five varieties except for Japonica 1. Meanwhile, for the rest of the genes, no presence of resistance alleles found in six varieties. In conclusions, most of tropical japonica varieties are lack of the major biotic stress resistant genes and seed quality genes (Sdr4 and qLG-9). Moreover, the results indicated that rapid deployment of a few major genes in the current tropical japonica rice varieties is urgent to increase durability and spectrum of biotic stress resistance and also seed dormancy/longevity which are essential traits for tropical environments.

쌀(백미, 현미) 시료로부터 고해상도 질량 분석 기반의 페놀산 유도체 동정 및 정량 평가 (Identification and Quantification of Phenolic Acid Derivatives from Three Korean White and Brown Rice Samples Based on High-Resolution Mass Spectrometry)

  • Reported positive ion fragmentation of phenolic acid derivatives in rice (Oryza sativa L.) were summarized based on the literature. A total of eight phenolic acids (4 derivatives of ferulic acid, 3 derivatives of sinapic acid and p-coumaric acid) were isolated and identified from rice (raw and steamed) using UPLC-DAD-QToF/MS. Results revealed that 6-O-feruloylsurose was the major component with 3'-O-sinapoylsucorse being tentatively identified in Oryza sativa L. for the first time as a new hydroxycinnamoyl derivative in rice grains. In our study, raw brown rice had the highest phenolic acid contents with Samkwang showing higher phenolic acid content than Saeilmi and Sindongjin (12.41 vs. 7.89 and 3.10 mg/100 g dry weight, respectively). Of all varieties, brown rice had higher phenolic acid contents than white rice. These contents decreased considerably when rice was steamed whereas, p-coumaric acid and ferulic acid contents were increased. Additionally, contents of rice (raw and steamed) can be used as a fundamental report for new rice varieties.

누대도태에 의한 벼멸구의 품종적응성의 변화 (Changes in the Fitness of Brown Planthopper, Nilapawata lugens Stal (Homoptera: Delphacidae) to Several Resistant Rice Varieties after Multi-generational Selection)

  • 벼멸구를 저항성 품종에서 도태시켰을 때 새로운 생태형으로 발전되는 속도와 벼멸구의 생태형과 벼의 품종저항성 간에 유전적으로 대응하고 있는지를 알아보고자, 감수성품종(동진벼)으로 사육하던 벼멸구를 Bph1 gene을 가진 저항성품종(청청벼)으로 각각 3세대 및 6세대 도태시킨 후, 같은 Bph1 gene을 가진 4개 품종(Mudgo, IR26, IR64, 청청벼)과, 다른 bph2 gene을 가진 2개 품종(밀양63호 및 가야벼)및 저항성이 없는 2개 품종(동진벼, TN-1)을 공시하여 각 계통의 벼멸구의 산란 및 섭식선호성, 수명, 발육기간, 사충율, 산란수 등을 조사하였다. 그 결과 감수성 벼멸구(생태형-1)를 저항성 Bph1 유전자를 갖인 청청벼에서 3세대(청청3세대계)및 6세대(청청6세대계)로 도태시켰을 때, 사육세대가 많아질수록 청청벼에서의 성충의 수명은 길어지고, 산란수는 많아지며, 또한 약충의 우화율이 높아지고, 약충기간이 짧아지는 등 빠르게 저항성을 극복하였다. 청청3세대계 및 6세대계 벼멸구는 청청벼와 같은 Bph1저항성 유전자를 가진 다른 품종들(Mudgo, IR26, IR64)에 대해서도 도태세대수가 많아지면서 선호성이 점점 높아지는 경향이었다. 성충의 수명과 산란수에 있어서도 청청3세대계, 청청6세대계 등으로 도태세대가 많아질수록 같은 저항성 Bph1유전자를 가진 Mudgo, IR26, IR64등에서 수명이 길어지고 산란수도 많아졌다. 벼멸구의 약충기간은 품종과 벼멸구 도태계통간에 큰 차이가 없었다. 그러나, 약충기간 중 사망률은 도태가 진행되면서 역시 Bph1저항성 gene의 4개 품종 모두에서 낮아졌고, 죽은 개체의 수명도 길어졌다. 밀양 63호(bph2 gene)와 가야벼(Bph1+bph2 gene) 등 2개 품종에서는 벼멸구의 3개 계통 모두 우화하지 못했다.

현미 저온저장이 쌀과 밥의 품질에 미치는 영향 (Effects of Low-Temperature Storage of Brown Rice on Rice and Cooked Rice Quality)

  • 현미저장 중 저장온도에 따른 쌀과 밥의 품질변화를 구명하고자 추청벼 등 14품종의 현미를 $15^{\circ}C$$35^{\circ}C$에 10주간 저장 후 현미 및 백미, 밥의 품질관련 특성을 조사하였다. 1. $15^{\circ}C$저장이 $35^{\circ}C$에 비하여 현미의 Tz값이 높아 배아 활성 정도가 높았으며 도정 후 백미 완전미율이 높아 $15^{\circ}C$ 저장 쌀의 품질이 양호하였다. 2. $35^{\circ}C$에 비하여 $15^{\circ}C$에 저장한 현미에서 쌀밥의 윤기치는 높았으나, 밥 색도 b 값, 밥 경도, 호화점도(최종점도, 강하점도)가 낮아 밥의 품질이 우수하였다. 3. 밥 색도 b값은 조생종이 중.만생종에 비하여 높았고, 밥 경도는 조생종이 낮았으며, 저장 현미의 도정 후 백미 완전미율은 $15^{\circ}C$에서 78%, $35^{\circ}C$에서 70%이었으며 $35^{\circ}C$에서 화성벼, 추청벼, 남평벼는 82%이상을 나타냈다. 4. $15^{\circ}C$ 저장에서 밥 색도 b값은 pH와 $-0.693^{**}$, 윤기치와 $-0.823^{**}$, L값과 $-0.772^{**}$, 지방산도와 $-0.548^*$, 백미 완전미율과 $-0.576*$, L값과 $-0.772^{**}$, a값과 $0.557*$ 의 유의성을 나타냈으며, 백미 완전미율은 밥색도 L값과 $0.638*$, 밥색도 b값과 $-0.576*$의 유의성을 나타냈다.

Study on the Geometrical Properties of Brown Rice on Shape Factors

  • Purpose: This study was conducted to investigate the optimal sorting factors in establishing an efficient sorting technology for brown rice. Methods: The brown rice varieties used in this study were Il Pum, Chu Cheong, Dong Jin, Un Gwang, Nam Pyeong, and Dae An. These were classified into whole grain, unriped grain, and green dead rice. The shape factors were analyzed based on length, width and thickness of the grains. Results: The results revealed that the maximum length among whole grain, unriped grain, and green dead rice was observed in Dae An variety while Chu Cheong variety showed the minimum. Further more, Il Pum brown rice showed the maximum width while Dong Jin variety showed the minimum. In the case of thickness, the maximum was observed in Un Gwang variety and that of the minimum among Nam Pyeong variety for both whole grain and unriped grain. Conclusions: The length and width can be used as determinants in sorting factors of whole grain and green dead rice, and the thickness can be considered as optimum sorting factor of whole grain and unriped grain.

현미와 백미의 품종간 아미노산 조성 (Amino Acid Composition of Milled and Brown Rices)

  • 본 연구는 표준 경작법으로 재배한 육성 품종 5종(광안, 대안, 대진, 수라, 화성)의 현미와 백미의 품종간 단백질과 아미노산 함량을 분석하고, 단백질의 화학적 질적 가치를 알아보고자 하였다. 단백질 함량은 현미에서는 대진벼(7.98%)>대안벼ㆍ화성벼(7.46%)>수라벼 (7.03%)>광안벼(6.70%) 순이었고, 백미에서는 대진벼(7.52%)>대안벼(6.90%)>화성벼(6.78%)>수라벼(6.64%)>광안벼(6.08%) 순으로 대진벼가 단백질 함량이 다른 품종에 비하여 가장 높았다. 총 아미노산에 대한 총 필수아미노산의 비율을 보면, 현미는 36.8∼38.3%이었으며, 백미는 35.8∼37.7%으로 화성벼가 가장 높았고, 대안벼가 가장 낮았다. 쌀의 모든 품종에서 가장 많이 함유된 아미노산인 Glx(glutamate+glutamine)은 현미에서 900.5∼965.9 mg/g N, 백미에서 1003.3∼1058.5 mg/g N으로 분석되었다. 현미와 백미의 아미노산 함량은 Glx>Asx>arginine>leucine>valine순이었다. A/E비 화학가는 현미에서 수라벼>화성벼>대진벼>대안벼>광안벼 순이었고, 백미에서 수라벼>광안벼>대안벼>대진벼>화성벼 순이었다. A/T비 화학가는 현미에서는 화성벼>대진벼>수라벼ㆍ광안벼>대안벼 순이었고, 백미에서는 광안벼>대진벼>수라벼>화성벼>대안벼 순이었으며, 제 1제한 아미노산은 lysine으로 판정 되었다. Protein score와 amino acid score에서 제 1제 한 아미노산은 lysine이었고, 제 2제 한 아미노산은 threonine, 그리고 제 3제한 아미노산은 isoleucine이었다.

Physicochemical and textural properties of germinated brown rice according to rice varieties

  • Germination is one of the techniques used to enhance the texture properties and nutritional value of the brown rice (BR). Therefore, germinated BR (GBR) has received significant attention during the last decade. Physicochemical and cooking properties of brown rice were examined before and after germination. Germination raised the cooking properties, such as water absorption, expanded volume and soluble solid of cooked BR (brown rice). The texture, measured using tensipresser, was significantly improved by germination. The hardness of cooked BR was decreased by germination, but the GBR was sticker. In RVA, all viscosity value (peak viscosity, break down, set back, and final viscosity) of germinated rice flour was also reduced while gelatinization temperature did not change. Amylose content and amylopectin chain length distribution of BR starch were slightly changed by germination. Overall results revealed that germination was an effective tool to improve texture and cooking properties of BR.

Contents of Phytic Acid and Minerals of Rice Cultivars from Korea

  • To elucidate the contents of phytic acid and minerals in rice produced in Korea, samples of 68 verieties of brown rice and 9 varieties of polished rice were analyzed for phytic acid by colorimetric method, and Ca, Fe, Mg and Zn contents by the atomic absorption spectrophytometer. Selenium was measured by fluorometry. Averages of phytic acid content of the 68 brown rice were 12.6 g/kg, whereas that of the 8 polished rice were 1.83 g/kg on dry matter. Averages of Ca, Fe, Mg and Zn contents on the 68 brown rice were 111, 7.4, 1,068 and 19.1 mg/kg, and those of the polished rice were 45.6, 2.1, 250 and 14 mg/kg, respectively. A averge of selenium content of the brown rice was 38.3 $\mu\textrm{g}$/kg.

품종별 현미의 수화와 취반에 관한 연구 (The Hydration Properties and the Cooking Qualities of Various Brown Rices.)

  • Five-brown-rice-variety, Akibare, Odaebyeo, Taebaegbyeo, Nonglim Na 1 and Hankangchalbyeo, was prepared and examinated the hydration kinetics and the cooking qualities. Before the hydration the L/W ratio of raw Taebaegbyeo was the biggest value among the five brown rices. The water uptake was directly proportional to the square root of soaking time. During the hydration water uptake of high yielding brown rices was bigger than those of traditional brown rices among the nonglutenious varieties but waxy brown rices were not. Generally volume increase constant was directly proportional to the water uptake constant, which were different a little with brown rices was hydrated which was inversed proportional to the water uptake degree. According to the instrumental result using the rheometer of cooked brown rice with increased soaking times that was decreased the hardness and was increased the adhesiveness. The sensory evaluation test indicated that the hardness and the stickiness value of cooked brown rices were proper after 15-hour-soaking time and it was identical result to the instrumental result using the rheometer.

큰눈벼와 일품벼의 발아에 의한 생리활성물질 함량 및 암세포 증식억제활성 변화 (Changes in Phytochemical Content and Antiproliferative Activity of Germinated Geunnun and Ilpum Rice Varieties)

  • 본 연구에서는 현미의 발아를 통해 큰눈벼와 일품벼 두 품종에서의 GABA(${\gamma}$-aminobutyric acid) 함량, 항산화성분 함량 및 암세포 증식억제 활성의 변화를 측정하고자 하였다. 연구 결과 두 품종의 현미는 발아 과정을 통해 GABA 함량이 증가하였으며, 특히 일품벼의 미강층에서 발아 후 가장 큰 증가율을 나타내었다. 항산화 성분의 경우, 큰눈벼에서는 발아 후 polyphenol 함량이 증가하였으나, flavonoid 함량의 경우 두 품종 모두 발아 후 감소하는 것으로 나타났다. 암세포 증식억제활성 측정 결과, 폐암 세포주의 경우 두 품종 모두 현미상태에서는 발아 후 암세포 증식 억제활성이 증가하였으며, 위암 세포주에서 발아 후에 암세포 증식 억제활성이 보다 증가하였다. 따라서 현미의 발아 시 항산화 성분이 용출되는 현상을 방지하는 조건이 확립된다면, 향후 발아현미는 건강기능식품 및 건강보조제 등을 위한 기능성 식품 소재로 이용될 수 있을 것으로 사료된다.
