Application of Cold-Osmotic Dehydration Method for Extending the Shelf Life during Frozen Storage of Filleted and Salted Fishes (염지어(鹽漬漁) 동결저장 중 Shelf life 연장을 위한 저온삼투압탈수법(低溫?透壓脫水法)의 적용)

  • The study was undertaken to extend the shelf life of filleted and salted fishes such as mackerel and jacopever. These filleted and salted fishes were dehydrated by dewatering sheet containing sodium polyacrylate resin at $5{\pm}1^{\circ}C$, wrapped with low density polyethylene film, and then stored at $-18{\pm}2^{\circ}C$. During the frozen storage, the change of brown pigment formation, peroxide value, carbonyl value, drip formation content in the cold-osmotic dehydrated fishes after salt dipping were much lower than those of non-dehydrated ones. Moreover, the proteins and Ca-ATPase in the cold-osmotic dehydrated fishes after salt dipping were more stable than those of non-dehydrated ones during frozen storage. It was supposed that the cold-osmotic dehydration pretreatment processing for filleted and salted fishes was useful in improvement of the frozen storage stability.

Comparison of Soyasapogenol A, B Concentrations in Soybean Seeds and Sprouts

  • Soybean seeds contain many biologically active secondary metabolites, such as proteins, saponins, isoflavones, phytic acids, trypsin inhibitors and phytosterols. Among them, saponins in soybeans have attracted considerable interest because of their health benefits. Soyasaponin A and B are the most abundant types of saponins found in soybeans along with soyasapogenol (aglycone), which is a precursor of soyasaponin. The main purpose of this experiment was to determine the concentration of soyasapogenol in soybean seeds and sprouts as a function of seed size, usage, seed coat color and seed cotyledon color. The 79 Korean soybean varieties were cultivated at Yesan of Chungnam in 2006 for the analysis of soyasapogenol using HPLC with Evaporative Light Scattering Detection (ELSD). The total average concentration of soyasapogenol was $1313.52{\mu}g\;g^{-1}$ in soybean seeds and $1377.22{\mu}g\;g^{-1}$ in soybean sprouts. Soybean sprouts were about 5% higher than soybean seeds in average total soyasapogenol concentration. In the process of sprouting, the average soyasapogenol A content decreased by approximately 1.6%, but soyasapogenol B and total soyasapogenol increased by 8.31% and 4.88%, based on the content of soybean seeds. When classified according to the size of seeds, the total soyasapogenol concentration of soybean seeds were not significantly different (p<0.05) On average, small soybean seeds were increased by as much as $103.14{\mu}g\;g^{-1}$ in sprouting process. As a function of the use of the seeds, The total soyasapogenol in soybean seeds were significantly different (p<0.05). While, the soybean sprouts were not significant different (p<0.05). Altogether, sprout soybean seeds show the greatest change in content during the germination process. When seeds with different coat colors were compared, the total soyasapogenol concentration of soybean with yellow seed coats ($1357.30\mu g\;g^{1}$) was slightly higher than that of soybean with black ($1260.30{\mu}g\;g^{-1}$) or brown ($1263.62{\mu}g\;g^{-1}$) seed coats. For the color of the cotyledon, the total soyasapogenol concentration was significantly increased in green cotyledon during the germination and seedling process. The results of this study suggest the functional characteristics of soybeans through quantitative analysis of soyasapogenol. In addition, the concentration of soyasapogenol exhibited a change during the germination process, which was evaluated by the nutritional value of the soybean sprouts.

Electrophoretic Patterns of Sarcoplasmic Proteins in Mid-West Korean Sea Fishes by Thin Layer Polyacrylamide Gel Isoelectrofocusing (등전점 전기영동에 의한 서해산 해산어의 근형단백질의 전기영동에 관한 연구)

  • In order to identify hard distinct 12 fish species(shiba shrimp Metapenaeus joyneri, fleshy shrimp Penaeus orientalis, ridgetail prawn Palaemon carinicauda, yellow croaker Pseudosciaena manchurica, croaker Niber albiflora, Colichthyes fragilis, brown sole Limanda herzensteini, frog flounder Pleuronichthys cornutrs, Areliscus rhomaleus, stone flounder Kareius bicoloratus, harvest fish Pampus argenteus, flag fish Goniistius zonatus) by seeing with naked eye in Kunsan coastal area, sarcoplasmic protein in the supernatant was used for isoelectric focusing. For getting supernatant, fish muscle tissue was blended with two times deionized water and centrifuged (at $4^{\circ}C$, 12,000rpm for 15min). Isoelectric focusing of sarcoploasmic protein carried out on a LKB Multiphor II using polyacrylamide gel plate (2mm thickness, pH $3.5~10^{\circ}C$, pH 5~8 gradient, at $10^{\circ}C$ for 1.5, 3 hours). In case of uncertain protein pattern, pH gradient was modified to narrow pH gradiet, and excuted 2-D electrophoresis using conventional polyacrylamide gel electrophoresis. Most of fishes except yellow croaker and Collichthyes fragilis were distingushed by isoelectric focusing. The protein maps of 2-D electrophoresis for analyzing two protein bands at aimilar positions(pH 5, 6) between the two fish species showed the diffeences of the estimated molecular weights, 11,700(pH5.0) and 87,000(pH6.0)

Effects of Ethanol Extract of Sargassum horneri on Adipocyte Differentiation and Adipogenesis in 3T3-L1 Preadipocytes (괭생이모자반 에탄올 추출물이 3T3-L1 지방전구세포의 분화 및 adipogenesis에 미치는 영향)

  • Sargassum horneri (Turner) C. Agardh is a marine brown algae widely distributed in the North Pacific Ocean. It is known for its anti-inflammatory and anti-atopic effects. In this study, we determined the effects of ethanol extract of Sargassum horneri (Turner) C. Agardh (EESH) on anti-obesity activities in 3T3-L1 preadipocytes. Our results indicated that treatment with EESH decreased the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by oil red O staining. The concentrations of cellular triglycerides were also reduced in 3T3-L1 cells after treatment with EESH. Triglyceride content was inhibited by 13%, 16%, and 23% after treatment with 250, 500, and $1,000{\mu}g/ml$ of EESH in 3T3-L1 cells, respectively. Western blotting analysis showed that EESH suppressed adipogenic transcription factor expression in a dose dependent manner. Specifically, it suppressed cytidine-cytidine-adinosine-adenosine-thymidine (CCAAT) /enhancer binding proteins $(C/EBP){\alpha}$, $C/EBP{\beta}$ and peroxisome proliferator-activated receptor $(PPAR){\gamma}$. This indicated that EESH could control the expression of adipogenic transcription factors and inhibit the differentiation of adipocytes. Taken together, these findings demonstrated that EESH showed anti-obesity effects and could have potential uses in the field of nutraceuticals.

  • Obesity is one of the metabolic diseases caused by excessive differentiation and accumulation of adipose tissue due to an imbalance between energy intake and consumption. In this study, we investigated the anti-obesity effect of SliMax, a natural-derived 6 compounds mixture, by using 3T3-L1 cells. As a result, SliMax showed the inhibitory effect on adipocyte differentiation through down-regulation of the PPARγ and C/EBPα expression, which are known to regulate the late adipogenesis stage. In the process of lipolysis on differentiated 3T3-L1 cells, SliMax accelerated decomposition of large-sized unilocular lipid droplet into numerous small-sized multilocular lipid droplets through up-regulation of the expression of lipolysis-related proteins ATGL and HSL. Finally, in order to confirm the effect of SliMax on induction of brown adipocyte, the expression of UCP-1 and the amount of mitochondria were confirmed by immunofluorescent staining, and as a result, SliMax increased the expression of UCP-1 and the amount of mitochondria in fat cells. Taken together, those results suggest that SliMax, a naturally-derived mixture, have a potential to be anti-obesity agent through exerting inhibitory effect on the formation of lipid droplet by suppression of adipogenesis and stimulation of lipolysis, and browning effect associated with generation of heat energy and energy consumption.

Non-Destructive Scientific Analysis of the Gold Fabric Excavated of Cheongsong Shim's Grave (청송심씨 묘에서 출토된 금직물의 비파괴 과학적 분석)

  • Using non-destructive analytical methods, we identified the material characteristics of two gold fabric artifacts excavated from the Cheongsong Sim clan (Bugeum Wonsam, Jikgeum Chima), including the artifact condition, fiber type, surface contamination, and metallic threads. We found that the artifacts were buried and had turned brown; thus, we were unable to determine their original color. The fiber type was determined to be silk from cocoons, based on scanning electron microscopy, Fourier transform infrared (FT-IR) analyses of Amide I, II, III, and IV peaks, and color reactions Further, the FT-IR and X-ray fluorescence (XRF) analyses identified the white and black stains as natural resin hydrolyzed substances, such as lipids and proteins, that occurred as microbial decomposition due to body decay. Finally, the XRF analyses identified the thin gold layer of the metallic yarn as gold (Au). According to the FT-IR data and the color reaction to the metallic yarn medium, the adhesive component of the medium was a product of-Amides I, II, III, and 3000 cm-1 within Amides A and B (an animal type), respectively. Thus, the medium was identified as Hanji (Korean paper), which is made from domestically produced Broussonetia kazinoki fibers.

Analysis of the Age-Dependent Change in the Blood Chemical Values from Hyline Brown Layer Chickens under Field Condition (하이라인 갈색 산란계의 일령별 혈액 화학치 변화 분석)

  • To evaluate the physiological status of laying flocks, the blood chemistry values were measured and analyzed in various ages under different feeding conditions. Total 671 birds from 48 Hyline brown hens flocks from 13 different poultry farms were bled at the ages of day(s) 1, 11, 21, 50, 80, 120, 180, 240, 300, 400, and 500. The 17 blood chemistries including glucose, lipids, proteins, enzymes, electrolytes and metabolic by-products were measured with an autoanalyzer. Blood glucose showed the highest at the hatching day not relate with the dietary carbohydrates and energy, but tended to decrease during the rest of growth stage in hens. Total blood protein, albumin and globulin increased depending on the ages even though dietary protein was decreased. Blood lipid was greatly changed at different growth stages. Cholesterol was the highest at hatching period and maintained consistently until the 120 days of age. It was increased in birds after 180 days of age. HDL was also highest in hatchery, but decreased greatly after 180 days of age. However, TG was the lowest at one day old, but was increased up to 10 times after 180 days of age compared to that of one day old. The enzyme activities were different. AST, ALT, and GGT showed comparatively contained consistently, whereas amylase was slowly decreased. Blood P, Na, K and Cl showed consistency, but Ca content was increased upto two times of the one day of age. The results from this study showed that the blood chemistry values were affected by the general metabolic status of the host with ages not by feeding conditions. Further, the standard data of age-dependent blood chemistry values in the laying flocks were obtained, which can be utilized for early detection of the changes in the physiological status occurred by the infectious or metabolic diseases. The results of these analyses seemed to be useful to increase the productivity of laying flocks through rapid and proper veterinary medical treatments.

Induction of Growth Inhibition and Apoptosis in Human Cancer Cells by a Brown Algae Extract (갈조류 추출물에 의한 인간 암세포 성장 억제 및 세포 사멸 유도)

  • In this study, we investigated the effects of Undaria pinnatifida (UP), Petalonia binghamiae (PB) and Punctaria latifolia (PL) extracts on the inhibition of proliferation and apoptosis in human gastric and breast cancer cells. AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml concentrations of the extracts to determine their anti-proliferative effects, using the MTT assay. The UP, PB and PL extracts inhibited proliferation of AGS, MDA-MB-231 and SK-BR-3 cells in a dose-dependent manner, and the PL extract was found to be the most effective. DAPI staining was also performed to determine changes in the cell nucleus. Further, the AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml of only the PL extract. DAPI staining showed increased chromatin condensation, which is indicative of apoptosis, in the 200 μg/ml group. The expression of the Bax, Bcl-2, and PARP proteins in AGS, MDA-MB-231 and SK-BR-3 cells treated with the PL extract was also determined by western blot analysis. The expression of Bax (a pro-apoptotic protein) and cleaved-PARP was increased, whereas the expression of Bcl-2 (an anti-apoptotic protein) was decreased compared with the control. These findings indicate that the PL extract may have potential as an alternative anticancer drug and nutraceutical.

Effects of Myelophycus Simplex Papenfuss Methanol Extract on Adipocyte Differentiation and Adipogenesis in 3T3-L1 Preadipocytes (바위수염 메탄올 추출물이 3T3-L1 지방전구세포의 분화에 미치는 영향)

  • Myelophycus simplex Papenfuss is distributed over the northern Pacific and southern coast of Korea, and is a member of the brown algae family. The objective of this study was to investigate the effect of M. simplex methanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes. Treatment with M. simplex methanol extract significantly suppressed terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by Oil Red O staining. Also, the M. simplex methanol extract significantly suppressed the triglyceride content of 3T3-L1 preadipocytes in a dose-dependent manner. Treatment with 300 and $500{\mu}g/ml$ of M. simplex methanol extract caused a 42% and 76% reduction in lipid droplet content, respectively. In order to understand the anti-adipogenic effects of M. simplex methanol extract, the changes in the expression of several adipogenic transcription factors, including peroxisome proliferator-activated receptor (PPAR) ${\gamma}$-cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding protein (C/EBP) ${\alpha}$ and ${\beta}$, were investigated using immunoblotting. M. simplex suppressed the expression of $PPAR{\gamma}$, $C/EBP{\alpha}$, and $C/EBP{\beta}$ proteins compared with control. Therefore, the results of this study suggest that M. simplex methanol extract inhibits adipocyte differentiation and thus may have applications as a potential source for an anti-obesity functional food agent.

Effect of Feeding Colored Barley and Whole Crop Barley on Performance, Egg Quality and Blood Composition of Laying Hens (유색보리 및 청보리의 급여가 산란계의 생산성과 계란의 품질 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effect of feeding colored barley (CB) and whole crop barley (WCB) on performance, egg quality and blood composition of laying hens. A total of 280, 35 weeks old Brown Nick laying hens were allocated into the individual cage for the period of four weeks. Experimental diets contained 2,750 kcal/kg ME and 16% CP, respectively. The treatments consisted of 5, 10, 15% CB and WCB as feed ingredients with control and there were five replications in each treatment. Higher egg production was noticed at 10% level of CB and 5% level of WCB in the diets, respectively. Feed intake was higher in WCB and CB treatments compared to those of the control (P<0.05), but the feed conversion was not different. Yolk color tended to increase both in WCB and CB groups. Haugh unit seemed to increase WCB treatments. When the CB and WCB were fed at 15% level, blood total proteins, albumin, total cholesterol and triglyceride contents were decreased than that of the control birds, but total cholesterol and HDL-cholesterol tended to be higher with the increasing level of WCB. As results, no significant differences were observed in performance and egg quality with different levels of dietary WCB and CB. Thus, CB and WCB can be substituted as a feed ingredient up to 15% level in the laying hens' diet. However, further studies are required by feeding more than 15% levels of CB and WCB in the diet of laying hens.
