Selection Procedure for DVT and Cis - abienol in Burley Tobacco (고 DVT 및 cis-abienol 함유 버어리종 계통 선발)

  • This study was carried out to obtain the basic information for breeding of aromatic burley tobacco, and to select the breeding lines with higher DVT (diterpenes $\alpha$-and $\beta$-4, 8, 13-duvatriene-1, 3-diols) and cis-abienol (Z-labda-12, 14-diene-8$\alpha$-ol) concentrations. The parental genotypes used to develop the Fl and Fl generations were Burley 21, Ky 17, male-sterile(MS) NC 107, TC 612, TC 613, Va 509 and Tl 1068. To estimated the modes of inheritances of DVT and cis-abienol known as the flavor and aroma compounds, the method of thin - layer chromatography (TLC) was used. It was considered that TLC procedure being relatively simple and reproducible in the selection of the breeding lines with higher DVT and cis-abienol. DVT was present in all of parents except MS NC 107, but cia-abienol presented only in Tl 1068. It was estimated from the TLC analysis of parents and Fl's that DVT and cis-abienol were each inherited by dominant gene or genes. But it was less clear to determine the presence or not of sucrose ester(SE) on the TLC plate. The eleven plants which had burley leaf color and also estimated to have DVT and cis-abienol were selected in the F2 population of three combinations.

Inheritance of Resistance to Nuclear Polyhedrosis Virus in Silkworm, Bombyx mori

  • Inheritance pattern of resistance to Bombyx mori nuclear polyhedrosis virus (BmNPV) was studied in an Indian silkworm stock TX by single back-cross test method. The resistant parent [TX], susceptible parent [HM], their Fl, F2, and Fl progeny back-crossed to TX [BC(R)] and HM [BC(S)] were inoculated per os with a fixed concentration of BmNPV($0.5{\times}10^{th} PIB/ml$) on the first day of second stadium. The cumulative mortality was recorded until day $10^{\times}$ post-inoculation. The results show that the resistance to BmNPV in TX fellow mono Mendelian inheritance pattern. The resistance dominated over the susceptibility at Fl. At F2, the resistant and susceptible offspring segregated in 3:1 ratio whereas at BC(S), the resistant and susceptible offspring segregated in 1:1 ratio. The response of BC(R) was more or less like the resistant parent TX which confirms the involvement of a major dominant gene conferring resistance to BmNPV in TX. The possible mechanism of inheritance of resistance in TX is discussed.

Conditions for Reintroduction of Captive-Bred Endangered Birds: A Review

  • Reintroduction programs have been promoted across the world to recover and rehabilitate endangered birds through ex situ captive breeding and releasing to the nature. The ultimate aims of these reintroduction programs are recovery of sustainable populations in natural habitats. To ensure the success of the reintroduction programs, it is necessary to examine the followings in order to increase the success rate of releasing along with the development of captive breeding techniques: 1) Adequate habitat provision, 2) adaptability of captive-bred individuals, 3) survivability of released individuals, and 4) social interest and will. Before releasing captive-bred individuals, it should be reviewed whether there are habitats in which the limiting factor is removed, and assessed their long-term safety. The quantity and quality of the released individuals must be considered to increase the chances for mate selection, maintain genetic diversity, and acquire the ability to adapt to the wild. The release method must be decided in consideration of characteristics of the target habitats and individuals, and rational means such as careful observation, evaluation, and feedback must be provided throughout the release process. For the long-term success of recovery projects, social awareness, sustainable support, and related experts are needed. Satisfying these criteria can help to increase the success rates of reintroduction programs. For the bird reintroduction program in the future, the feasibility of the methods and procedures must be closely reviewed before starting.

National genomic evaluation of Korean thoroughbreds through indirect racing phenotype

  • Objective: Thoroughbred horses have been bred exclusively for racing in England for a long time. Additionally, because horse racing is a global sport, a healthy leisure activity for ordinary citizens, and a high-value business, systematic racehorse breeding at the population level is a requirement for continuous industrial development. Therefore, we established genomic evaluation system (using prize money as horse racing traits) to produce spirited, agile, and strong racing horse population Methods: We used phenotypic data from 25,061 Thoroughbred horses (all registered individuals in Korea) that competed in races between 1994 and 2019 at the Korea Racing Authority and constructed pedigree structures. We quantified the improvement in racehorse breeding output by year in Korea, and this aided in the establishment of a high-level horse-fill industry. Results: We found that pedigree-based best linear unbiased prediction method improved the racing performance of the Thoroughbred population with high accuracy, making it possible to construct an excellent Thoroughbred racehorse population in Korea. Conclusion: This study could be used to develop an efficient breeding program at the population level for Korean Thoroughbred racehorse populations as well as others.

Analysis of energy-saving effects of recirculation aquaculture system using seawater source heat pumps and solar power generation (해수 열원 히트펌프와 태양광 발전을 이용한 순환여과식 양식장의 에너지 절감 효과 분석)

  • This study focuses on analyzing the energy-saving effects of the recirculation aquaculture system using seawater source heat pumps and solar power generation. Based on the thermal load analysis conducted using the transient system simulation tool, the annual energy consumption of the recirculation aquaculture system was analyzed and the energy-saving effects of utilizing the photovoltaic system was evaluated. When analyzing the heat load, the sea areas where the fish farms are located, the type of breeding tank, and the circulation rate of breeding water were taken into consideration. In addition, a method for determining the appropriate capacity for each operation time was examined when applying the energy storage system instead of the existing diesel generator as an emergency power, which is required to maintain the water temperature of breeding water during power outage. The results suggest that, among the four seas considered, Jeju should be estimated to achieve the highest energy-saving performance using the solar power generation, with approximately 45% energy savings.

  • Matting made of wools has a history which gose back even before B.C.7000, the time which is believed to be the starting point of the fabrication, production and usage of wools by the nomads in the Western and Central Asia who had made the living by breeding sheeps. The purpose of this thesis is to investigate the function of wool matting in Asia. The research period limited to 9C A.D. When classified by the method of production of wool matting, the pile method and felt method have been widely used by the nomads in Western Asia and the cattle breeding nomads in Central Asia consecutively. From ancient times, wool matting has been considered to be one of the necessities by the nomads for living in the wilderness, and even at present, continued to be used by the people both for the purpose of everyday use and decoration. Other than fore-mentioned purposes, wool matting have function that is expressed the incantation, authorization and hierarchy, emotion and desire, and cultural exchange between nations. In addition, wool matting had also been used as a mark to show ownership and for military purposes. Even a simple wool matting had a different symbolism and function by different region and people throughout Asia. However, by finding and studying further abut what wool matting had symbolized and how it had been used, the tradition and history of wool matting could continue to attract the interests which will make the tradition to continue. And also, in order for the tradition to continue, the utmost efforts to innovate and produce better quality and design wool matting to fulfil the needs of modern times are truly required.

Virus-induced Gene Silencing as Tool for Functional Genomics in a Glycine max

  • Virus-induced gene silencing (VIGS) is a recently developed gene transcript suppression technique for characterizing the function of plant genes. However, efficient VIGS has only been studied in a few plant species. In order to extend the application of VIGS, we examined whether a VIGS vector based on TRV would produce recognizable phenotypes in soybean. Here, we report that VIGS using the Tobacco rattle virus (TRV) viral vector can be used in several soybean cultivars employing various agro-inoculation methods including leaf infiltration, spray inoculation, and agrodrench. cDNA fragments of the soybean phytoene desaturase(PDS) was inserted into TRV RNA-2 vector. By agrodrench, we successfully silenced the expression of PDS encoding gene in soybean. The silenced phenotype of PDS was invariably obvious 3 weeks after inoculation with the TRV-based vector. Real-time RT-PCR analyses showed that the endogenous level of GmPDS transcripts was dramatically reduced in the silenced leaf tissues. These observations confirm that the silenced phenotype is closely correlated with the pattern of tissue expression. The TRV-based VIGS using agrodrench can be applied to functional genomics in a soybean plants to study genes involved in a wide range of biological processes. To our knowledge, this is the first high frequency VIGS method in soybean plants.

Prevalence of heartworm infection among dogs on breeding farms in southern Gyeongnam area (경남 남부지역 사육견에 대한 심장사상충 감염률 조사)

  • This study was attempted to survey the prevalence of heartworm infection among 253 dogs (male 167, female 86) on the 16 breeding farms in southern Gyeongnam area in May to December 2007. The results obtained from this experiment were summarized as follows; Fifty seven (22.5%) of the 253 examined dogs were heartworms-antigen positive, while twenty five dogs were Microfilaria positive by the modified Knott's method. The regional infection rates were of 25% in Tongyeong and 21% in Goseong, and on the breeding environment aspects, infection rates were lower in rural area than urban area farms. The infection rates were higher in female (26/86, 30.2%) than male dog (31/167, 18.6%). The infection rates by breeding size of farms were 31% (11/35) at <20 head, 26%(35/135) at 20~50 head and 13% (11/82) at >50 head. The infection rates of heartworm in examined dogs at the age of <2, 2~4 and >5 were 10.7% (8/75), 26% (40/154) and 37.5% (9/24), respectively. In the microfillaria-infected 25 dogs, identified Dirofilaria immitis of 21 (84%) using PCR. The dogs infected with heartworm showed the increased number of eosinophil.

Genetic Variability and Relationships of Native Japanese Chickens Assessed by Microsatellite DNA Profiling - Focusing on the Breeds Established in Kochi Prefecture, Japan -

  • Blood samples were collected from eight native Japanese breeds of chickens (Miyadi-dori, Ohiki, Onaga-dori, Shoukoku, Tosa-Jidori, Tosa-Kukin, Toutenkou and Uzurao) and two foreign breeds of chickens (White Leghorn and Rhode Island Red) to examine the genetic variability and relationships among the breeds by using a microsatellite DNA technique. Except for the Shoukoku breed, the other Japanese chicken breeds all originate from Kochi Prefecture. Ohiki, Onaga-dori, Tosa-Jidori, Toutenkou and Uzurao are fancy fowl, and Miyadi-dori and Tosa-Kukin are utility fowl. Among the fancy fowl, Ohiki, Onaga-dori, and Toutenkou males have thick and long feathers in the saddle and tail. Genetic variabilities of the 20 microsatellites examined, varied depending on the breed: the mean number of alleles per locus ranged from 2.05 (Miyadi-dori) to 3.90 (Rhode Island Red); proportion of polymorphic loci ranged from 0.75 (Miyadi-dori) to 1.00 (Rhode Island Red, Shoukoku and Uzurao); and mean expected heterozygosity ranged from 0.330 (Miyadi-dori) to 0.607 (Rhode Island Red). Unique microsatellite alleles were detected in each breed. Using the neighbour-joining method, phylogenetic trees were constructed based on the genetic distances of D$_{A}$ and D$_{ST}$. Among the breeds originating from Kochi Prefecture, fancy and utility breeds belonged to different clusters. Among the fancy breeds, those having thick and long feathers in the tail and saddle showed a close genetic relationship to the Shoukoku breed, which also has thick and long feathers in the tail and saddle.

Evaluation of Optimum Genetic Contribution Theory to Control Inbreeding While Maximizing Genetic Response

  • Inbreeding is the mating of relatives that produce progeny having more homozygous alleles than non-inbred animals. Inbreeding increases numbers of recessive alleles, which is often associated with decreased performance known as inbreeding depression. The magnitude of inbreeding depression depends on the level of inbreeding in the animal. Level of inbreeding is expressed by the inbreeding coefficient. One breeding goal in livestock is uniform productivity while maintaining acceptable inbreeding levels, especially keeping inbreeding less than 20%. However, in closed herds without the introduction of new genetic sources high levels of inbreeding over time are unavoidable. One method that increases selection response and minimizes inbreeding is selection of individuals by weighting estimated breeding values with average relationships among individuals. Optimum genetic contribution theory (OGC) uses relationships among individuals as weighting factors. The algorithm is as follows: i) Identify the individual having the best EBV; ii) Calculate average relationships ($\bar{r_j}$) between selected and candidates; iii) Select the individual having the best EBV adjusted for average relationships using the weighting factor k, $EBV^*=EBV_j(1-k\bar{{r}_j})$ Repeat process until the number of individuals selected equals number required. The objective of this study was to compare simulated results based on OGC selection under different conditions over 30 generations. Individuals (n = 110) were generated for the base population with pseudo random numbers of N~ (0, 3), ten were assumed male, and the remainder female. Each male was mated to ten females, and every female was assumed to have 5 progeny resulting in 500 individuals in the following generation. Results showed the OGC algorithm effectively controlled inbreeding and maintained consistent increases in selection response. Difference in breeding values between selection with OGC algorithm and by EBV only was 8%, however, rate of inbreeding was controlled by 47% after 20 generation. These results indicate that the OGC algorithm can be used effectively in long-term selection programs.
