Industrial strain Improvement is concerned with developing or modifying microorga-nisms used In production of commercially important fermentation products. The aim is to reduce the production cost by improving productivity of a strain and manipulating specific cilarafteristic such as the ability to utilize cheaper raw materials or resist bacteriophages. The traditional empiri-cal approach to strain improvement is mutation combined with selection and breeding techniques. It is still used by us to improve the productivity of organisms in amino acids. organic acids andenzymes production. The breeding of high L-lysine-producing strain Au112 is one of the outstanding examples of this approach. It is it homoserine auxotroph with AEC, TA double metabolicanalogue resistant markers. The yield reaches 100g/1. Resides, the citric acid-producing organism Aspergillus nuger, Co827, its productivity reches the advanced level in the world, is also the result of a series mutations expecially with Co Y-radiation. The thermostable a-amylaseroducing strain A 4041 is the third example. By combining physical and chemical multations. the strain ,A 4041becomes an asporogenous, catabolite derepressed mutant with rifamycin resistant and methionine, arginine auxotroph markers. The a-amylase activity reaches 200 units/ml. The fourth successful example of mutation in strain improvement is the glucoamylase-producing strain Aspergillus nigerSP56 its enzyme activity is 20,000 units/ml, 4 times of that of the parental strain UV_11. Recently recombinant DNA approach Provides a worth while alternative strategy to Industrial strain improve-ment. This technique had been used by us to increase the thermostable a-amylase production and on some genetic researches.
Genus Taraxacum has been widely used as a folkloric medicine for treatment of diverse diseases. The genetic diversity and relationship among 32 accessions belonging to five Taraxacum species (T. mongolicum T. coreanum, T. coreanum var. flavescens, T. officinale and T. laevigatum) which collected from field, mountain, island and seaside of Korea were evaluated using ISSR markers. A total of 142 ISSR loci detected in the overall species were all polymorphic loci (100%) and interspecies polymorphisms obtained from Korean native and naturalized species were 98.2% and 94.5%, respectively. The genetic similarity matrix (GSM) among 32 accessions ranged from 0.025 to 0.860 with an average of 0.303. According to the clustering analysis, the Korean native species and naturalized species were divided two major clusters. In addition, the different species were divided into independent groups except for the T. coreanum and T. coreanum var. flavescens, and all the 32 accessions could be classified into 7 categories. The study findings indicate that Taraxacum accessions have a high genetic diversity and the dandelion accessions as breeding materials can be effectively utilized for the improvement of Taraxacum breeding.
'Blue Edge' (Hosta minor) was induced by ethyle methane sulfonate (EMS) to obtain mutants. Among induced leaf variegated hosta plants, some with deep-green color pattern on the edge of the leaves was selected. They were cultivated via vegetative propagation. Assessment of botanical characteristics was conducted for three years since 2003. The major characteristic of these cultivars is that they maintain their deep-green color on the edge of the leaves all year round. These cultivars are useful as materials for pot plant or as ground cover plant.
Hosta 'Yellow Tiara' was induced using methyle methane sulfonate (MMS) to obtain mutagens. Among the induced leaf-variegated hosta plants, some with yellow-green leaves were selected. They were cultivated via vegetative propagation. Assessment of the botanical characteristics was conducted for three years since 2004. The major characteristic of these cultivars is that they maintain their yellow-green leaves uniformly all year round. These cultivars are useful as materials for pot plants or as ground cover.
Recently, giant embryonic rice and functional rice food are preferred by more consumers, which are attributed to the fact that the embryo has high concentrations of essential amino acids, fatty acids, and vitamins relative to other parts of rice grains. In this report, the heredity and stability of giant embryo mutations in successive generations were analyzed regarding a giant embryonic line, 'P47', induced by T-DNA insertion and a $F_2$ population from a cross between 'P47' and 'Junam'. The mutant lines with increases of 1.5, 1.7 and 1.8 times on embryo length, width and 100-embryo weight to those of the control showed stable inheritance across three generations. The continuous frequency distributions of embryo size in the $F_2$ population showed that the embryo size is a quantitative trait of polygene controlled. In addition, wide range of transgressive segregations of six traits affecting embryo size confirmed exchange of genetic materials and recombination between genes controlling embryo size. Five giant embryo mutant lines selected from the $F_2$ population will be used for artificial selection and improvement of giant embryonic varieties.
Rice consumption per capta, in South Korea, has been decreased dramatically, owing to the changes of living patterns. Because of not only the major energy source of Korean people but also major income source of Korean farmers, diversifying end-use-quality of rice has been demanded. To the context, 'Suweon 472', a high yielding and early mature japonica line and released as 'Namilbyeo' to framers in 2002, was treated with a chemical mutagen, Sodium Azide to find endosperm mutant types. A total of nine endosperm mutat lines, including five waxy, one dull, two floury, and one white core type, were identified from the 3,542 mutatagen treated lines. Amylose contents, iodine reaction, disintegration in alkali solution, gelatinization in urea solution and amylogram properties of those nine endosperm mutant lines were evaluated to address the possibility as new genetic materials for diversifying rice quality of Korean japonica cultivars. All embryo mutants were clearly differentiated from their wild type, 'Suweon 472', in terms of physic-chemical properties evaluated. The endosperm mutant lines would be very useful in expanding untiliztation of rice through opening new rice markets of processed foods from Korean japonica rice.

Amaranth is a nutritious and broadly adapted seed crop in high demand around the world. A preliminary approach for understanding the genetics of amaranth resources entails a morphologic characterization, which can provide the basis for breeding the first variety in Korea, leading to satisfying the needs of farmers and consumers. Therefore, this study aimed to evaluate the phenotypic characteristics of ten genetic amaranth accessions for the selection of outstanding accessions in terms of yield and grain quality. A randomized complete block design was used, with fifteen replications for each accession under field conditions. Five quantitative and three qualitative descriptors were evaluated with descriptive analysis. The results showed that the accessions with plant heights smaller than the average (>112.7 cm) presented lower yields and smaller seed sizes, thus decreasing the grain quality. The cluster analyses established groups of accessions with good yields (>30.1 g of seeds per plant) and stable morphological characteristics. Based on yield and morphological descriptors, the proposed selection index indicated four accessions as potential parents for amaranth breeding programs in Kora.
Kimchi cabbage (Chinese cabbage), radish and Cabbage are major Brassicaceae vegetables in Korea. Especially, we can easily develop whole plant from one microspore in Kimchi cabbage. To develop clubroot resistant doubled-haploid (DH) inbred lines, we pollinated a clubroot resistant turnip of 'IT 033820' with a Kimchi cabbage (Chinese cabbage) inbred of 'BP 079'. More than 85 DH inbred lines were developed from this combination. We screened about 400 materials including these DH inbred lines, commercial cultivars and breeding materials during 3 years using hydroponic system after inoculating single spore isolation race 4(SSI-04) inoculate. One inbred line derived from this combination selected as clubroot resistant and registered as 'Wonkyo20036ho'. We inoculated 26 DH inbred lines derived from 'Zoong-baek 2ho' using SSI-4, the percent of resistant plants varied from 0 to 83%. However the horticultural traits of highly resistant DH inbred line was poor. Thus we selected one DH line showing 77% resistant with yellow inner leaf and maid good head, was registered as 'Wonkyo20034ho'. Another DH inbred line derived from Korean variety of 'Wol-dong' showing 86% resistant was registered as 'Wonkyo20037ho'. Other DH inbred lines were derived from Chinese cultivar of 'Choon-hi-go-hang-wang' and 'Hwang-shim-zo48' showed 80 and 71% resistant, respectively, was also selected for registration. Even though DH inbred lines derived from turnip showed highly resistant to SSI-04 and provincial inoculate, they showed poor characteristics in horticultural traits. However, commercial seed companies showed interesting for adapting these DH inbred lines in commercial breeding.
Background: In order to prepare for the regulation about the limit of manure production, the status of manure management and the amount of nutrients in the compost discharged from Hanwoo breeding farm shall be known. This study aimed to find the practical amount of nutrients (volatile solids, VS; total nitrogen, T-N; total phosphorus, T-P) in manure, and compost samples collected from 40 Hanwoo breeding farms and the loss of the nutrients was calculated during the composting period, which supports to develop nutrient loading coefficients (NLCs) for each nutrient. Results: Although the addition of bedding materials for composting caused the increase of the VS amount before composting, the comparison of VS, N, and P amounts in between manure and compost showed the lower VS by 4 % as well as T-N and T-P amounts by 69 and 40 %, respectively, of which values were corresponded with the NLCs of 0.96, 0.31, and 0.60 for VS, N, and P, respectively, based on the questionnaire, and sample analyses. Considering with the environmental impacts including land application from Hanwoo manure, P loss should be zero before and after composting. In this regard, nitrogen loss of 50 % occurs and VS was increased by 30 %. In addition, feasible cases for the calculations based on the notification from Ministry of Environment were compared with this study. Conclusions: The development of NLCs from Hanwoo manure in this study implies that the loss of nutrients in manure occurs during the composting or storing period. The mass balances of N and P from livestock manure to land application may be overestimated over the practical values. It is necessary to build up the database about each livestock category other than Hanwoo.
