There are many joint fissures distributed in the engineering rock mass. In the process of geological history, the underground rock mass undergoes strong geological processes, and undergoes complex geological processes such as fracture breeding, expansion, recementation, and re-expansion. In this paper, the damage-stick-slip process (DSSP), an analysis model used for rock mass failure slip, was established to examine the master control and time-dependent mechanical properties of the new and primary fractures of a multi-fractured rock mass under the action of stress loading. The experimental system for the recemented multi-fractured rock mass was developed to validate the above theory. First, a rock mass failure test was conducted. Then, the failure stress state was kept constant, and the fractured rock mass was grouted and cemented. A secondary loading was applied until the grouted mass reached the intended strength to investigate the bearing capacity of the recemented multi-fractured rock mass, and an acoustic emission (AE) system was used to monitor AE events and the update of damage energy. The results show that the initial fracture angle and direction had a significant effect on the re-failure process of the cement rock mass; Compared with the monitoring results of the acoustic emission (AE) measurements, the master control surface, key blocks and other control factors in the multi-fractured rock mass were obtained; The triangular shaped block in rock mass plays an important role in the stress and displacement change of multi-fracture rock mass and the long fissure and the fractures with close fracture tip are easier to activate, and the position where the longer fractures intersect with the smaller fractures is easier to generate new fractures. The results are of great significance to a multi-block structure, which affects the safety of underground coal mining.
The effects of three different feeding systems on beef cattle production performance, rumen fermentation, and rumen digesta particle structure were investigated by using 18 Limousin (steers) with a similar body weight ($575{\pm}10kg$) in a 80-d experiment. The animals were equally and randomly divided into three treatment groups, namely, total mixed ration group (cattle fed TMR), SI1 group (cattle fed concentrate firstly then roughage), and SI2 group (cattle fed roughage firstly then concentrate). The results showed that the average daily gain was significantly higher in cattle receiving TMR than in those receiving SI1 and SI2 (p<0.05). Consumption per kg weight gain of concentrate, silage, and combined net energy (NEmf) were significantly decreased when cattle received TMR, unlike when they received SI1 and SI2 (p<0.05), indicating that the feed efficiency of TMR was the highest. Blood urea nitrogen (BUN) was significantly decreased when cattle received TMR compared with that in cattle receiving SI1 (p<0.05), whereas there was no difference compared with that in cattle receiving SI2. Ammonia nitrogen concentration was significantly lower in cattle receiving TMR than in those receiving SI1 and SI2 (p<0.05). The rumen area of cattle that received TMR was significantly larger than that of cattle receiving SI1 (p<0.05), but there was no difference compared with that of cattle receiving SI2. Although there was no significant difference among the three feeding systems in rumen digesta particle distribution, the TMR group trended to have fewer large- and medium-sized particles and more small-sized particles than those in the SI1 and SI2 groups. In conclusion, cattle with dietary TMR showed increased weight gain and ruminal development and decreased BUN. This indicated that TMR feeding was more conducive toward improving the production performance and rumen fermentation of beef cattle.
Objective: Jining Grey goat is a local Chinese goat breed that is well known for its high fertility and excellent meat quality but shows low meat production performance. Numerous studies have focused on revealing the genetic mechanism of its high fertility, but its highlighting meat quality and muscle growth mechanism still need to be studied. Methods: In this research, an integrative analysis of the genomics and transcriptomics of Jining Grey goats compared with Boer goats was performed to identify candidate genes and pathways related to the mechanisms of meat quality and muscle development. Results: Our results overlap among five genes (ABHD2, FN1, PGM2L1, PRKAG3, RAVER2) and detected a set of candidate genes associated with fatty acid metabolism (PRKAG3, HADHB, FASN, ACADM), amino acid metabolism (KMT2C, PLOD3, NSD2, SETDB1, STT3B, MAN1A2, BCKDHB, NAT8L, P4HA3) and muscle development (MSTN, PPARGC1A, ANKRD2). Several pathways have also been detected, such as the FoxO signaling pathway and Apelin signaling pathway that play roles in lipid metabolism, lysine degradation, N-glycan biosynthesis, valine, leucine and isoleucine degradation that involving with amino acid metabolism. Conclusion: The comparative genomic and transcriptomic analysis of Jining Grey goat and Boer goat revealed the mechanisms underlying the meat quality and meat productive performance of goats. These results provide valuable information for future breeding of goats.
Objective: Intramuscular fat (IMF) content plays an important role in meat quality. Identification of single nucleotide polymorphisms (SNPs) and genes related to pig IMF, especially using pig populations with high IMF content variation, can help to establish novel molecular breeding tools for optimizing IMF in pork and unveil the mechanisms that underlie fat metabolism. Methods: We collected muscle samples of 453 Chinese Lulai black pigs, measured IMF content by Soxhlet petroleum-ether extraction method, and genotyped genome-wide SNPs using GeneSeek Genomic Profiler Porcine HD BeadChip. Then a genome-wide association study was performed using a linear mixed model implemented in the GEMMA software. Results: A total of 43 SNPs were identified to be significantly associated with IMF content by the cutoff p<0.001. Among these significant SNPs, the greatest number of SNPs (n = 19) were detected on Chr.9, and two linkage disequilibrium blocks were formed among them. Additionally, 17 significant SNPs are mapped to previously reported quantitative trait loci (QTLs) of IMF and confirmed previous QTLs studies. Forty-two annotated genes centering these significant SNPs were obtained from Ensembl database. Overrepresentation test of pathways and gene ontology (GO) terms revealed some enriched reactome pathways and GO terms, which mainly involved regulation of basic material transport, energy metabolic process and signaling pathway. Conclusion: These findings improve our understanding of the genetic architecture of IMF content in pork and facilitate the follow-up study of fine-mapping genes that influence fat deposition in muscle.

Objective: The current research was aimed to profile the transcriptomic picture of the peripheral blood lymphocytes (PBLs) associated with immunity in Chinese Holsteins supplemented orally with coated folic acid during the periparturient period. Methods: The total of 123 perinatal cows were selected for this study and divided into three groups; group A (n = 41, 240 mg/500 kg cow/d), group B (n = 40, 120 mg/500 kg cow/d) and group C (n = 42, 0 mg/cow/d) based on the quantity of folic acid fed. Three samples of PBLs were selected from each folic acid treated group (high, low, and control) and RNA sequencing method was carried out for transcriptomic analysis. Results: The analysis revealed that a higher number of genes and pathways were regulated in response to high and low folic acid supplementation compared to the controls. We reported the novel pathways tumor necrosis factor (TNF) signaling, antigen processing and presentation, Staphylococcus aureus infection and nuclear factor (NF)-kappa B signaling pathways) and the key genes (e.g. C-X-C motif chemokine ligand 10, TNF receptor superfamily member 1A, cluster difference 4, major histocompatibility complex, class II, DQ beta, NF-kappa-B inhibitor alpha, and TNF superfamily 13) having great importance in immunity and anti-inflammation in the periparturient cows in response to coated folic acid treatment. Conclusion: Collectively, our study profiled first-time transcriptomic analysis of bovine lymphocytes and compared the involved cytokines, genes, and pathways between high vs control and low vs control. Our data suggest that the low folic acid supplementation (120 mg/500 kg) could be a good choice to boost appropriate immunity and anti-inflammation as well as might being applied to the health improvement of perinatal dairy cows.

Because of the various patterns of deep-water inrush and complicated mechanisms, accurately predicting mine water inflows is always a difficult problem for coal mine geologists. In study presented in this paper, the water inrush channels were divided into four basic water diversion structures: aquifer, rock fracture zone, fracture zone and goaf. The fluid flow characteristics in each water-conducting structure were investigated by laboratory tests, and multistructure and multisystem coupling flow analysis models of different water-conducting structures were established to describe the entire water inrush process. Based on the research of the water inrush flow paths, the analysis model of different water inrush space structures was established and applied to the prediction of mine water inrush inflow. The results prove that the conduction sequence of different water-conducting structures and the changing rule of permeability caused by stress changes before and after the peak have important influences on the characteristics of mine water-gushing. Influenced by the differences in geological structure and combined with rock mass RQD and fault conductivity characteristics and other mine exploration data, the prediction of mine water inflow can be realized accurately. Taking the water transmitting path in the multistructure as the research object of water inrush, breaking through the limitation of traditional stratigraphic structure division, the prediction of water inflow and the estimation of potentially flooded area was realized, and water bursting intensity was predicted. It is of great significance in making reasonable emergency plans.

Objective: Staphylococcus aureus (S. aureus) is one of the major microorganisms responsible for subclinical mastitis in dairy cattle. The present study was designed with the aim to explore the DNA methylation patterns using the Fluorescence-labeled methylation-sensitive amplified polymorphism (F-MSAP) techniques in a S. aureus-infected mouse model. Methods: A total of 12 out-bred Institute of Cancer Research female mice ranging from 12 to 13 weeks-old were selected to construct a mastitis model. F-MSAP analysis was carried out to detect fluctuations of DNA methylation between control group and S. aureus mastitis group. Results: Visible changes were observed in white cell counts in milk, percentage of granulocytes, percentage of lymphocytes, CD4+/CD8+ ratio (CD4+/CD8+), and histopathology of mice pre- and post-challenge with S. aureus. These findings showed the suitability of the S. aureus-infected mouse model. A total of 369 fragments was amplified from udder tissue samples from the two groups (S. aureus-infected mastitis group and control group) using eight pairs of selective primers. Results indicated that the methylation level of mastitis mouse group was higher than that in the control group. In addition, NCK-associated protein 5 (Nckap5) and transposon MTD were identified to be differentially methylated through secondary polymerase chain reaction and sequencing in the mastitis group. These observations might play an important role in the development of S. aureus mastitis. Conclusion: Collectively, our study suggests that the methylation modification in Nckap5 and transposon MTD might be considered as epigenetic markers in resistance to S. aureus-infected mastitis and provided a new insight into S. aureus mastitis research in dairy industry and public health.

To investigate the genetic diversity of six Chinese indigenous pig breeds in Shandong province (Laiwu Black, Dapulian Black, Licha Black, Yantai Black, Yimeng Black and Wulian Black), explain their genetic relationship and assess their integrity and degree of admixture with three Western commercial breeds (Landrace, Yorkshire and Duroc), 303 individuals from these breeds were genotyped for 26 microsatellite markers. In general, high genetic diversity (observed heterozygosity ranging from 0.5495 to 0.7746) and large breed differentiation ($F_{ST}$ = 0.188) were observed. The indigenous pig breeds in Shandong exhibited consistently higher levels of genetic diversity than the three Western breeds. However, compared with the Western breeds, which have an $F_{ST}$ value of 0.252, the indigenous breeds in Shandong have smaller $F_{ST}$ value of 0.145. The analysis of breed relationship indicated that the six indigenous breeds are classified into two groups. One includes four breeds, Licha, Yantai, Yimeng and Wulian, which have experienced large gene introgression of the Western breeds through progressive crossbreeding as well as gene flow among themselves. The other includes Laiwu and Dapulian, which are less influenced by the Western breeds and other indigenous breeds in Shandong in the recent past. The results show that some measures must be taken to effectively protect these indigenous pig breeds in Shandong.

This experiment was conducted to investigate the effect of fermented spent mushroom substrate (FSMS) on growth performance, blood profile, intestinal microflora and ammonia gas production of feces in broiler chickens. A total of three hundred sixty, 1-day-old male broiler chicks (Ross) were randomly divided into 3 groups with 6 replicates of 20 birds each. The treatments were control (free FSMS), 15% FSMS (basal diet with 15% FSMS) and 30% FSMS (basal diet with 30% FSMS). The final body weight and body weight gains were slightly improved in 30% FSMS than control (P<0.05). Feed intake and feed conversion were significantly improved as compared to those of the control groups. The Leukocytes of blood serum in FSMS groups were significantly decreased as compared to those of control groups. There are no significant differences among the groups in the contents of albumin (ALB), total cholesterol (TCHO), glucose (GLU), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in blood serum. The content of total glucose (TG) in 30% FSMS containing dietary groups was significantly decreased as compared to that of the control groups. The content of HDLC in 30% FSMS containing dietary groups was significantly increased as compared to that of the control group. The number of lactobacillus in the intestinal microflora were significantly increased in chicks fed FSMS groups. The ammonia gas production in FSMS groups was siginificantly decreased as compard to that the control groups. These results indicated that dietary FSMS exerted growth performance for feeding broiler.
Two separate trials were conducted to determine the effects of uterine priming prior to first breeding and quantify any changes in the reproductive efficiency of gilts. In trial I twelve (12) gilts were randomly assigned to 3 treatments:T1:infusion of distilled water (control), T2: single infusion of killed semen (KS1), and T3: double infusion of killed semen (KS2). Each treatment had 4 breeding gilts which were bred by natural insemination (NI). In trial II, another set of 12 breeding gilts were randomly allotted to the same treatments and were subsequently bred by artificial insemination (AI). Infusions, through the use of AI catheters, were done during the $2^{nd}$ estrous cycle for T1 and T2, whereas infusions for T3 were made during the $1^{st}$ and $2^{nd}$ cycles. Regular breeding was subsequently made during the $3^{rd}$ estrous cycle. All gilts that returned to cycle were rebred within the 30-day period. In trial I (natural breeding), total piglets born was higher (p<0.05) in T2 (12.75 piglets) and T3 (11.75 piglets) than in the control (10.5 piglets). T3 obtained the highest (p<0.05) litter size (10.25 piglets) and heaviest litter weight (74.12 kg) at 28 days weaning, followed by T2 (9.80 piglets and 65.0 kg, respectively). The control yielded the lowest (p<0.05) litter size (7.50) and the lightest litter weight (47.00 kg) at weaning. For Trial II gilts (artificially inseminated), T3 gave higher (p<0.05) litter size born alive (10.88 piglets), total piglets born (11.72 piglets) and live litter weight at birth (15.30 kg) than those of T2 and the control. These results indicate that prebreeding intrauterine infusion of killed boar semen, either single or double, improved the reproductive performance of gilts.
