The Effects of Bogimakseong-bang(補氣膜性方) Treatment on cBSA-induced Membranous Nephropathy in Mouse Model (보기막성방(補氣膜性方)이 Cationized Bovine Serum Albumin투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향)

  • Objective : We aimed to identify the effects of Bogimakseong-bang(BGMSB) treatment on cBSA-induced MN in a mouse model. Methods : We divided 20 mice into 4 groups. The normal group (NR) had no treatment. We used cBSA to induced MN to the other 3 groups. One group (CT) was treated with cBSA (7mg/kg i.p) only. The second (BG-250) was treated with cBSA (7mg/kg i.p) and BGMSB extract (250mg/kg, p.o). The third group (BG-500) was treated with cBSA(7mg/kg i.p) and BGMSB extract (500mg/kg, p.o). After cBSA and BGMSB extract treatment for 4 weeks, proteinuria, serum albumin, total cholesterol, serum creatinine, BUN, total nucleated spleen cell number and total infiltrated kidney cell number of all groups were measured. CD3e+/CD19+ and CD4+/CD8 cells ratio of peripheral blood, kidney and spleen of all groups were analyzed. $IL-1{\beta}$ and $TNF-{\alpha}$, IL-6, IgG, IgM, and $IFN-{\gamma}$ levels of all groups were gauged. Histological analysis of kidney tissue and immunohistochemical staining (CD4 CD8) of kidneys were observed. Results : Proteinuria significantly decreased and serum albumin increased in groups treated with cBSA and BGMSB extract compared with the control. Total cholesterol decreased but not significantly. CD3e+/CD19cells ratio of peripheral blood decreased. CD3e+/CD19+ and CD4+/CD8 cells percentage of kidney and spleen showed no significant change. Level of $IL-1{\beta}$, $TNF-{\alpha}$ and IL-6 significantly decreased. and $IFN-{\gamma}$ increased but has not significantly. Concentration of IgG and IgM significantly decreased compared with control. Thickness of GBM decreased on histological analysis of kidney. Deposition of CD4 and CD8 decreased on immunohistochemical staining of kidney. Conclusions : According to the above result, BGMSB had a significant effect for treating MN which is cBSA-induced.

Studies on Separation and Determination of Korean Bovine Serum Protein by Colorimetric Method (비색법에 의한 한우 혈청단백질의 분획정량 시험)

  • Serum Samples from adult of Korean cattles including 40 females and 20 males were analyzed by sodium salt precipitation and colorimetric method in the purpose of the determination of total serum protein, albumin, globulin, ${\alpha}$-globulin, ${\beta}$-globulin and ${\gamma}$-globulin. The results obtained arc summarized as follows: 1. Mean value of total serum protein showed a slight variation from 7.6%, and its regional and sex differences were not found to be significant. 2. Contents of albumin in serum showed lower level than that of globulin as low level of A/G ratio 0.4 in proportion. 3. Contents of Serum ${\alpha}$-globulin showed 1.4w/v% and $1.51{\pm}0.46$w/v% in each group of female, and $1.31{\pm}0.26$w/v%, in the group of male. 4. Contents of serum ${\beta}$-globulin showed 1.74w/v%, 1.95w/v%, in each group of female, and 1.82w/v% in the group of male. 5. Contents of serum ${\gamma}$-globulin showed 2.32w/v%, 2.30w/v% in each group of female, and 2.30w/v%, in the group of male.

Effect of Culture Medium and Additive on the Development of Bovine IVM/IVF Embryos (체외배양액과 첨가물질이 소 체외수정란의 체외발육에 미치는 효과)

  • The objective of this study were to investigate the effects of culture media and additives on the development of bovine in vitro matured(IVM) and in vitro fertilized(IVF) oocytes. In experiment 1, bovine oocytes were cultured in droplets of TC 199 supplemented with 10% fetal bovine serum(FBS) with or without hormones (5$\mu\textrm{g}$/ml FSH, 5$\mu\textrm{g}$/ml LH, 1$\mu\textrm{g}$/ml E2). Cleavage rates of embryos cultured for 40~44hrs after IVF were higher when embryos were cultured in TC 199 supplemented hormones (68.1%, 921/35) than without hormones (52.7%, 77/146), but the percentages of development beyond morulae stage were not difference (20.7%, 19.4%). In experiment 2, the effects of various media such as TC 199, synthetic oviduct fluid(SOF), CR1aa with different energy source (fatal bovine serum, FBS; bovine serum albumin, BSA) on developmental capacity of IVM/IVF bovine embryos were investigated. The developmental rates into morulae and blastocysts were 27.1, 10.7, 6.3 and 0%, respecitvely, in CR1aa plus 3mg/ml BSA, SOF plus 10% FBS, TC 199 plus 10% FBS, SOF plus 3mg/ml BSA. In experiment 3, the comparisons of bovine embryos developed to morulae and blastocysts in different culture media (TC 199, SOF, CR1aa, Menezo's B2) were investigated. The developmental capacity beyond morulae stage were 32.9, 26.6, 11.1 and 7.1%, respectively, in Menezo's B2 plus BSA, CR1aa plus BSA, SOF plus BSA, TC 199 plus FBS medium. The cell numbers of the blastocyst were not different in different cultrue media. In experiment 4, bovine embryos were co-cultured with vobine oviduct epithelial cells(BOEC) in TC 199 plus FBS, SOF plus BSA, CR1aa plus BSA, Menezo's B2 plus BSA. The morula and blastocyst rates were 44.7, 32.9, 26.0 and 23.3%, respectively, in CR1aa TC 199, SOF, and Menezo's B2 medium. The cell numbers of the blastocyst were similar to those of blastocyst developed in different culture media.

  • Ultrafiltration membranes based on anion charged poly(bis[4-(3-aminophenoxy)phenyl]sulfone pyromellite) imide derivatives (ACPI) were prepared by the phase inversion method. The polymers have good solubility in aprotic polar solvents. The composition of casting solution and the casting conditions played an important role in determining the permeation characteristics of membrane because the membrane structure could be controlled by the preparation conditions. The extent of fouling-repression was observed by the relative ratio of permeate flux ($J_t$)/pure water flux ($J_0$) and the membrane filtration index (MFI). The characteristics were measured by aqueous solution of bovine serum albumin (BSA) over a pH range of 2.5-9.0. The ACPI membrane having a hydrophilic property was less fouled than the membrane prepared from the original polyimide. With increasing the ion exchange capacity of ACPI membrane, th $\varepsilon$ relative ratio of flux was higher while the membrane filtration index was lower as compared with the original polyimide membrane. From the further away from isoelectric point of bovin serum albumin, the permeation was higher as well as the formation of fouling was more diminish. ACPI membranes having various their properties could be obtained. Further, it was proved that their permeation properties could be determined from the preparation conditions, various operating conditions, and dim $\varepsilon$ rent ion exchange capacity of anion charged polyimide derivatives.

  • The effects of ionic strength and pH on the binding of cefazolin to bovine serum albumin (BSA) were studied by UV difference spectrophotometry. As ionic strength at constant pH and temperature increases, the apparent bining constant decreased but the number of binding sites remained almost constant at 2. The constancy of the number of binding sites with increasing the ionic strength suggests that purely electrostatic forces between BSA and drug do not have great importance in the drug binding, even though there is a decrease in the apparent binding constant. Thus, the effect of ionic strength on the interaction between drug and BSA may be explained by the changes in ionic atmosphere of the aggregated BSA molecules and competitive inhibition by phosphate ions. In addition, the higher apparent binding constant at high ionic strength is explained by conformational changes of BSA from its aggregate forms into subunits. The pH effects on the afinity of interactions indicated that the binding affinity of cefazoline is higher in the neutral region than in the alkaline region. An d at high pH value, the number of binding sites decreased from 2 to 1 because of the conformational change of BSA in the alkaline region.

Effects of Interferents in Blood on the Blood Glucose Measurements by Using Polyphosphazene Diagnostic Membranes (포스파젠 고분자 진단막을 이용한 혈당 측정 시 혈액 중 장애성분이 미치는 영향)

  • Diagnostic membranes which were made of polyphosphazenes were prepared for the measurements of blood glucose concentration. The effects of possible interferents in human blood on the glucose concentration measurements were examined. Above those interferents, ascorbic acid (AA) showed 3~9% higher K/S values compared to the standard plasma solution, tolbutamide (TA) showed 11~13% lower K/S values than those of plasma, triglycerides and bovine serum albumin (BSA) showed 20~25% lower K/S values than those of plasma. However, most of the interferents except above materials did not show any serious effects on the blood glucose measurements.

  • The adsorption of a model protein bovine serum albumin (BSA) in expanded bed chromatography was undertaken by exploiting a commercially available expanded bed column (20 mm i.d.) from UpFront Chromatography and Streamline DEAE $(\rho=1.2g/cm^3)$ from Amersham Pharmacia Biotechnology. The influence of whole yeast cells on the adsorption capacity of column was explored by employing yeast cells in a concentration ranged of 0 to $15\%(w/v)$. Equilibrium isotherms for adsorption of BSA on Streamline DEAE were correlated by using Langmuir equation. The presence of yeast cells resulted in decreased of BSA binding capacity in both batch binding and expanded bed chromatography. Results indicated that the yeast cells act as competitor for proteins to bind to the sites on adsorbents.

Adsorption of Globular Proteins to Vaccine Adjuvants

  • The maximum adsorption/desorption conditions and the adsorption mechanism of globular proteins to vaccine adjuvants were determined. The maximum adsorption ratio of protein to the $Al^{3+}$ content of aluminum oxyhydroxide and the optimal adsorption pH are 2:1 (${\mu}g:{\mu}g$) for bovine serum albumin (BSA) at pH 6.0 and 2.5:1 (${\mu}g:{\mu}g$) for immunoglobulin G (IgG) at pH 7.0, respectively. The maximum adsorption ratio onto aluminum phosphate gel was 1.5:1 (${\mu}g$ Protein:${\mu}g$ $Al^{3+}$) at pH 5.0 for both BSA and IgG. Adsorption of the native globular proteins, BSA and IgG, to aluminum oxyhydroxide and aluminum phosphate gel was reversible as a function of pH. Complete desorption of these proteins from aluminum phosphate gel was observed at alkaline pH, whereas only 80~90% removal from aluminum oxyhydroxide was achieved with alkaline pH and 50 mM phosphate buffer. We conclude that electrostatic and hydrogen bonding interactions between the native proteins and adjuvants are important binding mechanisms for adsorption, and that the surface charge of the protein and the colloid components control the maximum adsorption conditions.

  • Periodate-oxidized soluble starch and maltohexaose, maltotetraose, maltose, and glyceraldehyde reacted with sweet potato ${\beta}-amylase$, wheat ${\beta}-amylase$, aldolase, bovine serum albumin, catalase, carboxypeptidase, ferritin and pronase. Electrophoretical mobility of modified proteins was different from that of native proteins, and modified proteins were stained with periodic acid-Schiff while native proteins did not stain. This results means that oxidized sugars attached on proteins. This bond is based on the Schiffs base between CHO group of oxidized sugar and ${\varepsilon}-NH_2$ group of lysine of protein. There is no changed UV absorption spectrum of sweet potato ${\beta}-amylase$ modified with oxidized soluble starch, in comparison with native enzyme.
