Influential Characteristics of Decoction Waters on Extraction of Components in Corydalis Tuber Analyzed by Multiple Factor Analysis (다중요인 분석에 의한 전탕용수에 따른 현호색 성분의 추출효과 비교)

  • The characteristics of decoction water are important for the extraction of herbal drugs. In this study, the relationship between the characteristics of water and extraction efficiency of bioactive compounds from Corydalis tuber was investigated. Six different types of decoction water, including three types of mineral water from Jeju Island and three types of bottled water sold in market, were used as decoction water for the extraction of bioactive compounds from Corydalis tuber. The contents of minerals in six types of water were analyzed by ICP-AES and ICP-MS; the contents of extracted bioactive compounds were analyzed by HPLC. Multivariate analysis of variance was used to analyze the differences in total extraction and bioactive compounds extracted with six types of water. Multiple factor analysis was used to analyze the relationship among the pH, content of mineral, anion, total extractions and content of bioactive components as factors. There was a significant difference among the six types of water in the total extracts and the bioactive compounds. The results proved that the content of extracted compounds from Corydalis tuber was influenced by the types of decoction water.

Identification of the Volatile Compounds in Polyethylene Terephthalate Bottles and Determination of Their Migration Content into Mineral Water (PET 생수병 내 휘발성 물질의 동정 및 이행량 분석)

  • This study was carried out to identify the volatile organic compounds (VOCs) in polyethylene terephthalate (PET) bottles and to determine the extent to which VOCs migrate into mineral water during the bottling process and storage. A greater amount of nonanal and decanal was generated from the PET bottles than from the PET preforms. Benzene, ethylbenzene, nonanal, and vinyl benzoate were identified from the PET bottles when the incubation temperature of the headspace solid-phase microextraction (HS-SPME) sampler was set to 60, 80, and $100^{\circ}C$. As the incubation temperature increased, the concentrations of nonanal, vinyl benzoate, and decanal increased significantly. When the high-density polyethylene (HDPE) PET bottle caps were extracted with dichloromethane, the level of Irgafos 168 was found to be $206{\pm}20.1\mu}g/g$. The concentration of 2,4-di-tert-butylphenol in water was $4.80{\pm}0.2{\mu}g/L$. Therefore, it is necessary to avoid exposing PET and HDPE resins to high temperatures during the manufacturing process and storage of bottled water.

Analysis of Acetaldehyde and Formaldehyde Migrated from Polyethylene Terephthalate Bottles into Mineral Water (페트병 생수 중 아세트알데하이드와 포름알데하이드의 이행 분석)

  • The objective of this study was to determine the extent to which acetaldehyde (AA) and formaldehyde (FA) migrated from polyethylene terephthalate (PET) bottles into mineral water during storage at 25 and 35$^{\circ}C$ for 240 days. Generally, AA and FA were observed to migrate more at 35$^{\circ}C$ than at 25$^{\circ}C$. The amount of FA in the samples irradiated under UV lamps was higher than in those stored in the dark over the storage period and vice versa for AA. Using a triangular test, the sensory panels distinguished a difference between one sample and the other two mineral water samples that had been exposed to UV lamps after 60 days of storage at 35$^{\circ}C$ (p<0.05). At this time, FA and AA concentrations were measured at 267.4 and 515.3 g/L, respectively. After 120 days, an independent mineral water sample from the other two samples in all treatments could be distinguished by the panel members at a significance level of p<0.001.

  • A practical and convenient method of rearing Eucyclops serrulatus in a microculture environment is described. A complete life cycle of E. serrulatus was maintained in a narrow space on a microscope slide glass on which a cover glass of $22{\times}40mm$ in size was mounted at a height of 0.8mm. The culture medium was constituted by bottled mineral water boiled with grains of Glycine max (soybean). Chilomonas paramecium, a free-living protozoan organism, was provided as live food. Growth of nauplii hatched from eggs to the first stage of copepodite took an average of 7.7 days, and the growth of copepodite 1 to the egg-bearing adult female took an average of 20.1 days in the microculture cell with an average life time of 44.7 days. Continuous passage of cope pods was successfully maintained as long as sufficient medium and food were provided. The microculture method enables an in situ microscopic observation on the growth and developmental process of helminth larvae experimentally infected to copepods as well as of copepod itself. Furthermore, it does not require anesthetization and, therefore, minimize the amount of stress exposed to cope pods during the handling process.

Economic Impact Analysis on High-yield Groundwater Development R&D Project in Jeju (제주도 청정 대용량 지하수개발 연구사업의 경제적 효과분석)

  • For economic impact analysis on a R&D project of high-yield groundwater development in Jeju conducted by KIGAM from 2004 to 2007, benefit/cost ratio(BCR), net present value(NPV), and internal rate of return(IRR) were calculated by contingent valuation method(CVM), production function analysis, domestic water market analysis and technology factor analysis. Measurable direct impact parameters among the major outputs of this R&D project consisted the estimation 4 high-yield and high mineral groundwater reserve in Jeju. Annual use of the reserve by piped water and bottled water was estimated as 12.23 million ton and its monetary value was calculated as 293.4 million dollar in 2006 year value applied of 5.5% discount rate. Economic impact of this R&D project in NPV of year 2006, with applying a discount rate of 5.5%, was identified and estimated as 13.66 million dollar in NPV, 4.05 points in BCR, and 22.74% in IRR, respectively. Additional early launch benefit was 5.58 million dollar. Even increased of the 1% discount rate, NPV of this R&D project was also positive as 12.18 million dollar and BCR was 3.71.

THE STUDY FOR THE MINERAL CONTENTS OF BOTTLED WATER (시판되는 생수 내 무기물 함량에 관한 연구)

  • Drinking water has lots of minerals, especially calcium, sodium, kalium, magnesium, and fluoride must be labelled on the bottle about their contents. Minerals like calcium, magnesium, and fluoride have influence to the tooth development. Appropriately taking some minerals, dental caries can be prevented somewhat degree. There is no guide line about innoxious minerals like calcium and magnesium, however, noxious mineral like fluoride, should be contained less than 2 mgF/L according to the current drinking water standard. Hereupon, it is necessary to recognize the concentration of fluoride in drinking water, so I studied 15 samples of domestic drinking water on sale about the concentration of calcium and magnesium, fluoride. The results obtained were as follows ; 1. 14 drinking waters in 15 samples showed various Ca concentration. The average Ca concentration is 34.68${\pm}$31.84 mg/L, the highest is 128.91${\pm}$1.85 mg/L and the lowest is 2.0${\pm}$0.02 mg/L. 2. 13 drinking waters in 15 samples indicate the Mg concentration. The average concentration is 9.22${\pm}$11.06 mg/L, the highest is 30.43${\pm}$0.75 mg/L and the lowest is 0.0 mg/L. 3. 11 drinking waters in 15 samples indicate the F concentration. The average concentration is 0.25${\pm}$0.33 mg/L, the highest is 1.13${\pm}$0.04 mg/L and the lowest is 0.01${\pm}$0.03 mg/L. All samples are satisfied the current drinking water standard, 2 mg F/L.

The Observation and a Quantitative Evaluation of Viable but Non-Culturable Bacteria in Potable Groundwater Using Epifluorescence Microscopy (형광현미경을 이용한 음용 지하수내 배양불능 세균의 관찰 및 정량적 평가)

  • The direct viable count (DVC) and plate count (PC) methods was used to measure the number of bacteria in potable groundwater samples collected from bottled water from the market, mineral water, and edible groundwater near the urban areas and the stock farming congested areas. As a result, the number of living bacteria by DVC was comprised 30~80% of the total direct count (TDC), whereas the number of living bacteria by PC was around l~30% of DVC. Such results show that viable but non-culturable (VBNC) bacteria exist in the potable groundwater with high percentages. On the other hand, upon measuring the value from the conventional nutrient broth (NB), $10^-2$ fold diluted nutrient broth (DNB), and R2A broth, the values from the DNB and R2A showed 2~50 times higher than the conventional NB medium. These results indicate that oligotrophic bacterial groups which can multiply in the low nutrient broth abundantly exist in the oligotrophic environment like potable groundwater.

Analysis of Off-flavor Generated from a Polyethylene Terephthalate Water Bottles and Caps by Using an Electronic Nose (MS-전자코를 사용한 페트(polyethylene terephthalate) 생수병과 뚜껑의 이취 이행 분석)

  • The objective of the study was to investigate the off-flavor generated from PET water bottles and their caps by using a mass spectrometry-based electronic nose. The ion fragment data obtained from the electronic nose were used for discriminant function analysis (DFA). In the case of increased concentrations of the contamination of water, the off-flavor pattern depended on the discriminant function second score instead of the discriminant function first score. To identify the cause of off-flavor in polyethylene terephthalate (PET) bottled water, the PET bottle and its cap were analyzed by DFA. The results showed that the cap generated more volatile compounds than the bottle or mineral water did. The substances causing the off-flavor were predicted to be 2,4-di-tert-butylphenol (2,4-DTBP), nonanal, and decanal when the main peak of the mass spectrum was compared with the major ion fragments of the electronic nose. Thus, using this method, we could determine whether the PET water bottle was contaminated and whether the off-flavor resulted from contamination of the bottle cap.

Concentration of Formaldehyde and Acetaldehyde depending on the Time of Storage into Mineral Water (먹는샘물의 보관 기간에 따른 formaldehyde 및 acetaldehyde의 농도 변화)

  • BACKGROUND: According to Korean regulations, bottled waters (BWs) can not be treated with chemical disinfectants like chlorine, so UV and ozone disinfection is applied. During the past several years, chemicals were detected in some BWs, and the public was concerned about the safety of BWs. METHODS AND RESULTS: Mineral waters were stored for 180 days at $25^{\circ}C$ and $50^{\circ}C$, tested acetaldehyde and formaldehyde by HPLC. When mineral waters were put in a PET bottles, the formaldehyde level ranged from 5 to $66{\mu}g/L$ during 180 days at $50^{\circ}C$. While the acetaldehyde level ranged from 31 to $221{\mu}g/L$, it was low than $16{\mu}g/L$ in glass bottle. CONCLUSION(s): This result showed that formaldehyde and acetaldehyde were detected higher in PET bottles than glass bottles, these also increased depending on the time of storage. Concentration of formaldehyde and acetaldehyde could be significantly influenced by the time of storage and temperature.

Rapid Simultaneous Determination of Anions in Mineral Water by Single Column Ion Chromatography (단일 컬럼이온크로마토그래피에 의한 광천수중 음이온의 신속한 동시정량)

  • Analytical conditions for single column ion chromatographic determination of five anions such as F-, Cl-, NO2-, NO3-, and SO42- were optimized with respect to analysis time and separation efficiency. At the optimum condition of 2.0 mM phthalic acid at pH 4.5, five anions were effectively separated in 10 minutes so that analysis time could be reduced by 40% compared to other recommended conditions. Under this condition all the calibration curves of five anions were linear with a correlation coefficient > 0.999. Analytical results for nine commercial bottled waters were presented.
