세 종류 줄기세포의 특성 분석과 지방유래 줄기세포의 심근세포로의 분화 (Characterization Analysis for Cardiogenic Potential of Three Human Adult Stem Cells)

  • 본 연구에서는 사람의 지방조직(human adipose tissue-derived stem cells, HAD), 탯줄(human umbilical cordderived stem cells, HUC), 그리고 양막(human amnion-derived stem cells, HAM)유래 줄기세포를 분리하여 세포의 형태 및 성장속도를 비교하고, 역전사 중합효소 연쇄반응과 면역세포화학 염색법을 이용하여 유전자와 단백질 발현을 비교 분석하였다. 또한 지방유래 줄기세포를 이용하여 심장근육세포로의 분화를 유도하였다. 본 연구 결과, 탯줄과 양막유래 줄기세포의 형태는 매우 유사하였으며, 지방유래 줄기세포의 형태는 다른 것으로 나타났다. 분열시간은 탯줄유래 줄기세포가 가장 빨랐으나 총 분열 횟수는 양막유래 줄기세포와 같았으며, 지방유래 줄기세포의 총 분열횟수가 가장 많았다. 세 종류 줄기세포의 유전자와 단백질 발현은 비슷한 양상을 나타냈다. 지방세포, 골세포, 연골세포로의 분화 유도 결과 세 종류의 줄기세포 모두 분화 유도되었다. 또한, 심장세포 특이 유전자의 발현 분석 결과 세 종류의 줄기세포에서 유사한 발현 양상을 나타냈다. 이 중 지방유래 줄기세포를 24시간 동안 $10\;{\mu}M$ 5-azacytidine 처리 후 기본 배양액에서 4주 동안 배양하거나 또는 5-azacytidine 처리 후 bone morphogenic protein-2(BMP-2)와 fibroblast growth factor-10(FGF-10) 또는 BMP-4와 FGF-4 또는 BMP-4와 FGF-8이 첨가된 배양액으로 4주 동안 배양하여 심근세포로의 분화를 유도하였다. 분화 유도 후 심장세포 특이 유전자 발현을 분석 결과 cardiac myosin light chain-1(Cmlc-1)과 L-type calcium channel ${\alpha}1C$ subunit(${\alpha}1C$) 유전자의 발현이 증가하였다. 그러나 troponin T(TnT), troponin I(TnI) 그리고 potassium channel Kv4.3 subunit (Kv4.3) 유전자의 발현은 증가하지 않았다. 본 연구 결과, 지방, 탯줄 및 양막유래 줄기세포는 특성이 매우 유사한 것으로 나타났으며, 심장 질환 치료를 목적으로 하는 세포 치료에 이용될 수 있을 것으로 사료된다. 또한, 적절한 배양조건 하에서 성장인자와 cytokine들을 처리하여 심장세포로의 분화 유도가 이루어진다면 임상적용에 유용한 세포로 사용될 수 있을 것으로 사료된다.

Laser-Induced Breakdown Spectroscopy (LIBS)를 이용한 방사성 스트론튬 오염물질에 대한 신속한 모니터링 기술 (Rapid Detection of Radioactive Strontium in Water Samples Using Laser-Induced Breakdown Spectroscopy (LIBS))

  • Cs-137 (반감기 : 30.17년)과 더불어 중요한 원자력발전소 주변 환경감시대상 방사성 핵종 가운데 하나인 순수한 베타방출체인 Sr-90 (반감기 : 28.8년)에 대한 신속하고 용이한 모니터링 방법을 연구하였다. 스트론튬은 칼슘과 같은 2족 알칼리 토금속에 속해 있어서 전자배치나 크기가 비슷하며 최외각전자를 2개 가지고 있기 때문에 화학적으로 칼슘과 치환될 수 있다. 이러한 유사한 화학적 성질로 인해서 환경으로 유출시 물, 토양 및 농작물을 통한 먹이사슬을 거쳐서 인체로 쉽게 유입될 수 있으며, 인체 유입 시 뼈에 쉽게 침적되어 장기간 (생물학적 반감기 : 약 50년) 동안 독성을 유발한다. 스트론튬은 매우 환원성이 있고 특이 반응으로 습식분석이 어려우며, 특히 원자력발전소에서 감시하고 있는 방사성 스트론튬은 복잡한 분석절차, 고가의 분석 장비 사용 및 화학 전처리약품 다량 사용 등으로 분석의 정확도 저하는 물론 고비용에 따른 문제를 안고 있다. 따라서 펄스에너지를 사용하여 시료에 플라즈마를 생성시켜 고유 스펙트럼을 이용해 시료내 원소를 분석하는 Laser-Induced Breakdown Spectroscopy (LIBS) 분석기법을 도입하여 전처리 과정 없이 수 초 내에 분석이 가능하고 현장에서 실시간으로 측정 가능한 스트론튬 원소의 정량분석 방법을 도출하였다. 다양한 분석에 필요한 시료기판을 개발하여 레이저, 파장 및 시간분해능의 최적화로 분석 감도를 향상시키고 방해이온에 대한 영향 평가로 액체시료의 정량분석을 가능하게 하여 신속한 모니터링 체계를 구축하게 하였다. 이는 원자력발전소로부터 방출되고 있는 방사성 폐수의 실시간 모니터링에 효과적으로 적용될 수 있으며, 더 나아가 후쿠시마 원전사고와 같은 비상시 모니터링 수단으로 적용 될 수 있다.

방사면역 측정법에 의한 25-OH-VitD와 25-OH-VitD3의 비교 평가 (Comparative Evaluation of 25-OH-VitD and 25-OH-VitD3 by Radioimmunoassay)

  • 비타민D는 체내에서 칼슘 대사를 조절하여 뼈의 건강을 유지하며, 세포의 증식 및 분화의 조절, 면역 기능 등에 관여하는 중요한 역할을 한다. 부족 시 구루병, 골다공증의 위험이 높아지고 심혈관계 질환, 당뇨병, 일부 암 등의 발병 위험이 증가하는 것으로 보고되고 있다. 특히 한국은 비타민D 부족의 인구 비율이 매우 높은 국가 중 하나이다. 따라서 비타민D 부족의 진단 및 치료를 위해 혈중 25-OH-VitD 또는 25-OH-VitD3의 정확한 측정이 요구된다. 이에 본원에서 시행하고 있는 25-OH-VitD와 25-OH-VitD3검사를 비교 평가하여 비타민D에 대한 정확한 진단 및 치료에 기여하고자 한다. 25-OH-VitD와 25-OH-VitD3의 결과를 전향적으로 측정하여 상관성, 재현성, 모집단 분포율을 구하였다. 또한 2017년 4월에서 2019년 6월까지 서울아산병원 핵의학과 혈액검사실에서 측정한 내부정도관리와 대한핵의학기술학회에서 주관하는 2018년 상반기, 하반기 외부정도관리(기관간 숙련도 평가)에 참여한 결과를 비교하였다. 그 결과 97개의 검체를 대상으로 25-OH-VitD는 25-OH-VitD3 × 0.9 + 0.3 (R >0.9)로 강력한 양의 상관관계를 보였다. 반복 측정을 통한 재현성 평가 결과 평균 diff(%) 값은 7.7%, 7.4%로 모두 우수한 성적을 나타내었다. 또한 모집단 분포를 비교한 결과 통계적으로 유의한 차이를 보이지 않았다(p>0.05). 2017년 4월에서 2019년 6월까지 서울아산병원 핵의학 혈액검사실에서 측정한 내부정도관리 결과값은 평균(CV%) 6.2%, 6.8%을 나타내었다. 2018년 상반기 외부정도관리(기관간 숙련도 평가) 수행도 평가 결과 Z 값이 25-OH-VitD는 저, 중, 고농도에 대해 -1.10, -1.00, -0.90이며 25-OH-VitD3는 0.22, 0.47, 1.10이고 하반기 수행도 평가 결과는 각각 -0.80, -0.30, -0.10과 -0.01, 1.50, 1.30으로 모두 Z ≤ 2.0의 적합한 결과를 얻었다. 방사면역 측정법에 의한 25-OH-VitD와 25-OH-VitD3 검사의 비교 평가 결과 상관성, 재현성, 모집단 분포율, 내부정도관리, 외부정도관리(기관간 숙련도 평가) 모두 우수한 성적을 보였다. 따라서 혈액 내 25-OH-VitD 또는 25-OH-VitD3로 단독 측정이 가능한 방사면역 측정법은 비타민D 부족에 대한 스크리닝 검사로 적합한 것으로 사료된다.

백봉오골계육의 이화학적 특성 (Physicochemical Characteristics of Silky Fowl(Gallus domesticus var. silkies))

  • 중국 최고의 의약서인 신농본초경(Shennong Classics of Material Medic)과 중국 명대($1368{\sim}1644$)에 이시진이 저술한 본초강목 등 여러 문헌에 기록으로 전해지고 있는 백봉오골계의 의약적 기능을 볼 때 백봉오골계육은 건강 기능 식품의 신소재로서 개발할 만한 충분한 가치가 있다. 본 연구에서는 약계인 백봉오골계가 우리나라 재래 오골계인 연산오골계와 해부학적 특성과 고기의 영양성분에 있어서 어떤 차이점이 있는가를 조사하였다. 1. 백봉오골계의 해부학적 특성 1) 백봉오골계는 깃털이 눈과 같이 희고 깃털의 우판(羽辦)은 가늘게 갈라져 있어 유연한 견사(silk)와 같다. 연산오골계의 깃털은 일반 닭과 같은 모양이고 검은색이다. 2) 백봉오골계의 벼슬은 phoenix crown이라는 별명을 가지고 있는데 일반 닭과는 모양이 다르며, 벼슬은 수컷 경우 장미모양, 산딸기 또는 오디 모양이며, 암컷의 경우 자주 빛깔의 산딸기 또는 오디 모양이다. 3) 백봉오골계의 부리(beak)는 짧고 단단하며 납빛 blue(leaden blue)이고 얼굴은 매끈하고 우아하며 육수는 자색을 띤 창백색이다. 4) 백봉오골계의 눈 홍채는 암갈색 내지는 검정색(dark dark brown)이고 가장 큰 특징 중의 하나는 공작 색(peacock green) 또는 터키 옥색(lightblue turquoise)과 같은 귓불(earlobes)을 가지고 있는 것이다. 연산오골계는 흑회색의 오디 모양의 벼슬을 가지고 있으며 부리(beak)는 길고 단단하며 얼굴은 크고 육수는 적자색을 띤다. 눈의 홍채는 검정색을 가지고 있다. 5) 백봉오골계의 꽁지는 짧은 편이고 많지 않은 부드러운 깃으로 되어 있고 연산오골계 꽁지는 일반 닭과 같다. 6) 백봉오골계의 머리에는 수술(tassel)이 있으며 너무 크게 자라서 앞이 보이지 않는 것도 있다. 턱수염이 있는 종(bearded)과 턱수염이 없는 종(non bearded)이 있다. 7) 백봉오골계의 발가락 수는 5개로 이루어져 있다. 다리는 짧고 흑회색이며 바깥쪽에 깃털이 나 있다. 연산오골계의 발가락 수는4개로 이루어져 있으며 다리 부분에 깃털이 없다. 8) 백봉오골계와 연산오골계의 총 신장을 비교해 볼 때 암수 모두 연산오골계가 백봉오골계 암수보다 훨씬 더 크다는 것을 알 수 있다. 9) 백봉오골계의 피부는 곱고 검푸른색 또는 흑회색이며, 모공은 연산오골계에 비해 모공수가 적고 모공이 가늘다. 연산오골계의 피부는 거칠고 짙은 흑회색을 띠며 백봉오골계보다 모공수가 더 많고 모공도 더 크다. 10) 백봉오골계와 연산오골계는 모두 일반 양계와는 달리 근육, 내장, 뼈 등이 일반적으로 흑회색을 띠고 있다. 2. 백봉오골계 육의 일반 성분과 Mineral 함량 1) 백봉오골계육은 연산오골계육과 일반 양계육에 비해 수분과 지방 함량은 적고, 단백질과 회분 함량이 높은 것이 특징 이다. 2) 백봉오골계는 칼슘(Ca), 인(P), 철(Fe), 칼륨(K), 아연(Zn)의 함량은 모두 다리살이 가슴살보다 더 높은 경향을 보였으며 특히 철(Fe)의 함량은 가슴살보다 약 5.6배, 아연(Zn)은 약 5.2배나 높았다. 그러나 연산오골계는 칼슘(Ca)과 철(Fe), 아연(Zn)만이 다리살이 가슴살 보다 더 높았고 인(P)과 칼륨(K)은 가슴살이 다리살보다 더 높은 경향을 보였다. 가슴살과 다리살 중의 Mineral 함량을 비교해 보았을 때 인(P), 철(Fe), 칼륨(K), 아연(Zn)의 함량은 백봉오골계가 연산오골계보다 높은 경향을 보였으며 칼슘(Ca)함량만이 백봉오골계보다 연산오골계가 높은 경향을 보였다. 3) Fe의 함량은 백봉오골계와 연산오골계 모두 다리살이 가슴살보다 더 높았으며, 다리살 중의 Fe 함량을 비교해 보았을 때 백봉오골계가 3.9 mg%, 연산오골계가 1.0 mg%로서 백봉오골계가 연산오골계보다 약 4.0배나 더 높았다. 4) Zn의 함량도 백봉오골계와 연산오골계 모두가 다리살이 가슴살보다 훨씬 더 높은 경향을 보여주었다. 연산오골계의 경우 Zn 함량은 가슴살이 0.6 mg%, 다리살이 2.3 mg%로서 다리살이 가슴살보다 약4배 높았다. 백봉오골계의 경우 Zn함량은 가슴살이 0.5 mg%, 다리살이 2.6 mg%로서 다리살이 가슴살보다 약 5배 정도 높았다. 5) K의 함량은 백봉오골계가 가슴살과 다리살 모두에서 연산오골계보다 높았다. 그리고 백봉오골계는 가슴살보다는 다리살이 연산오골계는 다리살보다는 가슴살이 K함량이 높았다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The authors suggests that this system based on down-sized computer could provide a progressive approach to total LIS based on local area network, and the implemented system could serve as a model for other hospital's LIS for routine hematology. 30609 T00401030609 ^x To develop an artificial bone substitute that is gradually degraded and replaced by the regenerated natural bone, the authors designed a composite that is consisted of calcium phosphate and collagen.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases.
