  • BACKGROUND/OBJECTIVES: Glutathione S-transferase (GST) forms a multigene family of phase II detoxification enzymes which are involved in the detoxification of xenobiotics by conjugating substances with glutathione. The aim of this study is to assess the antioxidative status and the degree of DNA damage in the subclinical hypertensive patients in Korea using glutathione S-transferase polymorphisms. SUBJECTS/METHODS: We examined whether DNA damage and antioxidative status show a difference between GSTM1 or GSTT1 genotype in 227 newly diagnosed, untreated (systolic blood pressure $(BP){\geq}130mmHg$ or diastolic $BP{\geq}85mmHg$) subclinical hypertensive patients and 130 normotensive subjects (systolic BP < 120 mmHg and diastolic BP < 80 mmHg). From the blood of the subjects, the degree of the DNA damage in lymphocyte, the activities of erythrocyte superoxide dismutase, the catalase, and the glutathione peroxidase, the level of glutathione, plasma total radical-trapping antioxidant potential (TRAP), anti-oxidative vitamins, as well as plasma lipid profiles and conjugated diene (CD) were analyzed. RESULTS: Of the 227 subjects studied, 68.3% were GSTM1 null genotype and 66.5% were GSTT1 null genotype. GSTM1 null genotype had an increased risk of hypertension (OR: 2.104, CI: 1.38-3.35), but no significant association in GSTT1 null genotype (OR 0.982, CI: 0.62-1.55). No difference in erythrocyte activities of superoxide dismutase, catalase, or glutathione peroxidase, and plasma TRAP, CD, lipid profiles, and GSH levels were observed between GSTM1 or GSTT1 genotype. Plasma levels of ${\alpha}-tocopherol$ increased significantly in GSTT1 wild genotype (P < 0.05); however, plasma level of ${\beta}-carotene$ increased significantly in GSTT1 null genotype (P < 0.01). DNA damage assessed by the Comet assay was significantly higher in GSTM1 null genotype than wild genotype (P < 0.05). CONCLUSIONS: These results confirm the association between GSTM1 null genotype and risk of hypertension as they suggest that GSTM1 null genotype leads to an increased oxidative stress compared with wild genotype.

  • Purpose: Lipopolysaccharide-binding protein (LBP) is a 65-kDa acute phase protein, derived from the liver, which is present in high concentrations in plasma. Data regarding the association between circulating plasma LBP levels and obesity-related biomarkers in the pediatric population are scarce. We aimed to determine whether there was a difference in plasma LBP levels between overweight/obese and normal-weight adolescents and to assess the correlation of circulating LBP levels with anthropometric measures and obesity-related biomarkers, including insulin resistance, liver enzyme levels, and lipid profiles. Methods: The study included 87 adolescents aged 12-13 years; 44 were overweight/obese and 43 were of normal-weight. We assessed anthropometric and laboratory measures, including body mass index (BMI), blood pressure, insulin resistance, liver enzyme levels, and lipid profiles. Plasma LBP levels were measured using an enzyme-linked immunosorbent assay. Results: The mean age of the participants was $12.9{\pm}0.3$ years. Circulating plasma LBP levels were significantly increased in overweight/obese participants compared with those in normal-weight participants ($7.8{\pm}1.9{\mu}g/mL$ vs. $6.0{\pm}1.6{\mu}g/mL$, P<0.001). LBP levels were significantly and positively associated with BMI, systolic blood pressure, aspartate aminotransferase, alanine aminotransferase, total cholesterol, low density lipoprotein-cholesterol, fasting glucose and insulin, and insulin resistance as indicated by the homeostatic model assessment of insulin resistance (HOMA-IR) (all P<0.05). In multivariate linear regression analysis, BMI and HOMA-IR were independently and positively associated with plasma LBP levels. Conclusion: LBP is an inflammatory biomarker associated with BMI and obesity-related insulin resistance in adolescents. The positive correlation between these parameters suggests a potentially relevant pathophysiological mechanism linking LBP to obesity-related insulin resistance in adolescents.

  • Sea tangle (Laminaria japonica) is well known as having anti-diabetic and hypolipidemic effects in animals as well as in humans. In this study, we evaluated the effects of sea tangle-added patty on postprandial blood glucose and lipid profiles in borderline-hypercholesterolemic (cholesterol ${\geq}200$ mg/dL) adults. Eleven subjects voluntarily participated in the experiment, and each subject provided written consent. Experimental patty (E) was made by adding 2.25 g of sea tangle powder as a substitution to 1.125 g each of pork and chicken. In the first week, 200 g of Control patty (C) was provided to each subject, who had fasted more than 12 hours. In the second week, the same amount of E patty was supplied under the same conditions. Serum glucose levels increased significantly less at 30, 60, and 120 min after consumption of E patty compared to the levels at all time points after eating C patty. Thus, the change in the area under curve (${\Delta}AUC$) of serum glucose levels through 120 minutes was lower when consuming E patty compared to C patty. Although serum C-peptide concentrations were not significantly different at all time points between the two patties, the ${\Delta}AUC$ of serum C-peptide concentrations through 120 minutes was lower when consuming E patty compared to C patty. However, there were no differences in serum levels of triglyceride, total cholesterol, LDL-C, and HDL-C at 30, 60, 120, 180, and 240 min between the two patties. Further, each ${\Delta}AUC$ of these lipid levels through 240 minutes was not significantly different between the two patties. The results indicate that sea tangle-added patty may decrease postprandial plasma glucose concentrations and reduce insulin secretion, although it might not ameliorate serum lipid profiles in adults with borderline-hypercholesterolemia.

  • Backgrounds: Blood stasis is hon as an important pathologic factor for vascular disorder in Oriental medicine. Despite its clinical importance there have been few objective tests for diagnosing blood stasis. Objectives: This study was designed to examine the relationship between blood stasis and arterial stiffness measured by cardio-ankle vascular index (CAVI). Methods: The subjects were 104 ischemic stroke patients with onset after 14 days. Their general characteristics, lipid profiles and uric acid were recorded. The degree of arterial stiffness was assessed by CAVI, and blood stasis was evaluated by diagnostic criteria. The data were analyze4 by chi-square test, student t-test, spearman correlation analysis, and pearson correlation analysis. Then, stepwise multiple logistic regression analysis was applied in order to exclude the interactions among several factors. Results: There were significant differences in right, left and higher CAVI between the blood stasis group and the non blood stasis group (p-value<0.01). Age, systolic blood pressure, triglyceride and CAVI had relationships with blood static scores. In multiple logistic regression analysis, the adjusted odds ratio of blood stasis for arterial stiffness with CAVI above 9 were 7.091 (95% confidence interval, $1.641\sim30.638$). Conclusions: The results demonstrated the relationship between blood stasis and arterial stiffness measured by CAVI. Therefore, we suggest that CAVI should be one of the objective tests for diagnosing blood stasis.

  • The purpose of this study was to investigate the relation among mineral intakes, blood pressure, and blood lipids for health management of Korean adults over 20 years old. Subjects were recruited from the district where they lived, which included rural (Yoju, n = 137) , coastal (Taean, n = 100) , and urban district (Suwon, n : 117) . hnthropornekical measurement, blood pressure measurement, dietary intake assessment using 24-hour recall method, and blood collection and analysis of blood lipids were conducted. The average age, height, weight, and BMI were 54.1 years, 168.5 cm, 67.3 kg, and 24.5 kg/$m^{2}$ for men and 53.9 years,153.8 cm,59.2 kg, and 25.0 kg/$m^{2}$ for women, respectively. The mean daily intakes of total food and energy were 1219.1 g and 1740.9 kcal for men and 1071.3 g and 1432.6 kcal for women. The mineral intakes of men and women were 448.5 mg and 400.4 mg for calcium,955.3 mg and 803.7 mg for phosphorus, 13.3 mg and 11.1 mg for iron, 4302.8 mg and 3393.3 mg for sodium,2588.0 mg and 2264.9 mg for potassium, and 8.9 mg and 7.3 mg for zinc. The daily intakes of energy, calcium, zinc, vitamin $B_{2}$, and vitamin I were $76.1\%$, $60.1\%$, $73.1\%$, $68.6\%$, and $80.4\%$ of RDAs, respectively. The intakes of energy, calcium, zinc, vitamin $B_{2}$ and I were not meet Korean RDAs. Especially, calcium, zinc, and vitamin $B_{2}$ were less than $75\%$ of RDAs. Blood pressure of the subjects was 128.3/75.5 mmHg for men and 124.1/73.7 mmHg for women. Serum lipids of men and women were 180.2 mg/dL and 184.9 mg/dL for total cholesterol, 160.8 mg/dL and 137.6 mg/dL for triglyceride,41.5 mg/dL and 44.7 mg/dL for HDL-cholesterol,106.6 mg/dL and 112.7 mg/dL for LDL-cholesterol, and 3.5 and 3.3 for atherogenic index. While serum triglyceride and AI of men was significantly higher, HDL-cholesterol was lower than those of women. In the relation among mineral intakes, blood pressure, and serum lipids, we found a significantly negative correlation of intakes of calcium, calcium/phosphorus, calcium density with systolic blood pressure. There were significantly negative correlation between intake of calcium/phosphorus and serum cholesterol, calcium density and serum cholesterol, sodium intake and serum HDL-cholesterol. Based on these results, we concluded that high intakes of calcium, calcium/phosphorus and low sodium intake were associated with favorable blood pressure and serum lipid profiles in adult men and women. • In humans, N-terminal pro-B-type natriuretic peptide (NT-proBNP) was shown to be inversely related to obesity; in addition, its association with contributing factors for obesity such as insulin, lipids, and glucose profiles has been demonstrated in the literature. However, this association between NT-proBNP and the severity of obesity has not been investigated in veterinary medicine. Our study hypothesis is that plasma levels of NT-proBNP may be related to body condition score (BCS) and contributing factors to obesity in dogs with heart diseases. To achieve our study goal, we collected blood samples from 73 client-owned dogs of small breeds at different stages of heart failure due to chronic mitral valvular insufficiency (CMVI). Fasting glucose concentrations, lipid profiles (i.e., total triglycerides [TG], total cholesterol [TC], high-density lipoprotein cholesterol [HDL-C], low-density lipoprotein cholesterol [LDL-C]), fructosamine, insulin and NT-proBNP concentrations were measured. The insulin/glucose ratio was also determined. NT-proBNP showed not only a significant correlation with the severity of CMVI related heart failure but also an inverse relationship to body condition scores (BCS), insulin plasma levels and fructosamine concentrations. We found the presence of an inverse relationship between plasma levels of NT-proBNP and the severity of obesity. In addition, NT-proBNP was associated with lower levels of contributing factors to obesity such as fructosamine and insulin, creating a possible link between the obesity and NT-proBNP in dogs with heart disease. This is also the first report demonstrating an inverse association between obesity and NT-proBNP in dogs with heart failure.

  • The purpose of this study was to investigate the effect of Monacolin-inoculated rice embryo on the body fat and serum lipid profiles in 61 obese elementary school students. The subjects divided into two groups 31 obese children had the experimental diet (rice embryo inoculated Monacolin) and 30 obese children had the control diet three times a day after meals. The mean age, height, weight, and BMI of 61 subjects were $10.5\pm0.5$ years, $143.6\pm6.8cm,\;55.0\pm8.9kg,\;and\;26.1 \pm3.9$, respectively. The changes of body fat mass (kg) in experimental group and control group after 6 weeks were -0.60kg and -0.03kg, respectively. The changes of body fat $(\%)$ in experimental group and control group after 6weeks were $-1.44\%\;and\;-0.25\%$, respectively. These changes of body fat in experimental group were significantly higher than in control group (p<0.05). The change of total-cholesterol in experimental group and control group during 6weeks were -17.52mg/dL and -1.70mg/dL, respectively. The change of LDL-cholesterol in experimental group and control group during 6 weeks were -17.06mg/dL and - 2.80mg/dL, respectively. The change of triglyceride in experimental group and control group after 6 weeks were -9.58mg/dL and 11.67mg/dL. Total-cholesterol, LDL- cholesterol and triglyceride of experimental group after 6 weeks significantly decreased compared to control group (p<0.05). After experimental diet (6 weeks), total-cholesterol was negatively correlated with body water contents, soft lean mass and fat free mass (p<0.05). Triglyceride showed a significantly positive correlation with body weight and body fat mass, however, it was negatively correlated with fat free mass (p<0.05). HDL-cholesterol showed a significantly positive correlation with fat free mass (p<0.05). These results show that Monacolin-inoculated rice embryo is effective in decreasing body fat and blood lipid in obese children.

  • Objectives: To investigate the relationship between nutrient intakes and blood biochemical parameters of Korean women classified by the Eight Constitutions. Methods: The constitutions of female subjects were determined by the methods of eight constitutional pulse formation. Anthropometric characteristics and blood biochemical parameters including fasting blood glucose (FSB), serum lipid profiles, homocysteine, and insulin were determined. Food frequency questionnaires were used to evaluate the nutrient intake of the subjects. Results: Subjects were classified as Hepatonia 16.8%, Cholecystonia 2.8%, Pancreotonia 58.9%, Pulmotonia 5.1%, Colonotonia 16.4%, respectively. Gastrotonia, Renotonia, and Vesicotonia individuals were not present in this study. BMI, total calorie and fat intakes of the Mok constitutions (Hepatonia and Cholecystonia) were higher than those of the Gum constitutions (Pulmotonia and Colonotonia) (p<0.05). Triglyceride, total cholesterol, and LDL-C concentrations for the Cholecystonia were the highest while those for the Pulmotonia (p<0.05) were lowest, which is in line with the results from nutrient intakes. Conclusions: Total calorie and fat intake among subjects with different constitutions were different. BMI and serum lipid profiles positively associated with calorie and nutrients intakes were significantly higher in Mok constitutions than Gum constitutions. Personalized diet plans seem to be needed for subjects on a special diet due to the individual constitutional differences.

  • To examine the combined effects of a high-protein diet and aerobic exercise on body weight and composition and blood lipid profiles in overweight women, 30 young women were recruited and placed into three groups: The high-protein diet and exercise group (HPE), the exercise-only group (EXO) and the control group (CON) (30$\pm$3%, 27$\pm$2%, and 29$\pm$3% body fat, respectively) for an 8-week experimental period. Daily diet included 25% isolated soybean protein (>90% protein, approximately 400 kcal) combined with each subject s usual diet for the HPE group. The exercise program consisted of aerobic-type exercises undertaken >3 times/wk and for>30 min/session at 50-60% of maximal capacity. Physical fitness, body composition, serum total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C), triglycerides (TG) and glucose were measured before and after the experiment. Maximal aerobic capacity increased by the end of experiment in both the HPE (from 27.2$\pm$3.5 to 35.l$\pm$5.9 ml/kg/min, p<0.01) and EXO (from 30.3$\pm$5.4 to 33.8$\pm$3.8 mㅣ/kg/min, p<0.05) groups. Percent body fat decreased by 3.3% (p<0.01) in the HPE group and by 1.5% (p<0.05) in the EXO group by the end of the experiment, but not in the CON group. Lower back strength and agility increased only in the HPE group. In the HPE group, TC decreased from 168$\pm$20 to 155$\pm$18 mg/dL and HDL-C increased from 57$\pm$l0 to 61$\pm$9 mg/dL in HPE (p<0.01). But TC and HDL-C did not change in the EXO and CON groups. TG and glucose did not vary among the groups. Although the EXO group showed a similar outcome to that of the HPE group, a favorable change in body composition and blood lipids as well as an improvement in aerobic capacity was more marginal in the latter group.
