Hepatoprotective and Blood Alcohol Lowering Effects of Fruit Peduncle Extract of Hovenia dulcis var. Koreana in the In Vitro and in Vivo Animal Models (헛개나무(Hovenia dulcis var. koreana) 과병 열수추출분획물의 간보호 및 혈중 알콜 저하 작용)

  • Hovenia dulcis which is distributed in Korea, China and Japan is known to show hepatoprotctive effect and reduce the acute alcohol toxicity. In this study, the hepatoprotective effect against the chemically induced experimental liver injury models and lowering effect of blood alcohol level in animal models acutely administered alcohol by the peduncle extracts of Hovenia dulcis var. koreana were investigated. HdfHW-1, and HdfM-1 which are the extracts of fruit peduncles and young branches with hot water or 70% methanol and followed with 100% methanol, were significantly reduced the $CCl_4$ or D-galactosamine/LPS induced damage in sliced liver. The hot water or methanol extracts of fruit peduncle protected dose-dependently against $CCl_4$ induced toxicity in primary hepatocyte culture and particularly, the amount of LDH release was reduced to the control level at 500 $\mu\textrm{g}$/ml of hot water extracts. HdfHW-1 also decreased the hepatotoxicity induced by $CCl_4$ in rats. The active components of HdfHW-1 seemed to be high molecular weights because 0.2 M NaCl HdfHW-1 fraction was the most effective among NaCl fractions of HdfHW-1 eluted with various concentrations of NaCl on DEAE 650C column chromatography. HdfM and HdfHW were significantly reduced the levels of blood alcohol in rats and mice administered 40% alcohol. These results indicated that the hot water or methanol extracts of fruit peduncle of Hovenia dulcis var. koreana have hepatoprotective effect and may be reduce alcohol toxicity.

Effect of Water Extract from Fugu xanthopterus on the Hyperuricemia in Alcohol-treated Rats (복어추출물이 Alcohol성 고요산혈증에 미치는 영향)

  • The present study was undertaken to investigate the effect of the water extract of the puffer fish Fugu xanthopterus(FXH) on the alcohol induced hyperuricemia. The normal group and the FXH treated group showed no sigbificant changes in the levels of blood uric acid but, the blood uric acid significantly decreased in the FXh treated rats with 100mg/kg for two weeks compared to the ethanol treated group. There were no significant changes in the activities of uricase, adenosine deaminase, guanine deaminase, and purine uncleoside phosphorylase, among all the test group. But the activitis of liver xanthine oxidase were recovered to the normal level in ethanol +FXH treated group comparing to the ethanol treated group. Furthermore, ethanol+FXH treated rats showed the similar pattern in the levels of blood uric acid and urinary allantoin with normal group. These results indicate that the decreased blood uric acid by the FXH treatment of the alcohol induced hyperuricemia rats may result from decreased activity of hepatic xanthine oxidase.

Effects of Water Extracts in fruits of Omija (Schizandra chinensis Baillon) on Alcohol Metabolism (오미자 열매의 물추출물이 알콜대사에 미치는 효과)

  • To assess the effects of water extracts in fruits of Omija (Schizandra chinensis Baillon) on alcohol metabolism, rats were orally administrated with alcohol (25% alcohol, 0.75g/200g B.W., 40% alcohol, 0.8g/200g B.W.). The level of metabolites and enzyme activities of the serum and liver were unchanged by the 25% ethanol or 40% ethanol treatment with acute orally administration. Blood alcohol level was markdely decreased by the treatment with water extracts in fruits of Omija. The serum level of Urea nitrogen, Free fatty acid, GPT and LDH were tended to decreased, level of GOT was unchanged. Contents of hepatic microsomal protein, glycogen, pyruvate in the liver were increased by water extracts in fruits of Omija. In conclusion, the present study clearly demonstrates that water extract in fruits of Omija promotes the overall metabolism and detoxication of alcohol.

  • To assess the effect of Panax ginseng on the detoxification of ethanol. we examined its effect on blood ethanol clearance in both man and experimental animals and on the rate of ethanol oxidation to carbon dioxide in experimental animals. Fourteen healthy male volunteers were subject to studies. The blood alcohol level in the test group receiving ginseng extract (3g/kg b.w.) along with alcohol (70g/65kg b.w.) was about $35\%$ lower than their control levels at 40 min after ethanol intake. When the blood alcohol level was compared on individual bases. blood alcohol concentrations in 10 subjects ranged from 32 to $51\%$ lower than their control values. The remaining 4 subjects appeared to have a high tolerance level. In experimental animals. the blood alcohol clearance was also much faster in test animals receiving ginseng along with ethanol. The rate of ethanol elimination was determined by the amount of $^{14}CO_2$ in exhaled air following the administration of [$^{14}C$] ethanol. During the first 7 1/4 hr (Phase I) after the ethanol administration. the $CO_2$ output was greater in test animals receving ginseng along with ethanol. whereas from beyond 7 1/4 hr to the near end (Phase II). the $CO_2$ output in control animals was over twice that in test animals. The present studies clearly demonstrate that ginseng promotes the overall metabolism of ethanol. resulting in an enhanced blood alcohol clearance and alcohol elimination.

  • The present study was designed to examine the effect of tested preparation, WY-91 (consisting of ginseng extract, ganoderma extract and Royal jelly) on alcohol-induced unusual metabolism(such as blood glucose(BG), triBlyceride(TG), and blood urea nitrogen(BUN) level in blood) and nrperinlental liver injury($CCl_4$ and high fat diet) in mice. WY-91 lowered the level of blood glucose (BG),triglyceride(TG) and blood urea nitrogen(BUN) induced by alcohol in a dose-dependent manner. This preparation could protect hepatic function from a damages caused by high fat diet and $CCl_4$ administration(p.o.).

Time-dependent Changes of Visual Acuity After Alcohol Ingestion (음주 후 시간경과에 따른 시력의 변화)

  • Purpose: This study was designed to investigate the time-dependent changes of visual acuity after alcohol ingestion. Methods: Both the subjective and objective refraction, distant contrast sensitivity, and three needle tests were carried out at 1 and 3 hours after alcohol ingestion. Results: It was found that the degree of myoptization increased as the blood alcohol level increased in the range of average -0.25D ~ -0.35D when tested 1 and 3 hours after alcohol ingestion. Under the same test conditions the increase in blood alcohol resulted in the reduction in the function of dynamic stereopsis. However, the variation in contrast sensitivity was found to be insignificant for all spatial frequencies. Conclusions: The optician has to be well aware of the fact that alcohol could affect the visual acuity and functional tests, therefore inquiring of the alcohol ingestion prior to visual test can be necessary.

Effects of Curcuma long gum preparation on the Blood Alcohol Level and Hangover (울금 검 제제가 알코올의 혈중농도와 숙취에 미치는 영향)

  • We conducted a single blind, randomized and crossover clinical trial in order to test the effect of curcuma longa herbal mixture on blood alcohol level and alcohol hangover in 19 healthy volunteers. The herbal mixture did not show a statistical significance in $C_{max}$, $T_{max}$ and AUC in alcohol disposition. The herbal mixture did not also ameliorate blood laboratory result after alcohol consumption. In contrast, the herbal mixture is shown effective on alcohol hangover. In behavior tests, the fewer subjects in the herbal mixture group were impaired with alcohol than in reference group. Moreover, the symptom severity score in the herbal mixture was lower than that in reference group. The symptom severity score was statistically especially in stomach pain, diarrhea, concentration disorder, memory and bad breath at drinking day and one day after drinking. These results indicate that the general symptoms seemed to be recovered as time goes on. From these results, it was suggested that the herbal mixture have a beneficial effect on modulating alcohol hangover.

Activity of Alcohol Dehydrogenase and Ethanol, Acetaldehyde Levels in Normal Adults Blood (정상인의 혈중 알코올 탈수소효소 활성도 및 에탄올, 아세트알데히드 농도에 관한 연구)

  • To identify normal levels of ethanol, acetaldehyde and alcohol dehydrogenase(ADH) activity in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood were measured in 97 subjects(male : 36, female : 61), 45 subjects(male : 21, female : 24) were not exposed to organic solvents and any other chemicals. Fifty two subjects(male : 15, female : 37) were exposed to organic solvents including toluene and xylene. The results were summerized as follows : 1. The blood ADH was not detected in exposed and non-exposed group. 2. The average blood ethanol level of non-exposed group was 0.0490 mg/dl, and exposed group was 0.0363 mg/dl. They were statistically significant(p<0.05). 3. The blood acetaldehyde levels in exposed group were significantly higher than that of non-exposed group was not statistically significant (p>0.05). 4. The average blood ethanol level of males in both groups was significantly higher than that of females, however, they were not statistically significant (p>0.05).

Effect of Ginseng on the Alcohol Metabolism in Alcohol Treated Rat (알톨 대사에 미치는 인삼의 영향)

  • After pretreatment with ginseng followed by induction of acute intoxication of alcohol, the activities of alcohol dehydrogenase (ADH), microsomal ethanol-oxidizing system (MEOS) and aldehyde dehydrogenase(Ald DH) increased respectively compared to the groups treated with alcohol alone. In case that ginseng was given to rats fed with 5% alcohol instead of water for 60 days, the activities of ADH and MEOS increased compared to the groups treated. On the contrary, the activity of Ald DH in mitochondrial fraction decreased to an extent of about 35% in chronic alcoholism, but after pretreatment of ginseng the activity was restored to the control level. On the other hand, the catalase activity was not significantly affected by either treatment. Ginseng butanol fraction significantly increased the serum isocitrate dehydrogenase activity which is inhibited by alcohol-treated in rat. Alcohol-induced lactate dehydrogenase activity was decreased to control level in liver by ginseng treatment. And the serum level of lactic acid also decreased by ginseng treatment in alcohol-intoxicated rat. Ginseng butanol fraction markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that ginseng reduced the blood concentration of uric acid on experimentally reduced hyperuricemia by alcohol treatment. Uricase activity was not affected by either treatment. Ginseng butanol fraction decreased the hepatic aniline hydroxylase activity which was induced by alcohol-treated rat. These results suggest that the treatment with ginseng can be promoted the recovery from alcohol intoxication and some therapeutic effect on alcoholinduced metabolic disease.

Blood Pressure and Serum Lipids in Relation to Serum Gamma-glutamyltransferase Activity in Men of Rural Area (농촌 지역 남성들에서 혈청 Gamma-glutamyltransferase 활성도와 혈압 및 혈청 지질의 관련성)

  • Objectives: To examine the association between serum gamma-glutamyltransferase(GGT) activity level and blood pressure, serum lipids in the male residents of rural area, we analyzed cross-sectionally the data of 379 male health examinees aged 40 years and older from rural areas in Gyeongsangnam-do prefecture in Korea. Methods: Blood pressure and concentration of serum lipids were compared between high and low level of serum GGT activity by t-test or Wilcoxon rank sum test. Possible confounding effects of age, body mass index and coffee drinking were adjusted by analysis of covariance. Results: Adjusted values of systolic and diastolic blood pressure were higher in the group of high level of serum GGT activity in non-drinkers(P=0.055 and P=0.074 respectively) and drinkers(P=0.284 and P=0.398) of alcohol. Adjusted serum total cholesterol level was also higher in the group of high level of serum GGT activity in non-drinkers(P=0.052) but not in drinkers(P=0.981) of alcohol. In serum triglyceride, adjusted level was significantly higher in the group of high level of serum GGT activity in both non-drinkers(P=0.035) and drinkers(P=0.002) of alcohol. Conclusions: These results suggest the association of serum GGT activity and serum triglyceride, total cholesterol, or blood pressure in non-drinkres of alcohol.
