헛개나무(Hovenia dulcis var. koreana) 과병 열수추출분획물의 간보호 및 혈중 알콜 저하 작용 (Hepatoprotective and Blood Alcohol Lowering Effects of Fruit Peduncle Extract of Hovenia dulcis var. Koreana in the In Vitro and in Vivo Animal Models)

  • Hovenia dulcis which is distributed in Korea, China and Japan is known to show hepatoprotctive effect and reduce the acute alcohol toxicity. In this study, the hepatoprotective effect against the chemically induced experimental liver injury models and lowering effect of blood alcohol level in animal models acutely administered alcohol by the peduncle extracts of Hovenia dulcis var. koreana were investigated. HdfHW-1, and HdfM-1 which are the extracts of fruit peduncles and young branches with hot water or 70% methanol and followed with 100% methanol, were significantly reduced the $CCl_4$ or D-galactosamine/LPS induced damage in sliced liver. The hot water or methanol extracts of fruit peduncle protected dose-dependently against $CCl_4$ induced toxicity in primary hepatocyte culture and particularly, the amount of LDH release was reduced to the control level at 500 $\mu\textrm{g}$/ml of hot water extracts. HdfHW-1 also decreased the hepatotoxicity induced by $CCl_4$ in rats. The active components of HdfHW-1 seemed to be high molecular weights because 0.2 M NaCl HdfHW-1 fraction was the most effective among NaCl fractions of HdfHW-1 eluted with various concentrations of NaCl on DEAE 650C column chromatography. HdfM and HdfHW were significantly reduced the levels of blood alcohol in rats and mice administered 40% alcohol. These results indicated that the hot water or methanol extracts of fruit peduncle of Hovenia dulcis var. koreana have hepatoprotective effect and may be reduce alcohol toxicity.

복어추출물이 Alcohol성 고요산혈증에 미치는 영향 (Effect of Water Extract from Fugu xanthopterus on the Hyperuricemia in Alcohol-treated Rats)

  • The present study was undertaken to investigate the effect of the water extract of the puffer fish Fugu xanthopterus(FXH) on the alcohol induced hyperuricemia. The normal group and the FXH treated group showed no sigbificant changes in the levels of blood uric acid but, the blood uric acid significantly decreased in the FXh treated rats with 100mg/kg for two weeks compared to the ethanol treated group. There were no significant changes in the activities of uricase, adenosine deaminase, guanine deaminase, and purine uncleoside phosphorylase, among all the test group. But the activitis of liver xanthine oxidase were recovered to the normal level in ethanol +FXH treated group comparing to the ethanol treated group. Furthermore, ethanol+FXH treated rats showed the similar pattern in the levels of blood uric acid and urinary allantoin with normal group. These results indicate that the decreased blood uric acid by the FXH treatment of the alcohol induced hyperuricemia rats may result from decreased activity of hepatic xanthine oxidase.

오미자 열매의 물추출물이 알콜대사에 미치는 효과 (Effects of Water Extracts in fruits of Omija (Schizandra chinensis Baillon) on Alcohol Metabolism)

  • To assess the effects of water extracts in fruits of Omija (Schizandra chinensis Baillon) on alcohol metabolism, rats were orally administrated with alcohol (25% alcohol, 0.75g/200g B.W., 40% alcohol, 0.8g/200g B.W.). The level of metabolites and enzyme activities of the serum and liver were unchanged by the 25% ethanol or 40% ethanol treatment with acute orally administration. Blood alcohol level was markdely decreased by the treatment with water extracts in fruits of Omija. The serum level of Urea nitrogen, Free fatty acid, GPT and LDH were tended to decreased, level of GOT was unchanged. Contents of hepatic microsomal protein, glycogen, pyruvate in the liver were increased by water extracts in fruits of Omija. In conclusion, the present study clearly demonstrates that water extract in fruits of Omija promotes the overall metabolism and detoxication of alcohol.

인삼의 알콜해독 효과 (Effects of Panax ginseng on Alcohol Detoxification)

  • 건강한 성인남자 14명을 대상으로 하여 혈중 알콜해독에 미치는 인삼의 효과 관찰 실험을 실시하였다. 술과 인삼을 동시에 마시는 시험군과 술만 마시는 대조군을 설정하여, 술을 마신후 40분이 경과하였을 때 혈중알콜농도를 측정하였다. 술 (70g/65kg 체중)과 동시에 인삼 (3g/65kg체중)을 먹었을 때의 혈중알콜농도 ($0.11\%$)는 대조군의 혈중알콜농도 ($0.18\%$)의 약 $6.5\%$이었고, 또 혈중알콜농도를 각 개인별로 비교해보면 실험대상자 14명중 인삼과 술을 동시에 마셨을때의 혈중 알콜농도가 술만 마셨을 때의 혈중 알콜농도의 $65\%{\~}49\%$에 불과한 사람이 10명이나 되었다. 인삼의 알콜배출속도에 미치는 영향을 관찰하기 위한 방사능이 표시된 알콜을 실험동물에 투여한 후 알콜의 대사물인 $^{14}CO_2$가 호흡을 통하여 배출되는 량을 측정비교 하였다. 알콜 투여후 2-7시간 사이의 $^{14}CO_2$ 배출속도는 알콜의 대사 및 배출속도를 증가시켜 알콜해독을 촉진한다는 것을 설명해주고 있다.

실험적 간장해에 대한 WY-91제제의 영향 (The Effect of WY-91 Preparation on Experimental Liver Injury in Mice)

  • The present study was designed to examine the effect of tested preparation, WY-91 (consisting of ginseng extract, ganoderma extract and Royal jelly) on alcohol-induced unusual metabolism(such as blood glucose(BG), triBlyceride(TG), and blood urea nitrogen(BUN) level in blood) and nrperinlental liver injury($CCl_4$ and high fat diet) in mice. WY-91 lowered the level of blood glucose (BG),triglyceride(TG) and blood urea nitrogen(BUN) induced by alcohol in a dose-dependent manner. This preparation could protect hepatic function from a damages caused by high fat diet and $CCl_4$ administration(p.o.).

음주 후 시간경과에 따른 시력의 변화 (Time-dependent Changes of Visual Acuity After Alcohol Ingestion)

  • 목적: 알코올 섭취 후 시간경과에 따른 시력변화를 알아보고자 하였다. 방법: 음주 1시간 및 3시간 후의 자각적 타각적 굴절검사, 원거리 대비감도 검사, 그리고 세막대 입체시 검사를 실시하였다. 결과: 혈중 알코올 농도의 상승과 비례하여 평균 -0.25D ~ -0.35D의 근시성 변화를 일으켰고, 동적입체시 기능이 저하되는 것으로 나타났다. 그러나 대비감도는 모든 공간주파수에서 큰 변화가 없는 것으로 나타났다. 결론: 알코올이 일반적인 시력검사 및 기능검사에 영향을 끼친다는 사실을 숙지하고 피검자의 검사 전 알코올 음용에 대해 문진할 필요성이 있다.

울금 검 제제가 알코올의 혈중농도와 숙취에 미치는 영향 (Effects of Curcuma long gum preparation on the Blood Alcohol Level and Hangover)

  • We conducted a single blind, randomized and crossover clinical trial in order to test the effect of curcuma longa herbal mixture on blood alcohol level and alcohol hangover in 19 healthy volunteers. The herbal mixture did not show a statistical significance in $C_{max}$, $T_{max}$ and AUC in alcohol disposition. The herbal mixture did not also ameliorate blood laboratory result after alcohol consumption. In contrast, the herbal mixture is shown effective on alcohol hangover. In behavior tests, the fewer subjects in the herbal mixture group were impaired with alcohol than in reference group. Moreover, the symptom severity score in the herbal mixture was lower than that in reference group. The symptom severity score was statistically especially in stomach pain, diarrhea, concentration disorder, memory and bad breath at drinking day and one day after drinking. These results indicate that the general symptoms seemed to be recovered as time goes on. From these results, it was suggested that the herbal mixture have a beneficial effect on modulating alcohol hangover.

정상인의 혈중 알코올 탈수소효소 활성도 및 에탄올, 아세트알데히드 농도에 관한 연구 (Activity of Alcohol Dehydrogenase and Ethanol, Acetaldehyde Levels in Normal Adults Blood)

  • To identify normal levels of ethanol, acetaldehyde and alcohol dehydrogenase(ADH) activity in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood were measured in 97 subjects(male : 36, female : 61), 45 subjects(male : 21, female : 24) were not exposed to organic solvents and any other chemicals. Fifty two subjects(male : 15, female : 37) were exposed to organic solvents including toluene and xylene. The results were summerized as follows : 1. The blood ADH was not detected in exposed and non-exposed group. 2. The average blood ethanol level of non-exposed group was 0.0490 mg/dl, and exposed group was 0.0363 mg/dl. They were statistically significant(p<0.05). 3. The blood acetaldehyde levels in exposed group were significantly higher than that of non-exposed group was not statistically significant (p>0.05). 4. The average blood ethanol level of males in both groups was significantly higher than that of females, however, they were not statistically significant (p>0.05).

알톨 대사에 미치는 인삼의 영향 (Effect of Ginseng on the Alcohol Metabolism in Alcohol Treated Rat)

  • After pretreatment with ginseng followed by induction of acute intoxication of alcohol, the activities of alcohol dehydrogenase (ADH), microsomal ethanol-oxidizing system (MEOS) and aldehyde dehydrogenase(Ald DH) increased respectively compared to the groups treated with alcohol alone. In case that ginseng was given to rats fed with 5% alcohol instead of water for 60 days, the activities of ADH and MEOS increased compared to the groups treated. On the contrary, the activity of Ald DH in mitochondrial fraction decreased to an extent of about 35% in chronic alcoholism, but after pretreatment of ginseng the activity was restored to the control level. On the other hand, the catalase activity was not significantly affected by either treatment. Ginseng butanol fraction significantly increased the serum isocitrate dehydrogenase activity which is inhibited by alcohol-treated in rat. Alcohol-induced lactate dehydrogenase activity was decreased to control level in liver by ginseng treatment. And the serum level of lactic acid also decreased by ginseng treatment in alcohol-intoxicated rat. Ginseng butanol fraction markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that ginseng reduced the blood concentration of uric acid on experimentally reduced hyperuricemia by alcohol treatment. Uricase activity was not affected by either treatment. Ginseng butanol fraction decreased the hepatic aniline hydroxylase activity which was induced by alcohol-treated rat. These results suggest that the treatment with ginseng can be promoted the recovery from alcohol intoxication and some therapeutic effect on alcoholinduced metabolic disease.

농촌 지역 남성들에서 혈청 Gamma-glutamyltransferase 활성도와 혈압 및 혈청 지질의 관련성 (Blood Pressure and Serum Lipids in Relation to Serum Gamma-glutamyltransferase Activity in Men of Rural Area)

  • 농촌 지역 남성 주민들을 대상으로 혈청 GGT 활성도와 혈압 및 혈청 지질의 관련성을 검토해 보기 위하여 경상남도 내 농촌 지역인 울주군 두동면과 창녕군 장마면에 거주하는 40세 이상의 남성으로서 1997년과 2000년에 실시한 주민 건강 검진에 참여하였던 사람들 중 379명의 자료를 분석하였다. 혈청 GGT 활성도의 수준을 중앙값을 기준으로 이분하여 두군에서의 수축기및 이완기 혈압, 혈청총 콜레스테롤, HDL-콜레스테롤의 평균치를 비교하였고 중성지방은 중앙값을 비교하였으며 통계학적 유의성은 t-검정과Wilcoxon rank sum 검정을 이용하여 검토하였다. 교란변수로 작용할 가능성이 있는 연령, 신체비만지수와 커피음용에의한영향은공분산분석으로보정하였다. 수축기 혈압과 이완기 혈압은 비음주자에서 GGT 활성도 수준이 높은 경우에 더 높은 값을 보였고 통계학적 유의수준에 접근하였다(P=0.055와 P=0.074). 음주자에서도 역시GGT 활성도가 높은 경우에 더 높은 값을 보였으나 통계학적 유의성은 없었다. 혈청 총 콜레스테롤은 비음주자에서 GGT 활성도가 높은 경우에 더 높은 값을 보였으며 통계학적 유의수준에 접근하였다(P=0.052). 음주자에서는 GGT 활성도 수준에 따라 비슷한 값을 보였다. 중성지방은 비음주자와 음주자 모두에서 GGT 활성도가 높은 경우에 더 높은 값을 보였다(P=0.035와 P=0.002). 이상의 소견으로 보아 혈청 GGT 활성도의 증가는 비음주자에서 혈청 중성지방, 총 콜레스테롤 및 혈압의 상승과 관련성이 있음을 암시한다.
