Saccharification of Fagopyrum esculentum by Amylase Treatments increases Phenolic Compound Content and Antioxidant Activity (아밀라아제 처리에 의한 메밀의 당화가 페놀화합물 함량과 항산화 활성의 증가에 미치는 영향)

  • This study evaluated the functional characteristics of saccharified buckwheat (Fagopyrum esculentum) following ${\alpha}$-amylase, ${\beta}$-amylase or glucoamylase treatment based on changes in soluble solid contents, rutin and quercetin contents, total polyphenols and DPPH radical scavenging activity. The results showed that the amylase treatments significantly influenced the saccharification time. Additionally, total polyphenol, rutin, and quercetin contents increased during the saccharification process; increase in phenolic compounds induced antioxidant activity. The present study demonstrated that buckwheat has a higher amount of functional compounds and higher antioxidant activity after saccharification. These results show that buckwheat saccharification can be used to increase antioxidant capacity and functional value for applications in functional food industries.

Strain Development for the Over-production of Alkaline Protease from Vibrio metschnikovii by Molecular Evolution (분자진화 기술을 통한 Vibrio metschnikovii 유래 고활성 알칼리성 단백질 분해효소 생산균주 개발)

  • Alkaline protease-overproducing strains of Vibrio metschnikovii were developed by using the molecular evolution from the classical mutants V. metschnikovii L12-23, N4-8, and KS1. Each vapK (Vibrio alkaline protease K) was obtained from the genomic DNAs of mutants by PCR to carry out the DNA shuffling. The modified vapK-1 obtained by DNA shuffling was used again as a template for the error-prone PCR to make the vapK-2. Both genes were cloned in the plasmid pKF3 to construct the recombinant plasmids which have one or two copies of the modified genes. The recombinant plasmids were back-transformed to V. metschnikovii KS1 to construct recombinant V. metschnikovii that expresses the alkaline protease. About 3.9-fold more protease activity was measured in the strain which has the plasmid containing two copies of vapK-2 when compared to strain KS1. When compared to wild type V. metschnikovii RH530, 43-fold more activity was achieved. Comparison of amino acids among vapK, vapK-1, and vapK-2 revealed that the active sites was highly conserved and not changed. However, many amino acids except the active sites were changed. These results suggested that the changes in amino acids might play an important role in the increase of protease activity by allowing the easy access of substrate to active sites of the protease. The fermentation of alkaline protease from the V. metschnikovii KS1 harboring the plasmid that contains two copies of vapK-1 showed the possibility of this strain to be used as industrial producer.

Anti-Oxidative and Anti-Inflammatory Effect of Water Extract from Perillae Semen in RBL-2H3 Cells (RBL-2H3 세포에서 차조기씨 물 추출물의 항산화 및 항염증 효과)

  • The present study was to investigate anti-oxidative and anti-inflammatory activity of Perillae semen in RBL-2H3 basophilic leukemia cells. Inhibitory effect of Perillae semen onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activities in vitro. Anti-inflammatory actions of Perillae semen extracts (100, 250, $500\;{\mu}g/mL$) were assessed by testing their effects on the degranulation of mast cells. For this, $\beta$-hexosaminidase released from RBL-2H3 cells was used and proinflammatory cytokines were measured by an ELISA kit. Our results indicated that Perillae semen water extracts effectively inhibited free radical generation. At the concentration of $500\;{\mu}g/mL$ of water extract, the degranulation of RBL-2H3 cells were inhibited by 42.1%. The IgE-antigen complex increased the accumulation of IL-4 and TNF-$\alpha$ secretion in RBL-2H3 cells and treatments with 250 and $500\;{\mu}g/mL$ of Perillae semen extracts suppressed the IgE induced secretion of IL-4 and TNF-$\alpha$ protein by 20.5, 26.9% and 14.5, 16.5% respectively. We observed that Perillae semen water extract reduced $\beta$-hexosaminidase, IL-4, and TNF-$\alpha$ secretion in RBL-2H3 cells. These results provide that Perillae semen may be beneficial in the treatment of allergic inflammatory disease.

Evaluation and Comparison of Meteorological Drought Index using Multi-satellite Based Precipitation Products in East Asia (다중 위성영상 기반 강우자료를 활용한 동아시아 지역의 기상학적 가뭄지수 비교 분석)

  • East Asia, which includes China, Japan, Korea, and Mongolia, is highly impacted by hydroclimate extremes such drought, flood, and typhoon recent year. In 2017, more than 18.5 million hectares of crops have been damaged in China, and Korea has suffered economic losses as a result of severe drought. Satellite-derived rainfall products are becoming more accurate as space and time resolution become increasingly higher, and provide an alternative means of estimating ground-based rainfall. In this study, we verified the availability of rainfall products by comparing widely used satellite images such as Climate Hazards Groups InfraRed Precipitation with Station (CHIRPS), Global Precipitation Climatology Centre (GPCC), and Precipitation Estimation From Remotely Sensed Information Using Artificial Neural Networks-Climate Data Record (PERSIANN-CDR) with ground stations in East Asia. Also, the satellite-based rainfall products were used to calculate the Standardized Precipitation Index (SPI). The temporal resolution is based on monthly images and compared with the past 30 years data from 1989 to 2018. The comparison between rainfall data based on each satellite image products and the data from weather station-based weather data was shown by the coefficient of determination and showed more than 0.9. Each satellite-based rainfall data was used for each grid and applied to East Asia and South Korea. As a result of SPI analysis, the RMSE values of CHIRPS were 0.57, 0.53 and 0.47, and the MAE values of 0.46, 0.43 and 0.37 were better than other satellite products. This satellite-derived rainfall estimates offers important advantages in terms of spatial coverage, timeliness and cost efficiency compared to analysis for drought assessment with ground stations.

The Flow Behavior of Skin Collagen (피부조직 콜라겐의 유동 특성)

  • To obtain the basic information for the effective use of collagen, the flow behavior of collagen extracted from skin tissue was studied. The viscosity of collagen varied with sex, age and the kinds of collagen by extraction method. Regardless of the kinds of collagen, the viscosity of collagen extracted from $6{\sim}12$ week old rat was relatively high. In case of the same age, the viscosity showed higher in female than in male rat and in acid soluble collagen than in insoluble collagen. The solution of the collagen showed the characteristics of Bingham plastic and thixotropic fluid, and the viscosity varied distinctly with temperature, pH, ethanol concentration and collagen concentration. As collagen concentration increased to 6%, the consistency of acid soluble- and insoluble collagen showed a tendency to increase linearly(r = 0.972 for acid soluble collagen, r = 0.957 for insoluble collagen). In that range of collagen concentration, the increasing velocity of consistency was higher in acid soluble collagen than in insoluble collagen. The consistency of collagen solution was decreased according to temperature rising. In case of acid soluble collagen, the consistency is decreased abruptly between $30{\sim}40^{\circ}C$. According to pH variation, the consistency of acid soluble collagen showed biphasic phenomenon, though the consistency of insoluble collagen was found not to be influenced by pH. The consistency of acid soluble- and insoluble collagen according to ethanol concentration showed high between $40{\sim}60%$ of ethanol concentration.

Antioxidant and Whitening Effects of Sorbus commixta HEDL Cortex Extract (정공피 추출물의 항산화 활성 및 미백효과에 관한 연구)

  • This study was performed to assess the antioxidant activities and whitening effects of Sorbus commixta HEDL cortex on melanin synthesis. The whitening effects of Sorbus commixta HEDL cortex water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effects of Sorbus commixta HEDL cortex water extracts on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects of Sorbus commixta HEDL cortex onto free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. Our results indicated that Sorbus commixta HEDL cortex water extracts effectively inhibited free radical generation. In DPPH radical scavenging activity, Sorbus commixta HEDL cortex water extracts had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Sorbus commixta HEDL cortex suppressed the expression of tyrosinase, TRP-1 and TRP-2 in B16BL6 melanoma cells. These results show that Sorbus commixta HEDL cortex inhibited melanin production on the melanogenesis. The underlying mechanism of Sorbus commixta HEDL cortex on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, TRP-2 expression. We suggest that Sorbus commixta HEDL cortex may be contain new natural active ingredients for antioxidant and whitening cosmetics.

Influence of Ammonium Phosphate on Mycelial Morphology during Submerged Cultivation of Ganoderma lucidum (영지의 액체배양에 있어서 균사체 형태에 미치는 Ammonium Phosphate의 영향)

  • The mycelial morphology during submerged cultivation of Ganoderma ludium using by air-lift fermenter system were analyzed by image processing system and the characterization of mycelial morphology were investigated. In submerged culture using medium with different ammonium phosphate concentrations, the various morphological forms of G. lucidum mycelium were observed. The filamentous forms such as non-branched long filamentous mycelium, non-branched short mycelium, branched long filamentous mycelium, branched short mycelium, entangled mycelium and clump were observed, and also, and also, the pelleted forms such as smooth pellet, rough pellet and hollow rough pellet were observed. The mycelial morphology was changed from the filamentous to the pelleted forms by addition of ammonium phosphate. The fractal dimensions of pelleted and filamentous forms were 1.05 and 1.3, respectively, while the fractal dimension of mixtures of pelleted and filamentous forms was 1.16. Therefore, the fractal dimension was found to be more effective index for the detection of the mycelial morphology and morphological change during batch cultivation. The circularity was also found to be useful for evaluating the surface growth of pelleted mycelium.

Modeling and Optimization of Dough Properties Using Response Surface Design (반응표면분석법을 이용한 반죽물성의 모델링 및 최적화)

  • The purpose of this study was to optimize dough properties using response surface methodology (RSM) and to demonstrate the performances of dough prepared under optimized conditions. Dough mixed with yeast, margarine, salt, sugar and wheat flour was prepared by fermentation process. Hardness, cohesiveness and springiness of dough were selected as critical quality attributes. The critical formulations (yeast and water) and process (fermentation time) variables were selected as critical input variables based on preliminary experiment. Box-Behnken design (BBD) was used as RSM. As a result, the quardratic, the squared and the linear model respectively provided the most appropriate fit ($R^2$>90) and had no significant lack of fit (p>0.05) on critical quality attributes (hardness, cohesiveness and springiness). The accurate prediction of dough characteristics was possible from the selected models. It was confirmed by validation that a good correlation was obtained between the actual and predicted values. In conclusion, the methodologies using RSM in this study might be applicable to the optimization of fermented foods containing various wheat flour and yeast.

Anti-Obesity Effects of Foeniculum fructus Water Extract (회향종자(Foeniculum fructus) 물 추출물의 항비만 효과)

  • We investigated the anti-obesity effects of Foeniculum fructus water extract on body weight, epididymal adipocyte size, plasma lipid levels and activities of key enzymes such as lipoprotein lipase (LPL), acyl-CoA synthetase (ACS) in high fat diet-induced obese mice. Experimental groups were normal diet group (ND), high fat diet group (HFD), high fat diet with 0.05% orlistat group (HFDO), and high fat diet with 0.5% Foeniculum fructus group (HFDF). Eleven-weeks feeding with HFD resulted in significant increase in lipid levels, body weight, liver and epididymal adipose tissue weight, compared with the ND group. Diet containing Foeniculum fructus water extract significantly reduced plasma total cholesterol, triglyceride and glucose concentrations as well as body weight, liver and epididymal adipose tissue weights. Plasma LDL cholesterol levels were significantly lower in the HFDF group than those in HFDO group. LPL activities elevated by a high fat diet were significantly decreased by Foeniculum fructus water extract administration. ACS activities decreased in the high fat diet group and markedly increased in the Foeniculum fructus water extract administered group. All things considered, Foeniculum fructus water extract efficiently inhibits the inflow of fatty acid into the cell, and activates metabolic process that uses fatty acids flowing as an energy source. Thus, Foeniculum fructus water extract may have great potential as a novel anti-obesity agent.
