Evaluation on the adsorption and desorption capabilities of filter media applied to the nonpoint source pollutant management facilities (비점오염 저감시설에 적용되는 여재의 흡착 및 탈착 능력 평가)

  • Urbanization causes many environmental, hydrological and ecological problems such as distortion of the natural water circulation system, increase in nonpoint source pollutants in stormwater runoff, degradation of surface water quality, and damage to the ecosystem. Due to the increase in impervious surface by urbanization, developed countries apply low impact development (LID) techniques as important alternatives to reduce the impacts of urbanization. In Korea, LID techniques were employed since 2012 in order to manage nonpoint source pollutants. LID technology is a technique for removing pollutants using a variety of physical, chemical and biological mechanisms in plants, microorganisms and filter media with the reduced effluence of stormwater runoff by mimicking natural water circulation system. These LID facilities are used in a variety of filter media, but an assessment has not been carried out for the comprehensive comparison evaluation of adsorption and desorption characteristics for the pollutant removal capacity. Therefore, this study was conducted to analyze the adsorption and desorption characteristics of various filter media used in the LID facilities such as sand, gravel, bioceramic, wood chips and bottom ash etc. in reducing heavy metals(Pb, Cu). In this study, the adsorption affinity for Pb in all filter media was higher than Cu. Pseudo second order equation and Langmuir-3 isotherm are more applicable in the adsorption kinetic model and adsorption isotherm model, respectively. As a result of the desorption experiment, the filter media does not exceed KSLT which is the hazardous substance leaching limit, showing the capability of the filter media in LID. The bioceramic and woodchip as filter medias were evaluated and exhibited excellent adsorption capacity for Pb.

Enhancement of Iron Oxidation Rate by Immobilized Cells in Chemo-biological Process for $H_2S$ Removal (화학.생물학적 황화수소 제거 공정에 있어서 고정화 세포를 이용한 철산화 속도 증진)

  • This study was aimed to enhance the Fe(II) oxidation rate using immobilized cells of Thiobacillus ferroxidans. For this purpose, a medium for the minimization of jarosite formation was developed first. Secondly, cell immobilization in celite beads was carried out. And then, repeated-batch and continuous operatons of Fe(II) oxidation by using immobilization cells were performed. In a series of flask cultures, three types of media were tested: media with a much lower salt concentration than that of the 9K medium; media which contained different nitrogen sources from that of the 9K medium, that is $(NH_4)_2HPO_4$, $NH_4Cl and HNO$_3$; media which contained $(NH_4)_2HPO_4$ as nitrogen and phosphate source, but without $K_2HPO_4$ as nitrogen and phosphate source in the 9K medium. As a result, the M16 medium which contained 3 g/L of $(NH_4)_2HPO_4$ as nitrogen and phosphate source was found to be the optimal one. It sustained good cell growth allowing no jarosite formation. In the repeated-batch operations, the rate of Fe(II) oxidation gradually increased to reach a maximum value as the batch was repeated. As a result of repeated-batch operations. a maximum Fe(II) oxidation rate was 2.33 g/L . h. In the continuous operations, the iron oxidation rate could be increased to 2.14 g/L .h at a dilution rate of 0.25 $h^{-1}$ which is greater than the maximum specific growth rate (0.12 $h^{-1}$) of the bacteria.

Biological Functions of N- and O-linked Oligosaccharides of Equine Chorionic Gonadotropin and Lutropin/Chorionicgonadotropin Receptor

  • Members of the glycoprotein family, which includes CG, LH, FSH and TSH, comprise two noncovalently linked $\alpha$- and $\beta$-subunits. Equine chorionic gonadotropin (eCG), known as PMSG, has a number of interesting and unique characteristics since it appears to be a single molecule that possesses both LH- and FSH-like activities in other species than the horse. This dual activity of eCG in heterologous species is of fundamental interest to the study of the structure-function relationships of gonadotropins and their receptors. CG and LH $\beta$ genes are different in primates. In horse, however, a single gene encodes both eCG and eLH $\beta$-subunits. The subunit mRNA levels seem to be independently regulated and their imbalance may account for differences in the quantities of $\alpha$ - and $\beta$ -subunits in the placenta and pituitary. The dual activities of eCG could be separated by removal of the N-linked oligosaccharide on the $\alpha$-subunit Asn 56 or CTP-associated O-linked oligosaccharides. The tethered-eCG was. efficiently secreted and showed similar LH-like activity to the dimeric eCG. Interestingly, the FSH-like activity of the tethered-eCG was increased markedly in comparison with the native and wild type eCG. These results also suggest that this molecular can implay particular models of FSH-like activity not LH-like activity in the eCG/indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion. A single-chain analog can also be constructed to include additional hormone-specific bioactive generating potentially efficacious compounds that have only FSH-like activity. The LH/CG receptor (LH/CGR), a membrane glycoprotein that is present on testicular Leydig cells and ovarian theca, granulosa, luteal, and interstitial cells, plays a pivotal role in the regulation of gonadal development and function in males as well as in nonpregnant and pregnant females. The LH/CGR is a member of the family of G protein-coupled receptors and its structure is predicted to consist of a large extracellular domain connected to a bundle of seven membrane-spanning a-helices. The LH/CGR phosphorylation can be induced with a phorbol ester, but not with a calcium ionophore. The truncated form of LHR also was down-regulated normally in response to hCG stimulation. In contrast, the cell lines expressing LHR-t63I or LHR-628, the two phosphorylation-negative receptor mutant, showed a delay in the early phase of hCG-induced desensitization, a complete loss of PMA-induced desensitization, and an increase in the rate of hCG-induced receptor down-regulation. These results clearly show that residues 632-653 in the C-terminal tail of the LHR are involved in PMA-induced desensitization, hCG-induced desensitization, and hCG-induced down-regulation. Recently, constitutively activating mutations of the receptor have been identified that are associated with familial male-precocious puberty. Cells expressing LHR-D556Y bind hCG with normal affinity, exhibit a 25-fold increase in basal cAMP and respond to hCG with a normal increase in cAMP accumulation. This mutation enhances the internalization of the free and agonist-occupied receptors ~2- and ~17-fold, respectively. We conclude that the state of activation of the LHR can modulate its basal and/or agonist-stimulated internalization. Since the internalization of hCG is involved in the termination of hCG actions, we suggest that the lack of responsiveness detected in cells expressing LHR-L435R is due to the fast rate of internalization of the bound hCG. This statement is supported by the finding that hCG responsiveness is restored when the cells are lysed and signal transduction is measured in a subcellular fraction (membranes) that cannot internalize the bound hormone.

  • 외국에서 ASM 모델의 BNR 적용 연구결과를 국내 하수에 적용하기에는 하수농도, 온도, 슬러지농도 등이 국내와는 달라 적용상 무리가 있다. 본 연구에서 BNR 시뮬레이션을 위한 입력 자료로 활용되는 인자들은 IAWPRC task group에서 제안하는 값들을 사용하되 국내 하수성상에서 필요로 하는 인자들은 직접 실험을 통하여 부분적으로 구해냄으로써 모델 시뮬레이션의 신뢰도를 높이고자 하였다. F/M비의 변화량과 1/SRT과의 관계로부터, 종속영양미생물 생산계수 $Y_H$값을 구한결과, 0.40mg VSS/mg COD였다. 이것을 ASM No.2d에 적용하기 위하여 mg cell COD formed/mg COD oxidized 단위로 환산한 결과 0.58을 나타냈다. H 하수처리장의 1차 침전지 하수를 이용하여 호기성상태에서 OUR Test를 통한 미생물에 의한 유기물 섭취시 산소섭취율 변화를 측정하였다. 호기성상태와 무산소상태에서 구한 쉽게 분해되는 용존성유기물(Ss)값을 비교해보면 각각 35.5mg/L와 39.9mg/L로 약간의 차이는 있으나 유사한 값을 보여주고 있다. 시뮬레이션을 위한 동력학적 계수 중 무산소 상태에서 종속영양미생물의 ${\mu}_{max,H}$$3.56d^{-1}$로 나타났고, 호기성상태에서는 구하면 ${\mu}_{max,H}$$4.2d^{-1}$로 산출되었다. 종속영양미생물의 사멸계수 $b_H$를 구하기 위한 실험에서 초기 OUR의 10%이내가 될 때까지 걸린 시간은 7일정도가 걸렸으며, 사멸률 $b_H$$0.043hr^{-1}$로 나타났다. 독립영양미생물의 최대비성장률 ${\mu}_{max,A}$는 최대암모니아 섭취률을 이용하여 구한 결과 $0.65d^{-1}$로 나타났다.EX>$60%{\sim}87%$가 수심 10m 이내에 분포하였고, 녹조강과 남조강이 우점하는 하절기에는 5m 이내에 주로 분포하였다. 취수탑 지점의 수심이 연중 $25{\sim}35m$를 유지하는 H호의 경우 간헐식 폭기장치를 가동하는 기간은 물론 그 외 기간에도 취수구의 심도를 표층 10m 이하로 유지 할 경우 전체 조류 유입량을 60% 이상 저감할 수 있을 것으로 조사되었다.심볼 및 색채 디자인 등의 작업이 수반되어야 하며, 이들을 고려한 인터넷용 GIS기본도를 신규 제작한다. 상습침수지구와 관련된 각종 GIS데이타와 각 기관이 보유하고 있는 공공정보 가운데 공간정보와 연계되어야 하는 자료를 인터넷 GIS를 이용하여 효율적으로 관리하기 위해서는 단계별 구축전략이 필요하다. 따라서 본 논문에서는 인터넷 GIS를 이용하여 상습침수구역관련 정보를 검색, 처리 및 분석할 수 있는 상습침수 구역 종합정보화 시스템을 구축토록 하였다.N, 항목에서 보 상류가 높게 나타났으나, 철거되지 않은 검전보나 안양대교보에 비해 그 차이가 크지 않은 것으로 나타났다.의 기상변화가 자발성 기흉 발생에 영향을 미친다고 추론할 수 있었다. 향후 본 연구에서 추론된 기상변화와 기흉 발생과의 인과관계를 확인하고 좀 더 구체화하기 위한 연구가 필요할 것이다.게 이루어질 수 있을 것으로 기대된다.는 초과수익률이 상승하지만, 이후로는 감소하므로, 반전거래전략을 활용하는 경우 주식투자기간은 24개월이하의 중단기가 적합함을 발견하였다. 이상의 행태적 측면과 투자성과측면의 실증결과를 통하여 한국주식시장에 있어서 시장수익률을 평균적으로 초과할 수 있는 거래전략은 존재하므로 이러한 전략을 개발 및 활용할 수 있으며, 특히, 한국주식시장에 적합한 거래전략은 반전거래전략이고, 이 전략의 유용성은 투자자가 설정한 투자기간보다 더욱 긴 분석기간의 주식가격정보에 의하여 최대한 발휘될 수 있음을 확인하였다.(M1), 무역적자의 폭, 산업의 생산

  • This experiment was conducted to study germination characteristics and the decrease cause of germination percent in Angelica gigas Nakai seed. The emergence percent of winter sowing was higher than that of spring sowing as 66.6% and 41.1%, respectively, and the first emergence date was also earlier in winter sowing. The seed germination speed, percent and coefficent showed the highest value at $20^{\circ}C$ of incubation temperature, but lower value at $10^{\circ}C\;and\;30^{\circ}C$. The water uptake speed was increased along with increasing water temperature. The weight of imbibed seed at germination was 3.4times higher based on the weight of intact dry seed and 2.3times on removal of seed coat. In terms of length of seed was large, the germination percent was higher. The germination percent of brown colored seeds showed higher value than that of green colored seeds. The prolonged storage period decreased germination percent. When A.gigas seeds stored at room temperatue for 2years, the seeds were lost their viability. The biological inhibition effect of methanol, water and ether extract on the germination and growth of A.gigas and lettuce seed showed the highest value in the methanol extract, followed by water extract and the least in ether extract.

Evaluation of Butyltin Compounds and its Distribution Among Seawater, Sediment and Biota from the Kwangyang Bay (광양만내 유기주석화합물의 해수, 퇴적물, 생물중 농도 및 분포 상관관계)

  • Seawater, sediment and biota in the Kwangyang Bay were analyzed by gas chromatography/quartz furnace atomic absorption spectroscopy (GC-QFAAS) to investigate concentrations and distribution pattern of butyltin compounds (TBT, DBT, MBT) during February, April and July, 1996, Marine biota analyzed were Tapes japcnicus and Crassostrea gigas. The concentrations of tributyltin (TBT) in seawater were in the range of ND-15.7 ng/L for the surface and ND-68.5 ng/L for the bottom. The highest concentration of TBT in seawater was detected in April for the both, surface and bottom water. The maximum value of $TBT_{(bottom)}/TBT_{(surface)}$, 3.6 in April showed the increased input of TBT from the surface water in April compared to February (2.1) and July (0.9). The concentrations of TBT in the sediment were in the range of ND-8.5 ng/g dry wt. The highest concentration of TBT in the sediment was measured in July, This result seems to attributed to the removal of TBT from water column via sorption onto particulate matters to the relatively undisturbed underlying sediment and increased input of TBT by increased fluxes of detritus of marine plankton after spring bloom, in July. The mean values of partitioning coefficient ($K_d$) of TBT between seawater and sediment were $3.0\times10^3$(February), $7.4\times10^3$(April) and $9.4\times10^3$(July). The concentrations of TBT in biosamples were in the range of ND-93.30 ng/g dru wt. (T. japonicus) and ND-138.53 ng/g dry wt. (C. gigas). The seasonal variation of TBT contents in biota was remarkable. The $K_d$ (biological concentration factor) was $7-41\times10^3$ for T. japonicus. and $5-34\times10^3$ for C. gigas. The measured TBT concentrations in seawater in the study area was sufficient to cause the imposex of shellfish and to retard the growth of aquatic organisms including oyster upon chronic exposure.

Radium Isotope Ratio as a Tracer for Estimating the Influence of Changjiang Outflow to the Northern Part of the East China Sea (라듐 동위원소 방사능비를 추적자로 사용한 동중국해 북부 해역에서 장강 유출수의 영향 추정)

  • In order to understand the present environmental condition and future impingement of Changjiang(Yangtze River) outflow upon the adjacent seas after the scheduled completion of the Sanxia (Three Gorges) Dam in 2009, we tried to estimate the mixing ratios among surface waters of three end-members: Changjiang Water (CW), Kuroshio Water (KW), and East China Sea Water (ECSW) using $^{228}Ra/^{226}Ra$ activity ratio and salinity as tracers. Water samples were collected from 32 stations in November 2005 (R/V Tamgu 3), from 20 stations in July 2006 (R/V Ocean 2000) and from 17 stations in August 2006 (R/V Ieodo) in the northern part of the East China Sea. Radium isotopes in ~300 liters of surface seawater were extracted onboard by filtering through manganese impregnated acrylic fibers and following coprecipitation as $Ba(Ra)SO_4$. Activities of radium isotopes were determined by a high purity germanium detector. Results show that the fraction of CW was in the range of 1-23% in the study area, while KW was in the range of 0-30 % and ECSW 58-100 %. The eastward plume of Changjiang outflow, commonly observed in satellite images during summer and also displayed by the eastward-decreasing CW fraction in this study, could be attributed to Ekman transport caused by the SE monsoon prevailing in this region during summer. Results of this study showed that in the drought season, there was a little or no fraction of CW in the study area. Concentration of dissolved inorganic nitrogen (DIN) showed strong positive relationship with the fraction of CW, suggesting Changjiang as the major source of nitrogen. The mixing curve of DIN indicates the removal of nitrate by biological uptake during the mixing of CW with ambient seawater in the study area.

Performance Characteristics of Agitated Bed Manure Composting and Ammonia Removal from Composting Using Sawdust Biofiltration System (교반식 축분 퇴비화 및 톱밥 탈취처리 시스템의 퇴비화 암모니아 제거 성능)

  • Sawdust biofiltration is an emerging bio-technology for control of ammonia emissions including compost odors from composting of biological wastes. Although sawdust is widely used as a medium for bulking agent in composting system and for microbial attachment in biofiltration systems, the performance of agitated bed composting and sawdust biofiltration are not well established. A pilot-scale composting of hog manure amended with sawdust and sawdust biofiltration systems for practical operation were investigated using aerated and agitated rectangular reactor with compost turner and sawdust biofilter operated under controlled conditions, each with a working capacity of approximately $40m^3\;and\;4.5m^3$ respectively. These were used to investigate the effect of compost temperature, seed germination rate and the C/N ratio of the compost on ammonia emissions, compost maturity and sawdust biofiltration performance. Temperature profiles showed that the material in three runs had been reached to temperature of 55 to $65^{\circ}C$ and above. The ammonia concentration in the exhaust gas of the sawdust biofilter media was below the maximum average value as 45 ppm. Seed germination rate levels of final compost was maintained from 70 to 93% and EC values of the finished compost varied between 2.8 and 4.8 ds/m, providing adequate conditions for plant growth.

Esterification Reaction of Animal Fat for Bio-diesel Production (바이오디젤 생산을 위한 동물성 오일의 에스테르화 반응)

  • In this study, the production of bio-diesel from animal oil by esterification and trans-esterification was investigated. There were three different extraction methods for oil extraction from raw animal fat. Heterogeneous catalysts such as Amberlyst-15 and Amberlyst BD-20 and a homogeneous catalyst such as sulfuric acid were used for esterification. Among three catalysts, the removal efficiency of Free Fatty Acid (FFA) was the highest in sulfuric acid. Response surface method was carried out to find the optimal esterification condition of sulfuric acid and methanol. After the esterification under the optimal condition, this animal fat was used for the trans-esterification. Animal oil used for trans-esterification was below 1% of FFA content and 0.09% of water content. The catalysts for trans-esterification were KOH, NaOH and $NaOCH_3$. To investigate the effects of catalyst type and amount on trans-esterification, The amount of catalyst were changed with 0.3, 0.6 and 0.9 wt%. The molar ratio of methanol/oil was changed with 4, 6, 9 and 12. The amount of catalyst was fixed to 0.8 wt%. The KOH catalyst showed the highest FAME conversion for trans- esterification, and the optimal methanol/oil weight ratio was 6. In the experiments of various catalysts and methanol molar ratios, the highest content of FAME is 96%. However, this FAME content was below Korean bio-diesel standard which is 96.5% of FAME content. After distillation, FAME content increased to 98%.
