Bioavailability of slow-desorbable naphthalene in a biological air sparging system

  • The bioavailability of sorbed organic contaminants is one of the most important factors used to determine their fate in the environment. This study was conducted to evaluate the bioavailability of slow-desorbable naphthalene in soils. An air sparging system was utilized to remove dissolved (or desorbed) naphthalene continuously and to limit the bacterial utilization of dissolved naphthalene. A biological air sparging system (air sparging system with bacteria) was developed to evaluate the bioavailability of the slow-desorption fraction in soils. Three different strains (Pseudomonas putida G7, Pseudomonas sp. CZ6 and Burkholderia sp. KM1) and two soils were used. Slow-desorbable naphthalene continuously decreased under air sparging; however, a greater decrease was observed in response to the biological air sparging system. Enhanced bioavailability was not observed in the Jangseong soil. Overall, the results of this study suggests that the removal rate of slow-desorbable contaminants may be enhanced by inoculation of degrading bacteria into an air sparging system during the remediation of contaminated soils. However, the enhanced bioavailability was found to depend more on the soil properties than the bacterial characteristics.

Characterization of Phage-Resistant Strains Derived from Pseudomonas tolaasii 6264, which Causes Brown Blotch Disease

  • Pseudomonas tolaasii 6264 is a representative strain that causes bacterial blotch disease on the cultivated oyster mushroom, Pleurotus ostreatus. Bacteriophages are able to sterilize the pathogenic P. tolaasii strains, and therefore, they can be applied in creating disease-free mushroom cultivation farms, through a method known as "phage therapy". For successful phage therapy, the characterization of phage-resistant strains is necessary, since they are frequently induced from the original pathogenic bacteria in the presence of phages. When 10 different phages were incubated with P. tolaasii 6264, their corresponding phage-resistant strains were obtained. In this study, changes in pathogenic, genetic, and biochemical characteristics as well as the acquired phage resistance of these strains were investigated. In the phylogenetic analyses, all phage-resistant strains were identical to the original parent strain based on the sequence comparison of 16S rRNA genes. When various phage-resistant strains were examined by three different methods, pitting test, white line test, and hemolytic activity, they were divided into three groups: strains showing all positive results in three tests, two positive in the first two tests, and all negative. Nevertheless, all phage-resistant strains showed that their pathogenic activities were reduced or completely lost.

New Species and Eight Undescribed Species Belonging to the Families Aspergillaceae and Trichocomaceae in Korea

  • During a survey of fungal diversity associated with insects, mud, soil, and freshwater niches in different areas in Korea, nine interesting fungal strains were isolated. Based on their morphological characteristics and molecular phylogeny analyses, using a combined data set of b-tubulin (BenA), calmodulin (CaM), and second largest subunit of RNA polymerase (RPB2) sequences, the strains CNUFC AM-44, CNUFC JCW3-4, CNUFC S708, CNUFC WT202, CNUFC AS1-29, CNUFC JCW3-5, CNUFC JDP37, and CNUFC JDP62 were identified as Aspergillus alabamensis, A. floridensis, A. subversicolor, Penicillium flavigenum, P. laevigatum, P. lenticrescens, Talaromyces adpressus, and T. beijingensis, respectively. The strain CNUFC JT1301 belongs to series Westlingiorum in section Citrina and is phylogenetically related to P. manginii. However, slow growth when cultivated on CYA, MEA, CREA is observed and the property can be used to easily distinguish the new species from these species. Additionally, P. manginii is known to produce sclerotia, while CNUFC JT1301 strain does not. Herein, the new fungal species is proposed as P. aquadulcis sp. nov. Eight species, A. alabamensis, A. floridensis, A. subversicolor, P. flavigenum, P. laevigatum, P. lenticrescens, T. adpressus, and T. beijingensis, have not been previously reported in Korea. The present study expands the known distribution of fungal species belonging to the families Aspergillaceae and Trichocomaceae in Korea.

Reaction Characteristics of Piggery Wastewater for Biological Nutrient Removal (생물학적 영양염류 제거를 위한 돈사폐수의 반응 특성)

  • This study was performed to investigate the reaction characteristics of piggery wastewater for biological nutrient removal. The reaction characteristics were discussed the fraction of organics, the behavior of nitrogen, nitrification, denitrification, and the behavior of phosphorus. The fraction of readily biodegradable soluble COD was 11-12 percent. The ammonia nitrogen was removed via stripping, nitrification, autotrophic cell synthesis, and heterotrophic cell synthesis. The removal percents by each step were 12.1%, 68.9%, 15.0%, and 4.0%, respectively. Nitrification inhibition of piggery wastewater was found to occur at an influent volumetric loading rate over 0.2 NH$_{3}$-N kg/m$^{3}$/d. Denitrification rates were the highest in the raw wastewater and the lowest in the anaerobic effluent. The denitritation of piggery wastewater came out to be possible, and the rate of organic carbon consumption decreased about 10 percent. The phosphorus removed was released in the form of ortho-p in the aerobic fixed biofilm reactor, it was caused by autooxidation. The synthesis and release of phosphorus were related to the ORP and the boundary value for the phase change was about 170mV. In the synthesis phase, the phosphorus removal rate per COD removed was 0.023mgP$_{syn}$/mgCOD$_{rem}$. The phosphorus contents of the microorganism were 4.3-6.0% on a dry weight basis.

Wang-ime Oreum Flora on Jeju Island (제주도 왕이메오름의 식물상)

  • This study investigated the flora of the Wang-ime oreum located on Seogwipo-si, Jeju-do, to basic data for the Jeju Island plant distribution. A total of 366 taxa were found with 94 families, 240 genera, 358 species, 1 subspecies, 6 varieties, and 1 forma. The floristic target species in Korea appeared as 2 taxa of grade V, 13 taxa of grade IV, 46 taxa of grade III, 5 taxa of grade II, and 49 taxa of grade I. There were 14 taxa for naturalized plants. The different plant life forms that appeared were large ground plants (M)(54 taxa), small land plants (N) (52 taxa), epiphyte (E) (6 taxa), indicator plants (Ch) (4 taxa), and semi-aquatic plants (H) (168 taxa), There were 34 and 48 taxa of plants(G) and annuals (Th) respectively. From these results, Wang-ime oreum, which is adjacent to ranches and grasslands, has little artificial interference, as more plants are distributed, and fewer naturalized plants are found than in Suwolbong and Dangsanbong. Accordingly, each oreum plays an important role in the flora of Jeju-do, thus a management plan tailored to the characteristics of the volcano is necessary.

Identification of 12 radiation-resistant bacterial species in the phylum Proteobacteria new to Korea

  • In 2019, after a comprehensive investigation of indigenous prokaryotic species in Korea, a total of 12 bacterial strains assigned to the phylum Proteobacteria were isolated from soil. With the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to independent, predefined bacterial species. This study identified two species in the family Burkholderiaceae, one species in the family Comamonadaceae, two species in the family Oxalobacteraceae, one species in the family Micrococcaceae, one species in the family Bradyrhizobiaceae, one species in the family Methylobacteriaceae, one species in the family Rhizobiaceae, one species in the family Rhodocyclaceae, and one species in the family Sphingomonadaceae. There is no official report about these 12 species in Korea, so are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are also described in the species description section.

Seventeen Unrecorded Species from Gayasan National Park in Korea

  • Macrofungi play important roles in forest ecology as wood decayers, symbionts, and pathogens of living trees. For the effective forest management, it is imperative to have a comprehensive overview of macrofungi diversity in specific areas. As a part of the National Institute of Biological Resources projects for discovering indigenous fungi in Korea, we collected macrofungi in Gayasan National Park from 2017 to 2018. These specimens were identified based on morphological characteristics and sequence analysis of internal transcribed spacer (ITS) or the nuclear large subunit rRNA (LSU) region. We discovered 17 macrofungi new to Korea: Butyrea japonica, Ceriporia nanlingensis, Coltricia weii, Coltriciella subglobosa, Crepidotus crocophyllus, Cylindrobasidium laeve, Fulvoderma scaurum, Laetiporus cremeiporus, Lentinellus castoreus, Leucogyrophana mollusca, Marasmius insolitus, Nidularia deformis, Phaeophlebiopsis peniophoroides, Phanerochaete angustocystidiata, Phlebiopsis pilatii, Postia coeruleivirens, and Tengioboletus fujianensis. We described their detailed morphological characteristics.

Meta-analysis Study on Microenvironmental Characteristics of Radon Concentration in Korea

  • Objective: The present study analyzed domestic and overseas academic journals to understand the research status and characteristics of radon concentration distribution in Korea in accordance with environmental conditions. Methods: As part of the meta-analysis, pooled average concentration was calculated using an inverse variance-weighted average of the arithmetic means and standard deviations among the investigated values. Using the obtained pooled average concentration, a Monte-Carlo simulation was performed to increase the reliability of the occurrence possibility of the calculated concentration distribution. A total of 38 research articles were selected, including 27 articles published in domestic academic journals and 11 articles published in foreign academic journals. Results: The comparison results showed differences in radon concentration distribution in accordance with regional and topographical characteristics. Conclusion: Currently, even though research into radon is steadily picking up the pace in Korea much remains to be done. Additional research is thus needed to establish a baseline for radon emissions in Korea.

Formal characteristics of an unrecorded freshwater diatom (Bacillariophyceae) in Korea

  • Diatoms, which are reported to have over 18,000 species worldwide and approximately 2,400 species in Korea, can be found in various environments including freshwater, seawater, and wetlands. They are particularly valuable for understanding global environmental changes throughout history due to their ability to maintain their shape for extended periods of time. Instead of collecting floating diatoms using nets, low-layer substrates such as gravel and leaves, as well as sedimentary surface layers, were gathered in order to identify attached diatom species. This is because attached diatoms demonstrate higher species diversity compared to floating diatoms. In this study, seven previously unrecorded diatoms were discovered in various domestic freshwater environments. Two species were found in reservoirs (Eunotia yanomami, Gomphonella pseudookunoi), two in parasitic cones (Eunotia karveerensis, Luticola minor), two in rivers (Cavinula maculata and Prestauroneis integra), and one (Surirella brebissonii var. kuetzingii) in a lagoon. The shapes, structures, and morphological characteristics of each diatom were identified using electron microscopy.

Monitoring of Pesticide Residues in Leafy Vegetables Collected from Wholesale and Traditional Markets in Cheongju (청주지역 도매 및 재래시장 유통 엽채류 중 잔류농약 모니터링)

  • In order to monitor the residual characteristics of the pesticides in leafy vegetables selling at wholesale markets and traditional markets in Cheongju, a total of 180 samples of 15 leafy vegetables, such as broccoli, celery, chard, chicory, Chinese vegetable, Chwinamul, crown daisy, Korean cabbage, leek, lettuce, perilla leaves, Shinsuncho, spinach, welsh onion and young radish, were purchased from the wholesale markets and traditional markets in June and August in 2010 and the pesticide residues in them were analyzed by multiresidue analysis method using GLC, HPLC and GC-MSD. Seven pesticides were detected from 12 samples out of total 180 samples collected, representing detection rate was 6.7%. In case of the samples collected from markets in June, four pesticides including tefluthrin were detected from six samples and in case of the samples collected from markets in August, three pesticides including pendimethalin were detected from three samples. The MRL-exceeding rate of pesticides detected from leafy vegetables was 0.6%. The pesticide exceeded its MRL was azoxystrobin detected from crown daisy and many pesticides were not registered to the crops, excepting that azoxystrobin detected from Chwinamul and tefluthrin from leek. Estimated daily intakes (EDIs) of the pesticides detected from leafy vegetables were less than 7% of their acceptable daily intakes (ADIs), representing that residue levels of the pesticides detected were evaluated as safe.
