자외선A로 손상된 DNA의 회복과 DNCB에 의한 알러지성 접촉피부염에 대한 표고버섯과 침향 추출 혼합물의 효과 (The Effects of Lentinula edodes and Aquilariae agallocha Extracts Combination on the Repair of UVA-Damaged DNA and DNCB-Induced Allergic Dermatitis)

  • The effects of extracts from Lentinula edodes (L. edodes) and Aquilariae agallocha (A. agallocha) on the DNA damage response in ultraviolet A (UVA)-exposed HaCaT cells and on the allergic contact dermatitis caused by 2,4-dinitro-chlorobezene (DNCB) were investigated. When UVA-exposed cells were incubated for 24 hours in medium containing L. edodes or A. agallocha extract, the level of 8-OHdG and CPD decreased in a concentration-dependent manner. The combined treatment with both extracts potentiated the decrease in UVA-induced 8-OHdG and CPD levels as compared with those following treatment with a single extract. In addition, the two extracts showed preventive effects against the UVA-induced reduction in collagen levels. Furthermore, the blood levels of IgE, IL-6, and histamine decreased more significantly upon combined treatment with L. edodes and A. agallocha extracts as compared with those following treatment with single extracts in DNCB-induced allergic contact dermatitis in the ICR mouse. The results of the present study suggest that the components with in the extracts of L. edodes and A. agallocha can help to prevent of UVA-induced genomic instability via a decrease in DNA damage, and to decrease the DNCB-induced allergic dermatitis via modulation of relevant proteins including IgE and IL-6. Further study is needed to clarify the purified components related to the preventative effects of the two extracts against UVA- or DNCB-induced genomic damage.

Biological Control of Pests of Non-Mulberry Silkworms and Its Host Plants in India

  • The protection of silkworm and its host plants from various kinds of pests parasite and predator is a chronic problem in sericulture. Silkworms and its primary food plants are heavily damaged by large number of pest. The major pests of primary tasar food plants (Terminalia arjuna and Terminalia tomentosa) are the gall insect (Trioza fletcheri minor). Various species of aphids (Eutrichosiphum sp.) have been recorded to damage oak tasar food plants whereas muga silkworm host plants (Machilus bombycina and Litsaea polyantha) are generally attacked by stem bores (Zeuzera multistrigata). Castor (Ricinus communis) is one of the primary host plant of eri silkworm and extensive damage is caused by the castor white fly (Trialeurodes ricini). Insects pests are major enemies of silkworms. Parasites (Blepharipa zebina, Exorista bombycis, Apateles glomeratus), predators (Canthecona furcellata, Sycanus collaris, Hierodulla bipapilla), wasps (Vespa orientalix) and ants (Oecophylla smargdina) continues to cause damage to silk industry. It is estimated that the losses due to parasites and predators are to an extent of 15-20 percent and varies from crop to crop. The complexities in the behaviour and life cycle of pest population existing in semi ecosystem warrant a special attention for their effective management specially in changing scenario for our modern sericulture. Though use of synthetic insecticides has provided us with effective control of almost all major pests and predators, yet their undesirable side effects limit their continued use. Biological control is one of the most important method which can be used to control the pests, parasites and predators population in sericulture. Various potential parasitoids, which can be utilized as an agent of biological control in sericulture have been screened. The natural enemies of the uzi fly (E. bombycis and B. zebina ) are already present in the nature. Nesolynx thymus, Trichria sp., Splangia endius, Dirhinus sp., Trichopria sp., Trichomalopsis apanteloctena and Pediobius sp. are the major parasitoids effective against uzi fly pupa. The scelionid Psix striaticeps and Trissolcus sp. are the Potential egg Parasitoids against stink bug (Canthecona furcellata). Various other native natural potential parasitoids have been screened and suitable strategies have been developed to check the population of pest insect in sericulture.

Role of oxygen in plasma induced chemical reactions in solution

  • Many researchers have paid attention to the studies on the interaction between non-thermal plasma and aqueous solutions for biomedical applications. The gas composition in the plasma is very important. Oxygen and nitrogen are the main gases of interest in biological applications. Especially, we focus on the oxygen concentration. In this experiment, we studied the role of oxygen concentration in plasma induced chemical reactions in solution. At first, the amount of ions are measured according to changing the oxygen concentration. And we checked the relationship between these ions and pH value. Secondly, when the oxygen concentration is changed, it identified the type and amount of radical generated by the plasma. In order to confirm the effect of these chemical property change to biological material, hemoglobin and RBCs are chosen. RBCs are one of the common basic biological cells. Thirdly, when plasma treated according to oxygen concentration in nitrogen feeding gas, oxidation of hemoglobin and RBC is checked. Finally, membrane oxidation of RBC is measured to examine the relation between hemoglobin oxidation and membrane damage through relative hemolysis and Young's modulus. Our results suggest that reactive species generated by the plasma differsdepending on the oxygen concentration changes. The pH values are decreased when oxygen concentration increased. OH decrease and NO increase are also observed. These reactive species makes change of chemical properties of solution. We also able to confirm that the difference in these reactive species to affect the oxidation of the Hb and RBCs. The Hb and RBCs are more oxidized with the high oxygen concentration conditions. But membrane is damaged more by plasma treatment with only nitrogen gas. It is shown that red blood cells membrane damage and oxidation of hemoglobin are not directly related.

허혈-재관류 유도 SH-SY5Y 모델에서 미토콘드리아 매개 Apoptosis 기전 제어를 통한 초석잠 추출물의 세포보호 효과 (Stachys sieboldii M iq. Protects SH-SY5Y Cells Against Oxygen-Glucose Deprivation/Reoxygenation-Induced Injury by Inhibition of Mitochondrion-Mediated Apoptosis Pathway)

  • Oxygen glucose deprivation/re-oxygenation (OGD/R) induces neuronal injury via mechanisms that are believed to mimic the pathways associated with brain ischemia. Stachys sieboldii Miq. (Chinese artichoke), which has been extensively used in oriental traditional medicine to treat of ischemic stroke; however, the role of S. sieboldii Miq. (SSM) in OGD/R induced neuronal injury is not yet fully understood. The present research is aimed to investigate the protective effect and possible mechanisms of SSM extract treatment in an in vitro model of OGD/R to simulate ischemia/reperfusion Injury. Pretreatment of these cells with SSM significantly attenuated OGD/R-induced production of reactive oxygen species (ROS) by increasing GPx, SOD, and decreasing MDA. SSM decreased mitochondrial damage caused by OGD/R injury and inhibited the release of cyt-c from mitochondrion to cytoplasm in SH-SY5Y cells. Furthermore, neuronal cell apoptosis caused by OGD/R injury was inhibited by SSM, and SSM could decrease apoptosis by increasing ratio of Bcl-2/Bax and inhibiting caspase signaling pathway in SH-SY5Y cells. SSM demonstrated a neuroprotective effect on the simulated cerebral ischemia in vitro model, and this effect was the inhibition of mitochondria-mediated apoptosis pathway by scavenging of ROS generation. Therefore, SSM may be a promising neuroprotective strategy against ischemic stroke.

팥(Phaseolus angularis) 열수 추출물의 산화적 DNA와 세포 손상 억제 효과 (Inhibitory Effect of Red Bean (Phaseolus angularis) Hot Water Extracts on Oxidative DNA and Cell Damage)

  • 본 연구에서는 열수 팥 추출물이 hydroxyl 라디칼에 의해 유도되는 산화적 스트레스에 미치는 영향을 알아보기 위하여 항산화활성과 DNA 및 세포의 산화적 손상 억제 효과를 조사하였다. 팥 열수 추출물의 DPPH 라디칼과 hydroxyl 라디칼의 제거능은 다소 낮았으나, $Fe^{2+}$-chelating과 과산화수소 제거효과는 높게 나타나 활성산소의 생성을 억제하는 데 효과적인 것으로 확인되었다. 또한 팥 열수 추출물의 in vitro DNA cleavage, DNA migration 및 H2AX의 인산화비 억제활성은 높은 활성을 보여주고 있어 라디칼에 의한 DNA 손상 억제에 효과적으로 작용하였다. 또한 지질과산화와 p21의 발현율을 통해 세포의 산화적 손상에 미치는 영향을 살펴보면 지질과산화 억제능과 p21의 발현율에 매우 효과적으로 작용하고 있어 라디칼에 의한 산화적 스트레스로부터 세포를 보호할 것으로 생각된다.

알코올로 유도된 간손상 모델 HepG2/2E1 세포에서 오리나무 잎 추출물의 간보호효과 (Hepatoprotective Effects of the Extracts of Alnus japonica Leaf on Alcohol-Induced Liver Damage in HepG2/2E1 Cells)

  • 활성산소종은 미토콘드리아 대사활동을 통해 생성되는 반응성이 강한 산소 함유 분자로서 특히 간에서 복잡한 세포 분자와 반응하여 산화적 스트레스를 유발하는 물질이다(Choi et al., 2012). 에탄올 대사 과정에서는 ROS가 생성되어 산화스트레스를 유발하며, 과도한 에탄올은 ROS를 제거하는 항산화 시스템을 억제함으로써 지방, 단백질 또는 DNA와 같은 생체 분자에 산화 손상을 유발하여 세포를 손상시킨다(Albano, 2006; Koch et al., 2004). 본 연구에서는 오리나무 잎 추출물(AJL)에서 알코올(EtOH)로 유발된 산화적 스트레스에 대한 간세포 손상의 보호 효과를 평가하기 위해 HepG2/2E1 세포에서 EtOH와 AJL 및 대조군silymarin을 각각 처리한 후 세포생존율을 평가하고 세포독성이 없는 범위를 설정하여 활성산소종 생성량, 항산화 관련 지표(DPPH, ABTS, MDA, GSH) 및 간 손상 지표(AST 및 ALT)를 측정하였다. 알코올에 대한 간 보호 효과에서는 알코올만 처리한 군의 세포생존율이 53.8%로 감소하였으나 AJL을 함께 처리 군에서 세포생존율이 85.1%의 유의적인 증가로 간 보호효과를 확인하였다. 다음으로 AJL에서 세포 내 ROS 생성 억제능은 에탄올 처리군에서 정상군에 비해 유의적으로 ROS 생성이 증가하였으나 AJL 농도별 처리시 정상군 수준으로 ROS 생성을 현저하게 감소시켰다. MDA 함량은 에탄올 처리군에서 무처리군에 비해 유의적으로 증가하였으나 AJL 처리시 농도 의존적으로 에탄올 처리군에 비해 유의적인 차이를 보이며 MDA 함량을 감소시켰다. GSH 함량은 에탄올 처리군에서 정상군보다 유의적으로 함량이 감소하였으나 AJL 농도별 처리군 모두 에탄올 처리군에 비해 GSH 함량을 증가시켰다. 또한 간손상 지표로 활용되는 AST 및 ALT 활성 변화 확인 시, 에탄올 처리에 따른 간세포 손상군에 비하여 AJL 처리군에서는 농도 의존적으로 현저하게 감소시켰으며, 대조군silymarin과 비교하여 유사한 감소된 결과를 관찰하였다. 이상의 결과로부터 종합해 볼때AJL은 간손상에 대한 보호 및 간손상 억제 가능성을 기대할 수 있으며, 간기능 개선 효과에 대한 건강기능식품개발의 기초자료로서 활용될 수 있을 것이라 사료된다.

꽈리허리노린재(Acanthocoris sordidus) 추출물이 산화적 손상에 미치는 억제 효과 (Inhibitory Effect of Extract from Acanthocoris sordidus on Oxidative Damage)

  • 본 연구는 꽈리허리노린재(Acanthocoris sordidus) 추출물이 산화적 스트레스에 의해 유발되는 세포와 DNA의 손상 억제력을 조사하였다. 꽈리허리노린재 추출물의 DPPH 유리 라디칼과 수산화 라디칼 제거능은 양성 대조군에 비해 각각 48.9%, 37.8%로 나타났으며, $Fe^{2+}$-chelating 효과는 80.0%로 조사되었다. 꽈리허리노린재 추출물이 활성산소에 의해 유도되는 세포손상에 미치는 억제 효과를 조사하기 위해 지질과산화의 상대적 수준과 p21 단백질의 발현율을 조사해보면 꽈리허리노린재 추출물은 라디칼 처리군에 비해 지질과산화를 거의 완벽하게 억제하고 있으며, p21 단백질의 발현은 대조군의 92.4%로 회복되는 것으로 조사되었다. 또한 꽈리허리노린재 추출물의 DNA 분절화 억제 활성은 대조군에 비해 53.3%로 나타나 산화적 스트레스에 의해 유발되는 DNA 분절화를 효율적으로 억제하고 있으며, H2AX 단백질의 인산화비는 라디칼 처리군의 39.0%에 해당하는 수준으로 조사되어 꽈리허리노린재 추출물은 히스톤 단백질의 인산화를 매우 효율적으로 억제하는 것으로 확인되었다. 이러한 결과로 보아 꽈리허리노린재 추출물은 활성산소에 대한 항산화 효과뿐만 아니라 세포와 DNA의 손상을 억제하는데 효과적인 것으로 사료된다.

현대사회의 환경변화에 따른 Bio-Terror의 위협요인 연구 (A Study on the Threat of Biological Terrorism in modern society)

  • In recent years, there is growing concern about the potential use of biological agents in war or acts of terrorism accompanied an increased realization that rapid preparedness and response are needed to prevent or treat the human damage that can be caused by these agents. The threat is indeed serious, and the potential for devastating numbers of casualties is high. The use of agents as weapons, even on a small scale, has the potential for huge social and economic disruption and massive diversion of regional and national resources to combat the threat, to treat primary disease, and to clean up environmental contamination. Biological weapons are one of weapons of mass destruction (or mass casualty weapons, to be precise. since they do not damage non-living entities) that are based on bacteria, viruses, rickettsia, fungi or toxins produced by these organisms. Biological weapons are known to be easy and cheap to produce and can be used to selectively target humans, animals, or plants. Theses agents can cause large numbers of casualties with minimal logistical requirements (in wide area). The spread of disease cannot be controlled until there is awareness of the signs of infection followed by identification of agents; and if the organism is easily spread from person to person, as in the case of smallpox, the number of casualties could run into the tens of thousands. Biological weapons could be used covertly, there can be a lot of different deployment scenarios. A lot of different agents could be used in biological weapons. And, there are a lot of different techniques to manufacture biological weapons. Terrorist acts that make use of Biological Agents differ in a number of ways from those involving chemicals. The distinction between terrorist and military use of Biological Weapon is increasingly problematic. The stealthy qualities of biological weapons further complicate the distinction between terrorism and war. In reality, all biological attacks are likely to require an integrated response involving both military and civilian communities. The basic considerations when public health agencies establish national defence plan against bioterrorism must be 1) arraying various laws and regulations to meet the realistic needs, 2)education for public health personnels and support of concerned academic society, 3)information collection and cooperative project with other countries, 4)Detection and surveillance(Early detection is essential for ensuring a prompt response to biological or chemical attack, including the provision of prophylactic medicines, chemical antidotes, or vaccines) and 5) Response(A comprehensive public health response to a biological or chemical terrorist event involves epidemiologic investigation, medical treatment and prophylaxis for affacted persons, and the initiation of disease prevention or environmental decontamination measures). The purpose of this paper is providing basic material of preparedness and response for biological terrorism in modern society.
