Bacillus subtilis S1-0210 was selected as a biological agent against Botrytis cinerea in strawberry. The isolate inhibited mycelial growth of B. cinerea in vitro tests. A wettable powder formulation of B. subtilis S1-0210 significantly reduced infection rates with lower than 5%, compared with higher than 70% of infection rates in untreated control. The formulation showed 85 to 89% control efficacies of gray mold incidences on fruits of strawberry in pots. Pre-treatment of the agent was more effective in controlling gray mold on fruits and leaves than post-treatment at the early stage of disease development. The formulation also showed 70% control efficacy of gray mold incidence on fruits of strawberry in a field trial. The results indicate that B. subtilis S1-0210 in the wettable powder formulation may be a potential biocontrol agent to control gray mold on strawberry.
Biological control of soilborne plant pathogens by the addition of antagonistic microorganisms to the soil may offer a practical supplement or alternative to existing disease management strategies that depend heavily on chemical pesticides. Soil amendment with antagonistic microbes was non-effective because of high cost, low efficacy, and inconvenient usage on the treatment course. Therefore, seed coating formulation for the application of biological seed treatments has been being to apply successful disease suppression for many important crops. The objectives of this study were to investigate the optimal condition for the spore production of biocontrol agent Bacillus subtilis YBL-7 and the liquid coating formulation that contained a suspension of a proper aqueous binder, as well as a ground fine solid particulate material. The maximum yield has been obtained from 60 hrs-old culture at 30$\circ$C in spore forming (SF) medium containing 0.8% nutrient broth, 0.05% yeast extract, 10$^{-1}$ M MgCl$^{2}$, 10$^{-4}$ M MnCl$^{2}$, 10$^{-5}$ M dipicolinic acid, and pH 6.5. The optimal condition of dried spore preparation was achieved when cells of B. subtilis YBL-7 was heat-dried with 50$\circ$C for 2 hrs.

Monochoria vaginalis var. plantaginea (Pontederiaceae) is one of the most problematic weed in the rice field in Korea. Tanysphyrus (Tanysphyroides) major was selected as a potential biological control agent for M. vaginalis. Continuous rearing of T. (T.) major was carried out from 2006 to 2007, and its morphological characteristics and ecological characteristics were investigated. This species has a single generation per year, over-wintering as an adult stage. The emergence of adults starts in later June and last until September. These observations indicate that T. (T.) major takes $22{\pm}0.7$ days to develop from egg to adulthood. Host specificity test showed that finally selected this species was suitable candidates for the biological control of M. vaginalis var. plantaginea since it showed negative host specificity against major 60 test crops.
Two strains of Cucumber mosaic virus (CMV) isolated in Kuwait were confirmed their infectivity based on symptomatology and host range on different cultivars of tomato (Lycopersicon esculentum), tobacco(Nicotiana tabacum L.) and squash (Cucurbita pepo). The pattern of symptoms differed for the two CMV strains in tomato and tobacco, showing severe stunting and mosaic symptoms with one strain designated KU2, and almost symptomless with the other strain designated KU1. A satellite RNA 5 (sat-RNA) was found to be associated with the KU1 strain and was characterized as a benign viral satellite RNA. Using reverse transcription and polymerase chain reaction (RT-PCR) with sat-RNA specific primers, an amplified PCR product of about 160bp was determined and analyzed by gel electrophoresis. This naturally occurring benign viral satellite RNA was successfully used as a biological control agent to protect tomato plants against the severe KU2 strain. Tomato plants grown in plant-growth chambers, were preinoculated with KU1 containing the benign viral satellite and then challenge inoculated with the severe KU2 strain at different time intervals. All plants challenged three weeks after preinoculation showed nearly complete protection from subsequent infection by the severe strain. This biological control technology using plant viruses was found protective and could be successfully established sooner after the preinoculation.
Biocontrol of plant pathogens provides an alternative means of reducing the incidence of plant diseases without the negative aspects of chemical pesticides. Nowdays, as the resistant fungi about the chemical fungicides have revealed and the concern of environment has increased, the biological control of phytopathogenic fungi by the antagonistic microorganisms is very much indispensable. For the selection of strong antagonistic bacterium for biological control agent of rice leafblast and cucumber gray mold rot, the antifungal strain KL-3 strain was selected among 120 strains isolated from the rhizosphere soils. And the strain was identified to be a species of Bacillus subtilis or closely related strain. In several biochemical and in vitro antibiosis tests, antifungal substances of Bacillus sp. KL-3 were presumed heat stable, micromolecular antibiotic substances. In vivo test and vinyl house field test, the antifungal substances of Bacillus sp. KL-3 represented excellent biocontrol ability aganist Alternaria mali, Phyricularia oryzae, and Alternaria kikuchiana as well as broad spectrum of other fungi. In particular, Bacillus sp. KL-3 strain showed more predominant activity than some chemical fungicides against fungi shown to resist chemcal fungicides.
A synergistic effect of biocontrol agent and chemicals on control of the large patch on turfgrass caused by Rhizoctonia solani AG2-2 was evaluated. Chemicals; mepronil, toclofos-methyl and iprodione inhibited 90∼100% mycelial growth of R. solani AG1 and AG2-2 in vitro. While on the other, the chemicals inhibited only 0∼5%, 18∼46% and 30∼67% of mycelial growth of the antagonists, respectively. In field application, toclofos-methyl was the best to suppress the disease in single application, however, the combination of mepronil and T. viride 41D showed highest disease control effect among combinations of antagonists and chemicals. Effect of the combination of mepronil and T. viride 41D was similar to that of toclofos-methyl and T. viride 41D stage of disease development, but became higher in six week late. Results indicated that the combined application of selected antagonistic agent and chemical increased control efficasy of turgrass large patch.
The golden apple snail, Pomacea canaliculata, is a freshwater snail native to tropical and subtropical South America. The species was introduced into Korea as a human food source in 1983 and was first applied as a weed control agent for the paddy fields in 1992. As the snail is well known as an environmentally friendly biological control agent for weeds, the area of cultivation in which the golden apple snail is used for biological control has been enlarged substantially each year. Currently, the species is observed in open water courses. It is possible that the snail may overwinter in these open water courses and may become a serious pest, as is already the case in many Asian countries. In this study, we determined the status of the overwintering golden apple snail based on a literature survey and investigated the potential distribution area of the snail, as a result from global warming in Korea. The potential distribution area of the overwintering golden apple snail would be enlarged under the influence of global warming; ranging from 45.5% of South Korea's land area in the 2020s to 88.4% in the 2080s.
Bacillus amyloliquefaciens strain NJZJSB3 has shown antagonism of several phytopathogens in vitro, especially Sclerotinia sclerotiorum. Both the broth culture and cell suspension of strain NJZJSB3 could completely protect the detached leaves of canola (Brassica napus) from S. sclerotiorum infection. In pot experiments, the application of strain NJZJSB3 cell suspension ($10^8CFU/ml$) decreased the disease incidence by 83.3%, a result similar to commercially available fungicide (Dimetachlone). In order to investigate the potential biocontrol mechanisms of strain NJZJSB3, the nonvolatile antifungal compounds it produces were identified as iturin homologs using HPLC-ESI-MS. Antifungal volatile organic compounds were identified by gas chromatography-mass spectrometry. The detected volatiles toluene, phenol, and benzothiazole showed antifungal effects against S. sclerotiorum in chemical control experiments. Strain NJZJSB3 also produced biofilm, siderophores and cell-wall-degrading enzymes (protease and ${\beta}$-1,3-glucanase). These results suggest that strain NJZJSB3 can be a tremendous potential agent for the biological control of sclerotinia stem rot.

An entomopathogenic fungus, Paecilomyces fumosoroceus SFP-198 was isolated in Kyungbuk province, Korea and screened out for the control of Trialeurodes vaporariorum nymphs. It showed 72.5% efficacy against second instars of T. vaporariorum nymphs at 6 days after treatment in the laboratory. To select an active ingredient for the mass production and the formulation, SFP-198 culture products were compared based on their insecticidal activities against T. vaporariorum nymphs. Among them, conidia and blastospores showed much higher insecticidal activity than supernatants. Furthermore, SFP-198 conidia were more heat-resistant than blastospores, and also powder form of conidia was more stable than their suspension form. SFP-198 conidia showed high pathogenicity on not only T. vaporariorum but also Tetranychus urticae in the glasshouse. This result suggested that SFP-198 conidia can be used as a multi-targeting biological control agent against sucking agri- cultural pests, such as whiteflies and mites.
To detect and quantitation residual antibiotics and antibacterial agents in meats, we performed a biological assay employing the three microorganisms Bacillus subtilis ATCC 6633, Micrococcus luteus ATCC 9341, and Bacillus cereus var. mycoides ATCC 11778 for the screening purpose and developed a Gas Chromatography-mass Spectrometry(GC/MS) analysis for the confirmation and quantiation. In the biological assay (paper disk method), three test solution are used depending on the character of the residual antibiotics and antibacterial agents, follow by a simple clean up procedure which includes homogenization with Mcilvaine buffer, defatting with includes homogenization with Mcilvaine buffer, defatting with hexane, extraction with chloroform, clean-up by Sep-Pak $C_{18}$ and Bakerbond SPE carboxylic acid column. The chloroform layer is used for the analysis of sulfa agents. macrolides antibiotics and antibacterial agents, Adsorbed materials in the Sep-Pak $C_{18}$ were also employed for th analysis of penicillins and tetracyclines. Effluents from the Sep-Pak $C_{18}$ were cleaned-up one more by Bakerbond 10 SPE COOH column and employed for the analysis of aminoglycosides. In the instrumental analysis by using the GC/MSD, residual antibiotics and antibacterial agent were quantitated by selected ion monitoring (SIM) mode after derivatization. A simultaneous analysis of six residual antibiotic and antibacterial agent such as oxytetracycline, penicillin, ampicillin, choliraphenicol and thiamphenicol was developed with simple cleanup procedures revealing good recovery and reproducibility. Also, simultaneous detection of macrolides antibiotics such as erythromycin, spiramycin, and oleandomycin was developed after acid hydrolysis due to their large molecular structures. Because of the high reproducibility and selectivity of these two methods, it is very desirable that the combination of the two methods be used in the bioassay for the screening of residual antibiotics and antibacterial agent and that GC/MSD analysis be used for the confirmation and quantitation.
