생물학적 고도처리 MLSS의 생응집성 및 침강성 파악 (Evaluation of bioflocculation and settling characteristics for MLSS from a Biological Nutrient Removal Plant)

  • Evaluating and designing secondary clarifier require to define characteristics of influent MLSS (Mixed Liquor Suspended Solids). In this study, bioflocculation and settling characteristics for MLSS from a Biological Nutrient Removal (BNR) plant located in near Seoul were measured. MLSS concentrations in bioreactor were about 2,500mg/L in summer and about 4,000mg/L in winter, respectively. Tests showed that there was not much bioflocculation occurred in secondary clarifier. Average ESS/DSS (Effluent Suspended Solids/Dispersed Suspended Solids) was 100%. From the settling tests, ZSV (Zone Settling Velocity) and settling constant (n) in Vesilind equation were estimated at different MLSS temperatures. SVI (Sludge Volume Index) and SSVI (Stirred Sludge Volume Index) were also measured at different temperatures. It was found that ZSV was positively correlated with temperature and n was inversely proportional to temperature. SVI and SSVI had very similar values at about $25^{\circ}C$ of MLSS, However, SVI had more than 2 times higher values than SSVI at below $20^{\circ}C$ of MLSS. Temperature effect must be considered to design and evaluate secondary clarifier.

Bioflocculant and Anticorrosive Activities of the Biopolymer Produced by Lysinibacillus macroides

  • The biopolymer produced by Lysinibacillus macroides can be useful as an emulsifying agent, flocculating agent, anti-oxidative agent, and anti-corrosive agent. The present work focused on the functional activities of this biopolymer such as bioflocculant and anti-corrosion. The results of the study for the biopolymer as a bioflocculating agent reveal that the biopolymer gave 72.3% flocculating activity against activated carbon at a concentration of 5 mg/l. In the optimization study, the highest bioflocculant activity was obtained in the presence of CaCl2 mineral at 15% biopolymer concentration, 7.0 pH, and 40℃ temperature. The biopolymer was also studied for its anti-corrosive nature and observed that it was able to prevent corrosion of steel paper clips in a 6% CaCl2 solution at a concentration of 0.5% after 7 days.

Effect of Carbohydrates to Protein Ratio in EPS on Sludge Settling Characteristics

  • Extracellular polymeric substances (EPS) are believed to play a role in the binding and formation of microbial flocs. However, the precise role is not well known. Sludge settling characteristics and the carbohydrate to protein ratio in EPS were tested with various airflow rates in this study. Sludge was collected from three modified sequencing batch reactors (SBRs), which were operated at 16$\^{C}$ with an airflow rate of 0.8L/min, 3L/min and 6L/min, respectively. During the operation, the reactor operated at an airflow rate of 0.8L/min showed sludge volume index (SVI) of 80 to 90ml/g and a constant ratio of carbohydrate to protein in the EPS, while a significant increase in the SVI was seen in the other reactors. Sludge bulking increased the amount of carbohydrate in the EPS, while kept protein almost constant in the airflow rate of 3L/min ad 6L/min. Surface charge also increased with increases in the carbohydrate to protein ratio in the EPS, which weakens the attraction between the EPS and multivalent cations. The ratio of carbohydrate to protein in the EPS was tween the EPS and multivalent cations. The ratio of carbohydrate to protein in the EPS was inferred to be essential for bioflocculation.

Novel Cationic Microbial Polyglucosamine Biopolymer from New Enterobacter sp. BL-2 and Its Bioflocculation Efficacy

  • A new bacterium BL-2 excreting a novel cationic polyglucosamine biopolymer was isolated from the spoiled leaves of Chinese cabbage and identified as Enterobacter sp. BL-2. The isolated Enterobacter sp. BL-2 was cultivated in pH-stat fed-batch culture using acetic acid as the feeding stock at pH 8.0, resulting in 17.11 g/l of cells and 1.53 g/l of an extracellular biopolymer after 72 h. The excreted biopolymer was purified by a three-step procedure, involving ethanol precipitation and deproteinizations, to a nearly homogeneous state, and its molecular weight was found to be 106 kDa. It was composed of glucosamine, rhamnose, and galactose at a molar ratio of 86.4:1.6:1.0, respectively, indicating a rarely found novel high-glucosamine-containing biopolymer. The FT-IR and $^{13}C-NMR$ spectra of the novel cationic polyglucosamine biopolymer PGB-l revealed a close identity with chitosan from crab shell. It can effectively flocculate various suspended solids, including kaolin clay, $Ca(OH)_2,\;Al_{2}O_3$, active carbon, microbial cells, and acidic dyes.
