랫트에 있어서 captafol의 독성에 대한 cysteine의 방어 작용 1. 혈액학 및 혈청 생화학적 성상에 미치는 영향 (Cysteine antagonism of captafol induced toxicities in rats 1. Effects on hematological and serum biochemical values)

  • 과일 및 채소류의 곰팡이 억제제로 개발된 captafol은 인간 및 동물에서의 독성작용은 많이 보고된 바 있으나 이들의 독작용을 완화시키거나 치료할 수 있는 제제에 대한 연구는 별로 알려진 바 없다. captafol에 의한 혈액학 및 혈청 생화화적 변화에 따른 cysteine의 방어 작용을 확인할 목적으로 랫트에 5mg/kg 용량의 captafol을 1회 복강 투여하고 동시에 5, 58, 290 및 580mg/kg cysteine을 각각 투여하고 체중의 변화, 부검소견, 절대장기중량, 혈액학 및 혈청 생화학적 소견을 비교 분석 하였다. captafol 단독 투여군에서는 체중 및 간 장기중량 감소, 복강내 장기들의 섬유소에 의한 유착 및 복수 등이 현저하였으며 적혈구 수, hemoglobin 농도, 혈청 AST의 증가가 인정되었으며 또한 혈청 인지질 함량 감소가 확인되었다. 한편, cysteine 58mg/kg 이상의 용량군에서는 복강내 장기들의 유착 및 복수가 확인되지 않았으며 장기중량, 혈액학 및 혈청 생화학적 소견에서도 정상을 나타내었다. 이 연구 결과는 captafol의 중독증상에 대해 cysteine 및 sulfhydryl군을 가진 물질은 그 증세를 감소시키거나 완화시킬 수 있는 효과가 있음을 보여주고 있다.

시험관내에서 captafol이 혈액학, 적혈구막 및 혈장생화학치에 미치는 영향 (The effect of captafol on the hematological value, erythrocyte membrane and plasma biochemical value in vitro)

  • Captafol은 phthalimide계통의 항진균제로써 difolatan이란 상품명으로 개발되어 과일, 채소의 탄저병 및 원목 등의 곰팡이 구제에 널리 사용되고 있는 것으로 국내에서도 두번째로 많이 생산, 사용되는 품목이나 독성실험에 관한 보고는 거의 없는 상태이다. 이에 본 연구에서는 captafol 자체에 대한 독성기전의 일부를 확인하고자 동물체내에서 일어나는 여러가지 대사작용이 배제된 시험관내에서 실험을 수행하였다. 랫트 혈액을 후대정맥으로 부터 채혈하여 4개의 시험관에 $2.5m{\ell}$씩 분주한 후 captafol 농도가 $0.1{\times}10^{-4}M$, $1{\times}10^{-3}M$, $1{\times}10^{-2}M$이 되도록 한 ethanol용액 $25{\mu}{\ell}$씩 넣고 $CO_2$ incubator에서 2시간 동안 부드럽게 혼합하여 혈액학적 성상 및 적혈구 취약성 시험을 수행하였다. 또한 원심분리하여 얻어진 혈장에서 생화학적 성상 등을 조사한 결과 다음과 같은 성적을 얻었다. 1. 적혈구수 및 hematocrit치는 captafol 투여량에 비례적으로 유의한(p<0.05) 감소를 나타내었다. 2. 적혈구막 취약성 실험에서 $1{\times}10^{-3}M$$1{\times}10^{-2}M$ 농도에서 대조군에 비하여 유의한(p<0.01) 증가를 나타내었다. 3. 혈장내 potassium이온의 농도는 captafol의 투여량에 비례적으로 증가하였다. 4. Total bilirubin 및 creatine kinase가 대조군에 비하여 각각 증가 및 감소하였다.

산양산삼약침의 지질강하 및 항산화효과에 대한 생화학적 및 분자생물학적 검토 (Effects of cultivated wild ginseng pharmacopuncture on lowering lipid and oxidative capacity in biochemical and molecular biological study in obese rats)

  • Objectives : This study was carried out to identified the effects of distilled cultivated wild ginseng pharmacopuncture to the obesity. Methods : Cultivated wild ginseng pharmacopuncture was administered on the points of chung-wan(CV12), $Ch'{\breve{o}}nch'u$(ST25), and Chok-samni(ST36) on lowering lipid and oxidative capacity in biochemical and molecular biological aspects were investigated in obese rats fed with high fat diet. Results : 1. The contents of plasma ${\beta}-lipoprotein$ showed a tendency to decrease in the pharmacopuncture groups compared to the control group. In the pharmacopuncture groups, the values of ST25 and ST36 pharmacopuncture groups showed lower value. 2. The contents of plasma free fatty acids showed a tendency to decrease in pharmacopuncture groups compared to the control group. However, in the pharmacopuncture groups, the values were not significantly different. 3. Plasma triglyceride and glucose showed lower value in the ST25 pharmacopuncture groups compared with the other groups. 4. The activity of AST showed a tendency to decrease in the pharmacopuncture groups. However, the activity of ALT was not significantly different in all the treatment groups. 5. Plasma total cholesterol and LDL-cholesterol showed lower value in the ST25 and ST36 pharmacopuncture groups and HDL-cholesterol showed higher value in the CV12 pharmacopuncture groups than that of the other treatment groups. 6. Liver total cholesterol values didn't show significant difference in all the treatment groups, and triglyceride showed lower value in the pharmacopuncture groups. 7. The contents of plasma TEARS showed lower value in the ST25 pharmacopuncture group and contents of liver TBARS showed a tendency to decrease in the pharmacopuncture groups. However these values didn't show significant difference in the pharmaco puncture groups. 8. Liver super oxide dismutase activity showed higher value in the ST25 and ST36 pharmacopuncture groups, and the value of liver glutathione peroxidase and catalase activity showed a tendency to increase in the pharmacopuncture groups. However, these values showed no significant difference in the pharmacopuncture groups. 9. Expression of apo-B and E mRNA in liver cells was lower in the ST25 pharmacopuncture group than that of the other treatment groups. However, expression of $TNF-{\alpha}$ and leptin mRNA in adipose cell showed no difference among all the treatment groups. 10. ST25 pharmacopuncture group showed a good histological character of liver. It showed similar to that of normal group. However other treatment groups and control group showed slight vasodilation and slight fat accumulation. Conclusion : These results indicate that distilled cultivated wild ginseng pharmacopuncture suppressed adipose tissue mass and lipid peroxidation, and increased antioxidant capacity.

부분동결시에 미생물의 생화학적 활성에 관한 연구 (Biochemical Activity of Microorganism Stored by the Method of Partial Freezing)

  • 부분동결의 유효성을 미생물학적인 측면에서 해명하고자 저온에서 부패의 주역을 담당하고 있는 몇종의 저온세균을 공시균으로 하여서 고등어육액즙에 대한 부패활성을 동일액상이면서 빙점부근에서의 근소한 온도차($0^{\circ}C$$-3^{\circ}C$ 과냉각) 및 $-3^{\circ}C$동일 온도에서 상변화(액상-고상)를 중심으로 균의 증식, 사멸, 부패활성 및 K값의 변화를 검토한 결과는 다음과 같다. (1) $25^{\circ}C$$0^{\circ}C$ 저장에서는 공시균 전부 강한 증식력 및 부패활성을 나타내었으나, 동일액상이면서 $0^{\circ}C$와 근소한 온도차인 $-3^{\circ}C$ 과냉각에서는 공시균 중 Pseudomonas III/IV-NH형의 증식력 및 부패활성이 현저히 억제되었으며, Moraxella 및 F/C도 실험기간을 통하여 점차 감소하는 경향을 나타내었고, 부패활성은 전혀 관찰되지 않았다. (2) $-3^{\circ}C$ 과냉각(액상)과 동일온도이면서 고상인 $-3^{\circ}C$ 동결에서는 전공시균의 증식 및 부패활성이 전혀 관찰되지 않았으며, 저장기간을 통하여 생균수가 감소하는 경향을 나타내었다. (3) 동일액상이면서 $0^{\circ}C$와 근소한 온도차인 $-3^{\circ}C$ 과냉각에서 K값의 증가가 억제되었으며, 더욱 $-3^{\circ}C$의 동온이상인 $-3^{\circ}C$동결(고상)에서는 상의 변화로 인하여 K값의 증가가 현저히 억제되었다. 이상의 결과로부터, $0^{\circ}C$와의 근소한 온도차 및 액상에서 친상으로의 상의 변화에 의한 효소황성의 저하(K값) 및 저온세균의 증식과 부패활성의 억제가 부분동결의 유효성의 큰 요인이 되리라 추측된다.

N-Acyl Taurates의 환경친화적인 제조공정 개발 및 이의 물성 연구 (Development of Environmental-friendly N-Acyl Taurates Manufacturing Process and Evaluation of their Physical Properties)

  • 기존 공정에 비하여 오염물 발생이 적고 수율이 60 % 이상의 환경친화적인 새로운 N-acyl taurate 단일축합공정을 개발하였다. 개발된 공정의 최적 운전조건은 석유계 정제유인 parasol 123 용매와 반응촉매로 sulfone산계 촉매와 인산계 촉매의 혼합촉매를 사용할 때 반응온도가 $210{\sim}230^{\circ}C$이며 반응시간이 6 ~ 9시간 이었다. 이렇게 제조된 제품은 기존 수입된 N-acyl taurate와 비교하여 산가, 아민가, 색상 등의 물성이 비슷하거나 더 좋고 부산물도 적게 배출되어 본 연구에서 개발된 공정이 기존 공정보다 우수한 공정이라고 할 수 있다. 그리고 본 연구에서 제조된 N-acyl taurate를 시판 음이온 계면활성제와 표면장력, 기포력, 기포안정성, 경수안정성, 유화력 등의 물성을 비교할 때 동등 이상의 물성을 보여 기존의 음이온 계면활성제를 충분히 대체할 수 있음을 확인할 수 있었다.

납의 생체내 세포독성 연구: 랫드에서 혈장 DNA와 혈액화학치 변화 (In Vivo Cytotoxicity of Lead Acetate: Changes of Plasma DNA Content and Blood Biochemical Values in Rats)

  • Changes of plasma DNA contents and serum biochemical values were measured in rats administered with lead acetate to investigate the in vivo cytotoxic effects of lead and examine the usefulness of these in vivo cytotoxicity changes as indicators of lead exposure and diagnosis of lead poisoning. Rats were given once intraperitonealy with lead acetate (1.6, 8, 40 and 200 mg/kg b.w) and the changes of plasma DNA contents and serum biochemical values were measured at the time of 2, 4, 8, 24, 48 and 72 hours after the administration of lead acetate. Plasma DNA contents began to increase at 2 hours after the administration of lead acetate in the treatment groups of 8, 40 and 200 mg/kg b.w dose-dependently and significantly compared with control group. These DNA increases of each dosage group were continued until 24, 48 and 72 hours and the maximum levels of DNA (4.02, 10.67 and 14.10 times of control) were arrived at 8, 8 and 4 hours after the each treatment, respectively. Among 10 serum biochemical indicators, the activities of creatine kinase were increased to maximum level (6.55 times of control) at 2 hours after the administration and remained to be significantly higher than that of control by 8 hours in the treatment group of 200 mg, however, after 48 hours, the levels in the treatment groups of 40 mg above were lower than that of control. The values of aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase were higher than that of control from 2 to 24 hours in the treatment group of 200 mg. Maximum levels of these enzymes were 3.34, 3.00 and 3.19 times of control, respectively. Both of alkaline phosphatase and triglyceride values in the treatment groups were decreased compared with control. In the case of alka-line phosphatse, the values were significanly decreased from 24 hours and more severely decreased until 72 hours in the treatment groups of 40 mg above (p<0.01). The minimum value was 0.36 times of control in the 200 mg group. The values of triglyceride were significantly decreased in the tratment groups of 40 mg above (p<0.01), but the values were not different significantly among the treatment groups. This study demonstrates that plasma DNA content and serum biochemical values such as aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, alkaline phosphatase and triglyceride are valuable as biomarkers for exposure assessment and diagnosis of lead poisoning.

Effects of Yeonlyeonggobon-dan on the Blood of Arsenic-poisoned Rats

  • Sodium arsenate and Yeonlyeonggobon-dan (nianlinggubendan) extract (YGD), a herbal restorative were treated p.o. 20 mg/kg and 500 mg/kg, respectively, and concurrently to rats, and examined the biochemical parameters in blood. The values of white blood cell (WBC), red blood cell (RBC), hemoglobin (Hgb) and hematocrit (Hct) in each group did not show significant variance. The value of aspartate aminotrasferase (AST) of arsenic-treated group was increased for 2 weeks significantly while that of the group of concurrent administration with YGD became low significantly compared with arsenic-treated group and the value of alkaline phosphatase (ALP) of arsenic-treated group was decreased while that of the group of concurrent administration with YGD was increased significantly compared with arsenic-treated group. In arsenic-treated groups, the value of glucose (Glu), and those of lactic dehydrogenase (LDH), blood urea nitrogen (BUN) and triglyceride (TG) were decreased at first but increased later while the groups of concurrent administration with YGD showed significant recovery from the toxicity of arsenic.

Valorization of refined sardine oil in canned Sardina pilchardus

  • This study aimed to use refined sardine oil as a covering juice in canned Sardina pilchardus. The oil's fatty acid profile, acidity, peroxide value, and p-anisidine value were analyzed. The biochemical composition, histamine content, and bacteriological stability of the canned sardines were determined. The results showed that canned sardines contained 51.40 g of moisture, 27.87 g of fat, 17.91 g of protein, and 1.03 g of salt per 100 g net weight. The fatty acid composition included 10.35% polyunsaturated fatty acids, 8.86% saturated fatty acids, and 6.27% monounsaturated fatty acids, with the most abundant fatty acids being eicosapentaenoic acid (EPA), palmitic acid, oleic acid, myristic acid, and docosahexaenoic acid (DHA). This recipe provided 8.88 g/100 g (net weight) of EPA and DHA. These findings support the interest in using refined fish oil to enrich foods with essential marine fatty acids, highly recommended for their health benefits and leading to encouraging prospects for sardine canneries to develop new and nutritious value products.

하계 경기만의 수질오염과 생산력에 관한 연구 1

  • The extent of water pollution was investigated at 4 stations in Kyonggi Bay during the summer seasons in 1970 and 1971. The concentrations of dissolved oxygen, total hardness, ammonia, nitrate, nitrite, phosphate, chemical oxygen demand, salinity, biochemical oxygen demand, coliform bacteria and facel coli were examined together with the measurement of pH and transparency. The relationship between the extent of pollution and the distance from the Inchon Bay also was examined. The concentrations of biochemical oxygen demand, chemical oxygen demand, ammonia, nitrate, nitrite, phosphate, coliform bacteria nad fecal coli were all highest at station 1, and lowest at station 4. Values were somewhat higher at low tide level in general. On the contrary, dissolved oxygen concentration was highest at station 4 and lowest at station 1. The highest and lowest values of biochemical oxygen demand were 10.88 ppm at station 1 and 0.27 ppm at station 4. The chemical oxygen demand concentrations at station 1 and 4 were 1.90 ppm and 0.63 ppm. Ammonia concnetration at station 1 was 0.43 pp, and was nearly 5 times as much as that at station 4. The values were $2.45{\times}10^{-4}$ ppm at station 1, and $6{\times}10^{-4}$ ppm at station 4. Nitrite concnetration at station 1 was $3{\times}10^{-4}$ppm and was the highest, while the lowest was $9.45{\times}10^{-5}$ ppm at station 4. Phosphate value at station 1 too was the highest and was about 4 times as much as that at station 4. Coliform bacteria were most abundant at station 1, and were counted to be 1.$1.7{\times}10^{-5}$cells/ml. At station 4, the number greatly reduced to 8 * 10$^{2}$ cells/ml. The number of fecal coli at station 1 was $2{\times}10^{-4}$ cells/ml. But at station 4, no fecal coli was detected at high tide level. At low tide level, 3 cells/ml were counted at station 4. In all of these, the highest data were obtained at low tide level, while most of the lowest value, at high tide level. Generally, values at statio 1 were 3-5 times as much as those at station 4. Concentration of dissolved oxygen at station 1 was 0.366 mg-atoms/1 and was the lowest. The highest value was 0.420 mg-atoms/1 and was recorded at station 4. The highest at station 4, which certainly were believed to be the result of the dilution by the fresh water of the Han river flowing into the Inchon Bay. As we can see from the data above, the extent of pollution was highest at station 1, the nearest from Inchon harbor, and lowest at station 4, where is the farthest, Station 1 and 2 were proved to be much polluted, but station 3 and 4, not.

Acarbose Effect for Dexran Synthesis, Acceptor and Disproportionation Reactions of Leuconostoc mesenteroides B-512FMCM Dextransucrase

  • Acarbose effectively inhibited the synthesis of dextran, and the inhibition pattern was a noncompetitive type with a $K_i$ value of 1.35 mM. It also inhibited the disproportionation reaction of dextransucrase with isomaltotriose and decreased the efficiency of the maltose acceptor reaction. Increased concentration of dextransucrase or maltose in reaction digests, however, decreased the degree of inhibition by acarbose.
