Metabolic profile according to the parity and stage of lactation of high-performance Holstein-Friesian cows

  • Objective: The aim of the study was to determine the effect of parity and the stage of lactation on the metabolic profile of cows based on the basic chemical milk components and the blood parameters. Methods: The study material consisted of high-yielding Holstein-Friesian cows. In total, 473 cows were examined. According to the parity, cows were divided into four groups: primiparous (P), and multiparous in the second (M2), in the third (M3), and in subsequent lactations (M4). The feeding of cows was based on total mixed ration (TMR) ad libitum. Milk and blood samples were collected individually from each cow three times per standard lactation period. Results: Greater exacerbation of changes in the dynamics of the blood plasma parameters examined was proved for multiparous cows. The highest value of β-hydroxybutyrate acid (0.946 mmol/L) was found for multiparous cows from group M3 at the beginning of lactation. However, it was still in the normal range. The results showed aspartate aminotransferase, and gamma-glutamyl transferase (GGT) activities in dairy cows during lactation had significant variations taking in to account stage of lactation. The highest activity of GGT was found in the group of the oldest cows and measured from 26.36 U/L at the beginning of lactation to 48.75 U/L at the end of the lactation period. Conclusion: The time-related changes in the concentrations of the biochemical parameters described differ markedly among lactating cows, though the housing conditions on the research dairy farm are highly standardised. This indicates that the ability to cope with metabolic stress is mainly affected by the individual predispositions of cows and feed nutrient supply in different stage of lactation. Especially, the feed nutrient supply (in net energy for lactation), which was the best in TMR 1 in comparison TMR 3.

Rodent peri-implantitis models: a systematic review and meta-analysis of morphological changes

  • Purpose: Rodent models have emerged as an alternative to established larger animal models for peri-implantitis research. However, the construct validity of rodent models is controversial due to a lack of consensus regarding their histological, morphological, and biochemical characteristics. This systematic review sought to validate rodent models by characterizing their morphological changes, particularly marginal bone loss (MBL), a hallmark of peri-implantitis. Methods: This review was conducted following the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines. A literature search was performed electronically using MEDLINE (PubMed), and Embase, identifying pre-clinical studies reporting MBL after experimental peri-implantitis induction in rodents. Each study's risk of bias was assessed using the Systematic Review Center for Laboratory animal Experimentation (SYRCLE) risk of bias tool. A meta-analysis was performed for the difference in MBL, comparing healthy implants to those with experimental peri-implantitis. Results: Of the 1,014 unique records retrieved, 23 studies that met the eligibility criteria were included. Peri-implantitis was induced using 4 methods: ligatures, lipopolysaccharide, microbial infection, and titanium particles. Studies presented high to unclear risks of bias. During the osseointegration phase, 11.6% and 6.4%-11.3% of implants inserted in mice and rats, respectively, had failed to osseointegrate. Twelve studies were included in the meta-analysis of the linear MBL measured using micro-computed tomography. Following experimental peri-implantitis, the MBL was estimated to be 0.25 mm (95% confidence interval [CI], 0.14-0.36 mm) in mice and 0.26 mm (95% CI, 0.19-0.34 mm) in rats. The resulting peri-implant MBL was circumferential, consisting of supra- and infrabony components. Conclusions: Experimental peri-implantitis in rodent models results in circumferential MBL, with morphology consistent with the clinical presentation of peri-implantitis. While rodent models are promising, there is still a need to further characterize their healing potentials, standardize experiment protocols, and improve the reporting of results and methodology.

Gynostemma pentaphyllum extract and its active component gypenoside L improve the exercise performance of treadmill-trained mice

  • BACKGROUND/OBJECTIVES: The effectiveness of natural compounds in improving athletic ability has attracted attention in both sports and research. Gynostemma pentaphyllum (Thunb.) leaves are used to make traditional herbal medicines in Asia. The active components of G. pentaphyllum, dammarane saponins, or gypenosides, possess a range of biological activities. On the other hand, the anti-fatigue effects from G. pentaphyllum extract (GPE) and its effective compound, gypenoside L (GL), remain to be determined. MATERIALS/METHODS: This study examined the effects of GPE on fatigue and exercise performance in ICR mice. GPE was administered orally to mice for 6 weeks, with or without treadmill training. The biochemical analysis in serum, glycogen content, mRNA, and protein expressions of the liver and muscle were analyzed. RESULTS: The ExGPE (exercise with 300 mg/kg body weight/day of GPE) mice decreased the fat mass percentage significantly compared to the ExC mice, while the ExGPE showed the greatest lean mass percentage compared to the ExC group. The administration of GPE improved the exercise endurance and capacity in treadmill-trained mice, increased glucose and triglycerides, and decreased the serum creatine kinase and lactate levels after intensive exercise. The muscle glycogen levels were higher in the ExGPE group than the ExC group. GPE increased the level of mitochondrial biogenesis by enhancing the phosphorylation of peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) protein and the mRNA expression of nuclear respiratory factor 1, mitochondrial DNA, peroxisome proliferator-activated receptor-δ, superoxide dismutase 2, and by decreasing the lactate dehydrogenase B level in the soleus muscle (SOL). GPE also improved PGC-1α activation in the SOL significantly through AMPK/p38 phosphorylation. CONCLUSIONS: These results showed that GPE supplementation enhances exercise performance and has anti-fatigue activity. In addition, the underlying molecular mechanism was elucidated. Therefore, GPE is a promising candidate for developing functional foods and enhancing the exercise capacity and anti-fatigue activity.

Comparative Analysis of Phenolic Compound of Mutant Lines of Sorghum (Sorghum bicolor)

  • Sorghum (Sorghum bicolor) is increasingly important as a biomass crop worldwide. Its genetic diversity provides a large range of biochemical composition suitable for various uses as bioplastics. Phenolic compounds are the main compounds of lignocellulosic residues, which can be used as a source of active components for their use in active packaging materials. In this research, we investigated the total phenolic content (TPC) and the total flavonoid content (TFC) among 60 mutant lines (early heading, high biomass and dwarfness) and their original cultivars. Sixty sorghum mutant lines were developed by treatment with gamma-ray or proton irradiation in 14 sorghum cultivars. The levels of TPC and TFC of 14 original cultivars were ranging from 3.27 to 11.54 mg/100 g and 2.39 to 6.74 mg/100 g, respectively. The TPCs of the mutant lines were ranging from 1.92 to 13.10 mg/100 g with average content of 6.35 mg/100 g. The TFCs of the mutant lines were ranging from 1.72 to 8.30 mg/100 g with average content of 4.20 mg/100 g. Three mutant lines derived from gamma-ray showed significant lower TPC and TFC than those of the original cultivar. While, five mutant lines showed significant higher TPC and TFC. These findings will be useful for the selection of sorghum genotypes with improved phenolic compounds.

Changes of biochemical components and physiological activities of coffee beans according to different roasting conditions (커피 볶음 정도에 따른 생화학적 성분 및 생리활성의 변화)

  • Four different kinds of coffee beans (CS, Colombia supremo; EY, Ethiopia yirgacheffee; IM, Indonesia mandheling; and IMM, India monsooned malabar) were roasted at 200 and $250^{\circ}C$ for 10, 15, and 20 min. To determine the optimum roasting conditions, various components of the coffee beans such as pyrazines produced during the roasting, and their antioxidant and antidiabetic effects were analyzed. The different roasting condition did not affect on the concentration of caffeine. However, the amount of 5-caffeoylquinic acid and the total phenolics decreased significantly, at a greater temperature and a longer roasting time. The greatest amount of pyrazines was produced from the IMM however, the amount of pyrazines decreased rapidly at $250^{\circ}C$ according to increasing in roasting time. The DPPH free radical scavenging activity was mostly 80% more effective than that of BHT and ${\alpha}$-tocopherol activities at the same concentration. In the case of the FRAP assay, the reducing power of the coffee slightly decreased at a greater temperature pand longer time. While the inhibitory effect on ${\alpha}$-glucosidase was negligible, the activity decreased by more than 80% when the coffee beans were roasted at $250^{\circ}C$ for 20 min. The inhibitory effect on ${\alpha}$-amylase showed similar results. Taken together, the optimum roasting conditions were determined to be $200^{\circ}C$ and 15 min, which provided the best physiological activity and nutty and chocolatey aromas from the pyrazine of coffee.

동면 동물의 혈액상에 관한 연구

  • 동면 상태에서 혈액성분의 생화학적 변화치를 조사하고자 전남 각지에 서식 또는 광주직할시 근교에서 양식하고 있는 뱀장어, 개구리, 살모사 그리고 다람쥐를 활동기(7월∼8월), 잠입기(10월∼11월), 동진기(12월∼1월) 및 각려기(3월∼4월)에 채집하였다. 이들 동물들의 혈청에서 Total protein, Albumin, Globulin, A/G ratio, Glucose, Calcium, Urea함유량을 조사하여 얻은 성적을 비교 검토하여 다음과 같은 결론을 얻었다. 1 . Total protein의 변화는 각 동물사이에서 상당한 차이를 보이되 잠입기에 다소 증가하였다. 특히 다람쥐에서 Total protein 함유량이 다른 동물들 보다 많았다. Albumin함유량은 잠입기에 상승하였고 동면기에는 약간의 감소를 보이다가 각성기에는 현저한 감소를 보였다. gloulin함유량은 잠입기에 상당히 상승하고 동면기에 약간의 감소를 나타내었으며 각성기 인 봄철에는 많은 증가 현상을 보였다. 2. 혈청 Urea의 함유량은 동면의 각과정에 따라 상당한 차이가 있었다. 즉 잠입기에 약간 상승하고 동면기에는 저하를 보였으며 각성기에는 전 실험동물에서 검출되지 않았다. 3. 혈당의 변화는 잠입기에 대체로 저하하고 동면기에는 심한 저혈당 수준에 이르렀다. 각성기인 봄철에는 다람쥐를 제외하고는 동면기의 혈희량과 유사하였다. 4. 혈청 Calcium함유량의 변화는 활동기에 비하여 잠입기에는 상당한 증가현상을 보였으나동면기에는 전 실험동물이 거의 같은 수준으로 감소를 보이며 각성기에 까지 감소하였다. In order to the biochemical change of the serological components of hibernating animals, eels(hnguillo joponica Temminck et Schlegel), frogs (Rana nigromacurata Hallowell), snakes (Agkistro-don holds breuicaudus Stejneserl, squirrels (Tamias striatus Linnaeus) which were collected around Gwangiu city and the Chunnam area were observed in accordance with the phases of hibernation. The author described the hibernating cycle as active (June-July), prehibematins(October-Novemberl, hibernating (December-January) and posthibemating phase (March-Aprill. The values of total protein, albumin, globulin, hJG ratlo, calcium, glucose and urea were chiefly investigated in their serum. The results are as following: Total protein was shown to be consider-able in eels, frogs, snakes, squirrels and increased in prehibemation. Especially, the mean value was high in sqirrels. Albumin values was increased during prehibemation. But a little decrease was shown during hibernating and an outstanding decrease during posthibemation. The globulin value increased considerably during prehlbemation but decreased a little during hibernation phase. Changes of urea were shown to be considerable according to the stage of each hibem3-tion. A little increase was shown during prehibemation and decrease during hibernating stage. but urea was not found in posthibemation in any experimental animals. The glucose value was generally decreased during prehibematlon. An extremely little increase in glucose was shown during hibernation (hypoglycemia). All hibernating values except those for squirrels in spring posthibemating phase were at almost the same level. The calcium value was shown to be considerable during prehibemation in comparison with active phase. The same level of decrease was represented in all kinds of animals and continued at the stage of posthibemation.

Biochemical Characteristics of Whole Soybean Cereals Fermented with Mucor and Rhizopus Strains (Mucor 및 Rhizopus속 균류를 이용한 콩알메주 발효의 생화학적 특성)

  • Whole soybean cereal was fermented with four fungal strains (Mucor and Rhizopus) in pilot meju fermentation system. The pH range of the fermented soybean cereal was $7.16{\sim}8.38$, the contents of reducing sugar and amino-nitrogen were $0.54{\sim}2.64%,\;93{\sim}312mg%$, respectively, and that of free fatty acid ranged $0.06{\sim}2.00%$. The components of the amino acid, organic acid, free sugars and fatty acid showed distinctive patterns among four groups of fermented soybean cereals. Amylase activity and carbohydrate degradation rate of R. oryzae, protease and protein degadation rate of R. stolonifer was higher than other strains. But lipase and lipid degradation rates of four strains were similar. The odor concentrates of the soybean cereals fermented with Mucor strains were similar to Aspergillus strains, but Rhizopus were possessed of the flavor components of Bacillus and Aspergillus. Soysauce, made from M. hiemalis and R. stolonifer fermented soybean cereal showed excellent sensory evaluation and it was proposed that the two strains will be useful in Korean soysauce process.

  • The ratio of the eatable flesh taken from the filefish and its chemical components were investigated from March 1980 to February 1981 and the results are as follows :1. The ratio of the eatable flesh taken from the fish was low during March to June and high during July to December. 2. The moisture content was $81.4-82.0\%$ during March to June, $77.5-80.1\%$ during July to December and $79.4-81.4\%$ during January to February. 3. The protein content was $15.4-15.7\%$ during March to June, $17.0-19.9\%$ during July to December and $16.1-17.0\%$ during January to February. 4. The Lipid content was $0.31-0.41\%$ during March to January, $0.46-0.84\%$ during July to January and $0.43\%$ during February. 5. The carbohydrate content was $0.3-0.4\%$ during March to July, $0.5-0.7\%$ during August to January and $0.3\%$ during February. In conclusion, the ratios of the eatable flesh taken from the filefish and its protein content were highest during July to December in one year.

Nutrition Survey in a Korean Mountainous Farming Area (산간지(山間地) 농촌주민(農村住民)의 영양실태조사(營養實態調査))

  • The main character of the Korean diet has been found to be low in protein both quantity and quality and high in carbohydrate. The purpose of this survey was to study the amount of salt intake related to the dietary pattern in Korea. The nutrition survey was conducted in a mountainous farming area located in Auhchun-ri, Gaebuk-myon, Changsoo-gun, Chunbuk Province, February 14-19 in 1973 (7 days). The precise weighing method was used in evaluating the kinds of foods and nutrients intake for 24 households during a three day period. The physical examinations were performed by a doctor on 120 persons and a detailed biochemical test on both blood and urine was made on 42 persons over 40 years old. The results obtained are summarized as follows: (1) Average nutrients intake of an adult per day: calorie intake was 2,446 Cal and its components-protein(61.1g) was 10 percent, fat(12.9g) was 5 percent and carbohydrate(521g) contributed 85 percent of the total calories. Other nutrients-calcium (443mg), thiamine(1.09mg), riboflavin (0.90mg), niacin (14.4mg) and vitamin C (63.2mg) were lower than the recommended daily allowance but vitamin A(2,083 I.U.), iron(11mg) and phosphorous(998mg) were slightly higher than that. (2) To evaluate the nutritional deficiences, clinical examinations were conducted. Angular stomatitis was present in 16.7 percent of those examiners. No edema was found. The rate of osteoarthritis, hepatomegaly diseases appeared in 20 percent of the total subjects and the symptoms appeared highest among those Iron 50 to 59 years old. (3) The following chemical components of blood serum were analyzed and found to be within the normal range: glucose, blood urea nitrogen, uric acid, total protein, albumin, globulin, bilirubin, total cholesterol, inorganic phosphate, alkaline phosphatase, sodium, potassium, chloride, calcium and lecithine dehydrase. One case of each of the following were found: hyperglycemia, hypocholesterolemia, renal problem, hypoproteinaemia and diabetes mellitus, and two persons were classified as showing hypoglycemia and hyponaturemia. (4) The sodium content in urine was 199.6 mEq/L, potassium content was 24.6 mEq/L. The sugar, pH and specific gravity in the urine was shown to be normal.

A Study on the Leaching of Heavy Metals by Municipal Solid Waste Landfill Leachate (폐기물 매립지 침출수에 의한 중금속 용출에 관한 연구)

  • Sanitary landfill is a general method as a final disposal of municipal solid waste(MSW), therefore leachate characteristics are very various as lime goes by because of highly concentrated organic acids are contained non biodegradable COD. So it is hard to abide by the mandatory standards of discharge eventhough applying the physicochemical and biological processes to treat the leachate. The process of treating leachate are determined by the degree of removal and components, but they are highly contained organic materials. It is a removal method to use jointly with the physicochemical process if the hard and fast rule is needed. The critical components of material are COD, ammonia, salts and heavy metals in the case of treating biologically. Biological process is to use metabolism of microorganism, therefore it is a desirable condition which heavy metals are not contained, because they acting as an inhibitor of enzyme. Of these are contained, organic decomposition and synthetic function of microorganisms decrease significantly. Consequently, this research paper lays emphasis on the concentration of heavy metals in leachate and for the purpose of forecasting the factors which are affecting the leaching of metalic waste in some degree, experimented the various reacting conditions. 1. When the concentration of heavy metals in leachate is in comparison with the level eluted after reaction, at pH 7.9 the result of reaction for PCB to CCL scrap showed that Zn, Mn, Cu was more eluted 11.6 times, 340.3 times, and 2,705.5 times respectively than the leachate undiluted solution. 2. At the condition of strong acid pH 4.7, the concentration of heavy metals in EM undiluted solution showed that Zn, Mn, Cu was more eluted 26.5 times, 147.3 times, and 3,656.3 times respectively than leachate undiluted solution. 3. When the ratio leachate to EM was 50 vs 50(V/V%), Mn was more eluted 198.7 times than leachate undiluted solution, but Zn and Cu do not show the meaningful results. 4. The color of landfill leachate was black-brown. And fulvic acid that is main ingredient of NBD COD contained, oxygen of 44~50%. For that reason, I estimated that the level of Zn, Mn, Cu was higher than the case of leachate. 5. COD of leachate from general landfill is difficult to remove. Because the solution of heavy metals is improved by the character of leachate(pH & ingredient of oxygen etc.) hence the Mn, Cu, Zn act as disturbing factor, the biochemical treatment is hard. Therefore the type of PCB & CCL scrap, iron, aluminum contained metals need to previously separate from general wastes as much as possible.
