The respiratory burst oxidase of neutrophils is a multicomponent enzyme, domant in resting cells, that catalyzes the reduction of oxygen to $O_{2}^{-}$ at the expense of NADPH. In the resting neutrophil, some of the components of the oxidase, including proteins p47 and p67, are in the cytosol, while the rest are in the plasma membrane. Recent evidence has suggested that at least some of the cytosolic oxidase components exist as a complex. The cytosolic complex with a molecular weight of ~240 kDa was found to bind to blue-agarose and 2',5'-ADP-agarose, which recognize nucleotide requiring enzymes. In order to identify the nucleotide binding component of the cytosolic complex we purified recombinant p47 and p67 fusion proteins using the pGEX system. Pure recombinant p47 was retained completely on 2',5'-ADP-agarose, whereas pure recombinant p67 did not bind to these affinity beads. On the basis of these results, we infer that p47 may contain the nucleotide binding site.
In order to assess controversial' proposals concerning the fatty acid-induced uncoupling of mitochondrial oxidative phosphorylation, we investigated the interaction of stearic acid with key mitochondrial proteins and measured the effect of stearic acid on the respiration of cytochrome c oxidase vesicles. Electron paramagnetic resonance spectra of spin-labeled stearic acid clearly demonstrated that cytochrome c oxidase interacts strongly with stearic acid. However, the respiration of detergent-solubilized cytochrome c oxidase was not altered significantly by stearic acid. Surprisingly, adenine nucleotide carrier, which was assumed to bind and translocate fatty acid anions in the Skulachev model of uncoupling, did not bind stearic acid at all. The respiration rate of cytochrome c oxidase vesicles was increased by ~70% in the presence of $20{\mu}m$ stearic acid and this uncoupling was attributed to a simple protonophoric effect of stearic acid.
Lactobacilli are normal constituents of the intestinal microbiota, and some strains show the capacity to bind to extracellular matrix proteins and components of the mucosal layer, which represents an adaptation to persist in this niche. A shotgun phage-display library of Lactobacillus casei BL23 was constructed and screened for peptides able to bind to fibronectin and collagen. Clones showing binding to these proteins were isolated, which encoded overlapping fragments of a putative transcriptional regulator (LCABL_29260), a hypothetical protein exclusively found in the L. casei/rhamnosus group (LCABL_01820), and a putative phage-related endolysin (LCABL_13470). The construction of different glutathione S-transferase (GST) fusions confirmed the binding activity and demonstrated that the three identified proteins could interact with fibronectin, fibrinogen, and collagen. The results illustrate the utility of phage display for the isolation of putative adhesins in lactobacilli. However, it remains to be determined whether the primary function of these proteins actually is adhesion to mucosal surfaces.
Synaptic vesicles (SVs) are key structures for synaptic transmission in neurons. Numerous membrane-associated proteins are sorted from the Golgi complex to the axon and the presynaptic terminal. Protein-protein and protein-lipid interactions are involved with SV targeting in neurons. Interestingly, many SV proteins have lipid binding capability, primarily with either cholesterol or phosphoinositides (PIs). As examples, the major SV protein synaptophysin can bind to cholesterol, a major lipid component in SVs, while several other SV proteins, including synaptotagmin, can bind to PIs. Thus, lipid-protein binding plays a key role for the SV targeting of synaptic proteins. In addition, numerous SV proteins can be palmitoylated. Palmitoylation is thought to be another synaptic targeting signal. Here, we briefly describe the relationship between lipid binding and SV targeting.
The effect of Ligand Binding energy in quantum rod (CdS/ZnS) plays a critical role in anisotropic growth. As mimicking large chain of ligands and using the head of the chain, I plan to bind the quantum rod and ligands so that it can grow well consequently. So the ultimate goal of this study is on how ligand binding can affect the growth of this quantum rod. There are preferred surfaces between the quantum rod and ligands, and we empirically know that ligands which bind the quantum rod; Phosphoric oxide (PO), Phosphoric acid(PA), Carboxylic acid(CA), Trimethylamine(TMA), have strong tendency to be attached on the surfaces of CdS/ZnS; ($11{\bar{2}}0$), ($10{\bar{1}}0$), ($000{\bar{1}}$), (0001). I virtually bond the surface and the ligands, and calculated the ligand binding energy after optimizing their structure, utilizing EDISON simulator. After all, I figured out how they are linked each other and how the quantum rod grows.
The action mechanism of hemolysin rendering virulency of Vibrio anguilarum has not clarified as yet, even though there were several possible factors explained. We have studied hemolytic kinetics performed by hemolysin from V. anguillarum strain V7 as well as binding of hemolysin to RBC membrane. Maximal rate of hemolysis and duration of lag phase were directly and inversly correlated to the concentration of hemolysin used. Hemolysin molecules are known to bind consumptively with proper diameter, while other protectants with smaller diameter could not. In conclusion, hemolysin should bind irreversibly to RBC membrane exert hemolysis distorting osmotic pressure. The binding could be hindered by spatial structure of the RBC surfacem which might be caused by sialic acid.

Thus, defining SOAP server objects and their methods are very important to provide services for web services. We define SOAP server objects for interaction databases such as BIND, MINT and DIP.
In this paper I develop a mechanism of interpreting Korean “wh-phrases”. The phrases have various readings depending on where they occur and they could be ambiguous even in the same structure. Yet their readings are subject to certain restrictions. I assume that the “wh-phrases” behave like variables and that there are three quantifiers to bind the phrases; $COMP_{wh},\;COMP_{conc}$ and derivational $\exists$-quantifier. Based on the assumptions I suggest derivational quantification, which consists of three conditions. 1) A quantifier can bind only when it merges into the derivation; 2) $\exists$-quantifier accompanies 〔-OP〕 complementizer but its activation is optional; 3) an instance of quantification makes the clause opaque to other instances of quantification. Scrambling data support derivational approach and across-the-board interpretation motivates the opacity condition. The opacity condition accounts for ATB- interpretations of reflexive pronouns. It can also explain the island effect of wh-islands without adopting covert wh-movement in Korean.
Transcription factors (TF) can bind tightly to specific DNA lesions formed by some anticancer agents. The formation these TF:(drug-modified DNA) complex may disrupt expression of genes critical for cell survival, and it was proved to be one of biochemical mechanisms of anticancer activity. Based on this model, we have designed programmable DNA Alkylating agents that can also attract TF, especially nuclear receptors. As a model compound, we designed drug molecules, RA-mustard and Am580-mustard, that enable to bind both retinoic acid receptor (RAR) and DNA by using molecular modeling techniques, and synthesized them by connecting chlorambucil and ligand for RAR with a linker unit. Objective: This study aimed to report the improvement in a patient with allergic contact dermatitis treated with Sihogyeji-tang based on the Shanghanlun disease pattern identification diagnostic system (DPIDS). Methods: According to the Shanghanlun DPIDS, the patient with allergic contact dermatitis was diagnosed with greater yang disease chest bind (Shanghanlun provision 146), and was administered Sihogyeji-tang for 90 days. The results were evaluated using the Dermatological Life Quality Index (DLQI) and a visual analog scale (VAS). Results: After taking Sihogyeji-tang for 90 days, the DLQI decreased from 28 points to 8 points and the VAS decreased from 10 points to 3 points. Conclusions: This case shows the potential of treatment through herbal medicine for allergic contact dermatitis using the Shanghanlun DPIDS.
