BCD를 섭취한 돼지의 부위별 콜레스테롤 함량 (Cholesterol Contents of Pork Fed Dietary β-Cyclodextrin)

  • 본 연구는 비육말기 돼지에서 베타사이클로덱스트린$\beta$-cyclodextrin의 급여가 돈육의 콜레스테롤 수준을 낮춘다는 것을 나타내고 있다. 120마리의 돼지에게 0, 5, 7, 10%의 $\beta$-cyclodextrin를 함유하는 사료를 35일 간 급여하였다. 혈액 총지질, 중성지방 및 총 콜레스테롤 함량은 $\beta$-cyclodextrin첨가구가 대조구에 비해서 낮게 나타났으며 처리구간 통계적인 유의차가 나타났다(p<0.05). 혈액 100 mL 당 지질은 대조구와 비교할 때 $\beta$-cyclodextrin 5.0%, $\beta$-cyclodextrin 7.0% 및 $\beta$-cyclodextrin 10.0% 첨가구에서 각각 63.22 mg, 73.98 mg, 82.12 mg의 유의적인 감소를 나타냈으며, 중성지방 56.24 mg, 55.48 mg, 60.02 mg, 총콜레스테롤 25.05 mg, 27.17 mg, 30.19 mg의 유의성 있는 감소경향을 보였다(p<0.05). 총스테로이드의 배설은 $\beta$-cyclodextrin 첨가수준이 증가할수록 대조구에 비해서 유의적으로 높게 나타났다(p<0.05). 돼지고기의 부위별, 등지방, 삼겹살, 등심 및 햄의 콜레스테롤 함량은 대조구에 비해서 $\beta$-cyclodextrin 첨가수준이 높아질수록 유의적인 감소경향을 나타냈다(p<0.05). 돼지고기의 콜레스테롤 함량 감소율은 $\beta$-cyclodextrin 첨가구가 대조구와 비교할 때 15.31% 이상의 높은 감소율을 나타냈다(p<0.05). 본 연구 결과는 비육말기 돼지사료 내 $\beta$-cyclodextrin를 혼합 급여하여서 콜레스테롤 함량이 낮아진 새로운 기능성 돈육을 생산할 수 있음을 시사해준다.

난황의 콜레스테롤 제거에 사용한 $\beta$-Cyclodextrin의 재활용 (Recycling of $\beta$-Cyclodextrin Used for Cholesterol Removal from Egg Yolk)

  • The method used to remove cholesterol from egg by using $beta$-cyclodextrin was relatively stable and efficient. The aim of this study was to cost down by recycling $\beta$-cyclodextrin used to remove cholesterol from egg yolk because $\beta$-cyclodextrin was expensive. The solvents used to separate $\beta$-cyclodextrin from $\beta$-cyclodextrin complex containing egg yolk cholesterol were butanol, chloroform, ether, hexane, methanol, 2-propanol and their mixture. The ratio of solvent and complex varied from 2 : 1 to 10 : 1. The condition of mixing time and temperature varied from 30 to 60$^{\circ}C$ and from 10 minutes to 3 hours to remove cholesterol from $\beta$-cyclodextrin complex. When the ratio of choloroform and methanol was 1 : 1, the removal efficiency of cholesterol was 98.8%. The efficiency of cholesterol removal was improved when the ratio of solvent : complex increased to 4 : 1. When mixing time and temperature was up to for 1hr, at 50$^{\circ}C$ respectively, the efficiency of cholesterol removal improved to 99%. It concluded that the efficiency of cholesterol removal of 50% renewed one contained $\beta$-cyclodextrin were 81.1% while the cholesterol removal efficiency of 100% renewed $\beta$-cyclodextrin was 24% if cholesterol removal efficiency of new $\beta$-cyclodextrin were 100%.

  • Data are present for the interaction of beta-cyclodextrin with 7 pharmaceuticals in aqueous solution. By means of the solubility method of analysis, definite interaction were found to occur with all the compounds tested. The amount of these pharmaceuticals in equilibrium with their respective solid phase increased progressively with beta-cyclodextrin. A concentration of 1.5 g of beta-cyclodextrin per 100 ml of water increased the solubility of most compounds tested (except one) by 15-120%. Hydrolysis in alkali solution of chrolothiazide was retarded in the presence of beta-cyclodextrin.

양귀비 세포 현탁배양계에서 Cyclodextrin을 이용한 Benzophenanthridine alkaloids의 생산성 증대 (Production Enhancement of Benzophenanthridine alkaloids in the Suspension Cultures of California poppy using Cyclodextrin)

  • 본 연구에서는, 최근 들어 관심의 대상이 되어 의 품 산업에 응용의 수가 늘고 있는 cyclodextrin을 이용하여 세포밖으로 배출된 식물세포 2차 대사물질을 신속히 포획하여 생산수율을 증대시키고자 하였다. 실험결과 양귀비 현탁배양에서 a-, y-cyclodex trin보다는 ${\beta}$-cyclodextrin이 benzophenan-thridine alkaloid를 생산하는데 더 효과적업을 알았다. 이때 ${\beta}$-cyclodextrin의 optimum concentration이 존재하였는데, 그 농도는 현탁배양배지의 1.5% (w/v) 부 근에서였다. 고체 배지에서 ${\beta}$-cyclodextrin의 첨가는 sangumarme의 생산성을 control의 경우보다 약 4 40배(포함), 약 17배(살포) 정도 증가시켰다. ${\beta}$-cyclodextrin은 현탁배양 배지의 수용액상에서 세포 외로 배출된 alkaloid를 신속히 포획하여 inclusion complex를 형생함으로써 product의 배출을 촉진하여 결과적으로, product pattern을 변화시켜 줌으로 써 생산과 분리가 통시에 이루어질 수 있는 연속공정의 가능성을 확인시켜주었다. 또한 $\beta$cyclodex trin 1.5%(w/v)을 첨가시기를 달리하여 실험한 결과 배양초기에 넣어주는 것이 효과적이었으며, 이때 ${\beta}$-cyclodextrin은 세포성장에 미치는 영향이 없음을 확인하였다. ${\beta}$-cyclodextrin에 의한 benzophenan­t thridine alkaloid의 엄청난 생산 중대 효과는 1) In­c clusion complex 형성에 의한 안정성 증대뿐만 아니라 2) elicitor로서의 역할 등의 복합적 인 요인을 생 각해 볼 수 있는데 2)의 경우에 대한 실험을 세포와 ${\beta}$-cyclodextrin간의 직접적인 접촉을 시키지 않고 해본 결과 미세한 영향을 나타내었으므로 elicitation 의 경우를 완전히 배제한다고 할 수 없었다. 그러나 uv에 의한 inclusion complex의 확인이 1)의 경우를 확인하여 주었으므로 본 실험에서 ${\beta}$-cyclodex­t trin의 역할은 inclusion complex에 의한 생산성 증대라 할 수 있다.

  • Inclusion complex of omeprazol with $\beta$-Cyclodextrin and Hydroxypropyl-$\beta$-Cyclodextrin were prepared by coprecipitation and freeze-drying method respectively. Effects of these inclusion complex on RBCs were monitored with a spectrophotometer by the method of Kahan et al. and the mutagenic activity based on the Ames plate incorporation test in the presence and absence of liver microsomal enzyme(S9 fraction) using Salmonella typhimurium TA98 and TA100. The RBCs hemolysis and mutagenic activity of these complex were not detected.

Water-Activity Depressor가 Pullulanase의 역반응에 의한 Maltosyl-$\beta$-Cyclodextrin의 합성에 미치는 영향 (Effect of Water-Activity Depressor on the Enzymatic Synthesis of Maltosyl-$\beta$-Cyclodextrin through the Reverse Reaction of Pullulanase)

  • Maltosy-$\beta$-cyclodextrin는 $\beta$-cyclodextrin에 maltose가 $\Alpha$-1,6 glycosidic bond로 결합된 분지환상결합체로서 pullulanase의 역반응(축합반응)을 이용하여 $\beta$-cyclodextrin과 maltose로부터 합성된다. Maltosyl-$\beta$-cycloextrin의 합성수율을 증가시키기 위하여 각종 water activity depressor인 각종 polyol, sugar 그리고 polyethylene glycol(PEG)등의 첨가의 영향을 검토하였다. 가장 적절한 water activity depressor는 PEG 6000로서, 첨가량 10%(w/w)의 경우 maltosyl-$\beta$-cyclodextrin 생성량과 합성수율은 크게 증가하여 3.02g/100ml 와 55.9%(w/w)로서, 첨가하지 않았을 경우보다 약 1.3배 증가하였다. Water activity는 PEG 6000을 20%(w/w) 첨가할 때 원래의 0.966에서 0.914로 감소하였으며, maltosyl-$\beta$-cyclodextrin 합성수율은 water activity에 반비례하여 증가하였다. Pullulanase의 역반응을 이용한 maltosyl-$\beta$-cyclodextrin 합성반응의 각종 열역학적 상수를 평가하였으며, $\Delta$H는 36.788kJ/mol, $\Delta$S는 0.067kJ/moleK, 그리고 $\Delta$G는 14.433kJ/mole이였다. PEG 6000의 분리회수에 적절한 ultrafiltration membrane의 pore size는 3K dalton이었으며, 여과액과 농축액의 적정 분획비는 1.0 : 9.0였다. Maltosyl-$\beta$-cyclodextrin 합성수율의 증가는 첨가한 PEG가 water activity를 감소시켜 합성된 maltosyl-$\beta$-cyclodextrin이 재분해되는 pullulanase이 정반응인 hydrolysis reaction을 억제하여 equilbrium state에 변화를 주기 때문인 것으로 유추된다.

[ ${\beta}-cyclodextrin$ ] inclusion properties with guest molecules using hetero-bi-functional reactive dye

  • Cyclodextrin is a cyclic oligosaccharid material which shows an ability to incorporate organic guest molecules inside their cavity area. Thus, this ${\beta}-cyclodextrin$ treatment on fiber substrates may provide the changed surface characteristics of the substrates such as solubility, chemical reactivity and spectral property. In this context, the aim of this present work is to make a bridge connection using hetero-bi-functional reactive dye between fiber substrates and ${\beta}-cyclodextrin$. In addition, the corresponding Berberine inclusion behaviors into the inner cavity of ${\beta}-cyclodextrin$ was examined. The %exhaustion of Berberine inclusion as a guest molecule within the ${\beta}-cyclodextrin$ was measured using UV-Vis spectrophotometer. The findings showed that the %exhaustion of Berberine inclusion increased with increasing the prepared dye bridge compound and ${\beta}-cyclodextrin$ host material.

Peppermint 세포 현탁배양에서 Cyclodextrin을 이용한 Menthol의 생산성 증대 (Production Enhancement of Menthol in Suspension Cultures of Peppermint Using Cyclodextrin)

  • The suspension cultures of Mentha piperita produce menthol which has very low solubility in water due to its hydrophobicity. This can be considered as a factor for its low production in the suspension suspension cultures. Cyclodextrin has the hydrophobic cavity inside the molecule in which menthol can be captured and allow to form a stable complex. The suspension culture of Mentha piperita showed 70% higher production enhancement in the medium containing 1.5%(w/v) $\beta$-cyclodextrin than the control. $\beta$-cyclodextrin had no adverse effect on the cell growth and showed the best result among $\alpha$-, $\beta$- and $\gamma$-cyclodextrins tested in terms of menthol production. We demonstrated that $\beta$-cyclodextrin can be used to enhance the production of menthol in the suspension cultures by capturing hydrophobic menthol into the cavity of cyclodextrin molecules.

Octyldimethl p-aminobenzoate 와 ${\beta}-Cyclodextrin$의 포접화합물(包接化合物)에 관(關)한 연구(硏究) (A Study on the Inclusion Complexation of Octyldimethyl p-aminobenzoate with ${\beta}-Cyclodextrin$)

  • Inclusion complex formation of octyldimethyl p-aminobenzoate with ${\beta}-cyclodextrin$in aqueous solution and in the solid state was studied by the solubility method, spectroscopic (UV, FT-IR) and X-ray diffractornetry. The solid complex of octyldimethyl p-aminobenzoate with ${\beta}-cyclodextrin$ was obtained in molar ratio of 1:2 (guest/host). A spatial relationship between host and guest molecule was clearly reflected in the magnitude of the apparent stability constant (K') and in the stoichiometry of the inclusion complex. Furthermore, a typical type Bs phase-solubility diagram was obtained for octyldimethyl p-aminobenzoate and ${\beta}-cyclodextrin$ in water at $25^{\circ}C$. The results indicated that the solubility of the guest molecule was higher by the formation of ${\beta}-cyclodextrin$ inclusion complex.

테르페나딘-${\beta}$-시클로덱스트린 포접화합물의 제조방법 및 물리화학적 특성 (Preparing Method and Physico-chemical Characteristics of $Terfenadine-{\beta}-Cyclodextrin$ Inclusion Compound)

  • Terfenadine, antihistaminic drug, is poorly soluble in water. The purpose of this study is to investigate the possibility of using $terfenadine-{\beta}-cyclodextrin$ inclusion compound, instead of terfenadine, as the active substance of solid dosage form by improving the solubility, dissolution and anti-histaminic activity of terfenadine. The solubility and binding characteristics of $terfenadine-{\beta}-cyclodextrin$ complex in pH $1.2{\sim}6.8$ were investigated. Furthermore, the preparing method of $terfenadine-{\beta}-\;cyclodextrin$ inclusion compound was setting up and its physico-chemical characteristics such as DSC curve, solubility, dissolution and anti-histaminic activity were investigated. In conclusion, the solubility of terfenadine was increasing ${\beta}-cyclodextrin$ and with the decreasing pH. $Terfenadine-{\beta}-cyclodextrin$ inclusion compound, whose yield is almost 100%, was prepared by neutralization method. This inclusion compound was 200-times as soluble as terfenadine in pH 1.2-6.8. In addition, it had the faster dissolution and anti-histaminic activity than terfenadine. Therefore, it is used to the active substance of solid dosage form such as tablet and capsule in stead of terfenadine.
