감과실의 성숙과 추숙중의 $\beta$-Galactosidase활성 변화 및 특성 (Characteristics and Activity Changes of $\beta$-Galactosidase during Maturation and Postharvest of Persimmon Fruits)

  • $\beta$-Galactosidase는 미숙감에서는 활성이 없었으나, 완숙감과 연시에서 각각 21.79, 380.23 unit/ 100g-fr. wt.이었으며 연시에서 현저히 증가하였다. $\beta$-Galactosidase는 isoenzyme이 존재하지 않았고, 분자량은 115,000dalton이었다. $\beta$-Galactosidase의 Km값은 1.84$\times$$10^{-2}$ mM, Vmax는 0.095 mmole p-nitrophenyl-$\beta$-galactoside degraded/$m\ell$/ 15min.이었고 최적 pH는 4.2. 최적온도는 45$^{\circ}C$이었으며, 6$0^{\circ}C$에서 10분간 열처리하였을 때 활성이 50%소실되었다. $\beta$-Galactosidase의 활성에 SDS는 저해하였으나 Mg++과 Ca++은 영향을 주지 않았다.

$TNF{\alpha}$ Increases the Expression of ${\beta}2$ Adrenergic Receptors in Osteoblasts

  • Tumor necrosis factor alpha ($TNF{\alpha}$) is a multifunctional cytokine that is elevated in inflammatory diseases such as atherosclerosis, diabetes and rheumatoid arthritis. Recent evidence has suggested that ${\beta}2$ adrenergic receptor (${\beta}2AR$) activation in osteoblasts suppresses osteogenic activity. In the present study, we explored whether $TNF{\alpha}$ modulates ${\beta}AR$ expression in osteoblastic cells and whether this regulation is associated with the inhibition of osteoblast differentiation by $TNF{\alpha}$. In the experiments, we used C2C12 cells, MC3T3-E1 cells and primary cultured mouse bone marrow stromal cells. Among the three subtypes of ${\beta}AR$, ${\beta}2$ and ${\beta}3AR$ were found in our analysis to be upregulated by $TNF{\alpha}$. Moreover, isoproterenol-induced cAMP production was observed to be significantly enhanced in $TNF{\alpha}$-primed C2C12 cells, indicating that $TNF{\alpha}$ enhances ${\beta}2AR$ signaling in osteoblasts. $TNF{\alpha}$ was further found in C2C12 cells to suppress bone morphogenetic protein 2-induced alkaline phosphatase (ALP) activity and the expression of osteogenic marker genes including Runx2, ALP and osteocalcin. Propranolol, a ${\beta}2AR$ antagonist, attenuated this $TNF{\alpha}$ suppression of osteogenic differentiation. $TNF{\alpha}$ increased the expression of receptor activator of NF-${\kappa}B$ ligand (RANKL), an essential osteoclastogenic factor, in C2C12 cells which was again blocked by propranolol. In summary, our data show that $TNF{\alpha}$ increases ${\beta}2AR$ expression in osteoblasts and that a blockade of ${\beta}2AR$ attenuates the suppression of osteogenic differentiation and stimulation of RANKL expression by $TNF{\alpha}$. These findings imply that a crosstalk between $TNF{\alpha}$ and ${\beta}2AR$ signaling pathways might occur in osteoblasts to modulate their function.

Kluyveromyces fragilis의 Beta- galactosidase에 의한 Oligosaccharide의 생성 (Production of Oligosaccharide by Beta-galactosidase of Kluyveromycces fragilis)

  • Kluyveromyces fragilis로부터 $\beta$-galactosidase의 생성조건과 조효소액의 성질 그리고 당 전이 반응에 의한 oligosaccharide 생성을 조사한 결과 다음과 같은 결과를 얻었다. \circled1 Peptone-Yeast extract 배지에 6% lactose를 첨가하였을 때 최대 효소생산을 보였다. \circled2 0.05 M potassium phosphate buffer (pH 7.0)에서 3% toluene를 첨가하여 37$^{\circ}C$ 5시간 배양하였을 때 K. fragilis로부터 효소가 최대로 추출되었다. \circled3 효소의 최적온도는 4$0^{\circ}C$이고 4$0^{\circ}C$ 이상의 열처리에서는 효소가 파괴되었으며, pH6-7에서는 상당히 안정하였다. \circled4 ONPG 기질로 사용하였을 때 Km 값은 2.5mM이었다. \circled5 당 전이 반응의 결과, 7개의 oligosaccharide가 생성되었다. 이상의 실험결과로 볼 때, 본 실험에 사용한 K. fragilis SKD 7001은 Beta-galactosidase의 생산을 위해서 이용 가치가 인정되었으며, 특히, 이 효소의 활성이 중성 pH에서 강하고 안정한 상태를 보이는 것은 시유나 원료우유의 lactose를 중성에서 해야 한다는 점을 고려할 때, 실용적 가치가 있다고 본다. 또, 이 효소의 작용과정에서 생성되는 oligosacchride는 장내 bifidus균의 생육을 촉진시키는 효과가 인정되고 있기 때문에 이용가치를 더욱 높여주는 것으로 생각된다.

Lithium Alumino Silicate계 유리에서 알카리 토류 첨가에 따른 $\beta$-quartz고용체의 형성에 관한 연구 (The formation of $\beta$-quartz solid silution in lithiu alumino silicate glasses)

  • 본 연구는 Lithium Alumino Silicate 기본계에 핵형성제로 $TiO_{2}$$ZrO_{2}$를 사용하여 기본유리를 제조하고 열처리에 따른 $\beta$-quartz고용체의 미결정 형성과정을 분석하였다. 또한 $\beta$-quartz고용체의 형성에 따른 열적 특성과 광학적 특성을 비교하였다. 그 결과, 기본 유리의 열팽창 계수는 $45\sim 55 \times 10^{-7} \textrm{cm}/^{\circ}C$였으며, 결정화 시편의 열팽창 계수는$ -8\sim +8 \times 10^{-7}\textrm{cm}/^{\circ}C$ ($25^{\circ}C \sim 525^{\circ}C$)였다. $900^{\circ}C$이하에서 열처리된 Glass Cermics의 결정은 $\beta$-quartz 고용체가 형성되었고, 결정의 크기가 0.21$\mu \textrm{m}$를 초과하지 않았으며, 최고의 투광도는 80%이상을 나타냈다. RO성분을 MgO와 ZnO를 사용한 경우보다 MgO 단독으로 사용한 시편을 결정입자의 크기가 온도에 의존하지 않고 일정함을 나타냈으며, ZnO의경우는 입자의 크기가 최소값(0.18$\mu \textrm{m}$)을 보이며 온도의존성이 크게 나타냈다.

Ti-6Al-2Sn-4Zr-6Mo 합금의 고온압축 변형거동 (High Temperature Compressive Deformation Behavior of Ti-6Al-2Sn-4Zr-6Mo Alloy)

  • Ti-6Al-2Sn-4Zr-6Mo(Ti6246) 합금의 고온 변형거동을 조사하기 위하여 $\alpha$+$\beta$ 영역 및 $\beta$영역의 온도에서 $10^0s^{-1}$에서 $10^{-3}s^{-1}$의 변형속도로 압축시험을 수행하였다. 유동응력은 변형속도가 증가하고 시험 온도가 감소함에 따라 증가하였다 90$0^{\circ}C$ 이하에서 시험한 결과로부터 얻어진 유동곡선은 가공연화 현상을 나타내었으나, 이 합금의$\beta$영역인 95$0^{\circ}C$ 이상에서는 유동응력이 지속적으로 증가하다가 정상 상태를 나타냈다. 압축시험 결과로부터 얻은 유동곡선 분석을 통하여 유동응력의 변형량, 변형속도 및 온도에 대한 관계로부터 이 합금에 대한 구성방정식을 구하였다.

  • 순수분리정제된 $\beta$-galactosidase의 분자량은 Sephadex G-200 gel filtration법에서 130,000이며 SDS-polyacrylamide gel electrophoresis에서 130,000외에 70,000이 확인되어 이 효소는 70,000인 2개의 subunit로 구성되어 있다고 판단되었다. 효소의 안정pH는 4.5~7.0이며 효소활성의 최적 pH는 4.7 최적온도는 5$0^{\circ}C$였다. 효소활성 및 안정제로 1가, 2가 금속ion을 필요로 하지않았으며 C $u^{++}$ 1mM에서 59%, galactose 100mM에서 48% 저해되었다. 5% lactose용액, 시유 및 10% skim milk 용액에 이 정제된 $\beta$-galactosidase 10units/$m\ell$를 사용하여 5$0^{\circ}C$에서 4시간 동안 반응시켰을 때 lactose가 각각 69.5%. 88.7% 및 72.6%가 glucose와galactose로 전환된 결과를 얻었다.

  • The $\beta$-SiC + $ZrB_2$ ceramic electroconductive composites were pressureless-sintered and annealed by adding 12wt% $Al_2O_3$ + $Y_2O_3$ (6 : 4wt%) powder as a function of sintering temperature. The relative density showed the highest value of 81.1% at 1900$^{\circ}C$ sintering temperature. The phase analysis of the composites by XRD revealed of $\alpha$-SiC(6H), $TiB_2$, $Al_5Y_2O_{12}$ and $\beta$-SiC(15R). Flexural strength showed the highest value of 230 MPa for composites sintered at 1900$^{\circ}C$. The vicker's hardness and the fracture toughness showed the highest value of increased with increasing sintering temperature and showed the highest of 9.88 GPa and 6.05 $MPa{\cdot}m^{1/2}$ at 1900$^{\circ}C$. The electrical resistivity was measured by the Pauw method from 25$^{\circ}C$ to 700$^{\circ}C$. The electrical resistivity of the composites showed the PTCR (Positive Temperature Coefficient Resistivity).

소염정장탕(消炎整腸湯)이 DSS로 유발(誘發)된 생쥐의 궤양성 대장염에 미치는 영향(影響) (The Effects of Soyumjungjang-tang on DSS-Induced Ulcerative Colitis in Mouse)

  • Objectives : This study was carried out to investigate the effects of Soyumjungjang-tang(SJT) on the experimental ulcerative colitis induced by dextran sulfate sodium(DSS) in mice. Methods : Ulcerative colitis was induced through supplying 4% DSS solution as the drinking water for 7 days in 6-week-old male ICR mice. The colitic mice were divided into three groups: the sample groups were orally administered SJT in doses of 25mg/kg(S25 group) or 100mg/kg(S100 group) once a day for 10 days, from 3 days before starting drinking the DSS solution, and the control(C) group was administered normal saline instead of SJT. The DSS solution or SJT was not administered to the normal(N) group. The length of colon, histologic finding, the activities of myeloperoxidase(MPO) and alkaline phosphatase(AP), and the expressions of $IL-1{\beta}$, IL-6, COX-2, $NF-{\kappa}B$, and $I{\kappa}B$ in colonic mucosa was checked using immunoblot, ELISA, etc. The activities of chondroitinase, tryptophanase, ${\beta}-glucuronidase$ and ${\beta}-glucosidase$ in stool were also measured. Results : The length of colon shortened, histologic finding deteriorated, the activities of MPO, AP, chondroitinase, tryptophanase, ${\beta}-glucuronidase$ and ${\beta}-glucosidase$, and the expressions of $IL-1{\beta}$, IL-6, COX-2, $NF-{\kappa}B$ increased, and the expression of $I{\kappa}B$ decreased in the C group. All measures, except $NF-{\kappa}B$, were restored in S25 group, but some measures deteriorated more in the S100 group than in the C group. Conclusions : According to the above results, it is supposed that SJT has a potential therapeutic effect on ulcerative colitis.

근육세포 분화에 대한 TGF-β1과 OP-1의 억제 효과 (The Inhibitory Effect of TGF-β1 and OP-1 onto the Myogenic Differentiation)

  • In order to investigate the effect of Transforming growth factor ${\beta}1$(below TGF-${\beta}1$) and osteogenic protein-1(below Op-1) onto the myogenic differentiation, C2C12 satellite myoblastic cell line was cultured and treated with both growth factors. At first morphological changes with microscopical examination were examined, and isolated total RNA to analyse mRNA expression of bone marker proteins, muscle regulatory proteins, TGF-${\beta}$ receptor and their ligands by Northern blot analysis. And cellular proliferative inducibility of both growth factors was also tested to C2C12 cells. Incubating the cell with $5ng/m{\ell}$ of TGF-${\beta}1$ until 4 days almost inhibited multinucleated myotube formation expressing muscular regulatory proteins, and induced decreasing Id proteins. However, no osteoblastic phenotypes was induced by TGF-${\beta}1$ in C2C12 cells. The mRNA expression of TGF-${\beta}$ receptors with TGF-${\beta}1$ was conversed after 48 hours cultured. Type I TGF-${\beta}$ receptor was seemed to play a role in negative signalling for inhibition of myogenic differentiation. OP-1 dose dependently induced ALP activity, osteopontine production and bone sialoprotein production at concentrations above $100ng/m{\ell}$ and osteocalcin production at concentrations above $300ng/m{\ell}$. The concentration of OP-1 required to induce these osteoblastic phenotypes was the same as that required to almost completely inhibit myotube formation. Incubation with above $100ng/m{\ell}$ OP-1 suppressed the expression of mRNA for muscular egulatory proteins from 2 days after incubation. Expression of Id-1, 2, 3 mRNA were stimulated by OP-1 at concentration above $300ng/m{\ell}$. When C2C12 cells were treated with both growth factors, TGF-${\beta}1$ potentiated the inhibitory effect of OP-1 on myotube formation and expression of mRNA for myogenin at 12 days. And TGF-${\beta}1$ reduced osteocalcin and bone sialoprotein production induced by OP-1 at 12 days in C2C12 cells. Both growth factor had no mitogenic effect. These results indicate that OP-1 converts the differentiation pathway of C2C12 myoblasts into that of osteoblastic lineage cells and it's not heritable, but TGF-${\beta}1$ does not and has reversible inhibitory activity on the myogenic differentiation. TGF-${\beta}1$ and OP-1 play a role in myogenic differentiation via different mechanism between them.
