담배 현탁배양 세포로부터 $\beta-1$,4-endoglucanase의 정제 및 성질 (Purification and Properties of $\beta-1$, 4-endoglucanase from Tobacco Suspension Cultured Cells)

  • $\beta$-1, 4-endoglucanase (EC 3, 2, 1, 4) was isolated and purified from Nicotiana tabaccum L. Var. Virginia 115 suspensin cultured cells. The molecular weight as estimated by Sephadex G-100 was about 14, 000. The optimum pH for activity was 5.4. The Km value for carboxymethyl cellulose was 0.18 mg/unit and the enzyme was quite resistant to heating. Polyamine did not affect the activity of $\beta$-1, 4-endoglucanase in vitro but the activity increased drastically when polyamines were added in the suspension culture medium. It is suggested that increase in $\beta$-1, 4-endoglucenase activity due to polyamine might be related to growth of the plant.

  • Trichoderma reesei KCTC 6952는 고분자 섬유소인 cellulose를 분해하는데 관여하는 3종류의 효소로 구성된 cellulase system을 보유하고 있다. 그 중 ${\beta}$-glucosidase는 glucose를 함유한 glycoside 유도체들로부터 glucose를 효율적으로 유리할 수 있는 것으로 알려져 있다. 본 연구에서는 변형된 Mandels의 배지를 사용하여 T. reesei KCTC 6952를 배양하면서 배지로 분비된 ${\beta}$-glucosidase를 조효소액으로 회수하여 조효소액내의 ${\beta}$-glucosidase 활성을 측정하고, 또한 효소활성이 온도와 pH에 대하여 어느 정도 안정한 지를 조사하였다. 그 결과 배양 4일째에 ${\beta}$-glucosidase의 효소활성이 최고(1.33 unit/mL)에 도달하였으며, 조효소액내의 ${\beta}$-glucosidase의 최적 반응 조건은 pH 5과 $70^{\circ}C$에서 10분간 반응하는 것이었으며, 그리고 효소의 안정성을 실험한 결과에 의하면 pH 4-5의 범위에서, $50^{\circ}C$ 이하에서는 안정적이었다. 그러므로 결론적으로 본 연구를 위하여 선택된 ${\beta}$-glucosidase 조효소액은 비교적 높은 온도와 넓은 pH범위에서 촉매반응을 일으킬 수 있었다.

Cyanoethyl화에 의한 제지용 섬유의 화학적 개질효과에 관한 연구 (A Study on Chemical Modification Effect of Papermaking Fiber by Cyanoethylation)

  • Since there are three hydroxyl groups on each anhydroglucose ring of the cellulose, the renewable resources, we can get various functional papers by the chemical modification of cellulose. The reaction involving the introduction of the ${\beta}$-cyanoethyl ($-CH_2-CH_2$-CN) group into organic substances containing reactive hydrogen atoms is known as cyanoethylation. Cellulose reacts with acrylonitrile in the presence of strong alkalis in a typical manner of primary and secondary alcohols to form cyanoethyl ethers. In cyanoethylation, important factors of reaction are temperature, concentration of the NaOH, and addition rate of acrylronitrile. FT-IR spectra of cyanoethyl group was confirmed at $2250cm^{-1}$, which corresponds the introduction of aliphatic nitrile group. Effect of cyanoethyl DS(degree of substitution) on strength properties was resulted that cyanoethylated BKP of DS 0.04 appeared to be the best choice for overall strength properties. Also, excellent thermal stability in aging characteristics was obtained.

Trichoderma harzianum 유래 ${\beta}$-mannanase에 의한 Locust Bean Gum 가수분해 올리고당의 동정 및 Bifidobacterium spp.에 대한 생육활성 (Identification and Growth Activity to Bifidobacterium spp. of Locust Bean Gum Hydrolysates by Trichoderma harzianum ${\beta}$-mannanase)

  • Trichoderma harzianum이 생산하는 ${\beta}-mannanase$의 최적 사면배지조성은 2.0% malt extract, 2.0% glucose, 0.1% peptone, 2.0% agar로, 효소생산 배지조성은 3.0% cellulose, 3.0% C.S.L, 1% $KH_2PO_4$, 0.2% $(NH_4){_2}SO_4$로 결정되어 $28^{\circ}C$, 130 rpm, 진탕배양법으로 조효소액을 생산하였다. ${\beta}-mannanase$의 최적 pH와 최적온도는 pH 4.5, $60^{\circ}C$에서 최대 상대활성을 나타내었다. Locust bean gum에 대한 효소적 가수분해 pattern을 TLC에 의해 검토한 결과 반응초기부터 반응말기까지 주요 생성물은 단당류, 중합도 4와 7의 올리고당으로 확인되었다. 중합도별 올리고당을 조제하기 위하여 activated carbon column chromatography에 250 ml/hr유속으로 tube당 20 ml씩 ethanol $0{\sim}50%$의 gradient법으로 분리 회수하였고, 분리된 각각의 올리고당 중 중합도4는 TLC $R_f$ value상으로 ${\beta}-1,4-mannotetraose$로, 중합도 7은 Methylation method에 의해 M-M-M-M-M 구조로 //G-G 동정되었다. (G-와 M-은 각각 ${\alpha}-1,6-D-galactosidic,\;{\beta}-1,4-mannosidic$ 결합을 나타냄). Bifidobacterium longum에 대한 생육활성을 기존의 MRS media와 탄소원으로 중합도 4와 7의 올리고당으로 대체하였을때 3.4배, 4.3배의 생육활성이 증가하였고, Bifidobacterium bifidum에 대해서는 1.2배, 1.1배의 낮은 활성을 나타내었다.

구상나무(Abies koreana Wilson)재(材)의 화학적(化学的) 조성(組成) (The Chemical Composition of Abies koreana Wilson Wood)

  • 한국(韓國)의 특산수종(特産樹種) 구상나무재(材)의 화학적(化学的) 조성분(組成分)을 분석조사(分析調査)하여 본바 그 결과(結果)를 요약(要約)하여 보면 아래와 같다. 1. 회분함량(灰分含量)의 범위(範囲)는 0.33~0.76%로서 평균(平均) 0.56%였다. 2. 냉수추출물(冷水抽出物)은 5.50~12.50% 범위(範囲)의 함량치(含量値)를 보였는데 평균(平均) 8.76%였다. 3. 열수추출물(熱水抽出物)은 4.80~13.65%로서 평균(平均) 10.16%의 함량치(含量値)를 보였다. 4. 염기추출물(塩基抽出物)은 최소(最少) 5.51%에서 최고(最高) 25.44%의 함량치(含量値)를 보였는데 평균(平均) 4.23%였다. 5. Alcohol-Benzol 추출물(抽出物)은 2.94~5.44%로서 평균(平均) 4.23%였다. 6. Holocellulose 함량(含量)은 73.68%에서 79.10%의 정량치(定量値)를 보였는데 평균(平均) 76.49%였다. 7. Cellulose 함량치(含量値)는 평균(平均) 56.30%로서 46.02~61.33%의 범위(範囲)였고 이중(中) ${\alpha}$-Cellulose가 78.34%, ${\beta}$-Cellulose가 7.66%, ${\gamma}$-Cellulose가 14.04%였다. 8. Klason lignin의 함량치(含量値)는 평균(平均) 25.30%로서 22.50~27.00% 의 범위(範囲)였다. 9. Pentosan 함량(含量)은 7.90~13.66%의 정량치(定量値)를 보였는데 평균(平均) 10.44%였다. 이상(以上)의 결과(結果)를 종합(綜合)하면 회분함량(灰分含量)과 각추출물(各抽出物)의 함량(含量)은 대체(大体)로 타쇠업수재(他釙業樹材)와 비슷하였고 Holl cellulose 함량(含量)은 76.49%로서 잣나무재(材)에서 보다 정량치(定量値)가 높고 소나무재(材)와는 비슷하였으며 특(特)히 Cellulose 함량 56.30% 중(中) 78.34%의 높은 ${\alpha}$-Cellulose 함량율(含量率)을 보여 우수(優秀)한 Pulp화학(化學) 공업재료(工業材料)인 것으로 사료(思料)된다.

전기영동분석법(電氣泳動分析法)에 의(依)한 치근단낭종((齒根端囊腫)과 치근단육아종(齒根端肉牙腫)의 감별(鑑別)에 관(關)한 연구(硏究) (A STUDY ON ELECTROPHORETIC DIFFERENTIATION OF RADICUAR CYSTS AND GRANULOMAS)

  • Cellulose acetate electrophoresis was used for differential diagnosis of radicular cysts and granulomas. Sixteen periapical lesions were excised from sixteen teeth with radiographic evidence of periapical pathosis, and two normal periodontal tissues were surgically removed from two wisdom teeth. The tissue samples were separated into two parts. Half was prepared for cellulose acetate electrophoresis, and the electrophoretic patterns were scanned with Helena densitometer. The other half was examined histologically, and the histologic diagnoses were then compared with the electrophoretic patterns and with the x-ray findings. The results were the following: 1. The histopathologic results showed the presence of 13 granulomas (81.8%) and 3 cysts (18.3%). 2. An albumin pattern, alpha-globulin pattern, beta-globulin pattern, fibrinogen pattern and gamma-globulin pattern were found in all eighteen cases. 3. All eighteen cases were divided into normal periodontal tissue group, dental granuloma group, and radicular cyst group by the histopathologic diagnosis. The electrophoretic pattern of dental granuloma group and that of normal periodontal tissue group resembled each other. The two groups showed the highest percentage in beta-globulin fraction, and the beta-globulin of dental granuloma group(47.17%) was higher than that of normal periodontal tissue group (40.54%) by 6.63%. On the contrary, beta-globulin fraction of radicular cyst group (28.00%) was much lower than those of the granuloma group (47.17%) and normal periodontal tissue group (40.54%), and alpha-globulin of radicular cyst group (34.16%) was much higher than those of the granuloma group (20.04%) and normal group (19.58%). 4. Five cases (31.3%) of the sixteen periodontal lesions showed different results between histopathologic diagnoses and radiographic findings.

Enzymatic Characteristics of a Highly Thermostable β-(1-4)-Glucanase from Fervidobacterium islandicum AW-1 (KCTC 4680)

  • A highly thermostable ${\beta}-(1-4)-glucanase$ (NA23_08975) gene (fig) from Fervidobacterium islandicum AW-1, a native-feather degrading thermophilic eubacterium, was cloned and expressed in Escherichia coli. The recombinant FiG (rFiG) protein showed strong activity toward ${\beta}-{\small{D}}-glucan$ from barley (367.0 IU/mg), galactomannan (174.0 IU/mg), and 4-nitrophenyl-cellobioside (66.1 IU/mg), but relatively weak activity was observed with hydroxyethyl cellulose (5.3 IU/mg), carboxymethyl cellulose (2.4 IU/mg), and xylan from oat spelt (1.4 IU/mg). rFiG exhibited optimal activity at $90^{\circ}C$ and pH 5.0. In addition, this enzyme was extremely thermostable, showing a half-life of 113 h at $85^{\circ}C$. These results indicate that rFiG could be used for hydrolysis of cellulosic and hemicellulosic biomass substrates for biofuel production.

식이섬유 급원 첨가하에서의 식이제한이 흰주의 장내 환경에 미치는 영향 (The Influence of Dietary Fiber on Intestinal Environment in Rats Controlled by Food Restriction)

  • The effects of food restriction on the fecal microflora, moisture, pH, indole, ${\beta}$-glucosidas, and ${\beta}$-glucuronidase in rate were studied for 4 weeks. Four groups of rats for feeding was allocated to the following experimental trials : (1) control containing 1% cellulose, (2) control with food restriction, (3) treatment of diet containing butterbur, (4) treatment of butterbur combined with diet restriction. Treatment of butterbur combined food restriction significantly (p<0.05) reduced the growth of Bacteroides, Peptococus, Streptococcus, Staphylococcus, and Escherchia coli, respectively. No remarkable changes in the ${\beta}$-glucosidase and ${\beta}$-glucuronidase activities were observed but indole content significanlty decreased. Based on these results, treatment of butterbur combined with diet restriction in rats had a significantly effect for preventing the growth of those pathogenic microorganims.

Pilot-Scale Production of Cellulase Using Trichoderma reesei Rut C-30 Fed-Batch Mode

  • Trichoderma reesei Rut C-30 produced high levels of ${\beta}$-glucosidase, endo-${\beta}$-glucosidase, endo-${\beta}$-1,4-glucanase, and exo-${\beta}$-1,4-glucanase. In pilot-scale production (50-1 fermentor), productivity and yield of CMCase (carborymethyl cellulose) and FPase (filter paper activity) were 273 U/ml and 35 U/ml, and 162 FPU/l.h and 437 FPU/g, respectively. The fed-batch techniques were used to improve enzyme activities with constant cell concentration. The acidity was an important parameter and controlled at pH 3.9 and 5.0 by automatic addition of ammonium hydroxide. Cellulase powder was prepared by ammonium sulfate precipitation and its CMCase and FPase activities were 3,631 U/g and 407 U/g, respectively.

감귤류 변패의 원인균인 Penicillium sp.-L4가 생성하는 식물세포벽 분해효소의 작용양상

  • Penicillium sp.-L4, a causative fungus of rot in citrus fruits, was isolated and its mode of hydrolytic enzyme production was investigated. Carboxymethylcellulase (CMCase), polygalacturonase(PGase), extra- & intra-cellular $\beta$-glucosidase and cellobiase were produced drastically by addition of substrates in minimal media. Production of the hydrolytic enzymes were induced efficiently by cellobiose and cellooligosaccharides which were the products of cellulose hydrolysis, but repressed by addition of mono-saccharide such as glucose, raffinose, galacturonic acid. The relative activity of p-nitrophenyl-$\beta$-D-glucopyranoside(PNPG) hydrolysis was higher than that of cellobiose hydrolysis in extracellular enzymes, and reverse is true in intracellular enzymes. Intact enzyme production of P. sp.-L4 on lemon peel lesion was sequential. $\beta$-Glucosidase and CMCase were produced first and followed by PGase. The enzyme productivities and pH in lesions were coincident with optimal pH of each enzyme activities.
