Severe soybean-sprout rot was found at the mass productive factory in 2000 and 2001 and it caused 10-20% loss of the production. Pythium sp. was isolated almost 90% by potato dextrose agar from rotted root and hypocotylsof the sprouts. And the pathogencity tests using test tubes with 2% water agar and small containers (30 ${\times}$ 30 ${\times}$ 50 cm, WxLxH) cultivation were shown a similar rot on roots and hypocotyls. The fungal mycelium grew rapidly on the water agar and it prevented the seed germination. Density of the Pythium sp. in the recycled water system at the factory was periodically measured using a selective medium, corn meal agar with Pimaricin 10 mg, Rifampicin 10 mg, Ampicillin 100 mg per 1 liter in order to check the contamination of recycled water. After fitering step using 5 and 1 ml in the recycled system was applied and it was effectively controlled Pythium rot. The daily yield of sprout was stable and the occurrenceof Pythium in the recycled water was much less after filtering. The fungal isolates were identified as Pythium deliense Meurs based on various mycological characteristics on corn meal agar and sucrose-asparagine bentgrass leaf culture medium. P. deliens oogonia were spherical, smooth, 19-23 urn in diameter, and their stalk bending toward antheridia. Antheridia were straw hat-shaped, curred club-shaped, therminal or intercalary, monoclinous, occasionally diclinous, 12∼15 ${\times}$ 8∼11 um, 1(∼2) per oogonium.
Antogonistic bacteria against Rhizoctonia solani and Pythium spp., causing serious damage to golf course grasses, were isolated from the top soil of several golf courses in Korea. The isolate of Limk0102 was selected as the biological agent by characterization of antifungal activity, large scale preparation, fungicides tolerance and ecological fitness to the targe environment. The isolate was identified as Bacillus subtilis by biochemical and physiological characterization, and 165 rDNA sequence analysis. The bacterial agent was formulated as a granule type by seeding it on fine sand. The formulated agent showed high recovery rate (more than 10$\^$8/ cells/g sand) even after 6 month-storage at room temperature with similar antifungal activity with that of original cells. In vitro, the biological agent successfully exhibited antagonistic performance on bentgrass inoculated with R. solani or Pythium spp. isolated from the diseased grasses on golf courses. Field evaluation on disease control activity and ecological fitness of the agent is now under going on several golf courses.
Mechanol extracts of three cold-tolerant eucalyptus trees-Eucalyptus darlympleana, E. gunnii and E. unigera were screened for antimicrobial activity against twenty two phyto-pathogenic fungi and six food-borne bacterial pathogens. E. unigera showed the antagonistic activity against all the tested pathogens. Among the tested fungal pathogens, Pythium species were highly sensitive to the leaf extracts. Especially, P. vanterpoolii, a causal agent of leaf blight in creeping bentgrass (Agrostis palustris), was completely inhibited by the extracts. The eucalyptus extracts were also effective in inhibiting the fungal growth of Botrytis cinerea and Phomopsis sp. isolated from the lesions of kiwifruit soft rot during post-harvest storage. Escherichia coli O-157 was less sensitive to the inhibition than the other bacterial pathogens tested. It was likely that Gram positive bacteria-Bacillus subtilis and Streptococcus mutans were more sensitive to the eucalyptus extracts than Gram negative bacteria-Escherichia coli, Salmonella enteritidis and Pseudomonas aeruginosa. Our findings suggest that the cold-tolerant eucalyptus species have antimicrobial properties that can serve the development of novel fungitoxic agents or food preservatives.

A causal fungus of turfgrass blight was isolated from the infected leaves of zoysiagrass (Zoysia japonica Steud.) and identified as Nigrospora sphaerica (Sacc.) Mason by using a light misroscope. Its conidia are large (14-20 ${\mu}{\textrm}{m}$ diameter), shiny, black, aseptate, and smooth-walled spheres. The fungus caused typical blighting symptoms on the two turfgrass plants of bermudagrass (Cynodon dactylon (L.) Pers.) and bentgrass (Agrostis palustris Huds.). The fungus was found to produce a phytotoxic subtance to be associated with the pathogenic mechanism. A phytotoxin was isolated from the liquid cultures of N. sphaerica by repeated silica gel column chromatography and its structure was determined to be 5, 6-dihydro-5-hydroxy-6-propenyl-2H-pyr-2-one (T-3 compound). It was not a host-specific toxin showing phytotoxic effects to various plants inclusing turfgrasses in the leaf-wounding assay, the whole plant test, and the cellular leakage test. The compound caused leaf tip dieback symptoms in turfgrass plants similar to those caused by the pathogen. Thus, T-3 compound is thought to be involved in the development of Nigrospora blight.
In use of pesticides in golf courses has been increased steadily. Environmental concern as well as decrease in efficiency led the turfgrass management into an alternate approach of using beneficial microorganism to deal with turfgrass pests. This study was focused on the use of such microorganisms for improving cultural environment and minimizing the use of pesticides. Microorganisms antagonistic to turfgrass diseases were applied to zoysiagrass fairways and creeping bentgrass greens in Yusung country club. Tharch accumulation, disease occurrence, and other cultural environments were compared among the combinations of microorganisms and suppliemental N applications. The application of microorganisms antagonistic to turfgrass diseases improved turf resiliency. Thatch thickness was 3.03cm in the control plot but it was 2.11cm in plots treated by microorganisms, indicating significant effects of microorganism application on reduction of thatch accumulation. Number of microorganism that can decompose of cellulose was higher at the plots treated with useful microbial products and it was considered that existence of higher population of microorganisms resulted in reduction of thatch accumulation. In the evaluation of relationship between thatch accumulation and disease occurrence, greater thatch accumulation was observed at the golf courses which have been frequently infested by large patch. However, the rate of thatch accumulation varied among surveyed golf courses regardless of the year of turf establishment. Therefore, management practice which can be effective for reduction of thatch could result in large patch suppression. The application of microorganisms on the established turfgrasses reduced the occurrence rate of pythium blight and yellow path diseases, whereas occurrence of brown patch and dollar spot increased.
Effect of an antagonistic bacterium AF-2001, Pseudomonas cepacia, on control of turfgrass diseases as brown patch, Pythium blight, dollar spot, and large patch were evaluated in vitro and in vivo. Results obtained in this study are summarized as follows. 1. Pseudomonas cepacia AF-2001 showed antagonism against to the pathogens causing brown patch, Pythium blighe, dollar spot and large patch. Especially, the biological agent showed strong antagonistic effect on the causal pathogens of brown patch, dollar spot, and anthracnose, but weak on Pythium blight 2. Populotion density of P. cepacia AR-2001 decreased ra;idly in turfgrass soils. Initial population of the agent was 2.4$\times$107 cfu/g soil, however, decreased to 1.4$\times$103, 6$\times$102 and 0 cfu/g soil on 10, 20, and 30 days after application, respectively. 3. Under the controlled controlled conditions of $27^{\circ}C$ and 95% RH, P. cepacia AR-2001 showed 100% control efficacy on brown patch either by pre-treatment or post-treatment of infection. However, Pythium blight was controlled about 94% by pre-treatment and only 29% by post-treatment 4. In field trials, P. cepacia AF-2001 did not suppress large patch and the control efficacy on other turfgrass diseases was lower than agro-chemicals such as tebuconazole and metalaxyl. Control efficacy of brown patch, Pythium and dollar spot by the biological agent was 57.4%, 40.4%, 61.5~87%, respectively. 5. Growth, color and texture of creeping bentgrass were not differ significantly between AF-2001 treatment and untreated control.
This Study was carried out to investigate on the environmental effects of golf courses and to find a solution to the negative effects in Korea. The results were ai follows. 1. There were 152 golf courses opened in December 2000. 113 golf courses were member-ship and 39 golf courses were public. Users (golfer) were over 12,000,000 in 2000. 2. Total area of golf courses was 0.8% as compared with total area of farmland in 1999. Total amount of agricultural chemicals used in golf courses was 0.3% of total amount of agricultural chemicals used in Korea. The remaining amount of agricultural chemicals in golf courses tested were almost below the permitted limits. 3. Recently, total amount of fertilizers in green decreased 5~7g/$m^2$/year as compared with the recommended. 4. The control by antagonistic microorganisms and the fertilizing of the organic matter or the organic fertilizer were effective to decrease the amount of agricultural chemicals used. To success the control of microorganisms, active antagonistic microorganisms had to be applied over the recommended dose eve교day. Though fertilizing of organic fertilizer, slow release fertilizer, was effective to suppress dollar spot, necrotic ring spot. fungicides were required as a supplementary means when disease symptom was very heavy. 5. Zoysiagrass was fertilized intensively from May to August. Cool-season grasses were fertilized intensively from March to May and September to October. The fall fertilizing of Zoysiagrass was carried out to the begining of October. The fall fertilizing of cool-season grasses were carried out to the end of November or the begining of December. The fertilizing amount of Zoysiagrass was 10g/$m^2$/year. The fertilizing amount of bentgrass was 25~27g/$m^2$/year.
Typhula incarnata에 의한 설부병 방제를 위한 살균제의 효과를 골프코스에서 평가하였다. 살균제의 살포효과, 살균제의 혼용효과, 살균제의 살포방법에 의한 효과 및 살포시기에 의한 설부병의 방제효과에 미치는 영향을 용평 골프장에서 평가하였다. 무처리구는 3종의 살균제처리구보다 병 피해가 더욱 심각하였다. 3종의 살균제(Azoxystrobin, propiconazole, tebuconazole)는 Kentucky bluegrass 종과 creeping bentrasss 종의 잔디에서 80% 이상의 설부병을 방제하는 효과가 있었다. 2종 이상의 혼용살포에 의한 설부병의 방제효과는 Kentucky bluegrass 종에서 93% 이상 방제효과가 있었다. Azoxystrobin 살균제를 관주와 경엽살포에 대한 설부병 방제 효과는 2종의 잔디에서 모두 유의성의 차이가 없었다. 살균제 살포시기에 따른 설부병 방제 효과는 tebuconazole 살포구에서 유의성 있었다. 이른 가을에 살포하는 것이 더 효과적이었다.
USGA green specification is currently accepted in construction method of Korea. This study was carried out to find the factors influencing growth of turfgrass associated with soil physical properties of soil root-zone on golf green constructed with USGA method. Three putting greens in poor turfgrass and one in good turfgrass condition were selected for investigation on one golf course site at mid-South Korean peninsula. Soil hardness, moisture content, root length, and turf density were measured on-site greens, and soil physical properties and soil chemical properties also analyzed in laboratory. As a result of on-site surveys and soil physical tests in laboratory, soil physical properties were most important factors which influenced on turfgrass growth at tested greens. The results of soil particle analysis on green No. 2, in good turf condition, matched USGA sand particle recommendations. But those greens such as Nos. 1, 11 and 16, in poor putting greens, showed high soil compaction and improper soil particle distribution. Those factors created low leaf density, poor root depth, and higher moisture content compared with lower part of topsoil. Such phenomena caused inadequate turfgrass growth with soil hardening associated with poor drainage. Therefore, declines of soil physical properties associated with improper particle distribution caused a major factor influencing on turfgrass growth in golf green. Adequate test of soil particle analysis by USGA specification and proper construction method followed by adequate turf maintenance should be performed to obtain optimal turf quality on putting green.
