Simultaneous Determination of Doxifluridine and 5-FU in Liver and Intestine Tissue Using LC/MS/MS (LC/MS/MS를 이용한 원숭이 및 비글견의 간 및 장관 조직에서의 Doxifluridine과 대사체 5-FU 동시분석법 개발)

  • A liquid chromatographic method with tandom spectrometric detection (LC/MS/MS) for the simultaneous determination of doxifluridine and its active metabolite, 5-fluorouracil (5-FU) was developed over the concentration range of $5{\sim}2000$ ng/ml, respectively. Doxifluridine, 5-FU and internal standard, 5-chlorouracil (5-CU), were extracted from liver and intestine tissue via protein precipitation. Acetonitrile was used as the extraction solvent and the supernatant was evaporated and reconstructed in mobile phase. Optimum chromatographic separation was achieved on a Agilent Zorbax $C_{18}$ ($100\;mm{\times}2.1\;mm$, $3.5\;{\mu}m$) column with mobile phase run in isocratic with methanol : water (20 : 80, v/v). The flow rate was 0.2 ml/min with total cycle time of 5 min. The lower limit of quantification was validated at 5.0 ng/ml of liver and intestine tissue, for both doxifluridine and 5-FU, respectively. The intra-day and inter-day precision and accuracy of quality control (QC) samples were <11% coefficient of variation and <7% relative error from theoretical concentration for both analytes. In addition, the special designed stability study was performed, because the metabolism of doxifluridine occurs spontaneously even in ice bath for monkey liver. The stability of doxifluridine in liver and intestine of monkey and beagle dog was compared. It was found that bioanalytical validation could not be performed for the monkey liver; however, beagle dog's liver has relatively low speed of metabolism compared to monkey liver and instead of monkey liver, beagle dog's liver could be used for the validation. Bioanalytical validation could be performed in monkey intestine. Eventually, this developed method for liver and intestine will be useful in support of the toxicokinetic and pharmacokinetic studies of doxifluridine and 5-FU.

  • 'The Voyage of the Beagle' , one of the best record of scientific travels ever made, written by Charles Darwin (1809-82), the greatest naturalist ever born, is reviewed in the viewpoint of the Earth Sciences. Various observations and interpretations on the Earth Sciences are told in the Voyage. First of all, Darwin understood the geological time much more longer than accepted then as well as the geological phenomena and processes such as orogeny, uplifting, subsidence, erosion, and deposition. And he also provided a perfect interpretation on the formation of coral reefs and a good theory on the evolution of organisms. Reasonable paleoenvironmental reconstructions and interpretations based on the fossils were given in the Voyage. His observations on meteorological phenomena were from the dust composed of organisms collected on the 'Beagle' in the Atlantic Ocean, to the movement of air and extreme clear dry condition experienced on the crest of the Andes, and etc. He made several observations on the general oceanography such as the discoloration of the sea, the lights on the sea surface, conchoidal provinces noted on the Galapagos Archipelago, the trees and plants found in the remotely separated islands, and etc. However relatively scarce observation was carried out on astronomy probably due to his relatively much land travelling. Most of his interpretations and suggestions are accepted in this time.

Guided Bone Regeneration with a PDGF-BB-Ioaded Resorbable Membrane in the Dehisced Implant of the Beagle Dog (치아매식술에 이용한 혈소판 유래 성장인자 함유 흡수성 차폐막의 골유도 재생효과)

  • 4 beagle dogs aged over one and half years and weighed 15 to 16 Kg were utilized in this study. Experimental dehiscent defects were made in the mandibular edentulous area after removal of lower premolar. e-PTFE membrane resorbable membrane, and PDGF-BB-loaded resorbable membrane were covered at the dehiscent defects around the dental implants respectively. Animal was sacrificed at 1, 2, 3 months respectively. Non-decalicifed specimens were made and mutiple-stained for light microscopic study. The results were as follows: 1. Dehiscent defects around the implant installed in the beagle dog were an excellent defect model for studying guided bone regeneration. 2. Fibroblasts penetrated into expanded-PTFE membrane was observed and inflammatory cell infiltration was also observed around the membrane. 3. Resorbable membrane was degradaded and resorbed at 1 month after application to the dehiscent defect. Though multinucleated giant cells were observed adjacent to the membrane, that had no reverse effect on the boe regeneration. 4. PDGF-BB-Ioaded resorbable membrane was same capability as the resorbable membrane and e-PTFE membrane in the guided bone regeneration. 5. PDGF-BB-Ioaded resorbable membrane-applied site was better than resorbable membrane-applied site in the speed and maturity of bone formation. Within the above results, it was suggested that PDGF-BB-Ioaded resorbable membrane might have same bone regeneration capacity as nonresorbable membrane in the dehisced implant of the beagle dog and potentiality to use in human subjects.

  • Gami-Honghwa-Tang (KH-19), a herbal prescription for reducing the side effect of radiotherapy, is composed of eight crude herbs such as Rehmanniae Radix Preparata, Angelicae Gigantis Radix, Cnidii Rhizoma, Paeoniae Radix, Corni Fructus, Moutan Cortex Radicis, Lycii Fructus, Carthami Flos, and Glycyrrhizae Radix. In this study, marker substances in KH-19 were analyzed by high performance liquid chromatography-diode array detector (HPLC-DAD) and safety evaluation of standardized KH-19 was evaluated by good laboratory practices (GLP) guideline of Korea Food and Drug Administration. HPLC-DAD was employed to determine the quantities and the qualities of several marker substances such as 5-hydroxymethyl-2-furaldehyde (5-HMF), paeonol, loganin, paeoniflorin, glycyrrhizin, and decursin in the KH-19. In acute oral toxicity study on rat, transient inhibition of body weight was shown, but change in general symptom was not detected. No dead animal was observed up to 5,000 mg/kg in both male and female animals. In acute oral toxicity study on Beagle dog, transient vomiting, diarrhea, anorexia, and body weight reduction were observed, However, no dead animal was observed up to 2,000 mg/kg in both male and female animals.

Concurrent Validity of a Universal Goniometer and a Double Meter Inclinometer for Passive Range of Motion in Beagle Dogs

  • The purpose of this study was to evaluate the concurrent validity of the double meter inclinometer (DMI) for passive joint range of motion (ROM) in beagle dogs and to compare these results to a universal plastic goniometer (UPG). Fifteen beagle dogs were recruited for this study. Joint ROM was evaluated twice with each device to calculate the intraobserver reliability. The intraclass correlation coefficient (ICC) values of the UPG were good to excellent (> 0.75) for all joint ROM tests. Similar results were obtained with the DMI. The ICC values of the DMI were good to excellent (> 0.75) except in extension of the tarsal joint (ICC = 0.69). The majority of the ICC results between each device were poor (< 0.50) with the exception of six joints. Our findings suggest that the inclinometer can be used for passive joint ROM in veterinary medicine. However, caution should be taken when comparing measured values of passive joint ROM obtained utilizing both the DMI and UPG.

Single Dose Toxicity Studies of C.1-50005 (Hepatitis A virus Vaccine) in Rats and Dogs (CJ-50005(A형 간염백신)의 Rat 및 Dog에서의 단회투여독성)

  • The acute toxicity of CJ-50005, an inactivated whole virus vaccine derived from hepatitis A virus (HM175) grown in human MRC-5 diploid fibroblast culture, was tested in Sprague Dawley (SD) rats and beagle dogs. CJ-50005 was orally and intramuscularly administered up to the maximum dose of 81$\mu\textrm{g}$/kg. as much as 3,000 times of the expected clinical dose, in SD rats and was intramuscularly administered up to 27 $\mu\textrm{g}$/kg, as much as 1,000 times of the expected clinical dose, in beagle dogs. In these experiments, there were no death and clinical changes which were related to CJ-50005 administration. In addition, there were no significant changes between control and treated groups in body weights and autopsy findings. In conclusion, the administration of CJ-50005 over 81$\mu\textrm{g}$/kg in SD rats and over 27$\mu\textrm{g}$/kg in beagle dogs was proved to be safe, and it is thought that CJ-50005 may not show any toxicity in its clinical use.

The effects of Magnoliae cortex and Zea Mays L. extract mixtures on experimentally induced periodontitis of beagle dog (후박추출물과 옥수수 불검화 추출물 혼합 경구용 제제가 비글견에서 실험적으로 유발된 치주염에 미치는 영향)

  • It has been reported that Magnoliae cortex extract has antibacterial and antimicrobial activity against pathogenic microbes and Zea Mays L. extract is effective for improving gingival tissue health. The purpose of this study was to examine the anti-inflammatory and antimicrobial effects of Zea Mays L. and Magnoliac cortex extract mixtures through experimental peridontitis induced beagle dog model. Nine beagle dogs with experimentally induced periodontitis were selected. Baseline clinical indices which includes plaque index, gingival index, probing pocket depth, clinical attachment level, gingival fluid flow rate were recorded and microbial assays were done. Magnoliac cortex and Zea Mays L., mixed at 2:l ratio in 105mg capsular dosage, were taken by 3 capsule (Group I) or 6 capsule dosages (Group II) three times a day. After 4,8,12 weeks, clinical indices were recorded. All data of clinical indices were compared through one-way ANOVA with 95% confidence level. Clinical indices of group I and II showed significantly better results than those of control group. There were no significant differences between group I and II. In conclusion, it was confirmed that mixture of Magnoliae cortex and Zea May L. (mix ratio 2:1) possessed clinical improving effects to periodontitis.

A Histological Study on the Bone Tissues Surrounding Coated and Non-Coated Titanium Implants in Beagle Dogs (Beagle Dog 에 식립된 순수한 타이타늄 임프란트 및 피막처리된 타이타늄 임프란트 주위의 골조직에 관한 조직학적 연구)

  • 본 연구에서는 악골에 식립된 순수한 타이타늄 임프란트와 타이타늄 플라즈마 처리된 타이타 늄 임프란트 및 수산화인회석으로 피막처리된 타이타늄 임프란트의 주위 골조직을 광학현미경 과 주사전자현미경을 이용하여 비교 관찾히였다. 3 마리의 beagle dog 에서 상하악 소구치를 편측으로 발거한 후 12 주의 치유기를 거쳐 임프란트 를 식립하였으며, 임프란트 식립 후 4주, 8주 및 12주에 실험동물을 각각 희생시켜서 조직 표본을 제작하였다. 임프란트 주위의 골조직에서 염증이나 이물반응은 관찰되지 않았으며, 정상적인 악골에서와 유사한 골조직 소견이 관찰되었다 상악골 및 하악골에 식립된 모든 임프란트의 4주 표본에서, 골과 임프란트 변의 직접적인 접촉이 관찰되었다. 동일한 치유기에서는, 상악골에 식립된 임프란트의 면에 비하여 하악골에 식립된 동종의 임프란트의 변에서 더 많은 양의 골접촉이 관찰되었으며, 순수한 타이타늄 임프란트의 면에 비하여 피막처리된 타이따늄 임프란트의 변에서 더 많은 양의 골접촉이 관찰되었다.

Tissue reactions to suture materials in the oral mucosa of beagle dogs

  • Purpose: The objective of this study was to compare and evaluate the inflammatory responses of three widely used suture materials in the keratinized gingiva and buccal mucosa of beagle dogs. Methods: Silk, polyglycolic acid, and nylon sutures were placed within the mandibular keratinized gingiva and maxillary buccal mucosa of four male beagle dogs. Biopsies were taken 3, 7, and 14 days after suturing. Specimens were prepared with hematoxylineosin stain for evaluation under a light microscope. Results: The suture materials placed in the oral mucosa elicited more inflammatory reactions than did those placed in the keratinized gingiva. The multifilament suture materials caused more inflammatory tissue reactions than did the monofilament suture materials in the oral mucosa. Conclusions: If oral hygiene is well maintained and suture materials are placed in the keratinized gingiva, silk, nylon, and polyglycolic acid are considered to be proper suture materials for oral surgery. However, it is advisable to use monofilament suture materials if the suture site is within the oral mucosa.

Comparison of Cardiopulmonary Effects and Recovery between Total Intravenous Anesthesia with Propofol and Volatile Induction/maintenance Anesthesia with Isoflurane in Beagle Dogs (비글견에서 Propofol 완전정맥마취와 Isoflurane 휘발성 유도/유지 마취 시 심폐기능과 회복의 비교)

  • To compare cardiopulmonary effects and recovery between total intravenous anesthesia (TIVA) with propofol (PRO group, n=5) and volatile induction/maintenance anesthesia (VIMA) with isoflurane (ISO group, n=5), we investigated changes of heart rate, $SpO_2$, arterial pressure, rectal temperature and respiratory rate during 60 minute anesthesia and 40 minute recovery period in beagle dogs, and investigated recovery (extubation, head lift, sternal position and righting) after 60 minute anesthesia. Rectal temperature was significantly low in ISO group (p<0.05) from 10 to 100 minute. Heart rate was significantly low in ISO group (p<0.05) at 40, 50, 60 minute. Respiratory rate was significantly low in PRO group (p<0.05) at induction and 70 minute. $SpO_2$ tendency was similar. Systolic arterial pressure (SAP) was significantly low in ISO group (p<0.05) at induction and during anesthesia. Recovery was similar in two groups. We concluded that TIVA with propofol is useful in stabilizing rectal temperature and arterial pressure during anesthesia and provide fast and stable recovery.
