Sequence Analysis of the Gene Encoding gp55 Protein of Suri Strain, an Attenuated Classical Swine Fever (Hog Cholera) Virus (Classical Swine Fever (Hog Cholera) Virus 약독순화주 (Suri 주)의 gp55 Gnen 염기서열 분석)

  • An attenuated classical swine fever virus (CSFV), Suri strain, is a variant derived from a vaccine virus, LOM strain. This study was performed to elucidate the molecular biologcal properties of CSFV Suri strain, and to obtain the basic data for molecular epidemiological approaches for the disease. The truncated form of gp55 gene without the C-terminal transmembrane domain, in size of 1,023bp, was amplified by RT-PCR and sequenced by dye terminator cyclic sequencing method, and inserted into BamHI site of pAcGP67B baculovirus vector, establishing a cloned pAcHEG plasmid. By the nucleotide sequences determined, 341 amino acid sequences were predicted. As compared the nucleotide and amino acid sequences of gp55 of Suri with the various CSFV, Suri strain showed the high homology over 99.1% with ALD and LOM strains, but comparably the lower homology with Alfort and Brescia. In comparison of amino acid sequence in variable domain of gp55 protein, the similar tendency of homology was observed. In hydrophobicity analysis, all of four CSFV strains revealed the analogous patterns of hydrophobicity. The numbers and locations of N-glycosylation site and cysteine residues in gp55 were analyzed, those of Suri strain being coincident with ALD and LOM strains. The results suggest that gp55 in Suri strain has the high similarity to those in ALD and LOM strains in terms of the nucleotide and amino acid sequences and the functional properties of gp55 protein.

  • There is a growing concern about the damage to aquatic ecosystem caused by immigration of non-indigenous species. It is estimated that more than 10,000 million tons of ballast water is transported by shipping activities annually, and ballast water has been recognised as a major vector for the transplant of aquatic species across bio-geographical boundaries. The problem of harmful aquatic organism in ballast water for the control and management of ships' ballast water and sediments was adopted and diplomatic conference of IMO at February 2004. At once ballast water is transported by shipping activities for organic compounds, nitrogen, phosphorus and heavy metal etc. but, It's fixing a focus only for the transported of pathogenic bacteria and non-indigenous species. Hence, this studies on the distribution of organic compounds, nitrogen, phosphorus and heavy metal in ballast water during the shipping activity with the basic data which is necessary to the systemic manage of ballast water.

  • This paper considers the solution of the stochastic differential equations (SDEs) with random operator and/or random excitation using the spectral SFEM. The random system parameters (involved in the operator) and the random excitations are modeled as second order stochastic processes defined only by their means and covariance functions. All random fields dealt with in this paper are continuous and do not have known explicit forms dependent on the spatial dimension. This fact makes the usage of the finite element (FE) analysis be difficult. Relying on the spectral properties of the covariance function, the Karhunen-Loeve expansion is used to represent these processes to overcome this difficulty. Then, a spectral approximation for the stochastic response (solution) of the SDE is obtained based on the implementation of the concept of generalized inverse defined by the Neumann expansion. This leads to an explicit expression for the solution process as a multivariate polynomial functional of a set of uncorrelated random variables that enables us to compute the statistical moments of the solution vector. To check the validity of this method, two applications are introduced which are, randomly loaded simply supported reinforced concrete beam and reinforced concrete cantilever beam with random bending rigidity. Finally, a more general application, randomly loaded simply supported reinforced concrete beam with random bending rigidity, is presented to illustrate the method.

An Empirical Study on Mutual Influence between Economic Index and Distribution Industry in Korean (한국 유통산업이 한국 경제에 미치는 상호영향력에 관한 실증적 연구)

  • Purpose - The objective of this paper is to discover if there exists a relationship between the economic index and distribution industry index in Korean. Because of the distribution industry boom in the recent years, a lot of interest in the relationship between the economic index and distribution industry index in Korean and the economy has been generated. This article examine on the mutual influence between economic index and distribution industry index in Korean. Research design, data, and methodology - For this purpose, we use the vector-auto regression model, impulse response function and variance decomposition of the economic index and distribution industry index, Granger causality test using weekly data on the economic index and distribution industry price index in korea. The sample period is covering from January 2, 2010 to August 31, 2019. The VAR model can also be linked to cointegration analysis. Cointegration Analysis makes possible to find a mechanism causing x and y to move around a long-run equilibrium (Engle and Granger, 1987). This equilibrium means that external shocks may separate the series temporarily at any particular time, but there will be an overall tendency towards some type of long-run equilibrium. If variables are found to have this tendency they are said to be cointegrated and a long-run relationship between these series is established. These econometric tools have been applied widely into economics and business areas to analyze intertemporal linkages between different time series. Results - This research showed following main results. First, from the basic statistic analysis of the economic index and distribution industry index in Korean, the economic index and the distribution industry index in korea have unit roots. Second, there is at least one cointegration between the economic index and distribution industry index in Korean. Finally, the correlation between of the economic index and the distribution industry index in korea is (+) 0.528876. Conclusions - We find that the distribution industry price index Granger cause the economic index in korea. As a consequence, the distribution industry index affect the economic index in Korean. The distribution industry index to the economic index is stronger than that from the economic index to the distribution industry index.

Optimal sensor placement under uncertainties using a nondirective movement glowworm swarm optimization algorithm

  • Optimal sensor placement (OSP) is a critical issue in construction and implementation of a sophisticated structural health monitoring (SHM) system. The uncertainties in the identified structural parameters based on the measured data may dramatically reduce the reliability of the condition evaluation results. In this paper, the information entropy, which provides an uncertainty metric for the identified structural parameters, is adopted as the performance measure for a sensor configuration, and the OSP problem is formulated as the multi-objective optimization problem of extracting the Pareto optimal sensor configurations that simultaneously minimize the appropriately defined information entropy indices. The nondirective movement glowworm swarm optimization (NMGSO) algorithm (based on the basic glowworm swarm optimization (GSO) algorithm) is proposed for identifying the effective Pareto optimal sensor configurations. The one-dimensional binary coding system is introduced to code the glowworms instead of the real vector coding method. The Hamming distance is employed to describe the divergence of different glowworms. The luciferin level of the glowworm is defined as a function of the rank value (RV) and the crowding distance (CD), which are deduced by non-dominated sorting. In addition, nondirective movement is developed to relocate the glowworms. A numerical simulation of a long-span suspension bridge is performed to demonstrate the effectiveness of the NMGSO algorithm. The results indicate that the NMGSO algorithm is capable of capturing the Pareto optimal sensor configurations with high accuracy and efficiency.

Hybrid Word-Character Neural Network Model for the Improvement of Document Classification (문서 분류의 개선을 위한 단어-문자 혼합 신경망 모델)

  • Document classification, a task of classifying the category of each document based on text, is one of the fundamental areas for natural language processing. Document classification may be used in various fields such as topic classification and sentiment classification. Neural network models for document classification can be divided into two categories: word-level models and character-level models that treat words and characters as basic units respectively. In this study, we propose a neural network model that combines character-level and word-level models to improve performance of document classification. The proposed model extracts the feature vector of each word by combining information obtained from a word embedding matrix and information encoded by a character-level neural network. Based on feature vectors of words, the model classifies documents with a hierarchical structure wherein recurrent neural networks with attention mechanisms are used for both the word and the sentence levels. Experiments on real life datasets demonstrate effectiveness of our proposed model.

  • Major advances in positive-sense RNA virus research have been facilitated by the development of reverse genetics systems. These systems consist of an infectious cDNA clone that encompasses the genome of the virus in question. This clone is then used as a template for the subsequent synthesis of infectious RNA for the generation of synthetic viruses. However, the construction of infectious cDNA for the Japanese encephalitis virus (JEV) has been repeatedly thwarted by the instability of its cDNA. As JEV is an important human pathogen that causes permanent neuropsychiatric sequelae and even fatal disease, a reliable reverse genetics system for this virus is highly desirable. The availability of this tool would greatly and the development of effective vaccines as well as facilitate studies into the basic biology of the virus, including the molecular mechanisms of viral replication, neurovirulence, and pathogenesis. We have successfully constructed a genetically stable infectious JEV cDNA containing full-length viral RNA genome. Synthetic RNA transcripts generated in vitro from the cDNA were highly infectious upon transfection into susceptible cells, and the cDNA remained stable after it had been propagated in E. coli for 180 generations. Using this infectious JEV cDNA, we have successfully expressed a variety of reporter genes from the full-length genomic and various subgenomic RNAs in vitro transcribed from functional JEV cDNAS. In summary, we have developed a reverse genetics system for JEV that will greatly facilitate the research on this virus in a variety of different fields. It will also be useful as a heterologous gene expression vector and aid the development of a vaccine against JEV.

  • In this paper, we discuss on the design of a current mode CMOS multiplier circuit over $GF(3^m)$. Using the standard basis, we show the variation of vector representation of multiplicand by multiplying primitive element α, which completes the multiplicative process. For the $GF(3^m)$ multiplicative circuit design, we design GF(3) adder and multiplier circuit using current mode CMOS technology and get the simulation results. Using the basic gates - GF(3) adder and multiplier, we build the $GF(3^m)$ multiplier circuit and show the examples for the case m=3. We also propose the assembly of the operation blocks for a complete $GF(3^m)$ multiplier. Therefore, the proposed circuit is easily extensible to other p and m values over $GF(p^m)$ and has advantages for VLSI implementation. We verify the validity of the proposed circuit by functional simulations and the results are provided.

Effects of Eicosapentaenoic Acid and Docosahexaenoic Acid on Mitochondrial DNA Replication and PGC-1α Gene Expression in C2C12 Muscle Cells

  • Mitochondrial biogenesis is a complex process requiring coordinated expression of nuclear and mitochondrial genomes. The peroxisome proliferator-activated receptor gamma co-activator 1-alpha (PGC-$1{\alpha}$) is a key regulator of mitochondrial biogenesis, and it controls mitochondrial DNA (mtDNA) replication within diverse tissues, including muscle tissue. The aim of this study was to investigate the effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on mtDNA copy number and PGC-$1{\alpha}$ promoter activity in $C_2C_{12}$ muscle cells. mtDNA copy number and mRNA levels of genes related to mitochondrial biogenesis such as PGC-$1{\alpha}$, nuclear respiratory factor 1 (NRF1) and mitochondrial transcription factor A (Tfam) were assayed by quantitative real-time PCR. The PGC-$1{\alpha}$ promoter from -970 to +412 bp was subcloned into the pGL3-basic vector, which includes a luciferase reporter gene. Both EPA and DHA significantly increased mtDNA copy number, dose and time dependently, and up-regulated mRNA levels of PGC-$1{\alpha}$, NRF1, and Tfam. Furthermore, EPA and DHA stimulated PGC-$1{\alpha}$ promoter activity in a dose-dependent manner. These results suggest that EPA and DHA may modulate mitochondrial biogenesis, which was partially associated with increased mtDNA replication and PGC-$1{\alpha}$ gene expression in $C_2C_{12}$ muscle cells.

A Homeotic Gene, Hoxc8, Regulates the Expression of Proliferating Cell Nuclear Antigen in NIH3T3 Cell

  • Hoxc8 is one of the homeotic developmental control genes regulating the expression of many downstream target genes, through which animal body pattern is established during embryonic development. In previous proteomics analysis, proliferating cell nuclear antigen (PCNA) which is also known as cyclin, has been implied to be regulated by Hoxc8 in F9 murine embryonic teratocarcinoma cell. When the 5' upstream region of PCNA was analyzed, it turned out to contain 20 Hox core binding sites (ATTA) in about 1.17 kbp (kilo base pairs) region ($-520{\sim}-1690$). In order to test whether this region is responsible for Hoxc8 regulation, the upstream 2.3 kbp fragment of PCNA was amplified through PCR and then cloned into the pGL3 basic vector containing a luciferase gene as a reporter. When the luciferase activity was measured in the presence of effector plasmid (pcDNA : c8) expressing murine Hoxc8, the PCNA promoter driven reporter activity was reduced. To confirm whether this reduction is due to the Hoxc8 protein, the siRNA against Hoxc8 (5'-GUA UCA GAC CUU GGA ACU A-3' and 5'-UAG UUC CAA GGU CUG AUA C-3') was prepared. Interestingly enough, siRNA treatment up regulated the luciferase activity which was down regulated by Hoxc8, indicating that Hoxc8 indeed regulates the expression of PCNA, in particular, down regulation in NIN3T3 cells. These results altogether indicate that Hoxc8 might orchestrate the pattern formation by regulating PCNA which is one of the important proteins involved in several processes such as DNA replication and methylation, chromatin remodeling, cell cycle regulation, differentiation, as well as programmed cell death.
