A protocol for in vitro propagation from pseudobulb sections of Lycaste armomatica (Graham ex Hook) Lindl., an ornamental and fragrant orchid, was developed. The effect of four cytokinins: kinetin (K), metatopolin (mT), $N^6$-benzyladenine (BA), and thidiazuron (TDZ), in equimolar concentrations, was investigated. Shoot formation from apical and basal pseudobulb sections was obtained in all treatments. A few medial sections cultured in media supplemented with BA formed protocorm-like bodies. Shoot formation was greater from the basal section than the apical, and mainly occurred in explants cultured in media containing TDZ. The highest average numbers of shoots per explant were achieved from basal sections cultured in media supplemented with TDZ at 4.4, 8.87 and 2.2 ${\mu}M$, forming on average 9.9, 8.6 and 7.3 shoots per explant, respectively. Since the medial pseudobulb section was the worst explant for propagation of L. aromatica, we recommend that pseudobulbs be divided into two sections; the basal half should be cultured in MS medium supplemented with TDZ at 4.4 ${\mu}M$ and the apical half with TDZ at 2.2 ${\mu}M$. Subculturing individual shoots in MS medium without plant growth regulators allows further development and rooting. A survival rate of more than 90% under greenhouse conditions was achieved. This research represents a direct contribution to the conservation and sustainable use of this valuable natural resource.
Giardia lamblia is a protozoan that causes diarrheal diseases in humans. Cytoskeletal structures of Giardia trophozoites must be finely reorganized during cell division. To identify Giardia proteins which interact with microtubules (MTs), Giardia lysates were incubated with in vitro-polymerized MTs and then precipitated by ultracentifugation. A hypothetical protein (GL50803_8405) was identified in the precipitated fraction with polymerized MTs and was named GlMBP1 (G. lamblia microtubule-binding protein 1). Interaction of GlMBP1 with MTs was confirmed by MT binding assays using recombinant GlMBP1 (rGlMBP1). In vivo expression of GlMBP1 was shown by a real-time PCR and western blot analysis using anti-rGlMBP1 antibodies. Transgenic G. lamblia trophozoites were constructed by integrating a chimeric gene encoding hemagglutinin (HA)-tagged GlMBP1 into a Giardia chromosome. Immunofluorescence assays of this transgenic G. lamblia, using anti-HA antibodies, revealed that GlMBP1 mainly localized at the basal bodies, axonemes, and median bodies of G. lamblia trophozoites. This result indicates that GlMBP1 is a component of the G. lamblia cytoskeleton.
This study was carried out to test the recycling post-harvest medium from other mushroom bottle cultivation as a secondary medium of the oyster mushroom. In the post-harvest medium from winter mushroom and king oyster mushroom cultivation, oyster mushroom varieties in Chunchu-2ho and Manchuri fruit bodies yields compared with control group tend to be low. After recycling the post-harvest medium, it was replaced by basal medium up to 50%, of which the fruit bodies with stable yield increase from 10% to 30% were increased.
Cryptoglena pigra Ehrenberg from Korea was a photosynthetic euglenoid alga, which had typical characteristics of the Euglenales. The ultrastructure examination of C. pigra revealed certain features which were distinctly photosynthetic euglenoid: one U-shaped chloroplast with thylakoid membranes; two paramylon grains appressed to both sides of the chloroplast; eyespot associated with the chloroplast but not part of it. Three flagellar roots were associated with the two basal bodies. The four-membered dorsal root arose from the dorsal body and extended anteriorly following the reservoir membrane. At the base of the reservoir the dorsal band was nucleated by the dorsal root and it ran anteriorly between the reservoir membrane and eyespot. The dorsal band was continued with the microtubules of the canal and the pellicle. The singlet dorsal microtubules at the transition level arranged into doublets by a successive linkage of the existing adjacent microtubules, and the doublets rearranged into the cytoskeletal microtubules that were continuous with four microtubules in pellicles. Finally, the sixteen ridges gave rise to the pellicular ridges. The five to six-membered ventral root extended anteriorly into a cytoplasmic pocket through the reservoir and lined a cytoplasmic pocket.
Foreign bodies in the airway are very import-tant and not uncommon conditions in the field of E.N.T., and practically there are many reports of them. Occasionally, it can be followed by some pulmonary complications and technical difficulty to remove. Therefore, foreign body in the trachea-bronchial tree is serious and, on occasion, fatal. Recently, the authors experienced 7 interesting cases of foreign bodies in the airway and a brief review of literature was made. Case 1. (1 year old male): A bit of walnut was coughed out spontaneously. Case 2. (2 years old male): A bit of raw chestnut was removed from the right main bronchus. Case 3. (21 Years old male): A pushpin was removed from the left main bronchus. Case 4. (1 year old male): A kind of bean was removed from the right main vronchus. Case 5. (4 years old male): A coiled steel spring (1.1cm$\times$2cm) was removed from the subglottic region. Case 6. (5 years old female): A shell of pine nut which had been impacted in the wall of right main bronchus was removed during right middle and lower lobectomy. Case 7. (2 years old male): A metal nail was removed from the rightposterior basal segmental bronchus during right lower lobectomy.

Dendrobium candidum Wallich ex Lindley is a traditional Chinese medicine plant and has been widely used for medicinal and ornamental purposes. In this study, several different factors affecting micro propagation of protocorm-like bodies (PLBs) such as basal media, plant growth regulators, and carbon sources. The proliferation PLB derived from seeds was the best in $H_3P_4$ basal medium containing $0.1mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$ Kinetin. PLB growth was the best when $10g{\cdot}L^{-1}$ sucrose was added to the carbon atoms in the medium. The rate of shoot formation from the propagated PLB was the highest in 1/4 MS or $H_1P_2$ medium containing $10g{\cdot}L^{-1}$ sucrose, and the shoot length was longer than the others.
Bulbophyllum auricomum Lindl. is very popular among orchid growers due to the attractive fragrance of its flowers and has become an endangered orchid in Myanmar. In this study, we carried out an aseptic technique that can be used to conserve this endangered orchid species. The seeds of B. auricomum Lindl. were obtained from artificial pollination and cultured in MS basal medium for seed germination. The effect of coconut water and BAP in MS basal medium on callus induction was investigated. The highest callus induction was found at 2.0 mg/L BAP. The maximum growth of protocorm-like bodies (PLBs) was evaluated, and the best response was observed on MS medium supplemented with 150 mL/L coconut water at pH 5.6. MS basal medium supplemented with 150 mL/L coconut water along with 2.0 mg/L BAP and 1.0 mg/L NAA (MCBN) showed the highest number of plantlets at 15℃ at the second week of culture. At the second and third week of culture, MS medium supplemented with 2.0 mg/L BAP and 1.0 mg/L NAA (MBN) showed the best result in terms of the number of leaves and the longest leaves at 15℃ and 25℃, respectively. The present study showed evidence of successful in vitro propagation of B. auricomum Lindl.
While conducting a floristic study of Korean hepatics, we discovered two unrecorded species, which were collected from wind holes near the Donggang River, Korea. Mannia fragrans (Balb.) Frye & L. Clark and Mannia androgyna (L.) A. Evans are hereby reported for the first time in Korea. M. androgyna is characterized by pale grayish oil bodies in both the aerenchyma and basal tissue, and saccate spores with a conspicuous proximal disc. M. fragrans is characterized by a gynoecial segment with a whitish apical brush of scales, a commonly aromatic smell, and areolate spores with a conspicuous proximal disc. Two unrecorded species are described and illustrated based on Korean material.
This study was conducted to test whether post-harvest medium of king oyster mushroom can be recycled for its bottle cultivation. Physical properties of the post-harvest medium and the resulting yield of fruit bodies revealed no significant difference compared with control. Recycling post-harvest medium can be replaced to 15% of the basal medium, thus decreasing the cost of raw materials up to 15%.
To obtain basic data for mass production of Isaria japonica, cultural characteristics of japonica were investigated by using liquid, solid media and silkworms pupa. Mycelia grew favorably at the temperature of $23{\sim}28^{\circ}C$ on MYG medium with pH 7.0. The fruiting-body of I. japonica was induced below $20^{\circ}C$ in MYG liquid medium (Malt yeast glucose) under fluorescent light. In MYG basal medium mixed with pupal powder of silkworms, the fresh weight of fruiting-bodies was increased with increasing concentration of pupal powder. The highest yield of fruiting bodies was obtained in carbon-rich medium supplemented with pupal powder of silkworm. Also, fruiting-bodies of I. japonica were produced massively on the silkworm pupa placed on the stainless tray in the shortest time. The structure and shape of fruiting-bodies were coral-like, many-branched types with numerous conidiospores.
