Screening of BCL-2 associated X protein gene polymorphism associated with scrotal hernia in domesticated swine using polymerase chain reaction-restriction fragment length polymorphism

  • Objective: This study was conducted to screen scrotal hernia in domesticated swine from selected breeders in the Philippines. This defect is associated with a cytosine to thymine mutation in the BCL-2 associated X protein (BAX) gene of swine. Methods: Genetic screening was done by DNA extraction followed by amplification and digestion using polymerase chain reaction-restriction fragment length polymorphism, amplifying the 416 bp region of the BAX gene that was subjected to digestion using the Ear I enzyme. Sequencing was also conducted to validate the results. Results: Results revealed that out of 538 samples tested, 411 (76.4%) of the samples were found to be normal whereas the remaining were carriers of the mutation in which 80 (14.9%) were heterozygous mutants and 47 (8.7%) were homozygous mutants. Pietrain breed was found to have the highest incidence. Conclusion: Having a scrotal hernia eliminates the chances of using the boar as a breeder stock because the following generations arising from it would most likely exhibit herniation. It is therefore advised to establish a genetic screening method for Scrotal Hernia in the Philippines to eliminate the negative gene from the herd.

MINERAL STATUS OF GRAZING PHILIPPINE GOATS I. THE NUTRITION OF CALCIUM, MAGNESIUM AND PHOSPHORUS OF GOATS IN LUZON ISLAND

  • Nutritional status of macro minerals (Ca, Mg and P) in goats grazed on the native pasture was investigated for 2 years in Luzon Island, Republic of the Philippines. Three regions in Luzon Island (Los Banos; Southern area, Munoz, Nueva Ecija; Central area and Baguio; Northern area) were objected to collect the samples of forages and blood of goats on the pasture. The average values in Ca, Mg and P contents of all the forage samples were 0.29-1.32, 0.13-0.56 and 0.15-0.45% of dry matter, respectively, and these figures almost exceeded the required dietary levels for sheep. There was no obvious changes in mineral content of forages between the seasons of rainy and dry, or among regions. In blood Ca level, more than a half of goats (57.9%) without supplement showed the level below the lower limit, and the low level was not improved even when they fed with some supplemental concentrates. The plasma levels of Mg and P were satisfactory, referring to the lower limit of normal level in goats. It is, therefore, necessary to decide the requirement and to establish a most suitable technique for supplying Ca to grazing goats in Luzon Island.

  • In days to calyx-flex stage test, flower development affected by light intensity, light duration and the prevailing temperature. As, 'Jacaranda' variety shorter in under 46 days of flower bud formation and 13 days of calyx-flex stage than the other variety with 'Queen Elizabeth' control variety. About cut flower only 'Grand Vicki Brown' (as within 46 days of flower formation, 13 days of calyx stage and 'Jacaranda', both with shorter plants than the other varieties produced cut flower classified as short and medium-stemmed cut flowers. 'Jacaranda', 'Vicki Brown', 'Golden Times', 'Champagne', 'Osiana', and 'Queen Elizabeth (control variety)' flowered earlier from 46.7 to 50.8 days from pruning. Flower development was likewise faster in all the above mentioned cultivars; while 'Tineke' had significantly delayed flower development reaching calyx-flex stage from 0.5 cm bud size after 51.0 days. The top ten high yielding varieties were 'American Orange', with a net income of P3,081,676.00; 'Yellow Island' with P1,572,874.00; 'Frosty Pink' with P1,085,183.00; 'Champagne' with P980,620.00; 'Coral Pink' with P959,669.00; 'Lara Pink' with P918,444.00; 'Lara Orange' with P714,252.00; 'Grand Gala' with P705,787.00; 'Golden Times' with P806,489.00; and 'Lady X' with P778,060.00 net returns.
