본 연구는 S. Typhimurium을 대상으로 paraformaldehyde 35%를 함유한 훈증소독제의 살균효과를 평가하기 위해 수행되었다. 예비 시험에서, S. Typhimurium의 현탁액 균수는 모두 $3.9{\times}10^8CFU/mL$이었으며, 훈증소독제에 노출시킨 모든 담체의 균수는 평판배지법과 여과법으로 배양한 시험균주 현탁액의 균수의 50%보다 많았다. 또한, 대조 담체로부터 회복된 S. Typhimurium 균수는 모두 $3.3{\times}10^6CFU/mL$이었다. 훈증소독제의 살균효과 시험에서는, 훈증 소독제를 처리한 담체의 S. Typhimurium의 감소 균수는 5.22 log CFU/mL로 나타났다. 이상의 결과로부터, paraformaldehyde를 주성분으로 하는 훈증소독제는 S. Typhimurium에 대해 효과적인 살균력을 갖는 것으로 확인되었으며, 병원성 세균에 오염된 식품재료 및 주방용품의 소독에 적용할 수 있을 것으로 사료된다.
PURPOSE. The aim of this study was to identify in vitro antimicrobial activity of the tissue conditioner containing silver nanoparticles on microbial strains, Staphylococcus aureus, Streptococcus mutans and Candida albicans. MATERIALS AND METHODS. Experimental disc samples ($20.0{\times}3.0$ mm) of tissue conditioner (GC Soft-Liner, GC cooperation, Tokyo, Japan) containing 0.1 - 3.0% silver nanoparticles (0%: control) were fabricated. Samples were placed on separate culture plate dish and microbial suspensions (100 ${\mu}L$) of tested strains were inoculated then incubated at $37^{\circ}C$. Microbial growth was verified at 24 hrs and 72 hrs and the antimicrobial effects of samples were evaluated as a percentage of viable cells in withdrawn suspension (100 ${\mu}L$). Data were recorded as the mean of three colony forming unit (CFU) numerations and the borderline of the antimicrobial effect was determined at 0.1% viable cells. RESULTS. A 0.1% silver nanoparticles combined to tissue conditioner displayed minimal bactericidal effect against Staphylococcus aureus and Streptococcus mutans strains, a 0.5% for fungal strain. Control group did not show any microbial inhibitory effect and there were no statistical difference between 24 hrs and extended 72 hrs incubation time (P > .05). CONCLUSION. Within the limitation of this in vitro study, the results suggest that the tissue conditioner containing silver nanoparticles could be an antimicrobial dental material in denture plaque control. Further mechanical stability and toxicity studies are still required.
This study was carried out to determine therapeutic the effect of ozonated water therapy on bovine pododermatitis. In addition, bactericidal effect of ozonated water on etiological agent of bovine pododermatitis was examined. The pathohistological examination for the pododermatitis, according to application with ozonated water and ozone ointment was investigated. Thirty healthy cattle were divided two groups(each of 15) : control group(povidone group), treatment group(ozone solution group). Various parameters were evaluated in terms of the lameness score, swelling score, lesion score, WBC count, neutrophil count, pathohistological finding, and antimicrobial action. The decrease of lameness and lesion score were shown in hoof lesions on 14 days after application of ozonated water. Significant decrease of swelling was shown in hoof lesions on 14 days 1Corresponding author after application of ozonated water(p<0.01). In hematological findings, WBC count revealed values within normal range. The number of neutrophils was slightly higher than that of normal, however, this was improved on 14 days after application of ozonated water. In pathohistological findings, recovery was rapid macroscopically and microscopically in the treatment with ozonated water on the hoof lesions and ozonated water was effective. In antimicobial action, bactericidal effect was observed in treatment with ozonated water on the hoof lesions and ozonated water was effective.
리메렐라 아나티페스티퍼 감염증은 오리와 거위에서 섬유소성 심막염, 간주위염증, 기낭염, 건락성난관염, 뇌막염을 특징으로 하는 급성 또는 만성 패혈증이다. 이 균은 혈청형 또는 분리주별로 병원성에 큰 차이가 나타나는 것으로 알려져 있다. 그럼에도 불구하고 지금까지 이러한 다양한 병원성과 그 이유에 대한 연구는 거의 이루어지지 않았다. 본 연구에서는 리메렐라 아나티페스티퍼의 병원성과 serum resistance 상관성을 구명하였다. 우리는 다양한 분리원으로부터 확보한 130주의 균주를 대상으로 serum resistance 특성을 분석하였다. 건강한 오리 인후두에서 분리된 균주들은 혈청에 대한 감수성이 높은 반면에 전신감염을 일으킨 균주들은 강한 serum resistance를 보였다. 또한 우리는 이러한 혈청의 살균효과가 혈청내 보체 성분에 의해 유도됨을 확인하였다. 강한 serum resistance를 유발하는 세균의 표면 유전자와의 관련성을 조사한 결과, 외막 단백질의 AS87_09335, AS87_00480, AS87_05195 유전자가 serum resistance와 관련 있음을 알 수 있었다. 본 연구 결과로 serum resistance 특성이 리메렐라 아나티페스티퍼의 병원성 결정 요소 중 하나라는 것을 확인하였다.
Background: The poor intracellular concentration of enrofloxacin might lead to treatment failure of cow mastitis caused by Staphylococcus aureus small colony variants (SASCVs). Objectives: In this study, enrofloxacin composite nanogels were developed to increase the intracellular therapeutic drug concentrations and enhance the efficacy of enrofloxacin against cow mastitis caused by intracellular SASCVs. Methods: Enrofloxacin composite nanogels were formulated by an electrostatic interaction between gelatin (positive charge) and sodium alginate (SA; negative charge) with the help of CaCl2 (ionic crosslinkers) and optimized by a single factor test using the particle diameter, zeta potential (ZP), polydispersity index (PDI), loading capacity (LC), and encapsulation efficiency (EE) as indexes. The formation mechanism, structural characteristics, bioadhesion ability, cellular uptake, and the antibacterial activity of the enrofloxacin composite nanogels against intracellular SASCVs strain were studied systematically. Results: The optimized formulation was comprised of 10 mg/mL (gelatin), 5 mg/mL (SA), and 0.25 mg/mL (CaCl2). The size, LC, EE, PDI, and ZP of the optimized enrofloxacin composite nanogels were 323.2 ± 4.3 nm, 15.4% ± 0.2%, 69.6% ± 1.3%, 0.11 ± 0.02, and -34.4 ± 0.8 mV, respectively. Transmission electron microscopy showed that the enrofloxacin composite nanogels were spherical with a smooth surface and good particle size distributions. In addition, the enrofloxacin composite nanogels could enhance the bioadhesion capacity of enrofloxacin for the SASCVs strain by adhesive studies. The minimum inhibitory concentration, minimum bactericidal concentration, minimum biofilm inhibitory concentration, and minimum biofilm eradication concentration were 2, 4, 4, and 8 ㎍/mL, respectively. The killing rate curve had a concentration-dependent bactericidal effect as increasing drug concentrations induced swifter and more radical killing effects. Conclusions: This study provides a good tendency for developing enrofloxacin composite nanogels for treating cow mastitis caused by intracellular SASCVs and other intracellular bacterial infections.
본 연구에서는 그람 양성의 호기성 유포자 세균 중 자연계에 널리 분포되어 식품에 문제를 일으키는 대표적인 균인 B. subtilis를 대상으로 365 nm UV-LED의 생육 억제 효과를 입증하였다. 또한 365 nm UV-LED 조사 시, 식품 포장재로 흔히 사용되고 있는 유리, LLD-PE, Nylon/LDPE 및 PS 등의 포장 조건에 따른 B. subtilis 생육 억제 효과를 확인하였다. 그 결과, B. subtilis의 생육 억제 효과가 가장 뛰어난 재질은 Nylon/LDPE와 LLD-PE로 확인되었고, 대조구의 생존율인 -log 5 값과 비교하여 각각의 생존율은 약 -log 2.5-2.9, -log 2.58-3.61로 나타났다. 이 때 재질의 두께가 미생물의 생육에 미치는 영향은 미미한 것으로 관찰되었고, 포장재질에 따라 365 nm UV-LED 투과력이 다르다는 것을 확인할 수 있었다. 통상적으로 log 3 이상 생균수가 감소하면 99.9% 살균효과가 있는 것으로 나타낼 수 있는데, 본 연구를 통해 365 nm UV-LED가 흔히 사용되고 있는 식품 포장재를 투과하여 균의 생육 억제에 영향을 줄 수 있음을 확인하였다. 이러한 결과를 통해 365 nm UV-LED의 사용이 식품보존과 식품산업 분야의 응용기술로써 잠재력이 있음을 시사하는 바이다.
This study aimed to evaluate the antimicrobial effects of Acanthopanax sessiliflorum fruit (ASF; Ogaza) extracts on Streptococcus mutans and Streptococcus sobrinus, which are agents that cause dental caries, and on Streptococcus mitis and Streptococcus salivarius, the microbial flora of the oral cavity. The ASF extracts obtained using 70% ethanol were fractionated in the order of ethyl acetate and n-Butanol, concentrated under reduced pressure, and lyophilized to give powdery solvent extracts. The antimicrobial activity of ASF extracts from each solvent was examined using the disk diffusion method. As a result, only those extracts obtained using an ethyl acetate solvent showed antimicrobial activity. These extracts were selected, and the minimum inhibitory concentration was measured by disk diffusion method at various extract concentrations. Results showed a minimum inhibitory concentration of 32 mg/ml. The viable cell count was measured to confirm the minimum bactericidal concentration. Results showed a minimum bactericidal concentration of 64 mg/ml. In the cytotoxicity test using normal human dermal fibroblast cells, the absorbance value of the test group was similar to that of the control group at 0.64, 1.28, and 6.4 mg/ml. The bacteria and their colonies were examined using a scanning electron microscope. Boundaries between the antimicrobial activity region and non-antimicrobial activity region were observed around the paper disk, which was immersed in the extract with 32 mg/ml concentration. Bacterial colonization was not observed in the area with antimicrobial activity. This finding suggests that ASF extracts can inhibit the growth of some microorganisms in the oral cavity, in addition to the effects of these extracts known to date. In particular, ASF extracts may be used as a preparation for preventing dental caries by adding the extract to the toothpaste or oral mouthwash.
Purpose : Hypochlorous acid (HOCl), a major inorganic bactericidal compound of innate immunity, is effective against a broad range of microorganisms. In particular, HOCl is well-known as a non-antibiotic antimicrobial substance. However, effects of HOCl as an antimicrobial agent are still needed to study these functions against various specific type of microorganisms. In this study, we investigated the antimicrobial effect of hypochlorous acid (HOCl) in S. mutans and A. actinomycetemcomitans to cause dental caries and periodontal disease. Experiments were conducted to observe whether HOCl become effective replacement of disinfectant. Methods : To observe antimicrobial effect of HOCl, stabilized HOCl is prepared in the form of a physiologically balanced solution in pre-conditioned and post-conditioned HOCl solution. As a control, commercially available disinfectant MAXCLEAN was used as positive control. Moreover, S. mutans and A. actinomycetemcomitans distribution in gagrin, filtered tap water, and culture media. Cell viability were measured by viable cell count methods and disk diffusion test. Results : Our results showed that treatment of HOCl have no effect against antimicrobial effect compare to control group especially gagrin in disk diffusion test. HOCl tended to reduced viability against S. mutans in group of post-conditioned than pre-conditioned of HOCl solution however, there was no significant difference as well as no effect in A. actinomycetemcomitans. Conclusion : HOCl showed tendency to reduce viability against S. mutans in group of post-conditioned of HOCl solution and no effect of antimicrobial effect. Although HOCl is well known as effective against a broad range of microorganisms, HOCl seems to have diversity following type of species to be used as antimicrobial drug following our results. Therefore, it is necessary to be rigidly controlled and regulated in using HOCl solution clinically.
This study aimed to characterize a lytic Salmonella Typhimurium-specific (ST) phage and its biofilm control capability against S. Typhimurium biofilm on polypropylene surface. ST phage was isolated, propagated, and purified from water used in a slaughterhouse. The morphology of ST phage was observed via transmission electron microscopy. Its bactericidal effect was evaluated by determining bacterial concentrations after the phage treatment at various multiplicities of infection (MOIs) of 0.01, 1.0, and 100. Once the biofilm was formed on the polypropylene tube after incubation at 37℃ for 48 h, the phage was treated and its antibiofilm capability was determined using crystal violet staining and plate count method. The phage was isolated and purified at a final concentration of ~11 log PFU/mL. It was identified as a myophage with an icosahedral head (~104 nm) and contractile tail (~90-115 nm). ST phage could significantly decrease S. Typhimurium population by ~2.8 log CFU/mL at an MOI of 100. After incubation for 48 h, biofilm formation on polypropylene surface was confirmed with a bacterial population of ~6.9 log CFU/cm2. After 1 h treatment with ST phage, the bacterial population in the biofilm was reduced by 2.8 log CFU/cm2. Therefore, these results suggest that lytic ST phage as a promising biofilm control agent for eradicating S. Typhimurium biofilm formed on food contact surfaces.
Objectives : Prunus mume has been used for the folk medicine from old times. The purpose of the study was to investigate the antimicrobial activity of prunus mume extract to various oral microbes. Methods : This study was carried out to examine the antimicrobial effect of Prunus mume extract against oral microbes. Data were collected using a Dentocult SM Strip mutans and Dentocult LB Strip mutans from April 5 to May 4, 2013. A total of 36 experimental and 32 control group were selected for this study. Results : The MIC of Prunus mume extract was tested for 0.39% in S. mutans, S. salivarius and S. auerus, 0.78% in S. mitis, S. equi and E. coli. In vivo, experimental group showed significantly the lower Streptococcus mutans levels by the use of the gum contained Prunus mume extract from 15 days compared with control group(p=0.012). The reduction was more significant in Lactobacilli level of the experimental group than the control group(p=0.022). Conclusions : These findings suggest that the oral products containing Prunus mume extracts is effective in preventing oral diseases.
